    Embryopathic Effects of Waterborne and Sediment-Accumulated Cadmium, Mercury and Zinc on Reproduction and Survival of Fish and Amphibian Populations in Kentucky

    Fish and amphibian egg cultures were used to determine the embryopathic effects of cadmium, mercury, and zinc released from natural and metal-enriched sediments, and to develop egg culture bioassay procedures suitable for monitoring bottom sediments for hazardous contaminants. Eggs of the narrow-mouthed toad (Gastrophryne carolinensis), goldfish (Carassius auratus), and rainbow trout (Salmo gairdnei) were cultured in contaminant-free water added to natural and metal-enriched sediments. Exposure was initiated after fertilization (toad, goldfish) or at 10 days prehatching (trout) and maintained continuously through 4-10 days posthatching. Sediments were enriched with 0.1-100 ppm cadmium and mercury and 1.0-1000 ppm zinc. Natural control sediments contained average concentrations of 0.052 ppm mercury, 1.0 ppm cadmium, and 108.2 ppm zinc. Substantial frequencies of mortality and teratogenesis occurred for all 3 animal species when eggs were cultured over natural elements further enriched with as little as 0.1-1.0 ppm cadmium or mercury and 1-10ppm zinc. Survival of trout embryos and alevins closely paralleled sediment test concentrations. The sediment TL50 concentrations for trout stages cultured from 10 days prehatching through 10 days posthatching were approximately 1ppm for mercury, 2.15 ppm for cadmium, and 210.6 ppm for zinc.     Long distance biotic dispersal of tropical seagrass seeds by marine mega-herbivores

    Terrestrial plants use an array of animals as vectors for dispersal, however little is known of biotic dispersal of marine angiosperms such as seagrasses. Our study in the Great Barrier Reef confirms for the first time that dugongs (Dugong dugon) and green sea turtles (Chelonia mydas) assist seagrass dispersal. We demonstrate that these marine mega-herbivores consume and pass in faecal matter viable seeds for at least three seagrass species (Zostera muelleri, Halodule uninervis and Halophila decipiens). One to two seagrass seeds per g DW of faecal matter were found during the peak of the seagrass reproductive season (September to December), with viability on excretion of 9.13% ± 4.61% (SE). Using population estimates for these mega-herbivores, and data on digestion time (hrs), average daily movement (km h) and numbers of viable seagrass seeds excreted (per g DW), we calculated potential seagrass seed dispersal distances. Dugongs and green sea turtle populations within this region can disperse >500,000 viable seagrass seeds daily, with a maximum dispersal distance of approximately 650 km. Biotic dispersal of tropical seagrass seeds by dugongs and green sea turtles provides a large-scale mechanism that enhances connectivity among seagrass meadows, and aids in resilience and recovery of these coastal habitats

    HI intensity mapping with the MIGHTEE survey: power spectrum estimates

    Intensity mapping (IM) with neutral hydrogen is a promising avenue to probe the large scale structure of the Universe. With MeerKAT single-dish measurements, we are constrained to scales >1>1 degree, and this will allow us to set important constraints on the Baryon acoustic oscillations and redshift space distortions. However, with MeerKAT's interferometric observation, we can also probe relevant cosmological scales. In this paper, we establish that we can make a statistical detection of HI with one of MeerKAT's existing large survey projects (MIGHTEE) on semi-linear scales, which will provide a useful complementarity to the single-dish IM. We present a purpose-built simulation pipeline that emulates the MIGHTEE observations and forecast the constraints that can be achieved on the HI power spectrum at z=0.27z = 0.27 for k>0.3k > 0.3 Mpc−1\rm{Mpc}^{-1} using the foreground avoidance method. We present the power spectrum estimates with the current simulation on the COSMOS field that includes contributions from HI, noise and point source models from the data itself. The results from our \textit{visibility} based pipeline are in good agreement to the already available MIGHTEE data. This paper demonstrates that MeerKAT can achieve very high sensitivity to detect HI with the full MIGHTEE survey on semi-linear scales (signal-to-noise ratio >7> 7 at k=0.49k=0.49 Mpc−1\rm{Mpc}^{-1}) which are instrumental in probing cosmological quantities such as the spectral index of fluctuation, constraints on warm dark matter, the quasi-linear redshift space distortions and the measurement of the HI content of the Universe up to z∼0.5z\sim 0.5.Comment: 12 pages, 8 figures, Submitted to MNRAS, comments welcom

    Artificial Metalloenzymes as Catalysts for Oxidative Lignin Degradation

    We report novel artificial metalloenzymes (ArMs), containing tris­(pyridylmethyl)­amine (TPA), for the atom economic oxidation of lignin β-O-4 model compounds, using hydrogen peroxide. The protein scaffold alters the selectivity of the reaction from a low yielding cleavage reaction when using the parent Fe-tpa complex to a high yielding benzylic alcohol oxidation when using the complex incorporated into a protein scaffold, SCP-2L A100C. Engineering the protein scaffold to incorporate glutamic acid was found to improve the ArM activity, showing that rational design of the protein environment using metal binding amino acids can be a first step toward improving the overall activity of an artificial metalloenzyme

    Quasar Candidates in the Hubble Deep Field

    We focus on the search for unresolved faint quasars and AGN in the crude combine images using a multicolor imaging analysis that has proven very successful in recent years. Quasar selection was carried out both in multicolor space and in "profile space," defined as the multi-parameter space formed by the radial profiles of the objects in the different images. By combining the dither frames available for each filter, we were able to obtain well-sampled radial profiles of the objects and measure their deviation from that of a stellar source. We also generated synthetic quasar spectra in the range 1.0 < z < 5.5 and computed expected quasar colors. We determined that the data are 90% complete for point sources at 26.2, 28.0, 27.8, 26.8 in the F300W, F450W, F606W and F814W filters, respectively. We find 41 compact objects in the HDF: 8 pointlike objects with colors consistent with quasars or stars, 18 stars, and 15 slightly resolved objects, 12 of which have colors consistent with quasars or stars. We estimate the upper limit of unresolved and slightly resolved quasars/AGNs with V < 27.0 and z < 3.5 to be 20 objects (16,200 per deg^2). We find good agreement among authors on the number of stars and the lack of quasar candidates with z > 3.5. We find more quasar candidates than previous work because of our more extensive modeling and use of all of the available color information.     Veterinary drug therapies used for undesirable behaviours in UK dogs under primary veterinary care

    Undesirable behaviours (UBs) in dogs are common and important issues with serious potential welfare consequences for both the dogs and their owners. This study aimed to investigate the usage of drug therapy for UBs in dogs and assess demographic risk factors for drug-prescribed UBs within the dog population under primary-care veterinary care in the UK in 2013. Dogs receiving drug therapy for UB were identified through the retrospective analysis of anonymised electronic patient records in VetCompass™. Risk factor analysis used multivariable logistic regression modelling. The study population comprised 103,597 dogs under veterinary care in the UK during 2013. There were 413 drug-prescribed UBs recorded among 404 dogs. The prevalence of dogs with at least one UB event treated with a drug in 2013 was 0.4%. Multivariable modelling identified 3 breeds with increased odds of drug-prescribed UB compared with crossbred dogs: Toy Poodle (OR 2.75), Tibetan Terrier (OR 2.68) and Shih-tzu (OR 1.95). Increasing age was associated with increased odds of drug-prescribed UB, with dogs ≥ 12 years showing 3.1 times the odds compared with dogs < 3 years. Neutered males (OR 1.82) and entire males (OR 1.50) had increased odds compared with entire females. The relatively low prevalence of dogs with at least one UB event that was treated with a drug in 2013 could suggest that opportunities for useful psychopharmaceutical intervention in UBs may be being missed in first opinion veterinary practice. While bodyweight was not a significant factor, the 3 individual breeds at higher odds of an UB treated with a behaviour modifying drug all have a relatively low average bodyweight. The current results also support previous research of a male predisposition to UBs and it is possible that this higher risk resulted in the increased likelihood of being prescribed a behaviour modifying drug, regardless of neuter status

    Engineering Thermostability in Artificial Metalloenzymes to Increase Catalytic Activity

    Protein engineering has shown widespread use in improving the industrial application of enzymes and broadening the conditions they are able to operate under by increasing their thermostability and solvent tolerance. Here, we show that protein engineering can be used to increase the thermostability of an artificial metalloenzyme. Thermostable variants of the human steroid carrier protein 2L, modified to bind a metal catalyst, were created by rational design using structural data and a 3DM database. These variants were tested to identify mutations that enhanced the stability of the protein scaffold, and a significant increase in melting temperature was observed with a number of modified metalloenzymes. The ability to withstand higher reaction temperatures resulted in an increased activity in the hydroformylation of 1-octene, with more than fivefold improvement in turnover number, whereas the selectivity for linear aldehyde remained high up to 80%

    Versatile Diphosphine Chelators for Radiolabeling Peptides with <sup>99m</sup>Tc and <sup>64</sup>Cu

    We have developed a diphosphine (DP) platform for radiolabeling peptides with 99mTc and 64Cu for molecular SPECT and PET imaging, respectively. Two diphosphines, 2,3-bis(diphenylphosphino)maleic anhydride (DP Ph) and 2,3-bis(di- p-tolylphosphino)maleic anhydride (DP Tol), were each reacted with a Prostate Specific Membrane Antigen-targeted dipeptide (PSMAt) to yield the bioconjugates DP Ph-PSMAt and DP Tol-PSMAt, as well as an integrin-targeted cyclic peptide, RGD, to yield the bioconjugates DP Ph-RGD and DP Tol-RGD. Each of these DP-PSMAt conjugates formed geometric cis/ trans-[MO 2(DP X-PSMAt) 2] + (M = 99mTc, 99gTc, natRe; X = Ph, Tol) complexes when reacted with [MO 2] + motifs. Furthermore, both DP Ph-PSMAt and DP Tol-PSMAt could be formulated into kits containing reducing agent and buffer components, enabling preparation of the new radiotracers cis/ trans-[ 99mTcO 2(DP Ph-PSMAt) 2] + and cis/ trans-[ 99mTcO 2(DP Tol-PSMAt) 2] + from aqueous 99mTcO 4 - in 81% and 88% radiochemical yield (RCY), respectively, in 5 min at 100 °C. The consistently higher RCYs observed for cis/ trans-[ 99mTcO 2(DP Tol-PSMAt) 2] + are attributed to the increased reactivity of DP Tol-PSMAt over DP Ph-PSMAt. Both cis/ trans-[ 99mTcO 2(DP Ph-PSMAt) 2] + and cis/ trans-[ 99mTcO 2(DP Tol-PSMAt) 2] + exhibited high metabolic stability, and in vivo SPECT imaging in healthy mice revealed that both new radiotracers cleared rapidly from circulation, via a renal pathway. These new diphosphine bioconjugates also furnished [ 64Cu(DP X-PSMAt) 2] + (X = Ph, Tol) complexes rapidly, in a high RCY (&gt;95%), under mild conditions. In summary, the new DP platform is versatile: it enables straightforward functionalization of targeting peptides with a diphosphine chelator, and the resulting bioconjugates can be simply radiolabeled with both the SPECT and PET radionuclides, 99mTc and 64Cu, in high RCYs. Furthermore, the DP platform is amenable to derivatization to either increase the chelator reactivity with metallic radioisotopes or, alternatively, modify the radiotracer hydrophilicity. Functionalized diphosphine chelators thus have the potential to provide access to new molecular radiotracers for receptor-targeted imaging. </p
