A comparative molecular approach to mesodermal patterning in basal deuterostomes: the expression pattern of Brachyury in the enteropneust hemichordate Ptychodera flava
This work concerns the formation of mesoderm in the development of an enteropneust hemichordate, Ptychodera flava, and the expression of the Brachyury gene during this process. Brachyury expression occurs in two distinct phases. In the embryo, Brachyury is transcribed during gastrulation in the future oral and anal regions of the gut, but transcripts are no longer detected by 2 weeks of development. Brachyury expression is not detected during the 5 months of larval planktonic existence. During this time, the adult coeloms begin to develop, originating as coalescences of cells that appear to delaminate from the wall of the gut. Brachyury expression cannot be detected again until metamorphosis, when transcripts appear in the mesoderm of the adult proboscis, collar and the very posterior region of the trunk. It is also expressed in the posterior end of the gut. At no time is Brachyury expressed in the stomochord, the putative homologue of the chordate notochord. These observations illuminate the process of maximal indirect development in Ptychodera and, by comparison with patterns of Brachyury expression in the indirect development of echinoderms, their sister group, they reveal the evolutionary history of Brachyury utilization in deuterostomes
The Current Role of Androgen Deprivation in Patients Undergoing Dose-Escalated External Beam Radiation Therapy for Clinically Localized Prostate Cancer
Purpose. To review existing literature on the role of androgen deprivation therapy (ADT) with dose escalated radiation therapy. Methods and Materials. A PubMed search was undertaken to identify relevant articles. Results. Multiple recent studies were identified examining the role of ADT in the current era of radiation dose-escalation. Among the reviewed studies, varying radiation doses and techniques, ADT regimens, and patient selection criteria were utilized. Conflicting results were reported, with some studies demonstrating a benefit of delivering a higher radiation dose with ADT. Other studies failed to show significant benefits with the addition of ADT to dose-escalated RT. Conclusions. The benefit of adding ADT to dose-escalated RT is still uncertain. Prospective randomized trials, several of which are ongoing, are necessary to more adequately examine this issue. In the interim, physicians and patients should continue to utilize the existing data to weigh the risks and benefits of each approach to therapy
Synthetic DNA immunotherapy in biochemically relapsed prostate cancer
Background: INO-5150 (PSA and PSMA) +/- INO-9012 (IL-12), a synthetic DNA immunotherapy, was assessed for safety, immunogenicity and efficacy in biochemically recurrent prostate cancer patients (pts).
Methods: Phase I, open-label, multi-center study in the US included pts with rising PSA after surgery and/or RT, PSA doubling time (PSADT) \u3e3 months (mos), testosterone \u3e150 ng/dL and no concurrent ADT. Safety, immunogenicity and efficacy (PSA kinetics, PFS) were evaluated in 4 treatment arms of 15 pts each. Arms A: 2mg INO-5150, B: 8.5 mg INO-5150, C: 2mg INO-5150 + 1mg INO-9012 and D: 8.5mg INO-5150 + 1mg INO-9012. Pts received 4 IM doses of vaccine followed by electroporation on day 0, wks 3, 12 and 24 and were followed for 72 wks.
Results: 50/61 (82%) pts completed all visits and treatments were well tolerated with no safety concerns. Median PFS for overall population [N = 61, baseline (D0) PSADT range (mos) 1.5-217.1, median 9.8] and for a subset of pts with D0 PSADT ≤12mos (N = 36) has not yet been reached (FU 3-19 mos). 86% of pts with D0 PSADT ≤12 mos were progression free through 19mos FU. 27 out of 36 (75%) pts with D0 PSADT≤ 12 mos had disease stabilization at wks 27 evidenced by significant improvement in log2PSA change over time (slope) and PSADT from D0 (Slope=0.19 declined to 0.1, PSADT=5.3 improved to 10.1 mos, p = \u3c0.0001). This effect was maintained at wk 72 (Slope=0.09, PSADT=10.6, p = \u3c0.0001). Immunogenicity was observed in 77% (47/61) of pts by multiple immunologic assessments. Patient immunogenicity to INO-5150 as determined by CD38 and Perforin + CD8 T cell immune reactivity correlated with attenuated % PSA rise compared to pts without reactivity (p = 0.05, n = 50).
Conclusions: INO-5150 +/- INO-9012 was safe, well tolerated and immunogenic. Clinical efficacy was observed in the patients with D0 PSADT≤ 12 mos as evidenced by a significant dampening of log2PSA change over time and increased PSADT up to 72 weeks FU. Additional genomic analyses are ongoing to further elucidate the correlation of immunologic efficacy and clinical benefit. (NCT02514213)
Soliton excitations in halogen-bridged mixed-valence binuclear metal complexes
Motivated by recent stimulative observations in halogen (X)-bridged binuclear
transition-metal (M) complexes, which are referred to as MMX chains, we study
solitons in a one-dimensional three-quarter-filled charge-density-wave system
with both intrasite and intersite electron-lattice couplings. Two distinct
ground states of MMX chains are reproduced and the soliton excitations on them
are compared. In the weak-coupling region, all the solitons are degenerate to
each other and are uniquely scaled by the band gap, whereas in the
strong-coupling region, they behave differently deviating from the scenario in
the continuum limit. The soliton masses are calculated and compared with those

Ultrafast Pulse Radiolysis Using a Terawatt Laser Wakefield Accelerator
We report the first ultrafast pulse radiolysis transient absorption
spectroscopy measurements from the Terawatt Ultrafast High Field Facility
(TUHFF) at Argonne National Laboratory. TUHFF houses a 20 TW Ti:sapphire laser
system that generates 2.5 nC sub-picosecond pulses of multi-MeV electrons at 10
Hz using laser wakefield acceleration. The system has been specifically
optimized for kinetic measurements in a pump-probe fashion. This requires
averaging over many shots which necessitates stable, reliable generation of
electron pulses. The latter were used to generate excess electrons in pulse
radiolysis of liquid water and concentrated solutions of perchloric acid. The
hydronium ions in the acidic solutions react with the hydrated electrons
resulting in the rapid decay of the transient absorbance at 800 nm on the
picosecond time scale. Time resolution of a few picoseconds has been
demonstrated. The current time resolution is determined primarily by the
physical dimensions of the sample and the detection sensitivity. Subpicosecond
time resolution can be achieved by using thinner samples, more sensitive

A generalization of Mehta-Wang determinant and Askey-Wilson polynomials
Motivated by the Gaussian symplectic ensemble, Mehta and Wang evaluated the determinant in 2000. When , Ciucu and Krattenthaler computed the associated Pfaffian with an application to the two dimensional dimer system in 2011. Recently we have generalized the latter Pfaffian formula with a -analogue by replacing the Gamma function by the moment sequence of the little -Jacobi polynomials. On the other hand, Nishizawa has found a q-analogue of the Mehta–Wang formula. Our purpose is to generalize both the Mehta-Wang and Nishizawa formulae by using the moment sequence of the little -Jacobi polynomials. It turns out that the corresponding determinant can be evaluated explicitly in terms of the Askey-Wilson polynomials
Implementing Parallel Differential Evolution on Spark
[Abstract] Metaheuristics are gaining increased attention as an efficient way of solving hard global optimization problems. Differential Evolution (DE) is one of the most popular algorithms in that class. However, its application to realistic problems results in excessive computation times. Therefore, several parallel DE schemes have been proposed, most of them focused on traditional parallel programming interfaces and infrastruc- tures. However, with the emergence of Cloud Computing, new program- ming models, like Spark, have appeared to suit with large-scale data processing on clouds. In this paper we investigate the applicability of Spark to develop parallel DE schemes to be executed in a distributed environment. Both the master-slave and the island-based DE schemes usually found in the literature have been implemented using Spark. The speedup and efficiency of all the implementations were evaluated on the Amazon Web Services (AWS) public cloud, concluding that the island- based solution is the best suited to the distributed nature of Spark. It achieves a good speedup versus the serial implementation, and shows a decent scalability when the number of nodes grows.[Resumen] Las metaheurísticas están recibiendo una atención creciente como técnica eficiente en la resolución de problemas difíciles de optimización global. Differential Evolution (DE) es una de las metaheurísticas más populares, sin embargo su aplicación en problemas reales deriva en tiempos de cómputo excesivos. Por ello se han realizado diferentes propuestas para la paralelización del DE, en su mayoría utilizando infraestructuras e interfaces de programación paralela tradicionales. Con la aparición de la computación en la nube también se han propuesto nuevos modelos de programación, como Spark, que permiten manejar el procesamiento de datos a gran escala en la nube. En este artículo investigamos la aplicabilidad de Spark en el desarrollo de implementaciones paralelas del DE para su ejecución en entornos distribuidos. Se han implementado tanto la aproximación master-slave como la basada en islas, que son las más comunes. También se han evaluado la aceleración y la eficiencia de todas las implementaciones usando el cloud público de Amazon (AWS, Amazon Web Services), concluyéndose que la implementación basada en islas es la más adecuada para el esquema de distribución usado por Spark. Esta implementación obtiene una buena aceleración en relación a la implementación serie y muestra una escalabilidad bastante buena cuando el número de nodos aumenta.[Resume] As metaheurísticas están recibindo unha atención a cada vez maior como técnica eficiente na resolución de problemas difíciles de optimización global. Implementáronse tanto a aproximación master-slave como a baseada en illas, que son as mais comúns. Tamén se avaliaron a aceleración e a eficiencia de todas as implementacións usando o cloud público de Amazon (AWS, Amazon Web Services), tirando como conclusión que a implementación baseada en illas é a mais acaida para o esquema de distribución usado por Spark.
