    Limited Regeneration Potential with Minimal Epicardial Progenitor Conversions in the Neonatal Mouse Heart after Injury

    The regeneration capacity of neonatal mouse heart is controversial. In addition, whether epicardial cells provide a progenitor pool for de novo heart regeneration is incompletely defined. Following apical resection of the neonatal mouse heart, we observed limited regeneration potential. Fate-mapping of Tbx18MerCreMer mice revealed that newly formed coronary vessels and a limited number of cardiomyocytes were derived from the T-box transcription factor 18 (Tbx18) lineage. However, further lineage tracing with SM-MHCCreERT2 and Nfactc1Cre mice revealed that the new smooth muscle and endothelial cells are in fact derivatives of pre-existing coronary vessels. Our data show that neonatal mouse heart can regenerate but that its potential is limited. Moreover, although epicardial cells are multipotent during embryogenesis, their contribution to heart repair through "stem" or "progenitor" cell conversion is minimal after birth. These observations suggest that early embryonic heart development and postnatal heart regeneration are distinct biological processes.     Follow-up of patients with COVID-19 by the Delta variant after hospital discharge in Guangzhou, Guandong, China

    The B.1.617.2 (Delta) variant of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has contributed to a new increment in cases across the globe. We conducted a prospective follow-up of COVID-19 cases to explore the recurrence and potential propagation risk of the Delta variant and discuss potential explanations for the infection recurrence. A prospective, non-interventional follow-up of discharged patients who had SARS-CoV-2 infections by the Delta variant in Guangdong, China, from May 2021 to June 2021 was conducted. The subjects were asked to complete a physical health examination and undergo nucleic acid testing and antibody detection for the laboratory diagnosis of COVID-19. In total, 20.33% (25/123) of patients exhibited recurrent positive results after discharge. All patients with infection recurrence were asymptomatic and showed no abnormalities in the pulmonary computed tomography. The time from discharge to the recurrent positive testing was usually between 1-33 days, with a mean time of 9.36 days. The cycle threshold from the real-time polymerase chain reaction assay that detected the recurrence of positivity ranged from 27.48 to 39.00, with an average of 35.30. The proportion of vaccination in the non-recurrent group was higher than that in the recurrently positive group (26% vs. 4%; χ2 = 7.902; P < 0.05). Two months after discharge, the most common symptom was hair loss and 59.6% of patients had no long-term symptoms at all. It is possible for the Delta variant SARS-CoV-2 patients after discharge to show recurrent positive results of nucleic acid detection; however, there is a low risk of continuous community transmission. Both, the physical and mental quality of life of discharged patients were significantly affected. Our results suggest that it makes sense to implement mass vaccination against the Delta variant of SARS-CoV-2

    Protective Effect of a Combined Glutamine and Curcumin Formulation on Alcoholic Gastric Mucosal Damage

    Objective: This study aimed to investigate the protective effect and underlying mechanism of a combined glutamine and curcumin formulation on ethanol-induced gastric mucosal damage in rats. Method: A total of fifty SPF-grade healthy SD male rats were randomly partitioned into five groups: A normal group, a model control group, a cimetidine group, a high-dose treatment group, and a low-dose treatment group. After a period of 30 days marked by oral gavage administration, all groups, with the exception of the normal group, were euthanized post anhydrous ethanol-induced modeling. The histopathological alterations in the gastric mucosa were observed via hematoxylin & eosin (H&E) staining. Furthermore, serum levels of malondialdehyde (MDA), nitric oxide (NO), and glutathione peroxidase (GSH-PX) were ascertained using a specific reagent kit. Concurrently, the concentration of prostaglandin E2 (PGE2) within the tissue and the expression levels of heme oxygenase-1 (HO-1), NADPH quinone oxidoreductase (NQO1), the antioxidant-related nuclear factor-E2-related factor 2 (Nrf2) gene, and glycogen synthase kinase-3β (GSK-3β) were evaluated. Results: In the cimetidine and high-dose treatment groups, the incidence of gastric mucosal bleeding and other forms of injury were noticeably mitigated (P<0.05) compared to the model control group, with the high-dose treatment group demonstrating a more pronounced effect. Moreover, the model control group exhibited a significant elevation in MDA content and GSH-PX activity and a concurrent decline in NO and PGE2 levels (P<0.05). The expression of antioxidant-related genes, namely, HO-1, NQO1, and Nrf2, was significantly suppressed (P<0.05), whereas GSK-3β expression was markedly increased. In contrast, in comparison to the model control group, the cimetidine and high-dose treatment groups manifested a significant reduction in MDA content and GSH-PX activity, while NO and PGE2 levels notably increased (P<0.05). The expression of the antioxidant-related genes HO-1, NQO1, and Nrf2 was significantly returned to normal (P<0.05), and GSK-3β expression was suppressed (P<0.05). Conclusion: The combined formulation appears to exert an inhibitory effect on ethanol-induced acute gastric mucosal damage. This effect is hypothesized to be associated with the Keap1-Nrf2-ARE oxidative stress signaling pathway

    Evidence of elevation-dependent warming from the Chinese Tian Shan

    The phenomenon in which the warming rate of air temperature is amplified with elevation is termed elevation-dependent warming (EDW). It has been clarified that EDW can accelerate the retreat of glaciers and melting of snow, which can have significant impacts on the regional ecological environment. Owing to the lack of high-density ground observations in high mountains, there is widespread controversy regarding the existence of EDW. Current evidence is mainly derived from typical high-mountain regions such as the Swiss Alps, the Colorado Rocky Mountains, the tropical Andes and the Tibetan Plateau–Himalayas. Rare evidence in other mountain ranges has been reported, especially in arid regions. In this study, EDW features (regional warming amplification and altitude warming amplification) in the Chinese Tian Shan (CTM) were detected using a unique high-resolution (1 km, 6-hourly) air temperature dataset (CTMD) from 1979 to 2016. The results showed that there were significant EDW signals at different altitudes on different timescales. The CTM showed significant regional warming amplification in spring, especially in March, and the warming trends were greater than those of continental China with respect to three temperatures (minimum temperature, mean temperature and maximum temperature). The significance values of EDW above different altitude thresholds are distinct for three temperatures in 12 months. The warming rate of the minimum temperature in winter showed a significant elevation dependence (p<0.01), especially above 3000 m. The greatest altitudinal gradient in the warming rate of the maximum temperature was found above 4000 m in April. For the mean temperature, the warming rates in June and August showed prominent altitude warming amplification but with different significance above 4500 m. Within the CTM, the Tolm Mountains, the eastern part of the Borokoonu Mountains, the Bogda Mountains and the Balikun Mountains are representative regions that showed significant altitude warming amplification on different timescales. This new evidence could partly explain the accelerated melting of snow in the CTM, although the mechanisms remain to be explored

    Cardiac Sca-1+ cells are not intrinsic stem cells for myocardial development, renewal and repair

    Background: For over a decade, Sca-1+ cells within the mouse heart have been widely recognized as a stem cell population with multipotency that can give rise to cardiomyocytes, endothelial cells and smooth muscle cells in vitro and after cardiac grafting. However, the developmental origin and authentic nature of these cells remain elusive. Methods: Here, we used a series of high-fidelity genetic mouse models to characterize the identity and regenerative potential of cardiac resident Sca-1+ cells. Results: With these novel genetic mouse models, we found that Sca-1 does not label cardiac precursor cells during early embryonic heart formation. Postnatal cardiac resident Sca-1+ cells are in fact a pure endothelial cell population. They retain endothelial properties and exhibit minimal cardiomyogenic potential during development, normal aging and upon ischemic injury. Conclusions: Our study provides definitive insights into the nature of cardiac resident Sca-1+ cells. The observations challenge the current dogma that cardiac resident Sca-1+ cells are intrinsic stem cells for myocardial development, renewal and repair and suggest that the mechanisms of transplanted Sca-1+ cells in heart repair need to be reassessed

    A Two-Year Surveillance of 2009 Pandemic Influenza A (H1N1) in Guangzhou, China: From Pandemic to Seasonal Influenza?

    In this two-years surveillance of 2009 pandemic influenza A (H1N1) (pH1N1) in Guangzhou, China, we reported here that the scale and duration of pH1N1 outbreaks, severe disease and fatality rates of pH1N1 patients were significantly lower or shorter in the second epidemic year (May 2010-April 2011) than those in the first epidemic year (May 2009-April 2010) (P<0.05), but similar to those of seasonal influenza (P>0.05). Similar to seasonal influenza, pre-existing chronic pulmonary diseases was a risk factor associated with fatal cases of pH1N1 influenza. Different from seasonal influenza, which occurred in spring/summer seasons annually, pH1N1 influenza mainly occurred in autumn/winter seasons in the first epidemic year, but prolonged to winter/spring season in the second epidemic year. The information suggests a tendency that the epidemics of pH1N1 influenza may probably further shift to spring/summer seasons and become a predominant subtype of seasonal influenza in coming years in Guangzhou, China

    RPL22 Overexpression Promotes Psoriasis-Like Lesion by Inducing Keratinocytes Abnormal Biological Behavior

    BackgroundKeratinocytes of psoriasis have anti-apoptotic properties including delayed apoptosis process, accelerated proliferation metabolism and postponed differentiation process. However, the specific mechanism leading to the abnormal biological behavior of keratinocytes remains unclear.ObjectivesWe investigated the role of increased RPL22 expression in regulating the abnormal biological behavior of keratinocytes and the mechanism of regulation of RPL22 expression in skin lesions of psoriatic patients.MethodsWe examined clinical samples and utilized cytokine-induced cell and IMQ-treated mouse models. We determined the expression and functions of RPL22 in vitro and in vivo.ResultsWe showed that RPL22 expression was significantly increased in the skin lesions of psoriasis patients and IMQ-treated psoriatic-like mice. Such increased expression is attributed to hyperacetylation of histone H3K27 in the promoter region of RPL22. Interestingly, overexpression of RPL22 enhanced keratinocyte proliferation by increasing cyclinD1 expression and accelerated CD4+T cells recruitment via upregulating CXCL10 expression. Finally, we demonstrated that RPL22 overexpression promoted psoriasiform phenotypes in IMQ-induced mouse skins.ConclusionsThese findings suggested that RPL22 regulates keratinocytes abnormal biological behavior and contributes to the development of psoriatic phenotypes. Thus, RPL22 might be a novel potential molecular target for treatment of psoriasis
