Towards a Formal Approach to Validating and Verifying Functional Design for Complex Safety Critical Systems
The quality and reliability of safety criticalsoftware systems are highly dependent on proper systemvalidation and verification. In model-driven softwaredevelopment, semi-formal notations are often used inrequirements capture. Though semi-formal notations possessadvantages, their major disadvantage is their imprecision. Atechnique to eliminate imprecision is to transform semi-formalmodels into an analyzable representation using formalspecification techniques (FSTs). With this approach to systemvalidation and verification, safety critical systems can bedeveloped more reliably. This work documents early experienceof applying FSTs on UML class diagrams as attributeconstraints, and pre- post-conditions on procedures. Thevalidation and verification of the requirements of a system tomonitor unmanned aerial vehicles in unrestricted airspace is theorigin of this work. The challenge is the development of a systemwith incomplete specifications; multiple conflicting stakeholders’interests; existence of a prototype system; the need forstandardized compliance, where validation and verification areparamount, which necessitates forward and reverse engineeringactivities
An Improved Algorithm for Generating Database Transactions from Relational Algebra Specifications
Alloy is a lightweight modeling formalism based on relational algebra. In
prior work with Fisler, Giannakopoulos, Krishnamurthi, and Yoo, we have
presented a tool, Alchemy, that compiles Alloy specifications into
implementations that execute against persistent databases. The foundation of
Alchemy is an algorithm for rewriting relational algebra formulas into code for
database transactions. In this paper we report on recent progress in improving
the robustness and efficiency of this transformation
Occurrence of PAHs in water samples of the Diep River, South Africa
Occurrence of polycyclic aromatic hydrocarbons (PAHs) in freshwater may aggravate the water crisis currently being experienced in the Western Cape Province of South Africa. However, there is dearth of data on the levels of PAHs, which is necessary for effective assessment of water quality as well as remediation strategies. This study therefore assessed levels of PAHs in the Diep River freshwater system of Western Cape Province, South Africa. A liquid-liquid extraction solid-phase extraction gas chromatography flame ionisation detection (LLE-SPEGC-FID) method was developed to simultaneously determine the 16 United States Environmental Protection Agency (USEPA) listed priority PAHs in water samples. The SPE-GC-FID method allowed an acceptable linearity (R2 > 0.999) within the calibration range of 1 to 50 μg/mL. Instrument detection limits ranged between 0.02 and 0.04 μg/mL and instrument quantification limits between 0.06 and 0.13 μg/mL. Recovery study results were also acceptable (83.69–96.44%) except for naphthalene, which had recovery of 60.05% in spiked water matrix. The seasonal averages of individual PAH detected at the studied sites ranged between not detected (nd) and 72.38 ± 9.58 μg/L in water samples.Keywords: PAHs, Diep River, GC-FID, freshwate
Use of medical, surgical and complementary treatments among women with fibroids
OBJECTIVE: To examine the use of medical management, uterus-preserving surgery (UPS), and complementary treatments among women with uterine fibroids.
STUDY DESIGN: Prospective cohort study of 933 premenopausal women ages 31-54 years with symptomatic fibroids who participated in the Study of Pelvic Problems, Hysterectomy, and Intervention Alternatives (SOPHIA) for an average of 4.3 years (SD 2.5 years). Incident use of fibroid treatments was determined through annual interviews. Linear regression models were used to compare changes in fibroid-related symptoms among women who underwent UPS versus those who did not undergo surgery.
RESULTS: Participants were racially and ethnically diverse, with a mean age of 43 years. During study follow-up, 531 participants (57%) did not undergo UPS or hysterectomy, 250 (27%) had at least one UPS, and 152 (16%) underwent hysterectomy. Complementary and alternative treatments were commonly used, including exercise (45%), diet (34%), herbs (37%), and acupuncture (16%): participants reported significant symptom improvement and few side effects with these interventions. In multivariable linear regression models, women who did not undergo surgery during the study reported improvement in dyspareunia (p<.001), pelvic pain (p<.001), and menstrual cramps (p<.001). However, women who underwent UPS reported greater overall resolution of "pelvic problems" compared with women who did not have surgical treatment (difference in change score 1.18 on a four-point Likert scale, p<.001).
CONCLUSION: UPS are effective treatments for women with fibroids, but many women use hormonal or complementary treatments and report significant symptom improvement without surgical intervention
Meditation smartphone application effects on prehypertensive adults' blood pressure: Dose-response feasibility trial
OBJECTIVE:
Essential hypertension (EH) is the most common chronic disease in the United States and a major cause of morbidity and mortality. Lifestyle interventions (e.g., diet, exercise, stress management) to reduce blood pressure (BP) are often complex with varying effectiveness. Breathing awareness meditation (BAM) is a stress management strategy with encouraging effects on BP, though widespread dissemination is hampered by the lack of an easy-to-use methodology to train and monitor BAM practices. A smartphone application (Tension Tamer [TT]) that implements BAM and tracks adherence has shown promise in addressing these gaps. This 6-month dose-response feasibility trial evaluated effects of the app on BP to further optimize BAM user guidelines.
METHODS:
Sixty-four adults with prehypertension were randomized to complete TT-guided BAM sessions for 5-, 10-, or 15-min intervals twice daily over 6 months. Continuous heart rate readings derived from the phone's video camera via reflective photoplethysmography were used as feedback and as an index of time-stamped adherence. Outcomes (resting BP, HR) were collected at baseline, 1-, 3-, and 6-months.
RESULTS:
Mixed modeling results showed a significant time effect for systolic BP (SBP) with a dose-response effect at Months 3 and 6. Adherence declined over time and was lowest in the 15-min dose condition, though SBP reductions were maintained. Generally, adherence was negatively associated with dose as the study progressed.
CONCLUSIONS:

Hepadnavirus DNA Is Detected in Canine Blood Samples in Hong Kong but Not in Liver Biopsies of Chronic Hepatitis or Hepatocellular Carcinoma
Chronic hepatitis and hepatocellular carcinoma (HCC) caused by the hepadnavirus hepatitis B virus (HBV) are significant causes of human mortality. A hepatitis-B-like virus infecting cats, domestic cat hepadnavirus (DCH), was reported in 2018. DCH DNA is hepatotropic and detectable in feline blood or serum (3.2 to 12.3%). Detection of HBV DNA has been reported in sera from 10% of free-roaming dogs in Brazil, whereas 6.3% of sera from dogs in Italy tested positive for DCH DNA by real-time quantitative PCR (qPCR). If DCH, HBV, or another hepadnavirus is hepatotropic in dogs, a role for such a virus in the etiology of canine idiopathic chronic hepatitis (CH) or HCC warrants investigation. This study investigated whether DCH DNA could be detected via qPCR in blood from dogs in Hong Kong and also whether liver biopsies from dogs with confirmed idiopathic CH or HCC contained hepadnaviral DNA using two panhepadnavirus conventional PCRs (cPCR) and a DCH-specific cPCR. DCH DNA was amplified from 2 of 501 (0.4%) canine whole-blood DNA samples. A second sample taken 6 or 7 months later from each dog tested negative in DCH qPCR. DNA extracted from 101 liver biopsies from dogs in Hong Kong or the USA, diagnosed by board-certified pathologists as idiopathic CH (n = 47) or HCC (n = 54), tested negative for DCH DNA and also tested negative using panhepadnavirus cPCRs. This study confirms that DCH DNA can be detected in canine blood by qPCR, although at a much lower prevalence than that reported previously. Reliability of maximal respiratory nasal pressure tests in healthy young adults
[Abstract] Introduction: Sniff nasal inspiratory (SNIP) and expiratory pressure (SNEP) may complement the assessment of respiratory muscle strength. Thus, specifying their reliability is relevant to improving the clinical consistency of both tests.
Objective: To assess the reliability of SNIP and SNEP in healthy young adults.
Methods: This cross-sectional study included self-reported healthy aged 18 to 29 years. SNIP was performed using a plug to occlude one nostril, while SNEP was conducted using a facemask. Participants performed 20 SNIP and SNEP maneuvers with 30-second intervals in between. The intraclass correlation coefficient (ICC), standard error of measurement (SEM), and minimum detectable change (MDC) assessed the reliability of SNIP and SNEP. Analyses were conducted between the highest peak pressure and the first reproducible maneuver in men and women.
Results: The total sample comprised 32 participants: 16 men and 16 women. The ICC, SEM, and MDC for SNIP maneuvers were 0.994 (95%CI 0.988 to 0.997), 1.820 cmH2O, and 5.043 cmH2O, respectively. For SNEP, these parameters were 0.950 (95%CI 0.897 to 0.976), 6.03 cmH2O, and 16.716 cmH2O. The SNIP and SNEP in men showed ICC of 0.992 (95%CI 0.977 to 0.997) and 0.877 (95%CI 0.648 to 0.957), SEM of 2.07 and 7.66 cmH2O, and MDC of 5.74 and 21.23 cmH2O. In women, SNIP and SNEP presented ICC of 0.992 (95%CI 0.977 to 0.997) and 0.957 (95%CI 0.878 to 0.985), SEM of 1.15 and 6.11 cmH2O, and MDC of 3.19 and 16.95 cmH2O. Also, 60% of the highest SNIPs occurred among the 11th and 20th maneuvers in men and women. In men, 55% of the highest SNEPs occurred among the 11th and 20th maneuvers; this value was 50% in women.
Conclusion: SNIP and SNEP showed excellent reliability. The reliability of SNIP and SNEP in men was good and excellent, respectively, whereas both tests had excellent reliability in women. Extrahypothalamic oxytocin neurons drive stress-induced social vigilance and avoidance
Oxytocin increases the salience of both positive and negative social contexts and it is thought that these diverse actions on behavior are mediated in part through circuit-specific action. This hypothesis is based primarily on manipulations of oxytocin receptor function, leaving open the question of whether different populations of oxytocin neurons mediate different effects on behavior. Here we inhibited oxytocin synthesis in a stress-sensitive population of oxytocin neurons specifically within the medioventral bed nucleus of the stria terminalis (BNSTmv). Oxytocin knockdown prevented social stress-induced increases in social vigilance and decreases in social approach. Viral tracing of BNSTmv oxytocin neurons revealed fibers in regions controlling defensive behaviors, including lateral hypothalamus, anterior hypothalamus, and anteromedial BNST (BNSTam). Oxytocin infusion into BNSTam in stress naïve mice increased social vigilance and reduced social approach. These results show that a population of extrahypothalamic oxytocin neurons plays a key role in controlling stress-induced social anxiety behaviors
