    A Comparison of Phone-Based and On-Site Assessment of Fidelity for Assertive Community Treatment in Indiana

    Objective: This study investigated the reliability and validity of a phone-administered fidelity assessment instrument based on the Dartmouth Assertive Community Treatment Scale (DACTS). Methods: An experienced rater paired with a research assistant without fidelity assessment experience or a consultant familiar with the treatment site conducted phone-based assessments of 23 teams providing assertive community treatment in Indiana. Using the DACTS, consultants conducted on-site evaluations of the programs. Results: The pairs of phone raters revealed high levels of consistency [intraclass correlation coefficient (ICC)=.92] and consensus (mean absolute difference of .07). Phone and on-site assessment showed strong agreement (ICC=.87) and consensus (mean absolute difference of .07) and agreed within .1 scale point, or 2% of the scoring range, for 83% of sites and within .15 scale point for 91% of sites. Results were unaffected by the expertise level of the rater. Conclusions: Phone-based assessment could help agencies monitor faithful implementation of evidence-based practices.     Hyperpolarizability and operational magic wavelength in an optical lattice clock

    Optical clocks benefit from tight atomic confinement enabling extended interrogation times as well as Doppler- and recoil-free operation. However, these benefits come at the cost of frequency shifts that, if not properly controlled, may degrade clock accuracy. Numerous theoretical studies have predicted optical lattice clock frequency shifts that scale nonlinearly with trap depth. To experimentally observe and constrain these shifts in an 171^{171}Yb optical lattice clock, we construct a lattice enhancement cavity that exaggerates the light shifts. We observe an atomic temperature that is proportional to the optical trap depth, fundamentally altering the scaling of trap-induced light shifts and simplifying their parametrization. We identify an "operational" magic wavelength where frequency shifts are insensitive to changes in trap depth.     Measuring the Recovery Orientation of Assertive Community Treatment

    BACKGROUND: Approaches to measuring recovery orientation are needed, particularly for programs that may struggle with implementing recovery-oriented treatment. OBJECTIVE: A mixed-methods comparative study was conducted to explore effective approaches to measuring recovery orientation of assertive community treatment (ACT) teams. DESIGN: Two ACT teams exhibiting high and low recovery orientation were compared using surveys, treatment plan ratings, diaries of treatment visits, and team leaderā€“reported treatment control mechanisms. RESULTS: The recovery-oriented team differed on one survey measure (higher expectations for consumer recovery), treatment planning (greater consumer involvement and goal-directed content), and use of control mechanisms (less use of representative payee, agency-held lease, daily medication delivery, and family involvement). Staff and consumer diaries showed the most consistent differences (e.g., conveying hope and choice) and were the least susceptible to observer bias but had the lowest response rates. CONCLUSIONS: Several practices differentiate recovery orientation on ACT teams, and a mixed-methods assessment approach is feasible

    Species specific differences in use of ANP32 proteins by influenza A virus

    Influenza A viruses (IAV) are subject to species barriers that prevent frequent zoonotic transmission and pandemics. One of these barriers is the poor activity of avian IAV polymerases in human cells. Differences between avian and mammalian ANP32 proteins underlie this host range barrier. Human ANP32A and ANP32B homologues both support function of human-adapted influenza polymerase but do not support efficient activity of avian IAV polymerase which requires avian ANP32A. We show here that the gene currently designated as avian ANP32B is evolutionarily distinct from mammalian ANP32B, and that chicken ANP32B does not support IAV polymerase activity even of human-adapted viruses. Consequently, IAV relies solely on chicken ANP32A to support its replication in chicken cells. Amino acids 129I and 130N, accounted for the inactivity of chicken ANP32B. Transfer of these residues to chicken ANP32A abolished support of IAV polymerase. Understanding ANP32 function will help develop antiviral strategies and aid the design of influenza virus resilient genome edited chickens

    SSI-AWARE: Self-Sovereign Identity Authenticated backup With Auditing by Remote Entities

    Part 5: CybersecurityInternational audienceThe self-sovereign identity (SSI) model entails the full responsibility and sovereignty of a user regarding his identity data. This identity data can contain private data which is solely known to the user. The user himself is therefore required to manage the whole lifecycle of his private data, including the backup and restore. We show that prior work on how to backup and restore the userā€™s identity data does not meet the requirements of the SSI setting, and we present the first solution which does meet the requirements. Authenticated backup with auditing by remote entities (AWARE) combines SSI sustaining aspects and extends them to create a truly self-sovereign backup-and-restore protocol. In AWARE, trusted, physically met humans, called custodians, hold a secure device. Custodians with a secure device offer an offline backup possibility and a secure channel. The backup and restore are audited by commits on a publicly accessible distributed ledger. These commits are answered by auditing services which are required during restore. Only some auditing services hold relevant data for a restore. The self sovereignty of the user lies in the exclusive information which auditing services hold relevant data. AWAREĀ  is the first backup-and-restore mechanism that fully complies with the SSI model. We perform an in-depth security-risk analysis of AWARE, showing a risk rating which is comparable to the best risk rating o related non-SSI-compliant backup-and-restore mechanisms. We instantiate the AWAREĀ protocol with cryptographic primitives providing a high security level of 256-bit. We show its implementation feasibility by providing a simulation of AWARE, and conclude with an estimated performance analysis on a microcontoller architecture based on our simulation and implementation results in the literature

    An E box comprises a positional sensor for regional differences in skeletal muscle gene expression and methylation

    AbstractTo dissect the molecular mechanisms conferring positional information in skeletal muscles, we characterized the control elements responsible for the positionally restricted expression patterns of a muscle-specific transgene reporter, driven by regulatory sequences from the MLC1/3 locus. These sequences have previously been shown to generate graded transgene expression in the segmented axial muscles and their myotomal precursors, fortuitously marking their positional address. An evolutionarily conserved E box in the MLC enhancer core, not recognized by MyoD, is a target for a nuclear protein complex, present in a variety of tissues, which includes Hox proteins and Zbu1, a DNA-binding member of the SW12/SNF2 gene family. Mutation of this E box in the MLC enhancer has only a modest positive effect on linked CAT gene expression in transfected muscle cells, but when introduced into transgenic mice the same mutation elevates CAT transgene expression in skeletal muscles, specifically releasing the rostral restriction on MLC-CAT transgene expression in the segmented axial musculature. Increased transgene activity resulting from the E box mutation in the MLC enhancer correlates with reduced DNA methylation of the distal transgenic MLC1 promoter as well as in the enhancer itself. These results identify an E box and the proteins that bind to it as a positional sensor responsible for regional differences in axial skeletal muscle gene expression and accessibility
