Immunohistochemical Study of Dendritic Cells and Kupffer Cells in Griseofulvin-Induced Protoporphyric Mice

To determine whether protoporphyric liver injury affects hepatic dendritic cells (DC) and Kupffer cells, we examined liver tissues of griseofulvin-induced protoporphyric mice using histological and immunohistochemical methods. After 1 week of griseofulvin feeding, the protoporphyric liver showed prominent hepatomegaly and a gradual increase in histopathological changes such as hepatocellular hypertrophy, focal necrosis and brown pigment deposits. After 4 weeks of treatment, marked ductular reaction was observed in the liver tissue. Immunohistochemical analyses indicated that the density of NLDC-145-positive hepatic DC gradually decreased during griseofulvin feeding. However, the index of the number of DC in the whole liver appeared to fall sharply after 6 weeks. In contrast, the density of F4/80-positive Kupffer cells gradually increased during griseofulvin feeding. In the spleen and lung, no significant differences were noted in the distribution of NLDC-145-positive DC between experimental and control mice. These results suggested that griseofulvin-induced protoporphyria leads to a specific decrease in the density of hepatic DC due to hepatomegaly until 4 weeks of treatment and is substantial after 6 weeks. This substantial decrease of hepatic DC might have been induced by some alterations in protoporphyric liver injury including ductular reaction at a later stage in this experiment. Since hepatic DC were reduced in number, they seemed to have no significant relation to the progression of griseofulvin-induced protoporphyric liver injury. However, the decrease of hepatic DC might affect the cellular immune response in protoporphyria

Rho-associated kinase inhibitor eye drop (Ripasudil) transiently alters the morphology of corneal endothelial cells

PURPOSE: Ripasudil (Glanatec), a selective Rho-associated coiled coil-containing protein kinase (ROCK) inhibitor, was approved in Japan in September 2014 for the treatment of glaucoma and ocular hypertension. The purpose of this study was to investigate the effect of ripasudil eye drops on corneal endothelial morphology, as ROCK signaling is known to modulate the actin cytoskeleton. METHODS: Morphological changes in the corneal endothelium were evaluated in human subjects by specular and slit-lamp microscopy, following topical administration of ripasudil. We also used a rabbit model to evaluate the effect of ripasudil on clinical parameters of the corneal endothelium. Twenty-four hours after ripasudil application, corneal specimens were evaluated by phalloidin staining, immunohistochemical analysis, and electron microscopy. RESULTS: Specular microscopy revealed morphological changes in human eyes, and slit-lamp microscopy showed guttae-like findings. The rabbit model showed morphological changes similar to those seen in human eyes after ripasudil administration. Electron microscopy demonstrated that these alterations are due to the formation of protrusions along the cell-cell borders, but this formation is transient. Expression of corneal endothelial function-related markers was not disrupted; corneal thickness and corneal volume were not changed; and no cell death was observed following ripasudil administration. CONCLUSIONS: Ripasudil induces transient guttae-like findings in humans, most likely due to protrusion formation along intracellular borders caused by the reduction in actomyosin contractility of the corneal endothelial cells. No severe adverse effects were observed. Physicians should be aware that ROCK inhibitors can cause these guttae-like findings, to avoid misdiagnosing patients as having Fuchs endothelial corneal dystrophy. Rheological Scaling of Ionic Liquid-Based Polyelectrolytes in the Semidilute Unentangled Regime from Low to High Salt Concentrations

Polymerized ionic liquids (PILs) are a special class of ion-containing polymers with ionic liquid structures. The viscoelastic properties of PILs in IL solutions are expected to be influenced by both polymer–polymer interaction and charge screening by IL ions, which becomes complex at high IL concentrations due to strong ionic correlations. In this work, we aim to understand the effect of the ionic correlations on the shear rheology of PIL in IL solutions in the semidilute unentangled (SU) polymer regime, with moderate polymer–polymer interactions. We conducted systematic rheological characterization of a PIL, poly(1-butyl-3-vinylimidazolium bis(trifluoromethanesulfonyl)imide), in a mixture of an IL, 1-butyl-3-methylimidazolium bis(trifluoromethanesulfonyl)imide, and a non-ionic solvent, dimethylformamide, to evaluate the specific viscosity ηsp and the longest relaxation time λ with varying IL concentrations cIL, at a fixed polymer concentration. We found that both ηsp and λ initially decreased with the increasing cIL, following the scaling theory of polyelectrolyte solutions in the low salt concentration regime. However, these values exhibited an upturn at high cIL. We explained the observed non-monotonic viscoelastic behavior by proposing a charge screening model for SU solutions, accounting for both modified screening length in concentrated IL solutions and complete charge screening. Our results demonstrate how the strong ionic correlation in concentrated IL solutions modifies the electrostatics of PILs and their corresponding viscoelastic properties in the presence of moderate polymer–polymer interactions

NMDAR2B tyrosine phosphorylation regulates anxiety-like behavior and CRF expression in the amygdala

Abstract

Background

Anxiety disorders are a highly prevalent and disabling class of psychiatric disorders. There is growing evidence implicating the glutamate system in the pathophysiology and treatment of anxiety disorders, though the molecular mechanism by which the glutamate system regulates anxiety-like behavior remains unclear.

Results

In this study, we provide evidence suggesting that tyrosine phosphorylation of the NMDA receptor, an ionotropic glutamate receptor, contributes to anxiety-like behavior. The GluN2B subunit of the NMDA receptor is tyrosine-phosphorylated: Tyr-1472 is the major phosphorylation site. Homozygous knock-in mice that express a Tyr-1472-Phe mutant of GluN2B, which prevents phosphorylation of this site, show enhanced anxiety-like behavior in the elevated plus-maze test. Expression of corticotropin-releasing factor (CRF), which is important for the regulation of anxiety-like behavior, is increased in the amygdala of the knock-in mice. Furthermore, injection of CRF receptor antagonist attenuated the enhanced anxiety-like behavior of the knock-in mice. We also show that elevated plus-maze exposure simultaneously induced de-phosphorylation of Tyr-1472 and increased CRF expression.

Conclusions

These data suggest that Tyr-1472 phosphorylation on GluN2B is important for anxiety-like behavior by negative regulation of CRF expression in the amygdala.</p

Three crystallization methods, including crystallization in the presence of a semi-solid agarose gel, top-seeded solution growth (TSSG) and a large-scale hanging-drop method, have previously been presented. In this study, crystallization has been further evaluated in the presence of a semi-solid agarose gel by crystallizing additional proteins. A novel crystallization method combining TSSG and the large-scale hanging-drop method has also been developed

Suitable Stereostructures of vibrioferrin probes for iron uptake of Vibrio parahaemolyticus was revealed. Stereoisomers of dansyl labeled vibrioferrin at the 2′′-position were synthesized and their uptake activities were evaluated. Vibrio parahaemolyticus take in both isomers at the 2′′-position. In addition to Vibrio parahaemolyticus, several bacteria have also taken up the (R)-isomer
