    Venom alkaloids against Chagas disease parasite: search for effective therapies

    Chagas disease is an important disease affecting millions of patients in the New World and is caused by a protozoan transmitted by haematophagous kissing bugs. It can be treated with drugs during the early acute phase; however, effective therapy against the chronic form of Chagas disease has yet to be discovered and developed. We herein tested the activity of solenopsin alkaloids extracted from two species of fire ants against the protozoan parasite Trypanosoma cruzi, the aetiologic agent of Chagas disease. Although IC50 determinations showed that solenopsins are more toxic to the parasite than benznidazole, the drug of choice for Chagas disease treatment, the ant alkaloids presented a lower selectivity index. As a result of exposure to the alkaloids, the parasites became swollen and rounded in shape, with hypertrophied contractile vacuoles and intense cytoplasmic vacuolization, possibly resulting in osmotic stress; no accumulation of multiple kinetoplasts and/or nuclei was detected. Overexpressing phosphatidylinositol 3-kinase—an enzyme essential for osmoregulation that is a known target of solenopsins in mammalian cells—did not prevent swelling and vacuolization, nor did it counteract the toxic effects of alkaloids on the parasites. Additional experimental results suggested that solenopsins induced a type of autophagic and programmed cell death in T. cruzi. Solenopsins also reduced the intracellular proliferation of T. cruzi amastigotes in infected macrophages in a concentration-dependent manner and demonstrated activity against Trypanosoma brucei rhodesiense bloodstream forms, which is another important aetiological kinetoplastid parasite.     The Antioxidant Capacity of Rosemary and Green Tea Extracts to Replace the Carcinogenic Antioxidant (BHA) in Chicken Burgers

    The present study aimed to evaluate the effect of natural extracts (rosemary and green tea extracts) in frozen storage of chicken burgers. Chicken burger treatments were prepared as follows: control (CON), 20 mg BHA/kg (BHA20), 10 mg green tea extract/kg (GT10), 38 mg green tea extract/kg (GT38), 18.6 mg rosemary extract/kg (RO18), and 480 mg rosemary extract/kg (RO480). Analysis of physicochemical parameters, color, TBAR index, and sensory acceptance were performed at 0, 30, 60, and 120 days of storage at −18°C in burgers packaged in LDPE plastic bags. The addition of natural antioxidants did not affect (p>0.05) the color and physicochemical parameters of the chicken burgers. After 120 days at −18°C, the RO480 sample showed a TBAR index similar (p>0.05) to BHA20 (0.423 and 0.369 mg, resp.). Sensory acceptance did not differ (p>0.05) among the treatments throughout the storage period (p>0.05)

    Biometric analysis of protein and oil contents of soybean genotypes in different environments

    The objective of this work was to identify by biometric analyses the most stable soybean parents, with higher oil or protein contents, cultivated at different seasons and locations of the state of Minas Gerais, Brazil. Forty-nine genotypes were evaluated in the municipalities of Viçosa, Visconde do Rio Branco, and São Gotardo, in the state of Minas Gerais, from 2009 to 2011. Protein and oil contents were analyzed by infrared spectrometry using a FT-NIR analyzer. The effects of genotype, environment, and genotype x environment interaction were significant. The BARC-8 soybean genotype is the best parent to increase protein contents in the progenies, followed by BR 8014887 and CS 3032PTA276-3-4. Selection for high oil content is more efficient when the crossings involve the Suprema, CD 01RR8384, and A7002 genotypes, which show high mean phenotypic values, wide adaptability, and greater stability to environmental variation

    Divergência em QTLs e variância genética para teores de proteína e óleo em soja

    Resumo: O objetivo deste trabalho foi avaliar a relação entre os parâmetros de divergência em regiões de QTLs e a variância genética em genótipos de soja, quanto aos teores de proteína e óleo nos grãos. Dois grupos de genótipos foram avaliados, em diferentes ambientes, quanto aos teores de proteína e óleo e genotipados com marcadores moleculares de regiões de QTLs. A partir de cada grupo, estabeleceram-se subgrupos por critérios pré-definidos e avaliou-se a relação entre os parâmetros, tendo-se comparado a divergência média e a variância genética entre os subgrupos. Os subgrupos foram definidos com base nos critérios de diferença em divergência média, homogeneidade e heterogeneidade nos subgrupos e proximidade em uma projeção tridimensional da matriz de distância. As percentagens de concordância entre maiores valores de divergência média e de variância genética para o total de subgrupos de cada grupo inicial foram de 72,5 e de 73,4%, respectivamente. Portanto, nestes genótipos, há relação positiva entre as estimativas de divergência em regiões de QTL e variância genética para os teores de proteína e de óleo dos grãos. As distâncias genéticas com base nos marcadores moleculares de regiões de QTLs são eficientes para a predição da variabilidade genética em genótipos de soja para os teores de proteína e de óleo dos grãos

    Caracterização da queixa escolar no Centro de Psicologia Aplicada da Universidade Federal de Juiz de Fora

    A queixa escolar vem se constituindo, historicamente, como um dos motivos mais frequentes de encaminhamentos de crianças para os serviços-escola de Psicologia, traduzindo, pela sua complexidade, um desafio para a formação e práticas no âmbito da psicologia escolar/educacional. Diante da necessidade de aperfeiçoar o atendimento à clientela e o processo de triagem, o presente estudo objetivou analisar os prontuários referentes à queixa escolar de pacientes atendidos no Centro de Psicologia Aplicada da Universidade Federal de Juiz de Fora, Minas Gerais, Brasil, num total de 1.590 documentos, no período de 1996 a 2009. Os resultados quanto à clientela evidenciaram a prevalência de meninos na faixa entre 9 e 13 anos de idade, com predomínio de encaminhamentos oriundos de escolas de periférias da cidade. Quanto à queixa, constatou-se expressiva coocorrência de problemas comportamentais e de aprendizagem. Os dados permitem uma reflexão crítica acerca da queixa escolar e de alguns fatores contextuais, bem como a necessidade de aperfeiçoamento da ficha de triagem

    Spatial analysis of avoidable hospitalizations due to tuberculosis in Ribeirao Preto, SP, Brazil (2006-2012)

    ABSTRACT OBJECTIVE To describe the spatial distribution of avoidable hospitalizations due to tuberculosis in the municipality of Ribeirao Preto, SP, Brazil, and to identify spatial and space-time clusters for the risk of occurrence of these events. METHODS This is a descriptive, ecological study that considered the hospitalizations records of the Hospital Information System of residents of Ribeirao Preto, SP, Southeastern Brazil, from 2006 to 2012. Only the cases with recorded addresses were considered for the spatial analyses, and they were also geocoded. We resorted to Kernel density estimation to identify the densest areas, local empirical Bayes rate as the method for smoothing the incidence rates of hospital admissions, and scan statistic for identifying clusters of risk. Softwares ArcGis 10.2, TerraView 4.2.2, and SaTScanTM were used in the analysis. RESULTS We identified 169 hospitalizations due to tuberculosis. Most were of men (n = 134; 79.2%), averagely aged 48 years (SD = 16.2). The predominant clinical form was the pulmonary one, which was confirmed through a microscopic examination of expectorated sputum (n = 66; 39.0%). We geocoded 159 cases (94.0%). We observed a non-random spatial distribution of avoidable hospitalizations due to tuberculosis concentrated in the northern and western regions of the municipality. Through the scan statistic, three spatial clusters for risk of hospitalizations due to tuberculosis were identified, one of them in the northern region of the municipality (relative risk [RR] = 3.4; 95%CI 2.7–4,4); the second in the central region, where there is a prison unit (RR = 28.6; 95%CI 22.4–36.6); and the last one in the southern region, and area of protection for hospitalizations (RR = 0.2; 95%CI 0.2–0.3). We did not identify any space-time clusters. CONCLUSIONS The investigation showed priority areas for the control and surveillance of tuberculosis, as well as the profile of the affected population, which shows important aspects to be considered in terms of management and organization of health care services targeting effectiveness in primary health care
