    Nucleic acid drugs: a novel approach

    Nucleic acid base sequence of proteins plays a crucial role in the expression of gene. The gene is responsible for the synthesis of proteins and these proteins, which are synthesized, are responsible for the biological process and also for dreadful diseases as well. Once if the nucleic acid sequence is altered, we would be able to block or transfer the message for protein synthesis, there by preventing the particular protein, which is responsible for the disease. These nucleic acids act as drugs by different mechanisms, they may bind with the synthesized proteins, and they can hybridize to a messenger RNA leading to translation arrest or may induce degradation to target RNA. In this way the nucleic acids act as drug for inhibiting gene expression or protein synthesis. This article examines about the different types of nucleic acid drugs, their mechanisms, advantages, drawbacks and their future prospects. Key Words: Aptamers, ribozymes, antisense oligonucleotides.     Hypoglycemic effect of white (Morus alba L.) and black (Morus nigra L.) mulberry fruits in diabetic rat

    The aim of the present study was to investigate the hypoglycemic effect of white (Morus alba L.) and black (Morus nigra L.) mulberry fruits either used individually or in a combination on alloxan diabetic rats. These fruits are reported to be rich in antioxidants, flavonoids and phenolics that can potentially fight against diabetes mellitus. Male albino rats were divided into 5 groups: normal control, alloxan-diabetic control, diabetic rats treated with white mulberry fruit powder at 5% in the diet, diabetic rats treated with black mulberry fruit powder at 5% in the diet and diabetic rats treated with mixture of white and black mulberry fruits powder at 5% in the diet. After 4 weeks of treatment, blood glucose level, liver and kidney enzymes activity, lipid profile, lipid peroxidation and histopathological studies on liver, kidney and pancreas were evaluated. The mixture of white and black mulberry fruits showed the most significant (p < 0.05) improvement in feed efficiency ratio with increasing body weight gain, as well as decrease in blood glucose level and liver-kidney dysfunction when compared with diabetic control rats. Significant decrease in serum cholesterol, triglycerides and low density lipoprotein cholesterol (LDLc) as well as significant increase in high density lipoprotein cholesterol (HDLc) in diabetic rats was observed with all treatments. Moreover, mulberry fruits administration caused significant inhibition in lipid peroxidation and α-amylase activity. In addition, the beneficial effect of all treatments was further confirmed with histopathological examination of liver, kidney and pancreas. This study reveals hypoglycemic and hypolipidemic effects of white and black mulberry fruits either used individually or in combination as a dietary supplement in alloxan diabetic rats

    Expression cloning of a candidate prostate cancer oncogene

    Genetic analysis, both karyotyping and comparative genomic hybridization, of prostate cancer cell lines and specimens have revealed multiple areas of concordant increases in DNA content. An increase of DNA in specific regions of the genome in cancer is often associated with the amplification of oncogenes. Based on these observations we have hypothesized that oncogenes are involved in the initiation or progression of prostate cancer. An expression cloning approach was utilized to identify candidate oncogenes in prostate cancer. A full-length, unidirectional cDNA expression library was constructed from DU145 prostate cancer cells. The cDNA library was screened using CP12, a rat prostate epithelial cell line. In soft agarose assays, CP12 (parental or vector transfected) do not form colonies. However, upon the introduction of a number of known oncogenes CP12 becomes anchorage independent in soft agarose. Based on this in-vitro phenotypic shift, a DU145 cDNA library was stably transfected into CP12, and selected for anchorage independence. Two hundred fifty nine anchorage independent clones were isolated. Some colonies contained more than one insert, bringing the candidate oncogene pool to approximately 400. Seven inserts were sequenced at random. Using the sequences obtained, GenBank was screened, and matches were found with p53, PARG1, a mitochondrial ATPase, RNF6, and three unknown genes that mapped to Unigene clusters. As the pool of cDNA inserts appeared promising, overexpressed genes were further selected. From 259 clones, 17 clones were overexpressed more than 6-fold in DU145 compared to Normal Prostate. From the 17 clones, 12 cDNA inserts were strongly expressed in DU145 and were isolated for sequencing. Two of the sequences, 1G6 and 3E9, were identical. Expression of 1G6/2G9/3E9 was tested by RT-PCR. 1G6/2G9/3E9 was not expressed in normal prostate, but was expressed in all prostate cancer cell lines tested as well as six prostate cancer samples. When retransfected into CP12, 1G6/2G9/3E9 induced the formation of foci and anchorage independent colonies. Thus, functional and expression data suggest that 1G6/2G9/3E9 may be a prostate cancer oncogene

    Machine learning for mathematical question difficulty classification

    This project is an experimental study on how machine learning models can be used for classification of GCE ‘A’ Level mathematical questions. Two levels of classification are carried out. First, the classification of questions to their respective topics and second, the classification of the questions to their difficulty level. The report will contain detailed explanations of the steps gone through during the experiment. The grading metrics used in this experiment are F1 Score, Precision, Recall and Accuracy. For data pre-processing three text vectorization methods, count vector, word level TF-IDF and N-gram level TF-IDF were used and tested. Four machine learning methods, Support Vector Machines, Naïve Bayes, Random Forest and Extreme Gradient Boosting, were then used to classify the data to their respective topic. Analysis was then done on the models’ performance on each topic. The same 4 machine learning methods were then again used to classify the difficulty of each question using the vectorized question and predicted topic.     Antidiabetic activity of flower buds of Michelia champaca Linn.

    Objective: To identify the antihyperglycemic activity of various extracts, petroleum ether (60-80 o ), chloroform, acetone, ethanol, aqueous and crude aqueous, of the flower buds of Michelia champaca , and to identify the antidiabetic activity of active antihyperglycemic extract. Materials and Methods: Plant extracts were tested for antihyperglycemic activity in glucose overloaded hyperglycemic rats. The effective antihyperglycemic extract was tested for its hypoglycemic activity at two-dose levels, 200 and 400 mg/kg respectively. To confirm its utility in the higher model, the effective extract of M. champaca was subjected to antidiabetic study in alloxan induced diabetic model at two dose levels, 200 and 400 mg/kg respectively. The biochemical parameters, glucose, urea, creatinine, serum cholesterol, serum triglyceride, high density lipoprotein, low density lipoprotein, hemoglobin and glycosylated hemoglobin were also assessed in the experimental animals. Results: The ethanolic extract of M. champaca exhibited significant antihyperglycemic activity but did not produce hypoglycemia in fasted normal rats. Apart from this extract, the crude aqueous and petroleum ether extracts were found active only at the end of the first hour. Treatment of diabetic rats with ethanolic extract of this plant restored the elevated biochemical parameters significantly (P< 0.05) (P< 0.01) and the activity was found dose dependent. Conclusion: This study supports the traditional claim and the ethanolic extract of this plant could be added in traditional preparations for the ailment of various diabetes-associated complications

    660-667Petroleum ether (60o-80oC), chloroform, acetone, ethanol, aqueous and crude hot water extracts of the whole plant of C. dactylon and the two fractions of aqueous extract were tested for antihyperglycaemic activity in glucose overloaded hyperglycemic rats and in alloxan induced diabetic model at two-dose levels, 200 and 400 mg/kg (po) respectively. The aqueous extract of C. dactylon and the non polysaccharide fraction of aqueous extract were found to exhibit significant antihyperglycaemic activity and only the non polysaccharide fraction was found to produce hypoglycemia in fasted normal rats. Treatment of diabetic rats with aqueous extract and non polysaccharide fraction of the plant decreased the elevated biochemical parameters, glucose, urea, creatinine, serum cholesterol, serum triglyceride, high density lipoprotein, low density lipoprotein, haemoglobin and glycosylated haemoglobin significantly. Comparatively, the non polysaccharide fraction of aqueous extract was found to be more effective than the aqueous extract
