    Effect of monoclonal anti-ANP antibodies on the acute functional adaptation to unilateral nephrectomy. The role of endogenous atrial natriuretic peptide (ANP) in the immediate response of sodium excretion to unilateral nephrectomy (UNX) was investigated in anesthetized euvolemic rats through measurement of UNX-induced change in plasma ANP concentration and the response of the remaining kidney to UNX following administration of monoclonal anti-ANP antibodies. The circulating ANP levels almost tripled (from 23 ± 4 to 66 ± 13 fmol/ml, P < 0.01) within two minutes after UNX, whereas no change resulted from sham intervention. In the control group receiving vehicle injection, UNX resulted in a twofold increase in urinary sodium excretion (from 1.39 ± 0.25 to 2.88 ± 0.28 µmol/min, P < 0.01) related to a decrease in the fractional reabsorption of sodium at both proximal and distal sites (estimated from fractional excretion of lithium). Urinary excretion of cyclic guanosine 3′-5′-monophosphate (cGMP) increased as well, but glomerular filtration rate did not change. In addition, UNX was associated with a short-lived (<20 min) rise in systemic arterial pressure and a transient fall in right atrial pressure. Administration of monoclonal anti-ANP antibodies totally prevented the UNX-associated natriuresis by blunting both proximal and distal tubular reabsorption of sodium, and suppressed the rise in urinary cGMP excretion following UNX. The duration of the post-UNX increase in arterial pressure was longer when compared to values observed in controls. These observations indicate that ANP release is stimulated after uninephrectomy. UNX-induced changes in arterial pressure and electrolyte excretion are altered by administration of anti-ANP antibodies, thus suggesting that ANP may be an important mediator of the acute adaptation to unilateral renal ablation

    Effect of cerium on structure modifications of a hybrid sol–gel coating, its mechanical properties and anti-corrosion behavior

    An organic–inorganic hybrid coating was developed to improve the corrosion resistance of the aluminum alloy AA 2024-T3. Organic and inorganic coatings derived from glycidoxypropyltrimethoxysilane (GPTMS) and aluminum tri-sec-butoxide Al(OsBu)3, with different cerium contents, were deposited onto aluminum by dip-coating process. Corrosion resistance and mechanical properties were investigated by electrochemical impedance measurements and nano-indentation respectively. An optimal cerium concentration of 0.01 M was evidenced. To correlate and explain the hybrid coating performances in relation to the cerium content, NMR experiments were performed. It has been shown that when the cerium concentration in the hybrid is higher than 0.01 M there are important modifications in the hybrid structure that account for the mechanical properties and anti-corrosion behavior of the sol–gel coating

    Nanomedicine represents a very significant contribution in current cancer treatment; in addition to surgical intervention, radiation and chemotherapeutic agents that unfortunately also kill healthy cells, inducing highly deleterious and often life-threatening side effects in the patient. Of the numerous nanoparticles used against cancer, gold nanoparticles had been developed for therapeutic applications. Inter alia, a large variety of den drimers, i.e. soft artificial macromolecules, have turned up as non-viral functional nanocarriers for entrapping drugs, imaging agents, and targeting molecules. This review will provide insights into the design, synthesis, functionalization, and development in biomedicine of engineered functionalized hybrid dendrimer-tangled gold nanoparticles in the domain of cancer theranostic. Several aspects are highlighted and discussed such as 1) dendrimer-entrapped gold(0) hybrid nanoparticles for the targeted imaging and treatment of cancer cells, 2) dendrimer encapsulating gold(0) nanoparticles (Au DENPs) for the delivery of genes, 3) Au DENPs for drug delivery applications, 4) dendrimer encapsulating gold radioactive nanoparticles for radiotherapy, and 5) dendrimer/dendron-complexed gold(III) nanoparticles as technologies to take down cancer cells.info:eu-repo/semantics/publishedVersio

    Optical elastic scattering for early label-free identification of clinical pathogens

    Full paper also available on https://arxiv.org/abs/1610.02980International audienceWe report here on the ability of elastic light scattering in discriminating Gram+, Gram-and yeasts at an early stage of growth (6h). Our technique is non-invasive, low cost and does require neither skilled operators nor reagents. Therefore it is compatible with automation. It is based on the analysis of the scattering pattern (scatterogram) generated by a bacterial microcolony growing on agar, when placed in the path of a laser beam. Measurements are directly performed on closed Petri dishes. The characteristic features of a given scatterogram are first computed by projecting the pattern onto the Zernike orthogonal basis. Then the obtained data are compared to a database so that machine learning can yield identification result. A 10-fold cross-validation was performed on a database over 8 species (15 strains, 1906 scatterograms), at 6h of incubation. It yielded a 94% correct classification rate between Gram+, Gram-and yeasts. Results can be improved by using a more relevant function basis for projections, such as Fourier-Bessel functions. A fully integrated instrument has been installed at the Grenoble hospital's laboratory of bacteriology and a validation campaign has been started for the early screening of SA and MRSA (Staphylococcus aureus, methicillin-resistant S. aureus) carriers. Up to now, all the published studies about elastic scattering were performed in a forward mode, which is restricted to transparent media. However, in clinical diagnostics, most of media are opaque, such as blood-supplemented agar. That is why we propose a novel scheme capable of collecting back-scattered light which provides comparable results

    International audienceA series of ZSM-5 zeolites (embryonic, microporous, hierarchical) is studied in the catalytic cracking of polypropylene in the framework of its chemical recycling. Two important zeolite features impact their catalytic performances and allow their design as efficient catalysts: porosity and acidity. They also play a key role in catalyst deactivation and regeneration. A detailed thermogravimetric and spectroscopic (operando FT-IR) analysis of the reaction, including catalyst coking and regeneration, shows the emergence of rules to design fit-for-purpose catalysts to be used in existing or grass-roots FCC units

    The VORTEX project: first results and perspectives

    (abridged) Vortex coronagraphs are among the most promising solutions to perform high contrast imaging at small angular separations. They feature a very small inner working angle, a clear 360 degree discovery space, have demonstrated very high contrast capabilities, are easy to implement on high-contrast imaging instruments, and have already been extensively tested on the sky. Since 2005, we have been designing, developing and testing an implementation of the charge-2 vector vortex phase mask based on concentric subwavelength gratings, referred to as the Annular Groove Phase Mask (AGPM). Science-grade mid-infrared AGPMs were produced in 2012 for the first time, using plasma etching on synthetic diamond substrates. They have been validated on a coronagraphic test bench, showing broadband peak rejection up to 500:1 in the L band, which translates into a raw contrast of about 6×1056\times 10^{-5} at 2λ/D2 \lambda/D. Three of them have now been installed on world-leading diffraction-limited infrared cameras (VLT/NACO, VLT/VISIR and LBT/LMIRCam). During the science verification observations with our L-band AGPM on NACO, we observed the beta Pictoris system and obtained unprecedented sensitivity limits to planetary companions down to the diffraction limit (0.10.1''). More recently, we obtained new images of the HR 8799 system at L band during the AGPM first light on LMIRCam. After reviewing these first results obtained with mid-infrared AGPMs, we will discuss the short- and mid-term goals of the on-going VORTEX project, which aims to improve the performance of our vortex phase masks for future applications on second-generation high-contrast imagers and on future extremely large telescopes (ELTs).Comment: To appear in SPIE proceedings vol. 914

    A novel statistical signal processing method to estimate effects of compounds on contractility of cardiomyocytes using impedance assays

    International audienceLabel free methods such as cell impedance assays are in vitro tests increasingly used in drug development and producing large and high-content data files. Since the current commercial software are not suited for fully automated analysis , there is a need to develop validated and rapid solutions to extract relevant information for biologists. This need is particularly obvious in the case of impedance signals analysis from cardiomyocytes. The proposed solution is based on three main steps. The first one consists in calculating five indices informing about the time variations of frequency (F), amplitude (A), shape (S) of beatings, trends (T) of the cardiomyocyte dependent on spreading, viability and attachment as well as irregularity (I) of the contractility. In a second phase, two summary statistics are proposed to test the concentration effect of drugs on the five FASTI indices. Results of the statistical tests are finally aggregated in a cardio-effect grade to compare the tested molecules in a cardio-impact scale graduated from 0 (no influence) to 10 (highly disturbed effects in cardiomy-ocytes). This innovative approach was tested using in vitro data obtained from cell impedance analysis of three known molecules (2 cardiotoxic and 1 non-cardiotoxic compounds). Results have clearly shown the ability of the proposed approach to identify significant effects on the contractility of cardiomyocytes. This solution speeds up the analysis of cardiomyocyte impedance data, takes into account all the kinetic data generated and is now available for biologists on a web-platform: i-Cardio TM developed by CYBERnano TM
