    Risk of elevated nitrate concentrations below forest in the region of Munich (South Bavaria) : regionalisation at the basis of remote sensing data and nested samples

    Eine zielgerichtete Bewirtschaftung der Wälder im Hinblick auf die fortschreitende N-Sättigung fordert entsprechende Geoinformationen. Auf der Basis der Ergebnisse der Nitratinventur Bayern (Gensior et al 2003b, Mellert et al. 2005a) wurde eine Karte des Risikos erhöhter Nitratkonzentrationen für das Land Bayern erstellt (Mellert 2005c). Die Bayernkarte liefert Informationen über die durchschnittliche Situation in den forstlichen Wuchsgebieten und dient der Identifizierung von Problemregionen. Als Übersichtskarte kann sie jedoch die Bedürfnisse auf regionaler Ebene, z.B. für ein Wuchsgebiet, kaum befriedigen. Hierzu ist eine räumliche Präzisierung der Geodaten in einem detaillierten Maßstab erforderlich. Die bayernweite Regionalisierung basiert auf der in den Jahren 2001/2002 durchgeführten Nitratinventur im 8 x 8 km Raster (Level-I/BZE) an 399 Punkten im Flachland. Die Anzahl von Inventurpunkten in den einzelnen Wuchsgebieten ist daher sehr begrenzt. Zur Informationsverdichtung der kleinmaßstäbigen Bayernkarte auf den größeren Maßstab der Karte für den Großraum München wurden detaillierte Informationen aus einer 1998 durchgeführten Sickerwasserstudie (Rothe & Mellert 2004) herangezogen. Im vorliegenden Beitrag wird die Möglichkeit eines Downscalings durch ein genestets Verfahren vorgestellt. Die auf einem logistischem Regressionsmodell basierende Regionalisierung auf bayerischer Ebene (Meller et al. 2005c) wird hierbei mit den regionalen Daten durch ein multiples Regressionsverfahren verknüpft. Dank einer ins Projekt integrierten Pilotstudie zur Fernerkundung von Waldtypen konnte eine geeignete Waldkarte für den Raum München durch Klassifikation von Landsat-Daten bereit gestellt werden.A nested model design at two different spatial scales is presented to predict the risk of elevated nitrate concentrations below the main rooting zone of forests in the region of Munich. The procedure combines a logistic regression on the Bavarian scale with a general linear model (Mellert et al. 2005c) are used as a link between both scales. This link of information on two spatial levels and the identification of predictors at different scales are the advantages of this approach, compared to a sole and independent model. At the Bavarian scale the ammonium deposition and the precipitation as well as the stand type and the site conditions predict the risk of elevated nitrate concentration. At the regional scale the degree of forestation and the stand age contribute considerably to explain the variance of the nitrate concentrations. Among the sources of nitrogen input into the forests especially regional ammonium emissions by agriculture seem to contribute to the occurrence of high nitrate concentrations. The central outcome of this study is a map indicating the risk of elevated nitrate concentrations. The results suggests that forestry could contribute to retard nitrogen saturation and nitrate leaching by conversation of spruce into mixed stands with a high percentage of broad-leaved species

    Sexual-dimorphism in human immune system aging.

    Differences in immune function and responses contribute to health- and life-span disparities between sexes. However, the role of sex in immune system aging is not well understood. Here, we characterize peripheral blood mononuclear cells from 172 healthy adults 22-93 years of age using ATAC-seq, RNA-seq, and flow cytometry. These data reveal a shared epigenomic signature of aging including declining naïve T cell and increasing monocyte and cytotoxic cell functions. These changes are greater in magnitude in men and accompanied by a male-specific decline in B-cell specific loci. Age-related epigenomic changes first spike around late-thirties with similar timing and magnitude between sexes, whereas the second spike is earlier and stronger in men. Unexpectedly, genomic differences between sexes increase after age 65, with men having higher innate and pro-inflammatory activity and lower adaptive activity.     Functional Dissection of the Neural Substrates for Sexual Behaviors in Drosophila melanogaster

    The male-specific Fruitless proteins (FruM) act to establish the potential for male courtship behavior in Drosophila melanogaster and are expressed in small groups of neurons throughout the nervous system. We screened ∼1000 GAL4 lines, using assays for general courtship, male–male interactions, and male fertility to determine the phenotypes resulting from the GAL4-driven inhibition of FruM expression in subsets of these neurons. A battery of secondary assays showed that the phenotypic classes of GAL4 lines could be divided into subgroups on the basis of additional neurobiological and behavioral criteria. For example, in some lines, restoration of FruM expression in cholinergic neurons restores fertility or reduces male–male courtship. Persistent chains of males courting each other in some lines results from males courting both sexes indiscriminately, whereas in other lines this phenotype results from apparent habituation deficits. Inhibition of ectopic FruM expression in females, in populations of neurons where FruM is necessary for male fertility, can rescue female infertility. To identify the neurons responsible for some of the observed behavioral alterations, we determined the overlap between the identified GAL4 lines and endogenous FruM expression in lines with fertility defects. The GAL4 lines causing fertility defects generally had widespread overlap with FruM expression in many regions of the nervous system, suggesting likely redundant FruM-expressing neuronal pathways capable of conferring male fertility.     A Microbe Associated with Sleep Revealed by a Novel Systems Genetic Analysis of the Microbiome in Collaborative Cross Mice.

    The microbiome influences health and disease through complex networks of host genetics, genomics, microbes, and environment. Identifying the mechanisms of these interactions has remained challenging. Systems genetics in laboratory mice (Mus musculus) enables data-driven discovery of biological network components and mechanisms of host-microbial interactions underlying disease phenotypes. To examine the interplay among the whole host genome, transcriptome, and microbiome, we mapped QTL and correlated the abundance of cecal messenger RNA, luminal microflora, physiology, and behavior in a highly diverse Collaborative Cross breeding population. One such relationship, regulated by a variant on chromosome 7, was the association of Odoribacter (Bacteroidales) abundance and sleep phenotypes. In a test of this association in the BKS.Cg-Dock7m +/+ Leprdb/J mouse model of obesity and diabetes, known to have abnormal sleep and colonization by Odoribacter, treatment with antibiotics altered sleep in a genotype-dependent fashion.     A complex regional intervention to implement advance care planning in one town's nursing homes: Protocol of a controlled inter-regional study

    <p>Abstract</p> <p>Background</p> <p>Advance Care Planning (ACP) is an emerging strategy to ensure that well-reflected, meaningful and clearly documented treatment preferences are available and respected when critical decisions about life-sustaining treatment need to be made for patients unable to consent. In Germany, recent legislation confirms that advance directives (AD) have to be followed if they apply to the medical situation, but implementation of ACP has not yet been described.</p> <p>Methods/Design</p> <p>In a longitudinal controlled study, we compare 1 intervention region (4 nursing homes [n/hs], altogether 421 residents) with 2 control regions (10 n/hs, altogether 985 residents). Inclusion went from 01.02.09 to 30.06.09, observation lasted until 30.06.10. Primary endpoint is the prevalence of ADs at follow-up, 17 (12) months after the first (last) possible inclusion. Secondary endpoints compare relevance and validity of ADs, process quality, the rate of life-sustaining interventions and, in deceased residents, location of death and intensity of treatment before death. The regional multifaceted intervention on the basis of the US program Respecting Choices<sup>® </sup>comprises training of n/h staff as facilitators, training of General Practitioners, education of hospital and ambulance staff, and development of eligible tools, including Physician Orders for Life-Sustaining Treatment in case of Emergency (POLST-E).</p> <p><it>Participation data: </it>Of 1406 residents reported to live in the 14 n/hs plus an estimated turnover of 176 residents until the last possible inclusion date, 645 (41%) were willing to participate. Response rates were 38% in the intervention region and 42% in the control region. Non-responder analysis shows an equal distribution of sex and age but a bias towards dependency on nursing care in the responder group. <it>Outcome analysis </it>of this study will become available in the course of 2011.</p> <p>Discussion</p> <p>Implementing an ACP program for the n/hs and related health care providers of a region requires a complex community intervention with the effect of nothing less than a cultural shift in this health care sector.     Regulation of the JNK3 signaling pathway during islet isolation: JNK3 and c-fos as new markers of islet quality for transplantation.

    Stress conditions generated throughout pancreatic islet processing initiate the activation of pro-inflammatory pathways and beta-cell destruction. Our goal is to identify relevant and preferably beta-specific markers to assess the activation of beta-cell stress and apoptotic mechanisms, and therefore the general quality of the islet preparation prior to transplantation. Protein expression and activation were analyzed by Western blotting and kinase assays. ATP measurements were performed by a luminescence-based assay. Oxygen consumption rate (OCR) was measured based on standard protocols using fiber optic sensors. Total RNA was used for gene expression analyzes. Our results indicate that pancreas digestion initiates a potent stress response in the islets by activating two stress kinases, c-Jun N-terminal Kinase (JNK) and p38. JNK1 protein levels remained unchanged between different islet preparations and following culture. In contrast, levels of JNK3 increased after islet culture, but varied markedly, with a subset of preparations bearing low JNK3 expression. The observed changes in JNK3 protein content strongly correlated with OCR measurements as determined by the Spearman's rank correlation coefficient rho [Formula: see text] in the matching islet samples, while inversely correlating with c-fos mRNA expression [Formula: see text]. In conclusion, pancreas digestion recruits JNK and p38 kinases that are known to participate to beta-cell apoptosis. Concomitantly, the islet isolation alters JNK3 and c-fos expression, both strongly correlating with OCR. Thus, a comparative analysis of JNK3 and c-fos expression before and after culture may provide for novel markers to assess islet quality prior to transplantation. JNK3 has the advantage over all other proposed markers to be islet-specific, and thus to provide for a marker independent of non-beta cell contamination

    ppk23-Dependent Chemosensory Functions Contribute to Courtship Behavior in Drosophila melanogaster

    Insects utilize diverse families of ion channels to respond to environmental cues and control mating, feeding, and the response to threats. Although degenerin/epithelial sodium channels (DEG/ENaC) represent one of the largest families of ion channels in Drosophila melanogaster, the physiological functions of these proteins are still poorly understood. We found that the DEG/ENaC channel ppk23 is expressed in a subpopulation of sexually dimorphic gustatory-like chemosensory bristles that are distinct from those expressing feeding-related gustatory receptors. Disrupting ppk23 or inhibiting activity of ppk23-expressing neurons did not alter gustatory responses. Instead, blocking ppk23-positive neurons or mutating the ppk23 gene delayed the initiation and reduced the intensity of male courtship. Furthermore, mutations in ppk23 altered the behavioral response of males to the female-specific aphrodisiac pheromone 7(Z), 11(Z)-Heptacosadiene. Together, these data indicate that ppk23 and the cells expressing it play an important role in the peripheral sensory system that determines sexual behavior in Drosophila

    Turning Males On: Activation of Male Courtship Behavior in Drosophila melanogaster

    The innate sexual behaviors of Drosophila melanogaster males are an attractive system for elucidating how complex behavior patterns are generated. The potential for male sexual behavior in D. melanogaster is specified by the fruitless (fru) and doublesex (dsx) sex regulatory genes. We used the temperature-sensitive activator dTRPA1 to probe the roles of fruM- and dsx-expressing neurons in male courtship behaviors. Almost all steps of courtship, from courtship song to ejaculation, can be induced at very high levels through activation of either all fruM or all dsx neurons in solitary males. Detailed characterizations reveal different roles for fruM and dsx in male courtship. Surprisingly, the system for mate discrimination still works well when all dsx neurons are activated, but is impaired when all fruM neurons are activated. Most strikingly, we provide evidence for a fruM-independent courtship pathway that is primarily vision dependent
