    Use of the LUS in sequence allele designations to facilitate probabilistic genotyping of NGS-based STR typing results

    Some of the expected advantages of next generation sequencing (NGS) for short tandem repeat (STR) typing include enhanced mixture detection and genotype resolution via sequence variation among non-homologous alleles of the same length. However, at the same time that NGS methods for forensic DNA typing have advanced in recent years, many caseworking laboratories have implemented or are transitioning to probabilistic genotyping to assist the interpretation of complex autosomal STR typing results. Current probabilistic software programs are designed for length-based data, and were not intended to accommodate sequence strings as the product input. Yet to leverage the benefits of NGS for enhanced genotyping and mixture deconvolution, the sequence variation among same-length products must be utilized in some form. Here, we propose use of the longest uninterrupted stretch (LUS) in allele designations as a simple method to represent sequence variation within the STR repeat regions and facilitate ā€“ in the nearterm ā€“ probabilistic interpretation of NGS-based typing results. An examination of published population data indicated that a reference LUS region is straightforward to define for most autosomal STR loci, and that using repeat unit plus LUS length as the allele designator can represent greater than 80% of the alleles detected by sequencing. A proof of concept study performed using a freely available probabilistic software demonstrated that the LUS length can be used in allele designations when a program does not require alleles to be integers, and that utilizing sequence information improves interpretation of both single-source and mixed contributor STR typing results as compared to using repeat unit information alone. The LUS concept for allele designation maintains the repeat-based allele nomenclature that will permit backward compatibility to extant STR databases, and the LUS lengths themselves will be concordant regardless of the NGS assay or analysis tools employed. Further, these biologically based, easy-to-derive designations uphold clear relationships between parent alleles and their stutter products, enabling analysis in fully continuous probabilistic programs that model stutter while avoiding the algorithmic complexities that come with string based searches. Though using repeat unit plus LUS length as the allele designator does not capture variation that occurs outside of the core repeat regions, this straightforward approach would permit the large majority of known STR sequence variation to be used for mixture deconvolution and, in turn, result in more informative mixture statistics in the near term. Ultimately, the method could bridge the gap from current length-based probabilistic systems to facilitate broader adoption of NGS by forensic DNA testing laboratories

    Using Economic Instruments to Develop Effective Management of Invasive Species: Insights From a Bioeconomic Model

    Economic growth is recognized as an important factor associated with species invasions. Consequently, there is increasing need to develop solutions that combine economics and ecology to inform invasive species management. We developed a model combining economic, ecological, and sociological factors to assess the degree to which economic policies can be used to control invasive plants. Because invasive plants often spread across numerous properties, we explored whether property owners should manage invaders cooperatively as a group by incorporating the negative effects of invader spread in management decisions (collective management) or independently, whereby the negative effects of invasive plant spread are ignored (independent management). Our modeling approach used a dynamic optimization framework, and we applied the model to invader spread using Linaria vulgaris. Model simulations allowed us to determine the optimal management strategy based on net benefits for a range of invader densities. We found that optimal management strategies varied as a function of initial plant densities. At low densities, net benefits were high for both collective and independent management to eradicate the invader, suggesting the importance of early detection and eradication. At moderate densities, collective management led to faster and more frequent invader eradication compared to independent management. When we used a financial penalty to ensure that independent properties were managed collectively, we found that the penalty would be most feasible when levied on a property\u27s perimeter boundary to control spread among properties. At the highest densities, the optimal management strategy was do nothing because the economic costs of removal were too high relative to the benefits of removal. Spatial variation in L. vulgaris densities resulted in different optimal management strategies for neighboring properties, making a formal economic policy to encourage invasive species removal critical. To accomplish the management and enforcement of these economic policies, we discuss modification of existing agencies and infrastructure. Finally, a sensitivity analysis revealed that lowering the economic cost of invader removal would strongly increase the probability of invader eradication. Taken together, our results provide quantitative insight into management decisions and economic policy instruments that can encourage invasive species removal across a social landscape

    TECHNIQUE SELECTION DURING LANDINGS IN ARTISTIC GYMNASTICS

    This study aimed to quantify the mechanical characteristics of two landing techniques performed by artistic gymnasts. Seven senior female artistic gymnasts performed 20 drop landings with different foot positions: feet together (womenā€™s technique) or hip width apart (menā€™s technique). Synchronised 3D kinematic (250 Hz) and kinetic data (1000 Hz) were collected for each trial and biomechanical variables associated with lower extremity injury during landing were analysed. Significant differences (Ī± \u3c 0.05) in ankle dorsiflexion and inversion at peak vertical ground reaction force (FPeakZ) were identified between techniques. The findings suggest the female landing style to be associated with increased FPeakZ, loading rate and reduced knee flexion. Whereas the male landing style was reported to exhibit significantly increased ankle inversion angles and reduced ankle dorsiflexion angles at FPeakZ. Results suggest that both strategies present characteristics associated with increased risk of differing injuries

    KINETIC STRATEGIES OF GYMNASTICS LANDINGS

    Organisation and magnitude of lower limb kinetics during landing can contribute to knowledge of injury. Our aim was to quantify leg stiffness and temporal organisation of joint moments of two different landing styles. Four female gymnasts performed 20 landings in the womenā€™s and menā€™s landing style. Synchronised kinematic (250 Hz) and kinetic (1000 Hz) data were used to describe the leg and joint stiffness, and the sequencing of the peak joint moments. Leg stiffness demonstrated lower variability compared to the associated joint level, suggesting joint level control. Group relative timing of the lower extremity joint moments presented distal-proximal sequencing, although individuals presented a variety of landing strategies. This study highlights the individualised signature strategies in landing, and consequently the importance of a single-subject design when examining injury risk

    The Sustainability Liability: Potential Negative Effects of Ethicality on Product Preference

    Manufacturers are increasingly producing and promoting sustainable products (i.e., products that have a positive social and/or environmental impact). However, relatively little is known about how product sustainability affects consumers\u27 preferences. Theauthors propose that sustainability may not always be an asset, even if most consumers care about social and environmental issues. The degree to which sustainability enhances preference depends on the type of benefit consumers most value for theproduct category in question. In this research, the authors demonstrate that consumers associate higher product ethicality with gentleness-related attributes and lower product ethicality with strength-related attributes. As a consequence of these associations, the positive effect of product sustainability on consumer preferences is reduced when strength-related attributes are valued, sometimes even resulting in preferences for less sustainable product alternatives (i.e., the sustainability liability ). Conversely, when gentleness-related attributes are valued, sustainability enhances preference. In addition, the authors show that the potential negative impact of sustainability on product preferences can be attenuated using explicit cues about productstrength

    Calmly coping: A Motivational Interviewing Via CoActive Life Coaching (MI-VIA-CALC) pilot intervention for university students with perceived levels of high stress

    Stress is a campus problem. This pre-post pilot study assessed the impact of Motivational Interviewing via Co-Active Life Coaching (MI-via-CALC) on the stress management of 30 full-time, English-speaking Canadian undergraduate students, aged 17-24 years. Participants were assessed using the Perceived Stress Scale and Hospital Anxiety and Depression Scale at pre-, mid-, and post-intervention. Three one-way, repeated-measures ANOVAs were completed. Statistically significant differences in stress reduction were found between pre-intervention to mid-intervention and between pre-intervention to post-intervention. No statistically significant differences occurred between mid- to post-intervention. Inductive content analysis of qualitative interviews at pre-, mid-, and post-intervention revealed participantsā€™ positive experiences
