    5-HT1A receptor blockade reverses GABAA receptor α3 subunit-mediated anxiolytic effects on stress-induced hyperthermia

    Stress-related disorders are associated with dysfunction of both serotonergic and GABAergic pathways, and clinically effective anxiolytics act via both neurotransmitter systems. As there is evidence that the GABA(A) and the serotonin receptor system interact, a serotonergic component in the anxiolytic actions of benzodiazepines could be present. The main aim of the present study was to investigate whether the anxiolytic effects of (non-)selective alpha subunit GABA(A) receptor agonists could be reversed with 5-HT1A receptor blockade using the stress-induced hyperthermia (SIH) paradigm. The 5-HT1A receptor antagonist WAY-100635 (0.1-1 mg/kg) reversed the SIH-reducing effects of the non-alpha-subunit selective GABA(A) receptor agonist diazepam (1-4 mg/kg) and the GABA(A) receptor alpha(3)-subunit selective agonist TP003 (1 mg/kg), whereas WAY-100635 alone was without effect on the SIH response or basal body temperature. At the same time, co-administration of WAY-100635 with diazepam or TP003 reduced basal body temperature. WAY-100635 did not affect the SIH response when combined with the preferential alpha(1)-subunit GABA(A) receptor agonist zolpidem (10 mg/kg), although zolpidem markedly reduced basal body temperature. The present study suggests an interaction between GABA(A) receptor alpha-subunits and 5-HT1A receptor activation in the SIH response. Specifically, our data indicate that benzodiazepines affect serotonergic signaling via GABA(A) receptor alpha(3)-subunits. Further understanding of the interactions between the GABA(A) and serotonin system in reaction to stress may be valuable in the search for novel anxiolytic drugs

    Infection of Semen-Producing Organs by SIV during the Acute and Chronic Stages of the Disease

    International audienceBACKGROUND: Although indirect evidence suggests the male genital tract as a possible source of persistent HIV shedding in semen during antiretroviral therapy, this phenomenon is poorly understood due to the difficulty of sampling semen-producing organs in HIV+ asymptomatic individuals. METHODOLOGY/PRINCIPAL FINDINGS: Using a range of molecular and cell biological techniques, this study investigates SIV infection within reproductive organs of macaques during the acute and chronic stages of the disease. We demonstrate for the first time the presence of SIV in the testes, epididymides, prostate and seminal vesicles as early as 14 days post-inoculation. This infection persists throughout the chronic stage and positively correlates with blood viremia. The prostate and seminal vesicles appear to be the most efficiently infected reproductive organs, followed by the epididymides and testes. Within the male genital tract, mostly T lymphocytes and a small number of germ cells harbour SIV antigens and RNA. In contrast to the other organs studied, the testis does not display an immune response to the infection. Testosteronemia is transiently increased during the early phase of the infection but spermatogenesis remains unaffected. CONCLUSIONS/SIGNIFICANCE: The present study reveals that SIV infection of the macaque male genital tract is an early event and that semen-producing organs display differential infection levels and immune responses. These results help elucidate the origin of HIV in semen and constitute an essential base to improving the design of antiretroviral therapies to eradicate virus from semen

Des considérations théoriques montrent que les méthodes de la goutte posée ou de la goutte pendante sont aussi utilisables pour ce type de mesure à la condition de pouvoir former de très petites gouttes. Ces méthodes présentent de plus l'avantage d'être plus facilement adaptables aux fortes pressions et aux températures élevées. Un appareillage pour ce type de mesure et des exemples de résultats sont présentés

    Gentlemen first? 'Broken stick' modelling reveals sex-related homing decision date in migrating seabirds

    Technical progress in animal-borne tracking and movement data analysis has facilitated the understanding of the interplay between successive periods in the life cycle of migratory animals. We investigated how sex differences on the constraints of homing may influence migration to breeding areas in crested penguins (genus Eudyptes). We used a novel approach to infer homing decision date, a precise point in time that translates statistically as a change point in the current distance of the animal to its colony (‘broken stick’ modelling approach, R codes provided here). We applied this approach to geolocation tracking data on migration in three Eudyptes species, from three localities in the southern Indian Ocean (five populations). Sex had a subtle and consistent influence on the temporal activity of the 66 animals during their migratory journey. Males began migration to the breeding localities earlier than females, by an average of 9.1 (range: 4.5–13.5) days. This difference was statistically significant in 4 of 5 populations, and occurred among all species, sites and years surveyed. Our study shows an original application of a recent modelling approach to detect change point in movement data. Our results suggest that sex-specific constraints related to breeding in migrating animals may also modify activity schedules well before breeding commences

    The Mozambique Channel (western Indian Ocean) is a dynamic environment characterised by strong mesoscale features, which influence all biological components of the pelagic ecosystem. We investigated the distribution, abundance and feeding behaviour of seabirds in the Mozambique Channel in relation to physical and biological environmental variables, with a specific interest in mesoscale features. Seabird censuses were conducted in summer and winter during 7 cruises in the southern and northern Mozambique Channel. Tropical species accounted for 49% of the 37 species identified and 97% of the individuals, and species from the sub-Antarctic region constituted 30% of the identifications. The typically tropical sooty tern (Onychoprion fuscata) was the dominant species during all cruises, and overall accounted for 74% of the species observations and 85% of counted birds. Outputs of Generalised Linear Models at the scale of the Mozambique Channel suggested that higher densities of flying and feeding birds occurred in areas with lower sea surface temperatures and lower surface chlorophyll a concentrations. Most of the flocks of feeding birds did not associate with surface schools of fish or marine mammals, but when they did, these flocks were larger, especially when associated with tuna. While tropical species seemed to favour cyclonic eddies, frontal and divergence zones, non-tropical species were more frequently recorded over shelf waters. Sooty terns foraged preferentially in cyclonic eddies where zooplankton, micronelcton and tuna schools were abundant. Among other major tropical species, frigatebirds (Fregata spp.) predominated in frontal zones between eddies, where tuna schools also frequently occurred and where geostrophic currents were the strongest. Red-footed boobies (Sula sub) concentrated in divergence zones characterised by low sea level anomalies, low geostrophic currents, and high zooplanlcton biomass close to the surface. Our results highlight the importance of mescoscale features in structuring the tropical seabird community in the Mozambique Channel, in addition to segregating tropical and non-tropical species. The mechanisms underlying the segregation of tropical seabirds seem to partially differ from that of other tropical regions, and this may be a consequence of the strong local mesoscale activity, affecting prey size and availability schemes. Beyond characterising the foraging habitats of the seabird community of the Mozambique Channel, this study highlights the importance of this region as a hot spot for seabirds; especially the southern part, where several endangered sub-Antarctic species over-winter

    A space oddity: Geographic and specific modulation of migration in Eudyptes penguins

    Post-breeding migration in land-based marine animals is thought to offset seasonal deterioration in foraging or other important environmental conditions at the breeding site. However the inter-breeding distribution of such animals may reflect not only their optimal habitat, but more subtle influences on an individual’s migration path, including such factors as the intrinsic influence of each locality’s paleoenvironment, thereby influencing animals’ wintering distribution. In this study we investigated the influence of the regional marine environment on the migration patterns of a poorly known, but important seabird group. We studied the inter-breeding migration patterns in three species of Eudyptes penguins (E. chrysolophus, E. filholi and E. moseleyi), the main marine prey consumers amongst the World’s seabirds. Using ultra-miniaturized logging devices (light-based geolocators) and satellite tags, we tracked 87 migrating individuals originating from 4 sites in the southern Indian Ocean (Marion, Crozet, Kerguelen and Amsterdam Islands) and modelled their wintering habitat using the MADIFA niche modelling technique. For each site, sympatric species followed a similar compass bearing during migration with consistent species-specific latitudinal shifts. Within each species, individuals breeding on different islands showed contrasting migration patterns but similar winter habitat preferences driven by sea-surface temperatures. Our results show that inter-breeding migration patterns in sibling penguin species depend primarily on the site of origin and secondly on the species. Such site-specific migration bearings, together with similar wintering habitat used by parapatrics, support the hypothesis that migration behaviour is affected by the intrinsic characteristics of each site. The paleo-oceanographic conditions (primarily, sea-surface temperatures) when the populations first colonized each of these sites may have been an important determinant of subsequent migration patterns. Based on previous chronological schemes of taxonomic radiation and geographical expansion of the genus Eudyptes, we propose a simple scenario to depict the chronological onset of contrasting migration patterns within this penguin group

    Although penguins are key marine predators from the Southern Ocean, their migratory behaviour during the inter-nesting period remains widely unknown. Here, we report for the first time, to our knowledge, the winter foraging movements and feeding habits of a penguin species by using geolocation sensors fitted on penguins with a new attachment method. We focused on the macaroni penguin Eudyptes chrysolophus at Kerguelen, the single largest consumer of marine prey among all seabirds. Overall, macaroni penguins performed very long winter trips, remaining at sea during approximately six months within the limits of the Southern Ocean. They departed from Kerguelen in an eastward direction and distributed widely, over more than 3.10(6) km(2). The penguins spent most of their time in a previously unrecognized foraging area, i.e. a narrow latitudinal band (47-49 degrees S) within the central Indian Ocean (70-110 degrees E), corresponding oceanographically to the Polar Frontal Zone. There, their blood isotopic niche indicated that macaroni penguins preyed mainly upon crustaceans, but not on Antarctic krill Euphausia superba, which does not occur at these northern latitudes. Such winter information is a crucial step for a better integrative approach for the conservation of this species whose world population is known to be declining
