    The Soviet Union had a number of satellite states, where communist puppet regimes were propped up in order to serve the interests of the Soviet Union. The Eastern Bloc was established with the goal of spreading the Soviet style of government, regardless of its unpopularity. The only reason that the communist regimes in these states were able to survive was because of Soviet support. This meant that the decline of the Soviet Union and the individual bloc states fed into each other. This is examined through the case of the German Democratic Republic and its relations with the Soviet Union

    The resilience of EU Member States to the financial and economic crisis What are the characteristics of resilient behaviour?

    This study presents an empirical analysis of the resilience of European countries to the financial and economic crisis that started in 2007. The analysis addresses the following questions: Which countries showed a resilient behaviour during and after the crisis? Is resilience related only to the economic dimension? Has any of the EU countries been able to use the crisis as an opportunity and 'bounce forward'? Is it possible to identify any particular country characteristics linked to resilience? The analysis is based on the JRC conceptual framework for resilience (Manca et al., 2017) which places at its core the wellbeing of individuals, thus going beyond the merely economic growth perspective. The study carefully selects a number of key economic and social variables that aim to capture the resilience capacities of our society. Resilience is measured by investigating the dynamic response of these variables to the crisis in the short and medium run. In particular, we define four resilience indicators: the impact of the crisis, the recovery, the medium-run, and the ‘bouncing forward’. Results from a narrow exercise focusing on macroeconomic and financial variables confirm the validity of the proposed measurement approach: Germany appears to be among the most resilient countries; Ireland, after having been severely hit, shows a good absorptive capacity; Italy seems to be still struggling with the recovery, while Greece remains the most affected. After measuring resilience, we identify underlying country characteristics that may be associated with resilient behaviour. As such, these could indicate entry points for policies to increase countries' resilience to economic and financial shocks.JRC.B.1-Finance and Econom

    Seamounts as hot-spots of large pelagic aggregations

    The distribution of four top predators in the Tyrrhenian Sea, a sub-basin of the Mediterranean Sea, was investigated by means of random forest regression considering depth, distance from the coast, seafloor slope, and distance from seamounts as habitat descriptors on a 2x2–nautical mile regular grid. RF results are processed to estimate variable importance and model performance. Random forest architecture reached optimal sensitivity and specificity, thus providing a consistent support tool for identifying suitable habitats. The considered species are characterized as having patched suitable habitats with a number of hot-spot areas where the different species' habitats overlap. These hot-spot areas' locations correspond to those of specific seamounts identifying the attraction effect of these topographic structures. The mean features typifying the most attractive seamounts are investigated and found to be shallow peak and base depths but wide base area and high relative elevation

    The Seabed Makes the Dolphins: Physiographic Features Shape the Size and Structure of the Bottlenose Dolphin Geographical Units

    The common bottlenose dolphin (Tursiops truncatus) is a cosmopolitan delphinid, regularly present in the Mediterranean Sea. According to previous studies, this dolphin tends to form resident geographical units scattered on the continental shelf. We investigated how the physiographic characteristics of the area of residence, with special reference to the size and shape of the continental shelf, affect the home range and the group size of the local units. We analysed and compared data collected between 2004-2016 by 15 research groups operating in different study areas of the Mediterranean Sea: the Alboran Sea, in the South-Western Mediterranean, the Gulf of Lion and the Pelagos Sanctuary for the marine mammals, in the North-Western Mediterranean, and the Gulf of Ambracia, in the North-Central Mediterranean Sea. We have found that in areas characterised by a wide continental platform, dolphins have wider home ranges and aggregate into larger groups. In areas characterized by a narrow continental platform, dolphins show much smaller home ranges and aggregate into smaller groups. The results obtained from this collective research effort highlight the importance of data sharing to improve our scientific knowledge in the field of cetaceans and beyond

    Akt1 Is Essential for Postnatal Mammary Gland Development, Function, and the Expression of Btn1a1

    Akt1, a serine-threonine protein kinase member of the PKB/Akt gene family, plays critical roles in the regulation of multiple cellular processes, and has previously been implicated in lactation and breast cancer development. In this study, we utilized Akt1+/+ and Akt1−/− C57/Bl6 female mice to assess the role that Akt1 plays in normal mammary gland postnatal development and function. We examined postnatal morphology at multiple time points, and analyzed gene and protein expression changes that persist into adulthood. Akt1 deficiency resulted in several mammary gland developmental defects, including ductal outgrowth and defective terminal end bud formation. Adult Akt1−/− mammary gland composition remained altered, exhibiting fewer alveolar buds coupled with increased epithelial cell apoptosis. Microarray analysis revealed that Akt1 deficiency altered expression of genes involved in numerous biological processes in the mammary gland, including organismal development, cell death, and tissue morphology. Of particular importance, a significant decrease in expression of Btn1a1, a gene involved in milk lipid secretion, was observed in Akt1−/− mammary glands. Additionally, pseudopregnant Akt1−/− females failed to induce Btn1a1 expression in response to hormonal stimulation compared to their wild-type counterparts. Retroviral-mediated shRNA knockdown of Akt1 and Btn1a1 in MCF-7 human breast epithelial further illustrated the importance of Akt1 in mammary epithelial cell proliferation, as well as in the regulation of Btn1a1 and subsequent expression of ß-casein, a gene that encodes for milk protein. Overall these findings provide mechanistic insight into the role of Akt1 in mammary morphogenesis and function

    The Prevalence and Cost of Unapproved Uses of Top-Selling Orphan Drugs

    Introduction: The Orphan Drug Act encourages drug development for rare conditions. However, some orphan drugs become top sellers for unclear reasons. We sought to evaluate the extent and cost of approved and unapproved uses of orphan drugs with the highest unit sales. Methods We assessed prescription patterns for four top-selling orphan drugs: lidocaine patch (Lidoderm) approved for post-herpetic neuralgia, modafinil (Provigil) approved for narcolepsy, cinacalcet (Sensipar) approved for hypercalcemia of parathyroid carcinoma, and imatinib (Gleevec) approved for chronic myelogenous leukemia and gastrointestinal stromal tumor. We pooled patient-specific diagnosis and prescription data from two large US state pharmaceutical benefit programs for the elderly. We analyzed the number of new and total patients using each drug and patterns of reimbursement for approved and unapproved uses. For lidocaine patch, we subcategorized approved prescriptions into two subtypes of unapproved uses: neuropathic pain, for which some evidence of efficacy exists, and non-neuropathic pain. Results: We found that prescriptions for lidocaine patch, modafinil, and cinacalcet associated with non-orphan diagnoses rose at substantially higher rates (average monthly increases in number of patients of 14.6, 1.45, and 1.58) than prescriptions associated with their orphan diagnoses (3.12, 0.24, and 0.03, respectively (p75%). Increases in lidocaine patch use for non-neuropathic pain far exceeded neuropathic pain (10.2 vs. 3.6 patients, p<0.001). Discussion In our sample, three of four top-selling orphan drugs were used more commonly for non-orphan indications. These orphan drugs treated common clinical symptoms (pain and fatigue) or laboratory abnormalities. We should continue to monitor orphan drug use after approval to identify products that come to be widely used for non-FDA approved indications, particularly those without adequate evidence of efficacy

    Phosphatidylinositol 3-Kinase Couples Localised Calcium Influx to Activation of Akt in Central Nerve Terminals

    The efficient retrieval of synaptic vesicle membrane and cargo in central nerve terminals is dependent on the efficient recruitment of a series of endocytosis modes by different patterns of neuronal activity. During intense neuronal activity the dominant endocytosis mode is activity-dependent endocytosis (ADBE). Triggering of ADBE is linked to calcineurin-mediated dynamin I dephosphorylation since the same stimulation intensities trigger both. Dynamin I dephosphorylation is maximised by a simultaneous inhibition of its kinase glycogen synthase kinase 3 (GSK3) by the protein kinase Akt, however it is unknown how increased neuronal activity is transduced into Akt activation. To address this question we determined how the activity-dependent increases in intracellular free calcium ([Ca(2+)](i)) control activation of Akt. This was achieved using either trains of high frequency action potentials to evoke localised [Ca(2+)](i) increases at active zones, or a calcium ionophore to raise [Ca(2+)](i) uniformly across the nerve terminal. Through the use of either non-specific calcium channel antagonists or intracellular calcium chelators we found that Akt phosphorylation (and subsequent GSK3 phosphorylation) was dependent on localised [Ca(2+)](i) increases at the active zone. In an attempt to determine mechanism, we antagonised either phosphatidylinositol 3-kinase (PI3K) or calmodulin. Activity-dependent phosphorylation of both Akt and GSK3 was arrested on inhibition of PI3K, but not calmodulin. Thus localised calcium influx in central nerve terminals activates PI3K via an unknown calcium sensor to trigger the activity-dependent phosphorylation of Akt and GSK3

    Heterogeneity from natural marks in bottlenose dolphin (Tursiops truncatus)

    Monitoring trends of animal population is known as an essential part of conservation issues. Although individuals of many cetacean species have distinctive natural variation in appearance which allows identification of individuals through field photography of free-ranging individuals, the use of mark-recapture methods involves several assumptions. The most important assumption is that every animal should have equal probability of being captured within any sampling occasion. In photo-id studies of bottlenose dolphins (Tursiops truncatus) photographs of dorsal fin have been used for matching. Marks on animal were not equally distributed in the population, due to the fact that some individuals are unmarked or with not so visible mark, while others have very large marks that make them easily recognized. The goal of this study was to identify potential heterogeneity in capture probabilities between females and the rest of population. 6550 photographs were taken over four year sampling in both Ligurian and Northern Tyrrhenian Sea. Only high quality pictures were selected from the catalog and used for examination. We analyzed 492 photographs (247 left sides, 245 right sides) making a distinction between adults female, identified as the individuals seen in strong association with a calf, and the other components of the population. We identified different type of natural marks: notch, scar, protuding piece, scrape, back indentation, hole, deformation, white coloured area. Finally the prevalence, abundance and intensity of each mark types have been assessed in order to underline heterogeneity between the two groups analysed

    Bottlenose Dolphins and Seabirds Distribution Analysis for the Identification of a Marine Biodiversity Hotspot in Agrigento Waters

    The aim of this study is to evaluate the presence of biodiversity hotspots in Agrigento waters (Mediterranean Sea) to define the conservation area for bottlenose dolphins (Tursiops truncatus) and seabirds (Calonectris diomedea, Puffinus yelkouan, and Hydrobates pelagicus), according to European directives. With this purpose, the maximum entropy algorithm (MaxEnt) was applied to the sighting points of the focal species. They co-occur in the study area and have been documented to forage behind trawlers. In this study, a fishing rate was designed and used as an explanatory variable of the species distribution, together with physiographic variables. Data were collected during 68 surveys in the waters off Agrigento province. MaxEnt models showed a strong predictive power, with distance from the coast being the greatest predicting variable, followed by slope, depth, and fishing rate. For all the species considered, the probability of presence increased as the fishing rate grew. Cartographic analysis revealed one area shared by the species, which occupies 529 km2, from the shoreline to 100 m depth. This study increases knowledge on the distribution and habitat preferences of the target species in the Sicilian waters. Evaluating the influence of fisheries is a promising method that needs further testing to apply effective management measures
