    Do Maladaptive Cognitions Mediate the Relationship between Perceived Parental Influences and Eating Behavior Among College Women?

    This thesis was submitted in partial fulfillment of the requirements for graduation with distinction in Psychology.Studies have shown that disordered eating is usually accompanied by other psychological disturbances that involve maladaptive cognitions such as perfectionism, depressive symptomatology, and negative affect. Evidence has been presented linking parental influences to these other maladaptive cognitions, showing a possible mediational relationship through maladaptive cognitions between parental influences and eating behavior. The present study is a preliminary investigation of this hypothesis. Specifically, parental influences to personality (i.e., attachment avoidance and anxiety) and eating behavior (i.e., family food rules) were expected to predict maladaptive cognitions (i.e., depression, negative affect, and maladaptive perfectionism) which were then expected to predict disordered eating. Among 107 women, all three parental influences predicted at least form of maladaptive cognitions, with attachment anxiety predicting all three. Depression and maladaptive perfectionism, in turn, predicted disordered eating. Implications for therapy to include parental influences for eating disordered clients are discussed.No embarg

    One Signal, Two Behaviors: Odor Discrimination in Unmated versus Mated Female Green Bottle Flies, Lucilia sericata

    The green bottle fly, Lucilia sericata, is of critical importance in the field of forensic entomology since it is one of the first insects to arrive at a freshly deceased carcass. These flies use a highly tuned and selective olfactory system to identify and locate the carcass; response to the decomposition odor usually occurs within minutes. Male flies use the carcass for a small protein meal and for locating females while females use the carcass for feeding (unmated) and egg-laying (mated). This divergence in behavior to a common odor cue is the subject of this thesis proposal: do unmated (feeding) females respond to the same odor cues as mated (egg-laying) females? The presence of two behaviors indicates that there may be an associated up or down regulation of gene expression of odor binding proteins during the two different stages of female sexual development. Olfactory response to selected decomposition odors in unmated versus mated females was determined using the electroantennogram (EAG) which measures the neuronal depolarization in antennae when an odor triggers a response. Fly heads were mounted on the EAG probes and exposed selected volatiles. Response was measured as the resultant change in voltage (a depolarization). Flies were also subjected to an odor choice behavioral assay. The results show a divergence in antennal response to VOC and choice of VOC at day 4 between the mated and unmated female flies. These results indicate that there are underlying molecular, biochemical, and physiological processes associated with fly response to odors

    Régulation du facteur de transcription FOXK1 par O-GlcNAcylation : implications dans la différenciation adipocytaire

    Les modifications post-traductionnelles telles que la phosphorylation, l’OGlcNAcylation et l’ubiquitination jouent des rôles critiques dans la coordination des fonctions protéiques et par conséquent influencent grandement de nombreux processus cellulaires. Il est à noter que ces modifications sont hautement dynamiques et finement regulées. Par exemple, l’ubiquitination peut être réversible via l’action des déubiquitinases comme le suppresseur de tumeurs BAP1. Parmis les gènes codant pour les déubiquitinases, BAP1 est la plus souvent mutée dans le cancer. Des études récentes ont démontré l’importance des dynamiques de modifications post-traductionnelles dans la régulation du complexe BAP1. En plus, BAP1 forme un complexe multi-protéiques contenant plusieurs régulateurs transcriptionnels comme la protéine polycomb OGT et les facteurs de transcription FOXK1 et FOXK2. OGT est une enzyme unique qui catalyze l’ajout d’un groupement O-GlcNAc sur ses substrats afin d’en moduler l’activité enzymatique, les interactions protéines-protéines et leur localisation cellulaire. Cette modification est aussi liée au métabolisme puisque son substrat donneur, l’UDP-GlcNAc, est dérivé de la voie biosynthétique des hexosamines. Parallèlement, FOXK1/2 ont aussi été démontrés comme étant critiques à des processus métaboliques telles que la myogenèse et l’autophagie. Lors de nos études, nous avons identifié FOXK1 comme un nouveau substrat d’OGT. De plus, les niveaux d’O-GlcNAcylation de FOXK1 fluctuent lors de l’entrée/sortie du cycle cellulaire. En outre, nous avons identifié l’importance de FOXK1 dans l’adipogenèse et observé que l’interaction FOXK1/BAP1 est affectée par le métabolisme cellulaire. En résumé, nos études ont révélé l’importance d’OGT dans la régulation de certaines composantes du complexe BAP1, ce qui aidera à la compréhension de l’effet suppresseur de tumeur de BAP1 ainsi que son mécanisme d'action dans différents processus tel que le remodelage de la chromatine.Post-translational modifications such as phosphorylation, O-GlcNAcylation and ubiquitination play critical roles in coordinating protein function and are therefore involved in diverse cellular processes. Of relevance here, ubiquitination may be removed by deubiquitinases such as the tumour suppressor BAP1, which represents the most mutated deubiquitinase gene in the human genome. Recent studies have revealed that important and dynamic post-translational modifications regulate several functions of the BAP1 complex. Indeed, BAP1 has been shown to form a multi-protein complex with several transcriptional regulators including the polycomb group protein OGT and the transcription factors FOXK1 and FOXK2. OGT is a unique enzyme that catalyzes the addition of an O-GlcNAc moiety to target proteins, which impacts protein function including enzymatic activity, protein-protein interactions and subcellular localization. This modification is also highly linked to cellular metabolism, as the donor substrate for the reaction, UDP-GlcNAc, is derived from the hexosamine biosynthesis pathway. Similarly, FOXK1 and FOXK2 have been shown to be implicated in metabolic processes such as myogenesis and autophagy. During our studies, we identified FOXK1 but not FOXK2 as a novel substrate of OGT. Further, we found that this OGlcNAcylation is modulated during the entry/exit of cell cycle. We also found that FOXK1 is critical for adipogenesis and that the interaction between FOXK1/BAP1 is compromised during nutrient starvation. Thus, our studies have revealed that OGT selectively modulates and regulates components of the BAP1 complex which may impact different cellular processes, notably chromatin remodelling and could help understanding how BAP1 acts as a tumor suppressor

    Environmental and Economic Assessment of Energy Renovation in Buildings, a Case Study in Greece

    The environmental and economic evaluation of energy renovation in buildings plays a crucial role in achieving sustainability goals and the decarbonization of the built environment. This paper presents a case study of a student house in Athens, Greece, to assess the environmental and economic impacts of energy renovation and seismic reinforcement with a steel exoskeleton. This study utilizes a comprehensive approach that combines life cycle assessment (LCA) and life cycle costing (LCC) methodologies using One Click LCA. The LCA assesses the environmental impacts associated with energy consumption and greenhouse gas emissions, while the LCC evaluates the economic aspects, both analyses being conducted for a lifespan of 25 years from now. The results provide an evaluation of what would happen in terms of greenhouse emissions and costs in two scenarios: with and without interventions. ProGETonE strategy results in an environmental impact with a GWP of 26.78 kgCO2eq/m2y with a reduction of 30% of the pre-renovation state. Economically, the actualized energy use costs for 25 years are 50% less in the post-renovation state, but the high construction costs make the strategy seem inconvenient. In this context, it is important to consider the non-economic benefits of seismic reinforcement, such as enhanced safety and the potential lives saved, which are critical in high seismic zones. These advantages complement the strategy's environmental and energy use impacts, underscoring the holistic value of integrated seismic and energy retrofitting approaches like ProGETonE. The study underscores the importance of LCA and LCC analyses when evaluating the feasibility of renovation projects and of an evidence-based decision-making process for policymakers, building owners, and stakeholders for energy-efficient retrofitting

    Building Information Modeling as an Effective Process for the Sustainable Re-Shaping of the Built Environment

    This paper focuses on the definition of a method supported by digital processes for a sustainable and user-orientated re-design of the existing building stock. Based on the analysis of the methodological and procedural aspects of the computational approach to architectural design in relation to different performance conditions, the research addresses the adoption of Building Information Modeling (BIM), intended as a powerful method for coordinating the complexity of the multiple, interdisciplinary and conflicting aspects involved in the rehabilitation of buildings. In addition to the advantages in terms of control and management, the BIM process has proven its effectiveness in tackling the issue of sustainability, allowing all actors involved in the research to share information and pro-actively control various outcomes of a building’s performance, such as energy and environmental quality. To show the opportunities and limitations of the digital management in information-based processes, the activities carried out in the framework of the European Horizon 2020 project “Pro-GET-onE—Proactive synergy of inteGrated Efficient Technologies on buildings’ Envelopes” are reported. The research, based on a case study method, which is applied to a student residence in Athens, demonstrates that BIM possesses great potentialities for developing effective and efficient construction and renovation processes toward buildings with high quality standards

    Applications of large language models in cancer care: current evidence and future perspectives

    The development of large language models (LLMs) is a recent success in the field of generative artificial intelligence (AI). They are computer models able to perform a wide range of natural language processing tasks, including content generation, question answering, or language translation. In recent months, a growing number of studies aimed to assess their potential applications in the field of medicine, including cancer care. In this mini review, we described the present published evidence for using LLMs in oncology. All the available studies assessed ChatGPT, an advanced language model developed by OpenAI, alone or compared to other LLMs, such as Google Bard, Chatsonic, and Perplexity. Although ChatGPT could provide adequate information on the screening or the management of specific solid tumors, it also demonstrated a significant error rate and a tendency toward providing obsolete data. Therefore, an accurate, expert-driven verification process remains mandatory to avoid the potential for misinformation and incorrect evidence. Overall, although this new generative AI-based technology has the potential to revolutionize the field of medicine, including that of cancer care, it will be necessary to develop rules to guide the application of these tools to maximize benefits and minimize risks
