    Building and exploiting a Digital Twin for the management of drinking water distribution networks

    [EN] Digital Twins (DTs) are starting to be exploited to improve the management of water distribution systems (WDSs) and, in the future, they will be crucial for decision making. In this paper, the authors propose several requirements that a DT of a water distribution system should accomplish. Developing a DT is a challenge, and a continuous process of adjustments and learning is required. Due to the advantages of having a DT of the WDS always available, during the last years a strategy to build and maintain a DT of the water distribution network of Valencia (Spain) and its Metropolitan Area (1.6 million inhabitants) was developed. This is one of the first DTs built of a water utility, being currently in operation.     Growth of InP on GaAs (001) by hydrogen-assisted low-temperature solid-source molecular beam epitaxy

    Direct heteroepitaxial growth of InP layers on GaAs (001) wafers has been performed by solid-source molecular beam epitaxy assisted by monoatomic hydrogen (H∗). The epitaxial growth has been carried out using a two-step method: for the initial stage of growth the temperature was as low as 200 °C and different doses of H∗ were used; after this, the growth proceeded without H∗ while the temperature was increased slowly with time. The incorporation of H∗ drastically increased the critical layer thickness observed by reflection high-energy electron diffraction; it also caused a slight increase in the luminescence at room temperature, while it also drastically changed the low-temperature luminescence related to the presence of stoichiometric defects. The samples were processed by rapid thermal annealing. The annealing improved the crystalline quality of the InP layers measured by high-resolution x-ray diffraction, but did not affect their luminescent behavior significantly

    Se realizó un estudio epidemiológico descriptivo, observacional y retrospectivo, analizando la incidencia y las características de las lesiones sufridas por jugadores de pádel españoles profesionales y no profesionales. Se registraron las lesiones sufridas por 478 jugadores entre el 1 de enero y el 31 de diciembre de 2016. Se registraron datos sobre el número de lesiones, lesiones más frecuentes, distribución, tipo, ubicación, el momento en que ocurrieron y la causa de las lesiones que afectan el sistema locomotor. La tasa de lesiones en esta población fue de 2,75 lesiones por 1000 horas de exposición al riesgo. Las lesiones leves y aquellas con mecanismos de lesión intrínseca fueron las más frecuentes, y tuvieron lugar el final de la práctica deportiva. La extremidad inferior fue el área corporal más lesionada, y el tejido más lesionado fue el sistema musculo-tendinoso. A su vez, la epicondilitis fue la patología más comúnAn epidemiological study was conducted using retrospective, observational and descriptive methodologies. We sought to analyse the incidence and features of injuries sustained by professional and non-professional Spanish Padel players. In total, 478 injuries were registered in 2016 from January 1st to December 31st. We collected data on the number of injuries, the most frequent injuries, their distribution, type, location, the moment at which these occurred and the cause of the injuries affecting the locomotor system. We found that the injury rate in this population was 2.75 injuries per 1000 hours of risk exposure. The most frequent injuries were those which were mild or with mechanisms of intrinsic injury, and most occurred towards the end any given game or practice.     The photosynthetic cytochrome c550 from the marine diatom Phaeodactylum tricornutum has been purified and characterized. Cytochrome c550 is mostly obtained from the soluble cell extract in relatively large amounts. In addition, the protein appeared to be truncated in the last hydrophobic residues of the C-terminus, both in the soluble cytochrome c550 and in the protein extracted from the membrane fraction, as deduced by mass spectrometry analysis and the comparison with the gene sequence. Interestingly, it has been described that the C-terminus of cytochrome c550 forms a hydrophobic finger involved in the interaction with photosystem II in cyanobacteria. Cytochrome c550 was almost absent in solubilized photosystem II complex samples, in contrast with the PsbO and Psb31 extrinsic subunits, thus suggesting a lower affinity of cytochrome c550 for the photosystem II complex. Under iron-limiting conditions the amount of cytochrome c550 decreases up to about 45% as compared to iron-replete cells, pointing to an iron-regulated synthesis. Oxidized cytochrome c550 has been characterized using continuous wave EPR and pulse techniques, including HYSCORE, and the obtained results have been interpreted in terms of the electrostatic charge distribution in the surroundings of the heme centre

    Synergistic activation of AMPK prevents from polyglutamine-inducedtoxicity inCaenorhabditis elegans

Supplementary material related to this article can be found, in the online version, at doi: https://doi.org/10.1016/j.phrs.2020.105105.Expression of abnormally long polyglutamine (polyQ) tracks is the source of a range of dominant neurodegenerative diseases, such as Huntington disease. Currently, there is no treatment for this devastating disease, although some chemicals, e.g., metformin, have been proposed as therapeutic solutions. In this work, we show that metformin, together with salicylate, can synergistically reduce the number of aggregates produced after polyQ expression in Caenorhabditis elegans. Moreover, we demonstrate that incubation polyQ-stressed worms with low doses of both chemicals restores neuronal functionality. Both substances are pleitotropic and may activate a range of different targets. However, we demonstrate in this report that the beneficial effect induced by the combination of these drugs depends entirely on the catalytic action of AMPK, since loss of function mutants of aak-2/AMPKα2 do not respond to the treatment. To further investigate the mechanism of the synergetic activity of metformin/salicylate, we used CRISPR to generate mutant alleles of the scaffolding subunit of AMPK, aakb-1/AMPKβ1. In addition, we used an RNAi strategy to silence the expression of the second AMPKβ subunit in worms, namely aakb-2/AMPKβ2. In this work, we demonstrated that both regulatory subunits of AMPK are modulators of protein homeostasis. Interestingly, only aakb-2/AMPKβ2 is required for the synergistic action of metformin/salicylate to reduce polyQ aggregation.     Intravitreal administration of recombinant human opticin protects against hyperoxia-induced pre-retinal neovascularization

    Opticin is an extracellular glycoprotein present in the vitreous. Its antiangiogenic properties offer the potential for therapeutic intervention in conditions such as proliferative diabetic retinopathy and retinopathy of prematurity. Here, we investigated the hypothesis that intravitreal administration of recombinant human opticin can safely protect against the development of pathological angiogenesis and promote its regression. We generated and purified recombinant human opticin and investigated its impact on the development and regression of pathological retinal neovascularization following intravitreal administration in murine oxygen-induced retinopathy. We also investigated its effect on normal retinal vascular development and function, following intravitreal injection in neonatal mice, by histological examination and electroretinography. In oxygen-induced retinopathy, intravitreal administration of human recombinant opticin protected against the development of retinal neovascularization to similar extent as aflibercept, which targets VEGF. Opticin also accelerated regression of established retinal neovascularization, though the effect at 18 h was less than that of aflibercept. Intravitreal administration of human recombinant opticin in neonatal mice caused no detectable perturbation of subsequent retinal vascular development or function. In summary we found that intraocular administration of recombinant human opticin protects against the development of pathological angiogenesis in mice and promotes its regression
