Stress rapidly suppresses in vivo LH pulses and increases activation of RFRP-3 neurons in male mice
Restraint stress is a psychosocial stressor that suppresses reproductive status, including LH pulsatile secretion, but the neuroendocrine mechanisms underlying this inhibition remains unclear. Reproductive neural populations upstream of gonadotropin-releasing hormone (GnRH) neurons, such as kisspeptin, neurokinin B and RFRP-3 (GnIH) neurons, are possible targets for psychosocial stress to inhibit LH pulses, but this has not been well examined, especially in mice in which prior technical limitations prevented assessment of in vivo LH pulse secretion dynamics. Here, we examined whether one-time acute restraint stress alters in vivo LH pulsatility and reproductive neural populations in male mice, and what the time-course is for such alterations. We found that endogenous LH pulses in castrated male mice are robustly and rapidly suppressed by one-time, acute restraint stress, with suppression observed as quickly as 12–18 min. This rapid LH suppression parallels with increased in vivo corticosterone levels within 15 min of restraint stress. Although Kiss1, Tac2 and Rfrp gene expression in the hypothalamus did not significantly change after 90 or 180 min restraint stress, arcuate Kiss1 neural activation was significantly decreased after 180 min. Interestingly, hypothalamic Rfrp neuronal activation was strongly increased at early times after restraint stress initiation, but was attenuated to levels lower than controls by 180 min of restraint stress. Thus, the male neuroendocrine reproductive axis is quite sensitive to short-term stress exposure, with significantly decreased pulsatile LH secretion and increased hypothalamic Rfrp neuronal activation occurring rapidly, within minutes, and decreased Kiss1 neuronal activation also occurring after longer stress durations
The search for novel analgesics: re-examining spinal cord circuits with new tools
In this perspective, we propose the absence of detailed information regarding spinal cord
circuits that process sensory information remains a major barrier to advancing analgesia.
We highlight recent advances showing that functionally discrete populations of neurons in
the spinal cord dorsal horn play distinct roles in processing sensory information. We then
discuss new molecular, electrophysiological, and optogenetic techniques that can be
employed to understand how dorsal horn circuits process tactile and nociceptive
information. We believe this information can drive the development of entirely new classes
of pharmacotherapies that target key elements in spinal circuits to selectively modify
sensory function and blunt pain
Constructing a United Disability Community: The National Council on Disability’s Discourse of Unity in the Deliberative System around Disability Rights
The National Council on Disability (NCD) is a federal agency that connects members of a broad disability community to federal policymakers within the deliberative system (Mansbridge, 2012) that constitutes the disability rights movement in the U.S. In this critical discourse analysis, the author considers the Council's depiction of the deliberative system in its publication Equality of opportunity: The making of the Americans with Disabilities Act (NCD, 2010). Paying particular attention to discourses of unity and difference within this history of the ADA and in NCD’s About Us web pages, the study looks to understand how the Council’s depiction of the disability community and portrayal of its own role within the deliberative system impacts their legitimacy within the disability rights movement. Interrogating the ways in which unity is privileged over diversity in NCD’s history of the ADA shows how the Council exhibits a consensus democratic orientation that presents the disability community as an unwavering force to be reckoned with, positions the National Council on Disability at a position of power within its deliberative system, and highlights the deliberative nature of NCD’s mission. However, the ways in which NCD’s history of the ADA downplays difference in favor of unity sidesteps stakeholder concerns and fails to engage with social difference as a resource for inclusion and collaboration. Further, NCD discourse works to define human worth in terms of work and deliberation in terms of consensus in ways that reinforce stigma around disability and exclude underrepresented groups. The author offers some practical suggestions aimed at helping the Council and other policymakers and leaders in social justice movements to incorporate more pluralist perspectives to address issues of exclusion
Analysis of Human Trafficking Cases in Rhode Island, 2009-2013
This presentation is an analysis of seven state and federal cases of human trafficking, including forced labor and sex trafficking, in Rhode Island from 2009 until 2013. In 2009, Rhode Island passed a comprehensive human trafficking law. Since then there have been six cases of sex trafficking and one case of forced labor. Sources for information on the human trafficking cases were police reports, witness statements, court documents and media reports. This presentation will briefly summarize the cases and discuss the similarities and difference among the cases and discuss of some key findings from these cases, which include: 1) Victims were trafficked to Rhode Island because of an actual or perceived environment of tolerance of prostitution 2) Mothers of victims were involved in finding their daughters on online prostitution advertising sites sites and making police reports that initiated investigations 3) Victims were identified and reported to police by educated professionals and aware citizens 4) Victims of sex trafficking were often runaway or missing teens 5) Traffickers used online prostitution sites to advertise victims of sex trafficking 6) The credibility of a domestic worker victim of forced labor differed between judges 7) Sex buyers were not arrested in connection to any of the sex trafficking cases

However, there are few convenient methods available to experimentalists for the systematic implementation of linkers in protein fusions. In this work, we describe a universal approach to the creation and insertion of focused linker libraries into protein fusions. This process, deemedprotaTETHER, utilizes reiterative oligomer design, PCR-mediated linker library generation, and restriction enzyme-free cloning methods in a straightforward, three-step cloning process. We utilize a fusion between the catalytic subunit of cAMP-dependent protein kinase A (PKAc) and green fluorescent protein (GFP) for the development of the protaTETHERmethod, implementing small linker libraries that vary by length, sequence, and predicted secondary structural elements. We analyze the impact of linker length and sequence on the expression, activity, and subcellular localization of the PKAc-GFP fusions, and use these results to select a PKAc-GFP fusion construct with robust expression and enzymatic activity. Psychometrics of the pre-schooler physical activity parenting practices instrument among a Latino sample
Background: Latino preschoolers (3-5 year old children) have among the highest rates of obesity. Low levels of physical activity (PA) are a risk factor for obesity. Characterizing what Latino parents do to encourage or discourage their preschooler to be physically active can help inform interventions to increase their PA. The objective was therefore to develop and assess the psychometrics of a new instrument: the Preschooler Physical Activity Parenting Practices (PPAPP) among a Latino sample, to assess parenting practices used to encourage or discourage PA among preschool-aged children. Methods: Cross-sectional study of 240 Latino parents who reported the frequency of using PA parenting practices. 95% of respondents were mothers; 42% had more than a high school education. Child mean age was 4.5 (±0.9) years (52% male). Test-retest reliability was assessed in 20%, 2 weeks later. We assessed the fit of a priori models using Confirmatory factor analyses (CFA). In a separate sub-sample (35%), preschool-aged children wore accelerometers to assess associations with their PA and PPAPP subscales. Results: The a-priori models showed poor fit to the data. A modified factor structure for encouraging PPAPP had one multiple-item scale: engagement (15 items), and two single-items (have outdoor toys; not enroll in sport-reverse coded). The final factor structure for discouraging PPAPP had 4 subscales: promote inactive transport (3 items), promote screen time (3 items), psychological control (4 items) and restricting for safety (4 items). Test-retest reliability (ICC) for the two scales ranged from 0.56-0.85. Cronbach’s alphas ranged from 0.5-0.9. Several sub-factors correlated in the expected direction with children’s objectively measured PA. Conclusion: The final models for encouraging and discouraging PPAPP had moderate to good fit, with moderate to excellent test-retest reliabilities. The PPAPP should be further evaluated to better assess its associations with children’s PA and offers a new tool for measuring PPAPP among Latino families with preschool-aged children
Demonstrating high-precision photometry with a CubeSat: ASTERIA observations of 55 Cancri e
ASTERIA (Arcsecond Space Telescope Enabling Research In Astrophysics) is a 6U
CubeSat space telescope (10 cm x 20 cm x 30 cm, 10 kg). ASTERIA's primary
mission objective was demonstrating two key technologies for reducing
systematic noise in photometric observations: high-precision pointing control
and high-stabilty thermal control. ASTERIA demonstrated 0.5 arcsecond RMS
pointing stability and 10 milliKelvin thermal control of its camera
payload during its primary mission, a significant improvement in pointing and
thermal performance compared to other spacecraft in ASTERIA's size and mass
class. ASTERIA launched in August 2017 and deployed from the International
Space Station (ISS) November 2017. During the prime mission (November 2017 --
February 2018) and the first extended mission that followed (March 2018 - May
2018), ASTERIA conducted opportunistic science observations which included
collection of photometric data on 55 Cancri, a nearby exoplanetary system with
a super-Earth transiting planet. The 55 Cancri data were reduced using a custom
pipeline to correct CMOS detector column-dependent gain variations. A Markov
Chain Monte Carlo (MCMC) approach was used to simultaneously detrend the
photometry using a simple baseline model and fit a transit model. ASTERIA made
a marginal detection of the known transiting exoplanet 55 Cancri e
(~\Rearth), measuring a transit depth of ppm. This is the
first detection of an exoplanet transit by a CubeSat. The successful detection
of super-Earth 55 Cancri e demonstrates that small, inexpensive spacecraft can
deliver high-precision photometric measurements.Comment: 23 pages, 9 figures. Accepted in A
Definition of the σW regulon of Bacillus subtilis in the absence of stress
Bacteria employ extracytoplasmic function (ECF) sigma factors for their responses to environmental stresses. Despite intensive research, the molecular dissection of ECF sigma factor regulons has remained a major challenge due to overlaps in the ECF sigma factor-regulated genes and the stimuli that activate the different ECF sigma factors. Here we have employed tiling arrays to single out the ECF σW regulon of the Gram-positive bacterium Bacillus subtilis from the overlapping ECF σX, σY, and σM regulons. For this purpose, we profiled the transcriptome of a B. subtilis sigW mutant under non-stress conditions to select candidate genes that are strictly σW-regulated. Under these conditions, σW exhibits a basal level of activity. Subsequently, we verified the σW-dependency of candidate genes by comparing their transcript profiles to transcriptome data obtained with the parental B. subtilis strain 168 grown under 104 different conditions, including relevant stress conditions, such as salt shock. In addition, we investigated the transcriptomes of rasP or prsW mutant strains that lack the proteases involved in the degradation of the σW anti-sigma factor RsiW and subsequent activation of the σW-regulon. Taken together, our studies identify 89 genes as being strictly σW-regulated, including several genes for non-coding RNAs. The effects of rasP or prsW mutations on the expression of σW-dependent genes were relatively mild, which implies that σW-dependent transcription under non-stress conditions is not strictly related to RasP and PrsW. Lastly, we show that the pleiotropic phenotype of rasP mutant cells, which have defects in competence development, protein secretion and membrane protein production, is not mirrored in the transcript profile of these cells. This implies that RasP is not only important for transcriptional regulation via σW, but that this membrane protease also exerts other important post-transcriptional regulatory functions
The cellular and synaptic architecture of the mechanosensory dorsal horn
The deep dorsal horn is a poorly characterized spinal cord region implicated in processing low-threshold mechanoreceptor (LTMR) information. We report an array of mouse genetic tools for defining neuronal components and functions of the dorsal horn LTMR-recipient zone (LTMR-RZ), a role for LTMR-RZ processing in tactile perception, and the basic logic of LTMR-RZ organization. We found an unexpectedly high degree of neuronal diversity in the LTMR-RZ: seven excitatory and four inhibitory subtypes of interneurons exhibiting unique morphological, physiological, and synaptic properties. Remarkably, LTMRs form synapses on between four and 11 LTMR-RZ interneuron subtypes, while each LTMR-RZ interneuron subtype samples inputs from at least one to three LTMR classes, as well as spinal cord interneurons and corticospinal neurons. Thus, the LTMR-RZ is a somatosensory processing region endowed with a neuronal complexity that rivals the retina and functions to pattern the activity of ascending touch pathways that underlie tactile perception
