    In-memory Databases in Business Information Systems

    In-memory databases are developed to keep the entire data in main memory. Compared to traditional database systems, read access is now much faster since no I/O access to a hard drive is required. In terms of write access, mechanisms are available which provide data persistence and thus secure transactions. In-memory databases have been available for a while and have proven to be suitable for particular use cases. With increasing storage density of DRAM modules, hardware systems capable of storing very large amounts of data have become affordable. In this context the question arises whether in-memory databases are suitable for business information system applications. Hasso Plattner, who developed the HANA in-memory database, is a trailblazer for this approach. He sees a lot of potential for novel concepts concerning the development of business information systems. One example is to conduct transactions and analytics in parallel and on the same database, i.e. a division into operational database systems and data warehouse systems is no longer necessary (Plattner and Zeier 2011). However, there are also voices against this approach. Larry Ellison described the idea of business information systems based on in-memory database as “wacko,” without actually making a case for his statement (cf. Bube 2010). Stonebraker (2011) sees a future for in-memory databases for business information systems but considers the division of OLTP and OLAP applications as reasonable. [From: Introduction

    In-Memory-Datenbanken halten den gesamten Datenbestand permanent im Hauptspeicher vor. Somit können lesende Zugriffe weitaus schneller erfolgen als bei traditionellen Datenbanksystemen, da keine I/O-Zugriffe auf die Festplatte erfolgen mĂŒssen. FĂŒr schreibende Zugriffe wurden Mechanismen entwickelt, die Persistenz und somit Transaktionssicherheit gewĂ€hrleisten. In-Memory-Datenbanken werden seit geraumer Zeit entwickelt und haben sich in speziellen Anwendungen bewĂ€hrt. Mit zunehmender Speicherdichte von DRAM-Bausteinen sind Hardwaresysteme wirtschaftlich erschwinglich, deren Hauptspeicher einen kompletten betrieblichen Datenbestand aufnehmen können. Somit stellt sich die Frage, ob In-Memory-Datenbanken auch in betrieblichen Anwendungssystemen eingesetzt werden können. Hasso Plattner, der mit HANA eine In-Memory-Datenbank entwickelt hat, ist ein Protagonist dieses Ansatzes. Er sieht erhebliche Potenziale fĂŒr neue Konzepte in der Entwicklung betrieblicher Informationssysteme. So könne beispielsweise eine transaktionale und eine analytische Anwendung auf dem gleichen Datenbestand laufen, d. h. eine Trennung in operative Datenbanken einerseits und Data-Warehouse-Systeme andererseits ist in der betrieblichen Informationsverarbeitung nicht mehr notwendig (Plattner und Zeier 2011).     A Comparative Study of Selected Physical and Biochemical Traits of Wild-Type and Transgenic Sorghum to Reveal Differences Relevant to Grain Quality

    Transgenic sorghum featuring RNAi suppression of certain kafirins was developed recently, to address the problem of poor protein digestibility in the grain. However, it was not firmly established if other important quality parameters were adversely affected by this genetic intervention. In the present study several quality parameters were investigated by surveying several important physical and biochemical grain traits. Important differences in grain weight, density and endosperm texture were found that serve to differentiate the transgenic grains from their wild-type counterpart. In addition, ultrastructural analysis of the protein bodies revealed a changed morphology that is indicative of the effect of suppressed kafirins. Importantly, lysine was found to be significantly increased in one of the transgenic lines in comparison to wild-type; while no significant changes in anti-nutritional factors could be detected. The results have been insightful for demonstrating some of the corollary changes in transgenic sorghum grain, that emerge from imposed kafirin suppression

    Repression of Sex4 and Like Sex Four2 Orthologs in Potato Increases Tuber Starch Bound Phosphate With Concomitant Alterations in Starch Physical Properties

    To examine the roles of starch phosphatases in potatoes, transgenic lines were produced where orthologs of SEX4 and LIKE SEX FOUR2 (LSF2) were repressed using RNAi constructs. Although repression of either SEX4 or LSF2 inhibited leaf starch degradation, it had no effect on cold-induced sweetening in tubers. Starch amounts were unchanged in the tubers, but the amount of phosphate bound to the starch was significantly increased in all the lines, with phosphate bound at the C6 position of the glucosyl units increased in lines repressed in StSEX4 and in the C3 position in lines repressed in StLSF2 expression. This was accompanied by a reduction in starch granule size and an alteration in the constituent glucan chain lengths within the starch molecule, although no obvious alteration in granule morphology was observed. Starch from the transgenic lines contained fewer chains with a degree of polymerization (DP) of less than 17 and more with a DP between 17 and 38. There were also changes in the physical properties of the starches. Rapid viscoanalysis demonstrated that both the holding strength and the final viscosity of the high phosphate starches were increased indicating that the starches have increased swelling power due to an enhanced capacity for hydration

    Downregulation of pyrophosphate: d-fructose-6-phosphate 1-phosphotransferase activity in sugarcane culms enhances sucrose accumulation due to elevated hexose-phosphate levels

    Analyses of transgenic sugarcane clones with 45–95% reduced cytosolic pyrophosphate: d-fructose-6-phosphate 1-phosphotransferase (PFP, EC 2.7.1.90) activity displayed no visual phenotypical change, but significant changes were evident in in vivo metabolite levels and fluxes during internode development. In three independent transgenic lines, sucrose concentrations increased between three- and sixfold in immature internodes, compared to the levels in the wildtype control. There was an eightfold increase in the hexose-phosphate:triose-phosphate ratio in immature internodes, a significant restriction in the triose phosphate to hexose phosphate cycle and significant increase in sucrose cycling as monitored by 13C nuclear magnetic resonance. This suggests that an increase in the hexose-phosphate concentrations resulting from a restriction in the conversion of hexose phosphates to triose phosphates drive sucrose synthesis in the young internodes. These effects became less pronounced as the tissue matured. Decreased expression of PFP also resulted in an increase of the ATP/ADP and UTP/UDP ratios, and an increase of the total uridine nucleotide and, at a later stage, the total adenine nucleotide pool, revealing strong interactions between PPi metabolism and general energy metabolism. Finally, decreased PFP leads to a reduction of PPi levels in older internodes indicating that in these developmental stages PFP acts in the gluconeogenic direction. The lowered PPi levels might also contribute to the absence of increases in sucrose contents in the more mature tissues of transgenic sugarcane with reduced PFP activity

    Variation in neurosurgical management of traumatic brain injury

    Background: Neurosurgical management of traumatic brain injury (TBI) is challenging, with only low-quality evidence. We aimed to explore differences in neurosurgical strategies for TBI across Europe. Methods: A survey was sent to 68 centers participating in the Collaborative European Neurotrauma Effectiveness Research in Traumatic Brain Injury (CENTER-TBI) study. The questionnaire contained 21 questions, including the decision when to operate (or not) on traumatic acute subdural hematoma (ASDH) and intracerebral hematoma (ICH), and when to perform a decompressive craniectomy (DC) in raised intracranial pressure (ICP). Results: The survey was completed by 68 centers (100%). On average, 10 neurosurgeons work in each trauma center. In all centers, a neurosurgeon was available within 30 min. Forty percent of responders reported a thickness or volume threshold for evacuation of an ASDH. Most responders (78%) decide on a primary DC in evacuating an ASDH during the operation, when swelling is present. For ICH, 3% would perform an evacuation directly to prevent secondary deterioration and 66% only in case of clinical deterioration. Most respondents (91%) reported to consider a DC for refractory high ICP. The reported cut-off ICP for DC in refractory high ICP, however, differed: 60% uses 25 mmHg, 18% 30 mmHg, and 17% 20 mmHg. Treatment strategies varied substantially between regions, specifically for the threshold for ASDH surgery and DC for refractory raised ICP. Also within center variation was present: 31% reported variation within the hospital for inserting an ICP monitor and 43% for evacuating mass lesions. Conclusion: Despite a homogeneous organization, considerable practice variation exists of neurosurgical strategies for TBI in Europe. These results provide an incentive for comparative effectiveness research to determine elements of effective neurosurgical care
