    Role of whole grains versus fruits and vegetables in reducing subclinical inflammation and promoting gastrointestinal health in individuals affected by overweight and obesity: a randomized controlled trial

    Background: Whole grains (WG) and fruits and vegetables (FV) have been shown to reduce the risk of metabolic disease, possibly via modulation of the gut microbiota. The purpose of this study was to determine the impact of increasing intake of either WG or FV on inflammatory markers and gut microbiota composition. Methods: A randomized parallel arm feeding trial was completed on forty-nine subjects with overweight or obesity and low intakes of FV and WG. Individuals were randomized into three groups (3 servings/d provided): WG, FV, and a control (refined grains). Stool and blood samples were collected at the beginning of the study and after 6 weeks. Inflammatory markers [tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), lipopolysaccharide binding protein (LBP), and high sensitivity C-reactive protein (hs-CRP)] were measured. Stool sample analysis included short/branched chain fatty acids (S/BCFA) and microbiota composition. Results: There was a significant decrease in LBP for participants on the WG (− 0.2 μg/mL, p = 0.02) and FV (− 0.2 μg/mL, p = 0.005) diets, with no change in those on the control diet (0.1 μg/mL, p = 0.08). The FV diet induced a significant change in IL-6 (− 1.5 pg/mL, p = 0.006), but no significant change was observed for the other treatments (control, − 0.009 pg/mL, p = 0.99; WG, − 0.29, p = 0.68). The WG diet resulted in a significant decrease in TNF-α (− 3.7 pg/mL; p \u3c 0.001), whereas no significant effects were found for those on the other diets (control, − 0.6 pg/mL, p =0.6; FV, − 1.4 pg/mL, p = 0.2). The treatments induced individualized changes in microbiota composition such that treatment group differences were not identified, except for a significant increase in α-diversity in the FV group. The proportions of Clostridiales (Firmicutes phylum) at baseline were correlated with the magnitude of change in LBP during the study Conclusions: These data demonstrate that WG and FV intake can have positive effects on metabolic health; however, different markers of inflammation were reduced on each diet suggesting that the anti-inflammatory effects were facilitated via different mechanisms. The anti-inflammatory effects were not related to changes in gut microbiota composition during the intervention, but were correlated with microbiota composition at baseline

In 2009, a team from The Australian National University (ANU), together with local people, partially excavated the site of Vasino, located close to the north coast of Timor-Leste, above the modern village of Moro-Parlamento (Figure 4.1). The site had been fortified with large stone walls and the aim was to provide more data on when, how and why these fortifications were used in the region. Two related questions guided the research. First, when was the main period of fort construction initiated? Secondly, what were the prevailing environmental and social conditions of those times

    Electrospun gelatin-based scaffolds as a novel 3D platform to study the function of contractile smooth muscle cells in vitro

    Contractile dysfunction of smooth muscle (SM) is a feature of chronic cardiovascular, respiratory and gastro-intestinal diseases. Owing to the low availability of human ex vivo tissue for the assessment of SM contractile function, the aim of this study was to develop a novel in vitro SM model that possesses the ability to contract, and a method to measure its contractility. A range of electrospun scaffolds were produced from crosslinked gelatin and methacrylated gelatin (GelMA), generating highly aligned scaffolds with average fibre diameters ranging from 200 nm to several micrometres. Young's moduli of the scaffolds ranged from 1x105 to 1x107 Pa. Primary aortic smooth muscle cells (AoSMCs; rat) cells readily adhered to and proliferated on the fibrous scaffolds for up to 10 days. They formed highly aligned populations following the topographical cues of the aligned scaffolds and stained positive for SM markers, indicating a contractile phenotype. Cell-seeded GelMA scaffolds were able, upon stimulation with uridine 5'-triphosphate (UTP), to contract and their attachment to a force transducer allowed the force of contraction to be measured. Hence, these electrospun GelMA fibres can be used as biomimetic scaffolds for SM cell culture and in vitro model development, and enables the contractile forces generated by the aligned three-dimensional sheet of cells to be directly measured. This will supplement in vitro drug screening tools and facilitate discovery of disease mechanisms

    Nrf2 activation in the human brain after stroke due to supratentorial intracerebral haemorrhage: a case–control study

    AIMS: Pharmacological activation of the antioxidative transcription factor nuclear factor erythroid 2-related factor 2 (Nrf2) improves outcomes in experimental models of intracerebral haemorrhage (ICH). However, the Nrf2 pathway has not been previously studied in humans after ICH. Our study aims to address this gap. METHODS: We selected cases with fatal ICH from a prospective community-based inception cohort study and age-matched and sex-matched controls who died suddenly of non-neurological disease. We used immunohistochemistry to quantify Nrf2 (% total area stained overall and % of nuclei stained) and CD68 expression in controls and perihaematomal, ipsilateral and contralateral brain tissue from cases. We measured downstream haem oxygenase-1 (HMOX1) and NAD(P)H dehydrogenase quinone 1 [NQO1] expression using RNA in situ hybridisation. RESULTS: 26 ICH cases (median age: 82 (IQR 76–86); 13 (50%) male) and eight controls (median age: 79 (IQR 77–80); 3 (37.5%) male) were included. We found no significant differences in overall % of Nrf2 staining between ICH cases and controls. However, the mean % of nuclei staining for Nrf2 seemed higher in perihaematomal compared with contralateral regions, although this was only statistically significant >60 days after ICH (25% (95% CI 17% to 33%) vs 14% (95% CI 11% to 17%), p=0.029). The percentage of perihaematomal tissue staining for CD68 was higher >60 days after ICH (6.75%, 95% CI 2.78% to 10.73%) compared with contralateral tissue (1.45%, 95% CI 0.93% to 1.96%, p=0.027) and controls (1.08%, 95% CI 0.20% to 1.97%, p=0.0008). RNA in situ hybridisation suggested increased abundance of HMOX1 and NQO1 transcripts in perihaematomal versus distant ipsilateral brain tissue obtained <7 days from onset of ICH. CONCLUSIONS: We found evidence of Nrf2 activation in human brain tissue after ICH. Pharmacological augmentation of Nrf2 activation after ICH might be a promising therapeutic approach

    Characterisation of ammonia emissions from gasoline and gasoline hybrid passenger cars

    Recent evidence suggests NH3 emissions from road vehicles play an important role in the formation of fine particulate matter, especially in urban areas. However, there is little data available for NH3 emitted from road vehicles under real driving conditions, in part due to its lack of regulation in vehicle emission legislation. In this study, we use 210,000 vehicle emission remote sensing measurements to evaluate the complex mix of factors affecting NH3 emissions from gasoline and gasoline hybrid passenger cars. The influence of vehicle model year and manufacturer on NH3 emissions is considered, as well as the effect of vehicle deterioration. It is found that the amount of NH3 emitted increases as vehicle mileage increases. A comparison of cold start and hot exhaust NH3 emissions reveals that on average, cold start emissions are a factor of 1.7 times higher. New NH3 emission factors are developed, in addition to speed-emission curves that are potentially useful for national inventories. A new application of remote sensing data is reported, whereby the proportion of failed CO2 measurements for hybrid vehicles provides unique insight into the real world battery use of both conventional hybrid electric and plug-in hybrid electric vehicles, which is used to refine the NH3 emission factors for these vehicles

    Underestimated Ammonia Emissions from Road Vehicles

    In this study, we use comprehensive vehicle emission remote sensing measurements of over 230,000 passenger cars to estimate total UK ammonia (NH3) emissions. Estimates are made using "top-down"and "bottom-up"methods that demonstrate good agreement to within 1.1% for total fuel consumed or CO2 emitted. A central component of this study is the comprehensive nature of the bottom-up emission estimates that combine highly detailed remote sensing emission data with over 4000 km of 1 Hz real driving data. Total annual UK NH3 emissions from gasoline passenger cars are estimated to be 7.8 ± 0.3 kt from the bottom-up estimate compared with 3.0 ± 1.7 kt reported by the UK national inventory. An important conclusion from the analysis is that both methodologies confirm that gasoline passenger car NH3 emissions are underestimated by a factor of about 2.6 compared with the 2018 UK National Atmospheric Emissions Inventory. Furthermore, we find that inventory estimates of urban emissions of NH3 for passenger cars are underestimated by a factor of 17

    Intercultural ethics: questions of methods in language and intercultural communication

    This paper explores how questions of ethics and questions of method are intertwined and unavoidable in any serious study of language and intercultural communication. It argues that the focus on difference and solution orientations to intercultural conflict has been a fundamental driver for theory, data collection and methods in the field. These approaches, the paper argues, have created a considerable consciousness raising industry, with methods, trainings and ‘critical incidents’, which ultimately focus intellectual energy in areas which may be productive in terms of courses and publications but which have a problematic basis in their ethical terrain. Dieser Artikel untersucht wie ethische und methodische Fragen nicht nur ineinander greifen, sondern in keiner ernstzunehmenden Studie ueber Sprache und interkulturelle Kommunikation ausgelassen werden duerfen. Es wird hier argumentiert, dass der Schwerpunkt auf Verschiedenheit und Problemorientierung im interkulturellen Konflikt einen wesentlichen Einfluss auf theoretische Entwicklungen, Datenerhebung und Methoden in diesem Bereich hatte. Dieser Artikel legt auch dar, wie diese Ansaetze eine betraechtliche ‘Bewusstseinsbildungs – Branche' erzeugt haben, mit Methoden, Trainings, und ‘kritischen Interaktionssituationen’, welche letztendlich allen intellektuellen Arbeitseifer auf Bereiche konzentriert hat, die zwar ertragreich sind in Bezug auf Kurse und Publikationen, jedoch eine problematische Grundlage im ethischen Bereich aufweisen

    Prevalence of HIV among blood donors at Juba Teaching Hospital Blood Bank, South Sudan

    Objectives: The aim of this study is to determine the prevalence of HIV among blood donors in Juba Teaching Hospital Blood Bank, South Sudan in 2013.Method and Materials: This is a retrospective study that involved the abstraction of data from registers at the blood bank. Data were collected onto data sheets and entered into a computer database. Statistical analysis was performed using SPSS Version 20 Software. A p value of &lt;0.05 was considered statistically significant.Results: Out of 1095 blood donors, 1074 (98.1%) were males and 21 (1.9%) were females. The mean age and the range for the whole group was 29+7.16 (15-69) yrs. The prevalence of HIV was higher among males than females 85 (7.9%) vs 1 (4.8%) respectively but this was not statistical significant (p=0.6). The 20 to 29 year age group had the highest prevalence of 49 (57%) with no statistical significance (p=0.3). The prevalence of HIV was 7. % (86) and there were co-infections between HIV and HBV, HCV and syphilis of14 (50%), 5 (18%), 9 (32%) with p=0.7, p=0.1, p=0.8 respectively. Blood group O positive had the highest percentage 58.1 % (n=50) and was the commonest group.Conclusion: In this study, HIV prevalence is very high among blood donors at the Juba Teaching Hospital blood bank

    Verification of a National Emission Inventory and Influence of On-road Vehicle Manufacturer-Level Emissions

    Road vehicles make important contributions to a wide range of pollutant emissions from the street level to global scales. The quantification of emissions from road vehicles is, however, highly challenging given the number of individual sources involved and the myriad factors that influence emissions such as fuel type, emission standard, and driving behavior. In this work, we use highly detailed and comprehensive vehicle emission remote sensing measurements made under real driving conditions to develop new bottom-up inventories that can be compared to official national inventory totals. We find that the total UK passenger car and light-duty van emissions of nitrogen oxides (NOx) are underestimated by 24-32%, and up to 47% in urban areas, compared with the UK national inventory, despite agreement within 1.5% for total fuel used. Emissions of NOx at a country level are also shown to vary considerably depending on the mix of vehicle manufacturers in the fleet. Adopting the on-road mix of vehicle manufacturers for six European countries results in up to a 13.4% range in total emissions of NOx. Accounting for the manufacturer-specific fleets at a country level could have a significant impact on emission estimates of NOx and other pollutants across the European countries, which are not currently reflected in emission inventories
