The Effect of topical application pandan extract gel on gingival wound. Post-gingivectomy wound is usually covered by periodontal dressing, which generally contains some chemical compounds to protect the wound. However, it can provide allergic effect on some patients. Pandan leaves (Pandanus amaryllifolius Roxb.) contain a number of active substances that have anti-inflammatory, antioxidant and antibacterial effect and play a role in wound healing. This study aims to determine the effect of topical application of Pandan leaf extract gel on gingival wound healing. Gingivectomy model was carried out on mandibular incisive gingival using the 2.5-mm punch biopsy. Thirty-six rats were randomly divided into 3 groups of treatment: negative control (CMC-Na), positive control (Aloclair), and 50% of pandan extract gel. The gels on each group were applied (twice in a day) to the wound area after gingivectomy. The observation of the wound healing process was also carried out on day 1, 3, 7, and 14 by making the histological preparations of gingival wound area. The number of blood vessels was observed using microscope and data was analysed using Two- Way Anova and LSD. The result showed that number of blood vessel increased on day 3 and the peak was on day 7. Anova and LSD test showed several significant differences comparison the number blood vessel between treatment and control. In conclusion, topical application Pandan leaves extract gel could accelerate gingival wound healing

Non-canonical features of the Golgi apparatus in bipolar epithelial neural stem cells.

Apical radial glia (aRG), the stem cells in developing neocortex, are unique bipolar epithelial cells, extending an apical process to the ventricle and a basal process to the basal lamina. Here, we report novel features of the Golgi apparatus, a central organelle for cell polarity, in mouse aRGs. The Golgi was confined to the apical process but not associated with apical centrosome(s). In contrast, in aRG-derived, delaminating basal progenitors that lose apical polarity, the Golgi became pericentrosomal. The aRG Golgi underwent evolutionarily conserved, accordion-like compression and extension concomitant with cell cycle-dependent nuclear migration. Importantly, in line with endoplasmic reticulum but not Golgi being present in the aRG basal process, its plasma membrane contained glycans lacking Golgi processing, consistent with direct ER-to-cell surface membrane traffic. Our study reveals hitherto unknown complexity of neural stem cell polarity, differential Golgi contribution to their specific architecture, and fundamental Golgi re-organization upon cell fate change

Malaria is one of the life-threatening diseases in the world. The spread of resistance to antimalarial drugs is a major challenge, and resistance to artemisinin has been reported in the Southeast Asian region. In the previous study, the active compound of Streptomyces hygroscopicus subsp. Hygroscopicus (S. hygroscopicus), eponemycin, has been shown to have antimalarial effects. To further analyze the effects of other active compounds on the Plasmodium parasite, identifying and analyzing the effectiveness of compounds contained in S. hygroscopicus through instrumentation of liquid chromatography/mass spectrometry (LC/MS) and in silico studies were very useful. This study aimed at identifying other derivative compounds from S. hygroscopicus and screening the antimalarial activity of the compound by assessing the binding affinity, pharmacokinetic profile, and bond interaction. The derivative compounds were identified using LC/MS. Protein targets for derivative compounds were found through literature studies, and the results of identification of compounds and protein targets were reconstructed into three-dimensional models. Prediction of pharmacokinetic profiles was carried out using Swiss ADME. Screening of protein targets for the derivative compound was carried out using the reverse molecular docking method. Analyzing bond interaction was done by LigPlot. One compound from S. hygroscopicus, i.e., 6,7-dinitro-2-[1, 2, 4]triazole-4-yl-benzo[de]isoquinoline-1,3-dione, was successfully identified using LC/MS. This compound was an isoquinoline derivative compound. Through literature studies with inclusion criteria, thirteen protein targets were obtained for reverse molecular docking. This isoquinoline derivative had the potential to bind to each protein target. The pharmacokinetic profile showed that this compound had the drug-likeness criteria. Conclusion. 6,7-Dinitro-2-[1, 2, 4]triazole-4-yl-benzo[de]isoquinoline-1,3-dione has antimalarial activity as shown by reverse molecular docking studies and pharmacokinetic profiles. The best inhibitory ability of compounds based on bond affinity is with adenylosuccinate synthetase

Background: The burden of Osteoarthritis (OA) is huge, with a sizable proportion of patients that have failed the analgesic treatment and are not candidates for surgery or have refused surgery. Intraarticular corticosteroid injections (IASI) are considered standard of care for pain relief and control of local inflammation in this category of OA patients. However, there is a wide variation in the duration of response to steroid injections. This study was designed to determine the duration of response to IASI in patients with osteoarthritis of the knee in our environment. Methods: Fifty-four patients aged between 30 and 80 years who have been diagnosed with osteoarthritis using the American College of Rheumatology criteria and have met the inclusion criteria were enrolled in the study and were given intra-articular steroid injections to the knee. Their responses were assessed using visual analogue scale (VAS) score, Western Ontario, and McMaster Universities Osteoarthritis Index (WOMAC) score at 2, 4 and 12 weeks. The patients were classified as responders if there was a fifty percent improvement in the WOMAC scores and a fifty percent reduction in the VAS score and non-responders if there is less than fifty percent improvement in the WOMAC score and less than fifty percent reduction in the VAS score. Results: Fifty-two patients completed the study; 78.4% and 100% of these had good WOMAC and VAS responses respectively at 2 weeks but this proportion gradually reduced to 47.9% and 56.3% for WOMAC and VAS respectively at 3 months. Conclusion: Intra-articular steroid injections provide sustained response in patients with osteoarthritis of the knee who have failed analgesic therapy and are not candidates for total knee replacement for up to three months and the response decreases with advancing age
