    Reproductive Cycle and Plasma Levels of Sex Steroids in Female Eurasian Perch Perca Fluviatilis

    From April 1995 to April 1996, the annual reproductive cycle of the Eurasian perch Perca fluviatilis was studied at the Fishfarming Lindre Center (Moselle, France). At monthly intervals (at intervals of 10 days during the periovulatory period), 5 females were caught and dissected. From sampled organs, the gonado-, hepato- and viscerosomatic indexes (GSI, HSI, VSI) were calculated, oocyte diameters (OD) and the plasma levels of testosterone (T), 17P-estradiol (E2), 17,20P-dihydroxy-4-pregnen-3-one (17,2Op-P) and protein-phosphorus (PPP) were measured. After the sexual resting period observed from May to August (GSI \u3c 1 %, OD \u3c 200 urn, VSI = 4-6 %), oogenesis began in September when the water temperature decreased from 26.4 to 14.1 “C. The GSI increased progressively until mid March (15 %), then rapidly until spawning (25 %, OD = 850 urn) which occurred in April (14- 15 “C). The plasma levels of T, E,, 17,2Op-P and PPP were low during the sexual resting period. E, and PPP levels increased significantly at the onset of the oogenesis in September, then the E, level raised abruptly in November (3-4 ng mL-‘). In December, the T level increased rapidly to 15-20 ng . mL-’ The testosterone, E, and PPP levels remained very high until spawning, indicating the existence of active vitellogenesis. The highest HSI (2.1-2.2 %) recorded in winter confirmed this. During the periovulatory period, a peak of E, (4 ng . mL-‘) appeared, whereas T level diminished. In this study, 17,2Op-P levels remained low (0.2-0.6 ng mL-‘) and relatively constant. No 17,2Op-P peak was observed during the periovulatory period. Sampling at 10 day intervals was probably inadequate to specify the hormonal variations related to the final oocyte maturation and the ovulation. 0 Ifremer-Elsevier, Paris

    From April 1995 to April 1996, the annual reproductive cycle of male Eurasian perch Perca fluviatilis was studied at the Lindre Center (Moselle, France). At monthly intervals, five males (mean body weight of 133 ± 43 g and total length of 175 ± 9 mm) were caught. From sampled organs, the gonadosomatic (GSI), hepatosomatic (HSI) and viscerosomatic indexes (VSI) were calculated and plasma testosterone (T) and 11-ketotestosterone (11KT) levels were measured. After the spawning period in mid-April, GSI and HSI dropped and VSI increased to 3.8 ± 0.1 %. In September, GSI reached its maximum (8.5 ± 1.8 %). At this time, spermatocytes, spermatids and spermatozoa were abundant, whereas in June only spermatogonia were observed. During winter, GSI was stable at approximately 5 %, HSI reached its maximum (1.9 ± 0.3 %) and VSI was low (2.6 ± 0.2 %). From April to November 1995, plasma T and 11KT concentrations were low (\u3c 0.5 ng·mL–1). Plasma T levels increased significantly in December and reached peak levels (12.3 ± 2.1 ng·mL–1) in January, then decreased in February and increased again until spawning in April (6.8 ± 2.1 ng·mL–1). This second elevation could be related to the beginning of a new spermatogenic cycle. Plasma levels of 11KT increased significantly from October to February 1996 (4.9 ± 1.1 ng·mL–1). From February to the spawning period (April 1996), plasma 11KT decreased significantly, but values were significantly higher than those measured in fall.     Premium food for offspring? Black-backed Woodpecker (Picoides arcticus) diet during breeding season in eastern Canada

    Knowledge on the diet of the Black-backed Woodpecker (Picoides arcticus Swainson, 1832) is fragmentary and relies on a limited number of studies. Gaps remain in our understanding of the plasticity of its diet, particularly in the eastern part of its range. The main objective of this study was to assess the diet of Black-backed Woodpeckers in burned and unburned habitats and among sexes and ages in Québec. We collected feces and fecal bags from unburned and burned habitats in the Central Laurentians ecoregion of the eastern boreal shield ecozone and assessed diets based on identified prey items. Buprestidae and Cerambycidae of the sub-family Lamiinae were the predominant prey for adult Black-backed Woodpeckers in burned habitats, and the Pythidae Pytho niger (Kirby, 1837) and Lamiinae were the most prevalent prey in unburned habitats. Lamiinae were the most predominant prey items provisioned to nestling in burned habitat, while P. niger was their predominant food in unburned habitat, followed by Cerambycidae (without Lamiinae) and Lamiinae. Our results present new insights into Black-backed Woodpecker diet where parents feed their offspring with the largest prey available, potentially providing higher fitness for their offspring. Furthermore, our study confirms that Black-backed Woodpeckers, at least in the eastern part of its range, are not restricted to feed on Lamiinae but are rather opportunistic in taking advantage from resource–pulse interactions provided by recently disturbed habitats, especially from recently burned habitats

    PGC-1-Related Coactivator Modulates Mitochondrial-Nuclear Crosstalk through Endogenous Nitric Oxide in a Cellular Model of Oncocytic Thyroid Tumours

    BACKGROUND:The PGC-1 related coactivator (PRC), which shares structural and functional features with PGC-1alpha, is believed to regulate several metabolic pathways as well as mitochondrial biogenesis. Its involvement in the early programming of cell proliferation suggests the existence of finely regulated crosstalk between mitochondrial functions and the cell cycle status. METHODOLOGY/PRINCIPAL FINDINGS:PRC-regulated pathways were explored in a cell-line model derived from mitochondrial-rich tumours with an essentially oxidative metabolism and specifically high PRC expression. The functional status of mitochondria was compared to the results of microarray analysis under conditions of temporal PRC inhibition. To specify the fine PRC regulation, the expression levels of the genes and proteins involved in the oxidative phosphorylation process were studied by real time quantitative PCR and western blotting. As in earlier studies on PGC-1alpha, we investigated the role of nitric oxide in PRC-regulated mitochondrial biogenesis and determined its action in the control of the phosphorylation status of the mitogen-activated protein kinase pathway. CONCLUSION/SIGNIFICANCE:We found that nitric oxide rapidly influences PRC expression at the transcriptional level. Focusing on mitochondrial energetic metabolism, we observed that PRC differentially controls respiratory chain complexes and coupling efficiency in a time-dependent manner to maintain mitochondrial homeostasis. Our results highlight the key role of PRC in the rapid modulation of metabolic functions in response to the status of the cell cycle

    QiSampler: evaluation of scoring schemes for high-throughput datasets using a repetitive sampling strategy on gold standards

    <p>Abstract</p> <p>Background</p> <p>High-throughput biological experiments can produce a large amount of data showing little overlap with current knowledge. This may be a problem when evaluating alternative scoring mechanisms for such data according to a gold standard dataset because standard statistical tests may not be appropriate.</p> <p>Findings</p> <p>To address this problem we have implemented the QiSampler tool that uses a repetitive sampling strategy to evaluate several scoring schemes or experimental parameters for any type of high-throughput data given a gold standard. We provide two example applications of the tool: selection of the best scoring scheme for a high-throughput protein-protein interaction dataset by comparison to a dataset derived from the literature, and evaluation of functional enrichment in a set of tumour-related differentially expressed genes from a thyroid microarray dataset.</p> <p>Conclusions</p> <p>QiSampler is implemented as an open source R script and a web server, which can be accessed at <url>http://cbdm.mdc-berlin.de/tools/sampler/</url>.</p

    HDAC Inhibition with Valproate Improves Direct Cytotoxicity of Monocytes against Mesothelioma Tumor Cells

    peer reviewedThe composition of the tumor microenvironment (TME) mediates the outcome of chemo- and immunotherapies in malignant pleural mesothelioma (MPM). Tumor-associated macrophages (TAMs) and monocyte myeloid-derived immunosuppressive cells (M-MDSCs) constitute a major fraction of the TME. As central cells of the innate immune system, monocytes exert well-characterized functions of phagocytosis, cytokine production, and antibody-dependent cell-mediated cytotoxicity (ADCC). The objective of this study was to evaluate the ability of monocytes to exert a direct cytotoxicity by cell-to-cell contact with MPM cells. The experimental model is based on cocultures between human blood-derived monocytes sorted by negative selection and mesothelioma cell lines. Data show (i) that blood-derived human monocytes induce tumor cell death by direct cell-to-cell contact, (ii) that VPA is a pharmacological enhancer of this cytotoxic activity, (iii) that VPA increases monocyte migration and their aggregation with MPM cells, and (iv) that the molecular mechanisms behind VPA modulation of monocytes involve a downregulation of the membrane receptors associated with the M2 phenotype, i.e., CD163, CD206, and CD209. These conclusions, thus, broaden our understanding about the molecular mechanisms involved in immunosurveillance of the tumor microenvironment and open new prospects for further improvement of still unsatisfactory MPM therapies

    Overview of grapevine trunk diseases in France in the 2000s

    The National Grapevine Trunk Disease Survey was conducted in France from 2003 to 2008 to monitor grapevine trunk diseases (GTDs), eutypa dieback and esca/black dead arm (BDA). Data collected from seven regions, 329 vineyards and 12 cultivars were analysed. There were great variations amongst regions in the incidence of GTDs. For esca/BDA, two groups were distinguished: vineyards in Jura and Charentes had greater incidence (93–95%) than those of Bordeaux, Alsace and Bourgogne (54–82%). Incidence increased in Charentes over the 6-year survey, with the highest values being recorded during the last 2 years. For eutypa dieback, all vineyards of Charentes were affected, with 17 to 25% of vines expressing symptoms; for the other regions, 52 to 80% of vineyards were affected, with incidences below 3%. Cultivars Savagnin and Trousseau in Jura were especially affected by esca/BDA. Instead, Ugni Blanc in Charentes was most affected by eutypa dieback. One cultivar could be significantly more affected in one region than in another. The global health status of the vineyards was also investigated. (i) For four regions, 82% (Jura) to 87% (Alsace) of the grapevines were healthy, but this percentage decreased steadily (67%) in Charentes. (ii) Plants infected by GTDs were 32 and 18% in Jura and Charentes respectively, and only 2.9% in the Bourgogne region. (iii) The unproductive plants, i.e. dead, missing, replanted or restored, represented a significant part of the losses (6.6% in Charentes to 9.9% in Jura). The extension of GTDs is discussed with regard to the abiotic and biotic factors that may favour the diseases
