    Japan’s New Trade Policy – A Policy Transformation Without Strategy?

    Japan’s trade policy has undergone major changes in recent years. This policy shift manifests itself in Japan’s turn towards bilateral and plurilateral trade agreements as a new pillar of its trade policy.In this book, the author identifies and analyzes the main driving forces and actors behind this policy change and discusses its potential political and economic ramifications. This book also raises the question on why Japan has failed to conduct a comprehensive and transparent evaluation of its new trade policy. The author draws on 25 expert interviews with policy makers, bureaucrats and other experts on Japanese trade policy to unveil deep insights into the complex process of trade policy-making in Japan.<br

    Characterizing Synchronization in Time Series using Information Measures Extracted from Symbolic Representations

    We present a methodology to characterize synchronization in time series based on symbolic representations. A symbol is linked to a sequence of numbers through the rank-order of its values. A representation of a time series results after mapping all sequences into symbols. We propose a transcription scheme between symbolic representations to study the dynamics of coupled systems. This scheme allows us to use elements of group theory and to derive information measures to assess the degree of synchronization. We apply our method to a prototype non-linear system which displays a rich coupled dynamics.Comment: 9 pages, 4 figure

    LRRML: a conformational database and an XML description of leucine-rich repeats (LRRs)

    Background: Leucine-rich repeats (LRRs) are present in more than 6000 proteins. They are found in organisms ranging from viruses to eukaryotes and play an important role in protein-ligand interactions. To date, more than one hundred crystal structures of LRR containing proteins have been determined. This knowledge has increased our ability to use the crystal structures as templates to model LRR proteins with unknown structures. Since the individual three-dimensional LRR structures are not directly available from the established databases and since there are only a few detailed annotations for them, a conformational LRR database useful for homology modeling of LRR proteins is desirable. Description: We developed LRRML, a conformational database and an extensible markup language (XML) description of LRRs. The release 0.2 contains 1261 individual LRR structures, which were identified from 112 PDB structures and annotated manually. An XML structure was defined to exchange and store the LRRs. LRRML provides a source for homology modeling and structural analysis of LRR proteins. In order to demonstrate the capabilities of the database we modeled the mouse Toll-like receptor 3 (TLR3) by multiple templates homology modeling and compared the result with the crystal structure. Conclusion: LRRML is an information source for investigators involved in both theoretical and applied research on LRR proteins.     Carbohydrate Targets for CAR T Cells in Solid Childhood Cancers

    Application of the CAR targeting strategy in solid tumors is challenged by the need for adequate target antigens. As a consequence of their tissue origin, embryonal cancers can aberrantly express membrane-anchored gangliosides. These are carbohydrate molecules consisting of a glycosphingolipid linked to sialic acids residues. The best-known example is the abundant expression of ganglioside GD2 on the cell surface of neuroblastomas which derive from GD2-positive neuroectoderm. Gangliosides are involved in various cellular functions, including signal transduction, cell proliferation, differentiation, adhesion and cell death. In addition, transformation of human cells to cancer cells can be associated with distinct glycosylation profiles which provide advantages for tumor growth and dissemination and can serve as immune targets. Both gangliosides and aberrant glycosylation of proteins escape the direct molecular and proteomic screening strategies currently applied to identify further immune targets in cancers. Due to their highly restricted expression and their functional roles in the malignant behavior, they are attractive targets for immune engineering strategies. GD2-redirected CAR T cells have shown activity in clinical phase I/II trials in neuroblastoma and next-generation studies are ongoing. Further carbohydrate targets for CAR T cells in preclinical development are O-acetyl-GD2, NeuGc-GM3 (N-glycolyl GM3), GD3, SSEA-4, and oncofetal glycosylation variants. This review summarizes knowledge on the role and function of some membrane-expressed non-protein antigens, including gangliosides and abnormal protein glycosylation patterns, and discusses their potential to serve as a CAR targets in pediatric solid cancers

    Analysing Large Scale Structure: I. Weighted Scaling Indices and Constrained Randomisation

    The method of constrained randomisation is applied to three-dimensional simulated galaxy distributions. With this technique we generate for a given data set surrogate data sets which have the same linear properties as the original data whereas higher order or nonlinear correlations are not preserved. The analysis of the original and surrogate data sets with measures, which are sensitive to nonlinearities, yields information about the existence of nonlinear correlations in the data. We demonstrate how to generate surrogate data sets from a given point distribution, which have the same linear properties (power spectrum) as well as the same density amplitude distribution. We propose weighted scaling indices as a nonlinear statistical measure to quantify local morphological elements in large scale structure. Using surrogates is is shown that the data sets with the same 2-point correlation functions have slightly different void probability functions and especially a different set of weighted scaling indices. Thus a refined analysis of the large scale structure becomes possible by calculating local scaling properties whereby the method of constrained randomisation yields a vital tool for testing the performance of statistical measures in terms of sensitivity to different topological features and discriminative power.Comment: 10 pages, 5 figures, accepted for publication in MNRA

    Interaction imaging with amplitude-dependence force spectroscopy

    Knowledge of surface forces is the key to understanding a large number of processes in fields ranging from physics to material science and biology. The most common method to study surfaces is dynamic atomic force microscopy (AFM). Dynamic AFM has been enormously successful in imaging surface topography, even to atomic resolution, but the force between the AFM tip and the surface remains unknown during imaging. Here, we present a new approach that combines high accuracy force measurements and high resolution scanning. The method, called amplitude-dependence force spectroscopy (ADFS) is based on the amplitude-dependence of the cantilever's response near resonance and allows for separate determination of both conservative and dissipative tip-surface interactions. We use ADFS to quantitatively study and map the nano-mechanical interaction between the AFM tip and heterogeneous polymer surfaces. ADFS is compatible with commercial atomic force microscopes and we anticipate its wide-spread use in taking AFM toward quantitative microscopy

    A Scaling Index Analysis of the WMAP three year data: Signatures of non-Gaussianities and Asymmetries in the CMB

    Local scaling properties of the co-added foreground-cleaned three-year Wilkinson Microwave Anisotropy Probe (WMAP) data are estimated using weighted scaling indices. The scaling index method (SIM) is - for the first time - adapted and applied to the case of spherical symmetric spatial data. The results are compared with 1000 Monte Carlo simulations based on Gaussian fluctuations with a best fit Λ\LambdaCDM power spectrum and WMAP-like beam and noise properties. Statistical quantities based on the scaling indices, namely the moments of the distribution and probability-based measures are determined. We find for most of the test statistics significant deviations from the Gaussian hypothesis. We find pronounced asymmetries, which can be interpreted as a global lack of structure in the northern hemisphere, which is consistent with previous findings. Furthermore, we detect a localized anomaly in the southern hemisphere, which gives rise to highly significant signature for non-Gaussianity in the spectrum of scaling indices. We identify this signature as the cold spot, which was also already detected in the first year WMAP data. Our results provide further evidence for both the presence of non-Gaussianities and asymmetries in the WMAP three-year data. More detailed bandand year-wise analyses are needed to elucidate the origin of the detected anomalies. In either case the scaling indices provide powerful nonlinear statistics to analyse CMB maps.Comment: submitted to MNRA
