    Measurements of CO2 exchange over a woodland savanna (Cerrado Sensu stricto) in southeast Brasil.

    The technique of eddy correlation was used to measure the net ecosystem exchange over a woodland savanna (Cerrado Sensu stricto) site (Gleba PĂŠ de Gigante) in southeast Brazil. The data set included measurements of climatological variables and soil respiration using static soil chambers. Data were collected during the period from 10 October 2000 to 30 March 2002. Measured soil respiration showed average values of 4.8 molCO m-2s-1 year round. Its 2 seasonal differences varied from 2 to 8 molCO m-2s-1 (Q = 4.9) during the dry (April to August) and wet season, respectively, and was concurrent with soil temperature and moisture variability. The net ecosystem CO2 flux (NEE) variability is controlled by solar radiation, temperature and air humidity on diel course. Seasonally, soil moisture plays a strong role by inducing litterfall, reducing canopy photosynthetic activity and soil respiration. The net sign of NEE is negative (sink) in the wet season and early dry season, with rates around -25 kgC ha-1day-1, and values as low as 40 kgC ha- 1day-1. NEE was positive (source) during most of the dry season, and changed into negative at the onset of rainy season. At critical times of soil moisture stress during the late dry season, the ecosystem experienced photosynthesis during daytime, although the net sign is positive (emission). Concurrent with dry season, the values appeared progressively positive from 5 to as much as 50 kgC ha-1day-1. The annual NEE sum appeared to be nearly in balance, or more exactly a small sink, equal to 0.1 0.3 tC ha-1yr-1, which we regard possibly as a realistic one, giving the constraining conditions imposed to the turbulent flux calculation, and favourable hypothesis of succession stages, climatic variability and CO2 fertilization

    The need to promote behaviour change at the cultural level: one factor explaining the limited impact of the MEMA kwa Vijana adolescent sexual health intervention in rural Tanzania. A process evaluation

    Background - Few of the many behavioral sexual health interventions in Africa have been rigorously evaluated. Where biological outcomes have been measured, improvements have rarely been found. One of the most rigorous trials was of the multi-component MEMA kwa Vijana adolescent sexual health programme, which showed improvements in knowledge and reported attitudes and behaviour, but none in biological outcomes. This paper attempts to explain these outcomes by reviewing the process evaluation findings, particularly in terms of contextual factors. Methods - A large-scale, primarily qualitative process evaluation based mainly on participant observation identified the principal contextual barriers and facilitators of behavioural change. Results - The contextual barriers involved four interrelated socio-structural factors: culture (i.e. shared practices and systems of belief), economic circumstances, social status, and gender. At an individual level they appeared to operate through the constructs of the theories underlying MEMA kwa Vijana - Social Cognitive Theory and the Theory of Reasoned Action – but the intervention was unable to substantially modify these individual-level constructs, apart from knowledge. Conclusion - The process evaluation suggests that one important reason for this failure is that the intervention did not operate sufficiently at a structural level, particularly in regard to culture. Recently most structural interventions have focused on gender or/and economics. Complementing these with a cultural approach could address the belief systems that justify and perpetuate gender and economic inequalities, as well as other barriers to behaviour change

    Generally covariant dynamical reduction models and the Hadamard condition

    We recall and review earlier work on dynamical reduction models, both non-relativistic and relativistic, and discuss how they may relate to suggestions which have been made (including the matter-gravity entanglement hypothesis of one of us) for how quantum gravity could be connected to the resolution of the quantum-mechanical measurement problem. We then provide general guidelines for generalizing dynamical reduction models to curved spacetimes and propose a class of generally covariant relativistic versions of the GRW model. We anticipate that the collapse operators of our class of models may play a r\^ole in a yet-to-be-formulated theory of semiclassical gravity with collapses. We show explicitly that the collapse operators map a dense domain of states that are initially Hadamard to final Hadamard states -- a property that we expect will be needed for the construction of such a semiclassical theory. Finally, we provide a simple example in which we explicitly compute the violations in energy-momentum due to the state reduction process and conclude that this violation is of the order of a parameter of the model -- supposed to be small. We briefly discuss how this work may, upon further development of a suitable semiclassical gravity theory with collapses, enable further progress to be made on earlier work one of us and collaborators on the explanation of structure-formation in a homogeneous and isotropic quantum universe and on a possible resolution of the black hole information loss puzzle

    A 1.3 mm SMA Survey of 29 Variable Young Stellar Objects

    Young stellar objects (YSOs) may undergo periods of active accretion (outbursts), during which the protostellar accretion rate is temporarily enhanced by a few orders of magnitude. Whether or not these accretion outburst YSOs possess similar dust/gas reservoirs to each other, and whether or not their dust/gas reservoirs are similar as quiescent YSOs, are issues not yet clarified. The aim of this work is to characterize the millimeter thermal dust emission properties of a statistically significant sample of long and short duration accretion outburst YSOs (i.e., FUors and EXors) and the spectroscopically identified candidates of accretion outbursting YSOs (i.e., FUor-like objects). We have carried out extensive Submillimeter Array (SMA) observations mostly at ∼\sim225 GHz (1.33 mm) and ∼\sim272 GHz (1.10 mm), from 2008 to 2017. We covered accretion outburst YSOs located at 3-σ\sigma significance. Detected sources except for the two cases of V883 Ori and NGC 2071 MM3 were observed with ∼\sim1"" angular resolution. Overall our observed targets show a systematically higher millimeter luminosity distribution than those of the M∗>M_{*}>0.3 M⊙M_{\odot} Class II YSOs in the nearby (≲\lesssim400 pc) low-mass star-forming molecular clouds (e.g., Taurus, Lupus, Upp Scorpio, and Chameleon I). In addition, at 1 mm our observed confirmed binaries or triple-system sources are systematically fainter than the rest of the sources even though their 1 mm fluxes are broadly distributed. We may have detected ∼\sim30-60\% millimeter flux variability from V2494 Cyg and V2495 Cyg, from the observations separated by ∼\sim1 year

    Dominant Negative Mutants of Bacillus thuringiensis Cry1Ab Toxin Function as Anti-Toxins: Demonstration of the Role of Oligomerization in Toxicity

    BACKGROUND:Bacillus thuringiensis Cry toxins, that are used worldwide in insect control, kill insects by a mechanism that depends on their ability to form oligomeric pores that insert into the insect-midgut cells. These toxins are being used worldwide in transgenic plants or spray to control insect pests in agriculture. However, a major concern has been the possible effects of these insecticidal proteins on non-target organisms mainly in ecosystems adjacent to agricultural fields. METHODOLOGY/PRINCIPAL FINDINGS:We isolated and characterized 11 non-toxic mutants of Cry1Ab toxin affected in different steps of the mechanism of action namely binding to receptors, oligomerization and pore-formation. These mutant toxins were analyzed for their capacity to block wild type toxin activity, presenting a dominant negative phenotype. The dominant negative phenotype was analyzed at two levels, in vivo by toxicity bioassays against susceptible Manduca sexta larvae and in vitro by pore formation activity in black lipid bilayers. We demonstrate that some mutations located in helix alpha-4 completely block the wild type toxin activity at sub-stoichiometric level confirming a dominant negative phenotype, thereby functioning as potent antitoxins. CONCLUSIONS/SIGNIFICANCE:This is the first reported case of a Cry toxin dominant inhibitor. These data demonstrate that oligomerization is a fundamental step in Cry toxin action and represent a potential mechanism to protect special ecosystems from the possible effect of Cry toxins on non-target organisms

    Enterobactin-Mediated Delivery of β-Lactam Antibiotics Enhances Antibacterial Activity against Pathogenic Escherichia coli

    The design, synthesis, and characterization of enterobactin–antibiotic conjugates, hereafter Ent-Amp/Amx, where the β-lactam antibiotics ampicillin (Amp) and amoxicillin (Amx) are linked to a monofunctionalized enterobactin scaffold via a stable poly(ethylene glycol) linker are reported. Under conditions of iron limitation, these siderophore-modified antibiotics provide enhanced antibacterial activity against Escherichia coli strains, including uropathogenic E. coli CFT073 and UTI89, enterohemorrhagic E. coli O157:H7, and enterotoxigenic E. coli O78:H11, compared to the parent β-lactams. Studies with E. coli K-12 derivatives defective in ferric enterobactin transport reveal that the enhanced antibacterial activity observed for this strain requires the outer membrane ferric enterobactin transporter FepA. A remarkable 1000-fold decrease in minimum inhibitory concentration (MIC) value is observed for uropathogenic E. coli CFT073 relative to Amp/Amx, and time-kill kinetic studies demonstrate that Ent-Amp/Amx kill this strain more rapidly at 10-fold lower concentrations than the parent antibiotics. Moreover, Ent-Amp and Ent-Amx selectively kill E. coli CFT073 co-cultured with other bacterial species such as Staphylococcus aureus, and Ent-Amp exhibits low cytotoxicity against human T84 intestinal cells in both the apo and iron-bound forms. These studies demonstrate that the native enterobactin platform provides a means to effectively deliver antibacterial cargo across the outer membrane permeability barrier of Gram-negative pathogens utilizing enterobactin for iron acquisition.Pacific Southwest Regional Center of Excellence for Biodefense and Emerging Infectious DiseaseKinship Foundation.
