    An enzymatic method for the detection of human serum albumin

    Albumin is the most abundant protein in human serum. A dye-binding method is commonly used in clinical laboratories for its estimation using different types of dyes. However, all these dye methods were interfered by a variety of compounds. Here we present a method for the detection of albumin in human serum and other biological fluids. The principle is based on the fact that lactate dehydrogenase isoenzyme-5 (LDH-5) binds specifically to Dextran-Blue (DB). Albumin inhibits the binding of LDH-5 with DB. Absence of LDH activity in DB fraction after gel filtration indicates the presence of albumin in sample and vice versa

    Diagnosis of pulmonary embolism with helical CT scan

    Objective: To determine the role of helical C.T. scan in the diagnosis of pulmonary embolism.SETTING: Radiology department, Aga Khan University Hospital. One year prospective study May 2001 to May 2002.Methods: Sixty patients suspected of having pulmonary embolism were included in this study. Helical C.T. scan chest was performed from aortic arch to lung bases with intravenous contrast medium. Images were acquired in mediastinal and lung windows. Image interpretation was performed on the console and hard copies for main branch and segmental thrombus.Results: Fifteen patients were diagnosed of having pulmonary embolism on C.T. scan. Clinical correlation and follow up with pulmonary angiography and Doppler ultrasound of leg was carried out. Pulmonary angiography was performed in one patient with embolectomy done after confirmation of diagnosis. 8 patients had associated DVT confirmed on Doppler sonography. Sensitivity of helical C.T. is 93.75%, specificity 100% and positive predictive value 95.65%.CONCLUSION: Helical CT scan is a quick, easy and accurate imaging modality for diagnosing pulmonary embolism

    Detection of Four Novel Polymorphisms in PrP gene of Pakistani sheep (Damani and Hashtnagri) and goats (Kamori and Local Hairy) breeds

    Scrapie is a fatal neurodegenerative disorder of sheep and goats caused by post-translational conformational change in the host-encoded prion protein (PrPC). Susceptibility or resistance to scrapie has been associated with the presence of polymorphisms in the prion protein (PrP) gene. In the present study, we analyzed the PrP gene sequence to determine the frequency of polymorphisms in 56 sheep (28 each from Damani and Hashtnagri breeds) and 56 goats (28 each from Kamori and Local Hairy breeds). A total of 7 amino acid polymorphisms were detected in the PrP gene for sheep and 4 for goats. These amino acid polymorphisms were combined in 13 alleles and 15 genotypes in sheep and 5 alleles and 6 genotypes in goats. The overall frequency of the most sheep scrapie-resistant polymorphism (Q171R) was calculated to be 0.107. The most scrapie-susceptible polymorphism (A136V) was not detected in any of the studied sheep. The overall frequency of scrapie-associated polymorphism (H143R) in goats was found to be 0.152. Along with already known amino acid polymorphisms, two novel polymorphisms were also detected for each of sheep (Q171N and T191I) and goats (G22C and P63L). However, the overall frequency of these polymorphisms was extremely low

    [NiFe]-hydrogenase maturation in vitro: analysis of the roles of the HybG and HypD accessory proteins

    [NiFe]-hydrogenases (Hyd) bind a nickel-iron-based cofactor. The Fe ion of the cofactor is bound by two cyanide ligands and a single carbon monoxide ligand. Minimally six accessory proteins (HypA–HypF) are necessary for NiFe(CN)2CO cofactor biosynthesis in Escherichia coli. It has been shown that the anaerobically purified HypC–HypD–HypE scaffold complex carries the Fe(CN)2CO moiety of this cofactor. In the present study, we have purified the HybG–HypDE complex and used it to successfully reconstitute in vitro active Hyd from E. coli. HybG is a homologue of HypC that is specifically required for the maturation of Hyd-2 and also functions in the maturation of Hyd-1 of E. coli. Maturation of active Hyd-1 and Hyd-2 could be demonstrated in extracts derived from HybG- and HypD-deficient E. coli strains by adding anaerobically purified HybG–HypDE complex. In vitro maturation was dependent on ATP, carbamoylphosphate, nickel and reducing conditions. Hydrogenase maturation was prevented when the purified HybG–HypDE complex used in the maturation assay lacked a bound Fe(CN)2CO moiety. These findings demonstrate that it is possible to isolate incompletely processed intermediates on the maturation pathway and to use these to activate apo-forms of [NiFe]-hydrogenase large subunits

    The lack of access to safe medicines and quality healthcare services in peri-urban and rural areas is a major challenge driving a health system to innovate new models of care. This commentary will discuss the implementation and impact of the “Guddi baji” tele-pharmacy model, a project piloted by doctHERs, one of Pakistan’s leading telemedicine organizations. This innovative model has described the reintegration of women into the workforce by leveraging technology to improve the level of primary health care services and contributes to safe medication practice in a remote area. Our intervention proposed the deployment of technology-enabled, female frontline health workers known as the Guddi baji (meaning The Good Sister) in a rural village. They serve as an “access point to health care” that is linked to a remotely located health care professional; a licensed doctor or a pharmacist within this model

    A novel category detection of social media reviews in the restaurant industry

    Social media platforms have enabled users to share their thoughts, ideas, and opinions on different subject matters and meanwhile generate lots of information which can be adopted to understand people’s emotion towards certain products. This information can be effectively applied for Aspect Category Detection (ACD). Similarly, people’s emotions and recommendation-based Artificial Intelligence (AI)-powered systems are in trend to assist vendors and other customers to improve their standards. These systems have applications in all sorts of business available on multiple platforms. However, the current conventional approaches fail in providing promising results. Thus, in this paper, we propose novel convolutional attention-based bidirectional modified LSTM by combining the techniques of the next word, next sequence, and pattern prediction with ACD. The proposed approach extracts significant features from public reviews to detect entity and attribute pair, which are treated as a sequence or pattern from a given opinion. Next, we trained our word vectors with the proposed model to strengthen the ACD process. Empirically, we compare the approach with the state-of-the-art ACD models that use SemEval-2015, SemEval-2016, and SentiHood datasets. Results show that the proposed approach effectively achieves 78.96% F1-Score on SemEval-2015, 79.10% F1-Score on SemEval-2016, and 79.03% F1-Score on SentiHood which is higher than the existing approaches

    A New CuSe-TiO2-GO Ternary Nanocomposite: Realizing a High Capacitance and Voltage for an Advanced Hybrid Supercapacitor

    A high capacitance and widened voltage frames for an aqueous supercapacitor system are challenging to realize simultaneously in an aqueous medium. The severe water splitting seriously restricts the narrow voltage of the aqueous electrolyte beyond 2 V. To overcome this limitation, herein, we proposed the facile wet-chemical synthesis of a new CuSe-TiO2-GO ternary nanocomposite for hybrid supercapacitors, thus boosting the specific energy up to some maximum extent. The capacitive charge storage mechanism of the CuSe-TiO2-GO ternary nanocomposite electrode was tested in an aqueous solution with 3 M KOH as the electrolyte in a three-cell mode assembly. The voltammogram analysis manifests good reversibility and a remarkable capacitive response at various currents and sweep rates, with a durable rate capability. At the same time, the discharge/charge platforms realize the most significant capacitance and a capacity of 920 F/g (153 mAh/g), supported by the impedance analysis with minimal resistances, ensuring the supply of electrolyte ion diffusion to the active host electrode interface. The built 2 V CuSe-TiO2-GO||AC-GO||KOH hybrid supercapacitor accomplished a significant capacitance of 175 F/g, high specific energy of 36 Wh/kg, superior specific power of 4781 W/kg, and extraordinary stability of 91.3% retention relative to the stable cycling performance. These merits pave a new way to build other ternary nanocomposites to achieve superior performance for energy storage devices
