    Tainted ores and the rise of tin bronzes in Eurasia, c. 6500 years ago

    The earliest tin bronze artefacts in Eurasia are generally believed to have appeared in the Near East in the early third millennium BC. Here we present tin bronze artefacts that occur far from the Near East, and in a significantly earlier period. Excavations at Pločnik, a Vinča culture site in Serbia, recovered a piece of tin bronze foil from an occupation layer dated to the mid fifth millennium BC. The discovery prompted a reassessment of 14 insufficiently contextualised early tin bronze artefacts from the Balkans. They too were found to derive from the smelting of copper-tin ores. These tin bronzes extend the record of bronze making by c. 1500 years, and challenge the conventional narrative of Eurasian metallurgical development

    Diagnosing mucopolysaccharidosis IVA

    Mucopolysaccharidosis IVA (MPS IVA; Morquio A syndrome) is an autosomal recessive lysosomal storage disorder resulting from a deficiency of N-acetylgalactosamine-6-sulfate sulfatase (GALNS) activity. Diagnosis can be challenging and requires agreement of clinical, radiographic, and laboratory findings. A group of biochemical genetics laboratory directors and clinicians involved in the diagnosis of MPS IVA, convened by BioMarin Pharmaceutical Inc., met to develop recommendations for diagnosis. The following conclusions were reached. Due to the wide variation and subtleties of radiographic findings, imaging of multiple body regions is recommended. Urinary glycosaminoglycan analysis is particularly problematic for MPS IVA and it is strongly recommended to proceed to enzyme activity testing even if urine appears normal when there is clinical suspicion of MPS IVA. Enzyme activity testing of GALNS is essential in diagnosing MPS IVA. Additional analyses to confirm sample integrity and rule out MPS IVB, multiple sulfatase deficiency, and mucolipidoses types II/III are critical as part of enzyme activity testing. Leukocytes or cultured dermal fibroblasts are strongly recommended for enzyme activity testing to confirm screening results. Molecular testing may also be used to confirm the diagnosis in many patients. However, two known or probable causative mutations may not be identified in all cases of MPS IVA. A diagnostic testing algorithm is presented which attempts to streamline this complex testing process

    Evidence of causal effect of major depression on alcohol dependence: findings from the psychiatric genomics consortium

    BACKGROUND Despite established clinical associations among major depression (MD), alcohol dependence (AD), and alcohol consumption (AC), the nature of the causal relationship between them is not completely understood. We leveraged genome-wide data from the Psychiatric Genomics Consortium (PGC) and UK Biobank to test for the presence of shared genetic mechanisms and causal relationships among MD, AD, and AC. METHODS Linkage disequilibrium score regression and Mendelian randomization (MR) were performed using genome-wide data from the PGC (MD: 135 458 cases and 344 901 controls; AD: 10 206 cases and 28 480 controls) and UK Biobank (AC-frequency: 438 308 individuals; AC-quantity: 307 098 individuals). RESULTS Positive genetic correlation was observed between MD and AD (rgMD−AD = + 0.47, P = 6.6 × 10−10). AC-quantity showed positive genetic correlation with both AD (rgAD−AC quantity = + 0.75, P = 1.8 × 10−14) and MD (rgMD−AC quantity = + 0.14, P = 2.9 × 10−7), while there was negative correlation of AC-frequency with MD (rgMD−AC frequency = −0.17, P = 1.5 × 10−10) and a non-significant result with AD. MR analyses confirmed the presence of pleiotropy among these four traits. However, the MD-AD results reflect a mediated-pleiotropy mechanism (i.e. causal relationship) with an effect of MD on AD (beta = 0.28, P = 1.29 × 10−6). There was no evidence for reverse causation. CONCLUSION This study supports a causal role for genetic liability of MD on AD based on genetic datasets including thousands of individuals. Understanding mechanisms underlying MD-AD comorbidity addresses important public health concerns and has the potential to facilitate prevention and intervention efforts

    Genome-wide association and epidemiological analyses reveal common genetic origins between uterine leiomyomata and endometriosis

    Uterine leiomyomata (UL) are the most common neoplasms of the female reproductive tract and primary cause for hysterectomy, leading to considerable morbidity and high economic burden. Here we conduct a GWAS meta-analysis in 35,474 cases and 267,505 female controls of European ancestry, identifying eight novel genome-wide significant (P < 5 × 10−8) loci, in addition to confirming 21 previously reported loci, including multiple independent signals at 10 loci. Phenotypic stratification of UL by heavy menstrual bleeding in 3409 cases and 199,171 female controls reveals genome-wide significant associations at three of the 29 UL loci: 5p15.33 (TERT), 5q35.2 (FGFR4) and 11q22.3 (ATM). Four loci identified in the meta-analysis are also associated with endometriosis risk; an epidemiological meta-analysis across 402,868 women suggests at least a doubling of risk for UL diagnosis among those with a history of endometriosis. These findings increase our understanding of genetic contribution and biology underlying UL development, and suggest overlapping genetic origins with endometriosis

    Ancient raw copper from primary smelting sites in Cyprus

    Finds of metallic copper from various primary smelting sites in the Sia valley in Cyprus have been analysed by ICP-OES for their composition and by optical and electron microscopy for metallography. Results show a characteristic pattern of impurities for each of the sites examined which allow an assignment to specific types of ore body and geological matrix. Different zones of the Cyprus Ophiolite Complex were exploited in different periods in antiquity, but these results show different types could be exploited contemporaneously within a specific period, especially during the first millennium BC. One location in this area, Agia Varvara-Almyras, an Iron Age copper smelting site with the only complete chain of operation recorded in ancient Cypriote metallurgy, is used to show how analytical work can guide future field surveys to find ancient furnaces, slag heaps and mines. The ultimate goal of the project is to extend it to reconstruct the complete history of copper production in a well-defined mining district over the last 4000 years

    Scanning Auger microscopy studies of an ancient bronze

    Scanning Auger microscopy (SAM) has been used to study the surface and interface microchemistry of a sheet bronze belt from the Urartian kingdom in NE Syria of the early first millennium B.C. We find that the patina contains no copper species at all (decuprification), whereas carbonaceous species, Ca-silicates and N-bearing species are detected, the last being tentatively identified as organic (primarily amine-like) residues deriving from the soil. A textured grain, which we qualify as a second phase of bronze originated by an imperfect alloying of the two major metals (i.e., consisting of Cu-rich and Sn-rich domains) is observed on the metallic side lying beneath the patina. SAM imaging with a submicron spatial resolution highlights the presence of SnO2 oxide inside what appears to be the hollow veins of the grain, whereas a Cu2O-like oxide is confined exclusively to the flat regions of the grain. We explain these results by noting that the hollow veins, offering a higher exposure to external fluids. are likely to have promoted preferential formation of the more stable tin oxide over copper oxide. In another region of the metal side we studied the chemistry of grain boundaries and their surrounding areas. We find that S species lie exclusively inside the grain boundaries., whereas Sn and Zn species accumulate just outside the boundary channels, and this lateral chemical inhomogeneity is highlighted with a similar to 200 nm spatial resolution. Lateral segregation of Cu and Sn domains is imaged in another region with a spatial resolution of similar to 15 nm. This result marks the best spatial resolution any analytical method has yet achieved in highlighting chemical heterogeneities of ancient bronzes. Although archaeomaterials lie outside the mainstream applications of Auger techniques, this study provides convincing evidence that SAM can greatly advance our understanding of these materials, as it provides clues relating to corrosion and palmation phenomena. as well as manufacturing processes that are not easily reachable by other microscopies or microanalytical techniques.     PIXE and RES as a tool for the analysis of historic copper halfpennies

    RES and PIXE microbeam analysis is used to investigate the trace element concentrations as well as the surface corrosion of counterfeited copper halfpennies from the mid 18th century. PIXE elemental mapping was used to identify regions with a minimum of light element contamination which could be used for accurate trace element determinations. RES analysis of selected points on the map could be used to confirm the absence of corrosion or contamination, and in addition yielded valuable information on the composition and structure of the corrosion layers.
