A two-component pre-seeded dermal-epidermal scaffold
We have developed a bilayered dermal-epidermal scaffold for application in the treatment of full-thickness skin defects. The dermal component gels in situ and adapts to the lesion shape, delivering human dermal fibroblasts in a matrix of fibrin and cross-linked hyaluronic acid modified with a cell adhesion-promoting peptide. Fibroblasts were able to form a tridimensional matrix due to material features such as tailored mechanical properties, presence of protease-degradable elements and cell-binding ligands. The epidermal component is a robust membrane containing cross-linked hyaluronic acid and poly-l-lysine, on which keratinocytes were able to attach and to form a monolayer. Amine-aldehyde bonding at the interface between the two components allows the formation of a tightly bound composite scaffold. Both parts of the scaffold were designed to provide cell-type-specific cues to allow for cell proliferation and form a construct that mimics the skin environment.D.S.K. acknowledges funding from the Biotechnology Research Endowment from the Department of Anesthesiology at Boston Children's Hospital. The NEXT White (NEW) detector
Conceived to host 5 kg of xenon at a pressure of 15 bar in the fiducial volume, the NEXT-White apparatus is currently the largest high pressure xenon gas TPC using electroluminescent amplification in the world. It is also a 1:2 scale model of the NEXT-100 detector for Xe-136 beta beta 0 nu decay searches, scheduled to start operations in 2019. Both detectors measure the energy of the event using a plane of photomultipliers located behind a transparent cathode. They can also reconstruct the trajectories of charged tracks in the dense gas of the TPC with the help of a plane of silicon photomultipliers located behind the anode. A sophisticated gas system, common to both detectors, allows the high gas purity needed to guarantee a long electron lifetime. NEXT-White has been operating since October 2016 at the Laboratorio Subterraneo de Canfranc (LSC), in Spain. Functional mechanisms underlying pleiotropic risk alleles at the 19p13.1 breast-ovarian cancer susceptibility locus
A locus at 19p13 is associated with breast cancer (BC) and ovarian cancer (OC) risk. Here we analyse 438 SNPs in this region in 46,451 BC and 15,438 OC cases, 15,252 BRCA1 mutation carriers and 73,444 controls and identify 13 candidate causal SNPs associated with serous OC (P=9.2 × 10-20), ER-negative BC (P=1.1 × 10-13), BRCA1-associated BC (P=7.7 × 10-16) and triple negative BC (P-diff=2 × 10-5). Genotype-gene expression associations are identified for candidate target genes ANKLE1 (P=2 × 10-3) and ABHD8 (P<2 × 10-3). Chromosome conformation capture identifies interactions between four candidate SNPs and ABHD8, and luciferase assays indicate six risk alleles increased transactivation of the ADHD8 promoter. Targeted deletion of a region containing risk SNP rs56069439 in a putative enhancer induces ANKLE1 downregulation; and mRNA stability assays indicate functional effects for an ANKLE1 3′-UTR SNP. Altogether, these data suggest that multiple SNPs at 19p13 regulate ABHD8 and perhaps ANKLE1 expression, and indicate common mechanisms underlying breast and ovarian cancer risk
Unravelling pine response to Fusarium circinatum through Raman spectroscopy
Pine Pitch Canker (PPC), caused by the fungus Fusarium circinatum, is associated to significant economic and ecological losses worldwide. The effectiveness of PPC monitoring, early detection in nurseries and plantations, and the identification of resistant plant material relies on the development of objective, non-destructive, and cost-effective tools. This study analyzed the potential of employing Raman Spectroscopy (RS) for the early detection of biochemical changes associated with PPC in Pinus spp. with different susceptibilities to F. circinatum (highly susceptible Pinus radiata vs relatively resistant Pinus pinea), while unveiling possible mechanisms of action on these pathosystems. Our results indicate lignin as a key constitutive component of pine resistance against PPC and thus the potential of using this technology for the selection of PPC resistant trees. Moreover, we demonstrate the power of RS-based approaches for the rapid detection of the disease in susceptible species. Early spectral variations were found in P. radiata upon inoculation with F. circinatum from 3 days post-inoculation (dpi) onwards, whereas changes in histological analysis, relative internal stem necrosis measurements, and visual disease symptoms were only visible at 6, 8, and 9 dpi, respectively. These spectral changes have been associated to cell wall degradation and induction of phenolic compounds synthesis upon infection in P. radiata. Altogether, we believe that RS is an innovative promising tool able to reduce disease detection time in pine species and providing an appealing alternative for the development of new and eco-friendly disease control measures
Atributos químicos do solo e produção do feijoeiro com a aplicação de calcário e manganês Soil chemical atributes and common bean yield as affected by lime and manganese rates
O feijão (Phaseolus vulgaris L.) constitui a base alimentar da população brasileira. Muitos fatores determinam a baixa produtividade do feijoeiro, desde problemas políticos até fatores técnicos, dentre eles a adubação e nutrição. O objetivo foi avaliar os atributos químicos do solo e produção do feijoeiro (cv. Pérola) de acordo com a aplicação de calagem e Mn. O experimento foi realizado em vasos com Latossolo Vermelho-Amarelo distrófico típico fase cerrado, em delineamento inteiramente casualizado com três repetições, em esquema fatorial 2 x 6, envolvendo duas doses de uma mistura de CaCO3 e MgCO3, na relação molar 3:1 (de calcário, calculadas) para elevar a saturação por base para 50 % e para 70 % e seis doses de Mn (0, 20, 40, 60, 80 e 100 mg dm-3). Na análise dos resultados, verificou-se decréscimo no teor foliar de Mn de acordo com o aumento da saturação por base, não sendo observado o mesmo efeito no solo após o cultivo. O teor de Mn no solo para produção máxima de grãos mostrou-se semelhante para saturação por base de 50 e 70 %; no entanto, a dose do micronutriente aplicada para essa produção foi de 28 mg dm-3 e 57 mg dm-3, respectivamente.<br>Common bean is one of the most important staples in Brazil. The objective of this study was to evaluate the common bean cv. Pérola yield and changes in soil chemical attributes as influenced by liming and manganese rates. The experiment was carried out in pots containing dystrophic Red Yellow Latosol (Oxisol). Treatments consisted of a 2 x 6 factorial combination of two limestone (CO3 and MgCO3 in a 3:1 molar ratio) rates, calculated to increase base saturation to 50 and 70 %, respectively, and six Mn rates (0, 20, 40, 60, 80 and 100 mg kg-1), using MnSO4.H2O as source. There were three repetitions. Results showed that a higher base saturation reduced leaf Mn concentration, but soil Mn concentration after harvest was not altered. The soil Mn concentration sufficient to reach the maximum grain yield was similar for both 50 and 70 % base saturation, but the doses to reach such critical soil concentration were 28 and 57 mg dm-3, respectively
Profundidade de semeadura e crescimento inicial de espécies forrageiras utilizadas para cobertura do solo Depth of sowing and initial growth of forage species used for soil coverage
O uso do consórcio entre plantas de cobertura e culturas anuais, no Cerrado, tem sido prática cada vez mais difundida, pela possibilidade de implantação das forrageiras durante o desenvolvimento das culturas anuais, através da diferença de profundidade e sistema de semeadura. Objetivou-se, neste trabalho avaliar a emergência e algumas características agronômicas de quatro espécies de plantas de cobertura, em diferentes profundidades, visando ao seu emprego na consorciação e sobressemeadura em culturas anuais. O experimento foi conduzido em casa-de-vegetação, no delineamento experimental de blocos ao acaso, em arranjo fatorial 4 x 7, composto pelas plantas de cobertura Brachiaria brizantha, B. decumbens, B. ruziziensis e Panicum maximum cv. Tanzânia, e por seis profundidades de semeadura e um sistema de cobertura de sementes (0 sem cobertura, 0 com cobertura, 1, 4, 8, 10 e 15 cm). As sementes foram semeadas em vasos e as plantas cultivadas por 40 dias. Avaliou-se o índice e o tempo de emergência, a altura de plantas, a fitomassa verde e seca da parte aérea, a área foliar, a fitomassa seca radicular e a densidade de comprimento radicular. O maior desenvolvimento das plantas estudadas foi detectado nas semeaduras até 1 cm de profundidade. O P. maximum não apresentou aptidão para uso em consórcio com culturas anuais.Não se recomenda a semeadura das plantas de cobertura a partir de 10 cm.<br>The use of the consortium between cover crops and annual crops in the Cerrado, is an increasingly widespread practice, because of the possibility of deployment of cover crops during the development of annual crops by the difference in depth and seeding system. The objective was to evaluate the emergency and some agronomic characteristics of four species of cover crops at different seeding depths, to use them in consortium and oversown in annual crops. The experiment was conducted in a greenhouse, and the experimental design of randomized blocks in factorial arranged 4 x 7, where it was four cover crops: Brachiaria brizantha, B. decumbens, B. ruziziensis and Panicum maximum cv. Tanzania, and six depths of sowing and a cover of seed system (0 cm without any mulch, 0 cm with a mulch of soybean leaves, 1, 4, 8, 10 and 15 cm). The cover crops were cropped in vase for 40 days. We evaluated emergence index, emergence time, plant height, green biomass and dry biomass of the above-ground part, leaf area, root dry biomass and root length density. The largest development of the cover crops was detected in the sowing up to 1 cm of depth. The P. maximum showed no ability to use in consortium with annual crops. The sowing of the cover crops from 10 cm is not recommended

Os fertilizantes utilizados para suprir micronutrientes podem apresentar metais pesados tóxicos em sua composição que podem proporcionar severas consequências ao meio ambiente. Plantas cultivadas na presença de determinadas concentrações de elementos tóxicos podem oferecer risco de contaminação, pois os acumulam em seus tecidos. Desse forma, neste trabalho objetivou-se avaliar a disponibilização dos metais pesados tóxicos Cd, Pb, e Cr para o solo e tecido foliar da cultura da soja, cultivada a campo e fertilizada com um formulado N:P2O5:K2O e diferentes fontes de Zn. O delineamento experimental utilizado foi o de blocos ao acaso, em esquema fatorial [(5+1)x2], com três repetições. Os tratamentos foram constituídos de cinco diferentes fertilizantes NPK+Zn (quatro fontes com Zn e uma sem Zn), um tratamento sem adubação e duas doses de adubação (uma vez e o seu dobro). Com base nos resultados obtidos foi concluído que a aplicação dos tratamentos disponibilizou Cd, Pb e Cr para as plantas de soja e para o solo do experimento nas duas doses de adubação utilizadas.<br>Fertilizers used to supply micronutrients may have toxic heavy metals in their composition, which can have severe consequences for the environment. Plants grown in the presence of certain concentrations of toxic elements may present a risk of contamination because the toxins accumulate in their tissues. This study aimed to measure the presence of the toxic heavy metals Cd, Pb and Cr in the soil and leaf tissue of field-grown soybean plants, fertilized with a formulated N:P2O5:K2O and different sources of Zn. The experimental design utilized randomized blocks in a factorial scheme [(5+1)x2], with three replications. The treatments consisted of five different fertilizers NPK + Zn (four sources with Zn and one without Zn), a treatment without fertilizer and two fertilizer levels (a single and double application). Based on obtained results it was concluded that the double application of the treatments released Cd, Pb and Cr into the soybean plants and experiment
