    An epizootic causing mortality among cultured summer flounder Paralichthys dentatus occurred in summer of 1998 at a land-based facility on Narragansett Bay, Rhode Island, USA. The disease, flounder infectious necrotizing enteritis (FINE), was characterized by reddening around the anal area, distended abdomens filled with opaque serosanguineous fluid, enteritis and necrosis of the posterior intestine. In extreme cases of the disease, the posterior intestine was detached from the anus and was observed coming out the vent. The intestine of individuals that recovered from the dsease ended in a blind-sac; the abdomens of these fish were distended, due to food and water inside the intestinal blind-sac. A bacterium was isolated from ascites fluid and kidney of moribund flounder and identified as the causative agent in challenge experiments. The pathogen was identified as Vibno carchariae by morphological and biochemical characteristics and sequence of the 16S rRNA. The LD50 estimate was 5 x 105 colony-forming units injected intraperitoneally into 100 to 200 g summer flounder

    Whether your interests lie in scientific arenas, the corporate world, or in government, you have certainly heard the praises of big data: Big data will give you new insights, allow you to become more efficient, and/or will solve your problems. While big data has had some outstanding successes, many are now beginning to see that it is not the Silver Bullet that it has been touted to be. Here our main concern is the overall impact of big data; the current manifestation of big data is constructing a Maginot Line in science in the 21st century. Big data is not lots of data as a phenomena anymore; the big data paradigm is putting the spirit of the Maginot Line into lots of data. Big data overall is disconnecting researchers and science challenges. We propose No-Boundary Thinking (NBT), applying no-boundary thinking in problem defining to address science challenges

    Monitoring activities of teenagers to comprehend their habits: study protocol for a mixed-methods cohort study

    Abstract: Background: Efforts to increase physical activity in youth need to consider which activities are most likely to be sustained over time in order to promote lifelong participation in physical activity. The Monitoring Activities of Teenagers to Comprehend their Habits (MATCH) study is a prospective cohort study that uses quantitative and qualitative methods to develop new knowledge on the sustainability of specific physical activities. Methods/design: Eight hundred and forty-three grade 5 and 6 students recruited from 17 elementary schools in New Brunswick, Canada, are followed-up three times per year. At each survey cycle, participants complete self-report questionnaires in their classroom under the supervision of trained data collectors. A sub-sample of 24 physically active students is interviewed annually using a semi-structured interview protocol. Parents (or guardians) complete telephone administered questionnaires every two years, and a health and wellness school audit is completed for each school. Discussion: MATCH will provide a description of the patterns of participation in specific physical activities in youth, and enable identification of the determinants of maintenance, decline, and uptake of participation in each activity. These data will inform the development of interventions that take into account which activities are the most likely to be maintained and why activities are maintained or dropped

    The German National Registry of Primary Immunodeficiencies (2012-2017)

    Introduction: The German PID-NET registry was founded in 2009, serving as the first national registry of patients with primary immunodeficiencies (PID) in Germany. It is part of the European Society for Immunodeficiencies (ESID) registry. The primary purpose of the registry is to gather data on the epidemiology, diagnostic delay, diagnosis, and treatment of PIDs. Methods: Clinical and laboratory data was collected from 2,453 patients from 36 German PID centres in an online registry. Data was analysed with the software Stata® and Excel. Results: The minimum prevalence of PID in Germany is 2.72 per 100,000 inhabitants. Among patients aged 1–25, there was a clear predominance of males. The median age of living patients ranged between 7 and 40 years, depending on the respective PID. Predominantly antibody disorders were the most prevalent group with 57% of all 2,453 PID patients (including 728 CVID patients). A gene defect was identified in 36% of patients. Familial cases were observed in 21% of patients. The age of onset for presenting symptoms ranged from birth to late adulthood (range 0–88 years). Presenting symptoms comprised infections (74%) and immune dysregulation (22%). Ninety-three patients were diagnosed without prior clinical symptoms. Regarding the general and clinical diagnostic delay, no PID had undergone a slight decrease within the last decade. However, both, SCID and hyper IgE- syndrome showed a substantial improvement in shortening the time between onset of symptoms and genetic diagnosis. Regarding treatment, 49% of all patients received immunoglobulin G (IgG) substitution (70%—subcutaneous; 29%—intravenous; 1%—unknown). Three-hundred patients underwent at least one hematopoietic stem cell transplantation (HSCT). Five patients had gene therapy. Conclusion: The German PID-NET registry is a precious tool for physicians, researchers, the pharmaceutical industry, politicians, and ultimately the patients, for whom the outcomes will eventually lead to a more timely diagnosis and better treatment

    Metamorphosis in the summer flounder \u3cem\u3eParalichthys dentatus\u3c/em\u3e: changes in gill mitochondria-rich cells

    Salinity tolerance changes during larval development and metamorphosis in the summer flounder (Paralichthys dentatus) and other teleosts. The physiological mechanisms responsible for osmoregulation during these early stages of development are not well understood. This study characterized changes in ultrastructure, intracellular membranes and immunoreactive Na+/K+-ATPase of mitochondria-rich cells (MRCs) in the gills of summer flounder during metamorphosis. Gill ultrastructure at the start of metamorphosis revealed only one type of MRC, which had weak reactivity to osmium and lacked a well-defined apical pit. In juveniles, two types of MRCs were observed: light-staining MRCs (LMRCs) with weak reactivity to osmium, and dark-staining MRCs (DMRCs) with strong reactivity to osmium and positioned adjacent to LMRCs. Compared with MRCs at the start of metamorphosis, the mitochondria of juvenile MRCs appeared smaller, with more transverse cristae and electron-dense matrices. Changes in MRCs during metamorphosis were also accompanied by increased immunoreactive Na+/K+-ATPase. These findings suggest that gill MRCs develop during the metamorphosis of summer flounder as the gill takes on an increasingly important osmoregulatory role

    Developmental changes in stomach, intestine, and skin glycoconjugates in summer flounder (Paralichthys dentatus): A lectin histochemical study

    This study asked whether the glycoconjugates of stomach, intestine, and skin epithelia and their mucous secretions change during metamorphosis in summer flounder. Larvae and juveniles were assayed histochemically with biotinylated lectins, and scored for differences in staining intensity and specificity between the two developmental stages. In the stomach epithelium, the number of positive lectins changed little from larvae to juveniles; however, the secreted mucus increased in staining with DBA, ECL, GSA II, PNA, and SBA lectins. In the intestine, a developmental decrease in the binding intensity of the intestinal brush border was noted for DSL, RCA120, VVA, SNA, and UEA I lectins, while intestinal goblet cells underwent a large increase in binding for GSA II. The skin mucous cells of the larvae were negative for DBA, DSL, ECL, PNA, RCA120, and SNA, but became positive in the juveniles. In conclusion, this study showed that changes in glycoconjugate expression occur during metamorphosis in the stomach, intestine, and skin of summer flounder. The changes in the gut glycoconjugates correlate developmentally with shifts in bacterial flora composition found by other studies, and therefore might be relevant to the study of bacteria-host relationships in summer flounder. © 2005 Elsevier B.V.
