    Effects of Symptom Perception Interventions on Trigger Identification and Quality of Life in Children with Asthma

    Background. Management of individual triggers is suboptimal in practice. In this project, we investigated the impact of symptom perception interventions on asthma trigger identification and self-reported asthma quality of life. Methods. Children with asthma ( = 227) participated in three asthma education sessions and then were randomized first to one of three home monitoring conditions (symptom monitoring and peak flow training with feedback, peak flow training without feedback, or no peak flow training) and then subsequently to one of three resistive load discrimination training conditions (signal detection training with feedback, signal detection training without feedback, or no training). Triggers were reported at enrollment, following home monitoring, and following discrimination training; quality of life was measured after home monitoring and after resistive load testing. Results. Symptom perception interventions resulted in increases in reported triggers, which increased reliably as a function of home monitoring, and increased further in participants who completed discrimination training with feedback. Increases in the number of reported asthma triggers were associated with decreases in quality of life. Discussion. Patients may benefit from strategies that make trigger-symptom contingencies clear. Complementary strategies are needed to address changes in the perceived burden of asthma which comes from awareness of new asthma triggers

    Mg2+-dependent Gating and Strong Inward Rectification of the Cation Channel TRPV6

    TRPV6 (CaT1/ECaC2), a highly Ca2+-selective member of the TRP superfamily of cation channels, becomes permeable to monovalent cations in the absence of extracellular divalent cations. The monovalent currents display characteristic voltage-dependent gating and almost absolute inward rectification. Here, we show that these two features are dependent on the voltage-dependent block/unblock of the channel by intracellular Mg2+. Mg2+ blocks the channel by binding to a site within the transmembrane electrical field where it interacts with permeant cations. The block is relieved at positive potentials, indicating that under these conditions Mg2+ is able to permeate the selectivity filter of the channel. Although sizeable outward monovalent currents were recorded in the absence of intracellular Mg2+, outward conductance is still ∼10 times lower than inward conductance under symmetric, divalent-free ionic conditions. This Mg2+-independent rectification was preserved in inside-out patches and not altered by high intracellular concentrations of spermine, indicating that TRPV6 displays intrinsic rectification. Neutralization of a single aspartate residue within the putative pore loop abolished the Mg2+ sensitivity of the channel, yielding voltage-independent, moderately inwardly rectifying monovalent currents in the presence of intracellular Mg2+. The effects of intracellular Mg2+ on TRPV6 are partially reminiscent of the gating mechanism of inwardly rectifying K+ channels and may represent a novel regulatory mechanism for TRPV6 function in vivo

    Mass balance of the Greenland ice sheet from 1958 to 2007

    We combine estimates of the surface mass balance, SMB, of the Greenland ice sheet for years 1958 to 2007 with measurements of the temporal variability in ice discharge, D, to deduce the total ice sheet mass balance. During that time period, we find a robust correlation (R2 = 0.83) between anomalies in SMB and in D, which we use to reconstruct a continuous series of total ice sheet mass balance. We. find that the ice sheet was losing 110 ± 70 Gt/yr in the 1960s, 30 ± 50 Gt/yr or near balance in the 1970s-1980s, and 97 ± 47 Gt/yr in 1996 increasing rapidly to 267 ± 38 Gt/yr in 2007. Multi-year variations in ice discharge, themselves related to variations in SMB, cause 60 ± 20 more variation in total mass balance SMB, and therefore dominate the ice sheet mass budget.     Field Observations on the Effect of a Mannan Oligosaccharide on Mortality and Intestinal Integrity of Sole (Solea senegalensis, Kaup) Infected by Photobacterium damselae subsp. piscicida

    This study was conducted in order to investigate the effect of a mannan oligosaccharide (MOS) on the intestinal morphology of sole (Solea senegalensis, Kaup) reared under commercial conditions. The dietary inclusion rate for MOS was 0.4% and it was used either alone or in combination with a vaccination regime against bacterial diseases (Pasteurella spp. and Vibrio spp.). One week after the start of the experimental period, a natural outbreak of pasteurellosis, caused by Photobacterium damselae subsp.piscicida,occurred in all the groups of fish.A two-way ANOVA showed that only MOS supplementation reduced fish mortality by ca. 8% (P = 0.050). Additionally, light microscopy examination of the intestine revealed that MOS supplementation significantly increased the mucosal folding by 29% (P = 0.016) in the anterior intestinal region and by 33% (P = 0.002) in the posterior intestinal region. Scanning electron microscopy demonstrated that both MOS supplementation and vaccination significantly increased microvilli density on the enterocyte surfaces in the anterior intestinal regionby 13% (P = 0.028) and 30% (P = 0.001) respectively. In the posterior intestinal region neither MOS supplementation nor vaccination significantly affected the microvilli density (P = 0.005).The present study suggests that dietary MOS supplementation protects the intestinal morphology of infected sole and hinders the development of pathogenic infection, possibly by binding with Photobacterium damselae subsp. piscicida, resulting in reduced mortality of infected fish
