    Humans are essentially human politics (zoon politikon), so that all the dynamics of human life must always be related to politics, both as a political subject and as an object driven by politics itself. It can be clearly stated that the dynamics of human life in a country must be in circulation of the subject and object at once. Ideally, whether it is entrusted as a political leader or as a society controlled by politics itself, both are political subjects. That is, those who are entrusted with directing and regulating state politics are clearly a subject that always deals with politics in a concrete way, but the general public is also a subject, meaning that all aspects of life and community activities influence the political world both directly and indirectly, or with in other words it can be said that every activity of the community both in the world of education, religion, social, law, etc. is a political responsibility to organize, control, and direct all aspects of life for the common good, so that briefly it can be said that society is the determinant politics itself and as a function of political control, both directly and indirectly. AbstrakManusia pada hakikatnya adalah manusia politik (zoon politikon), sehingga seluruh dinamika kehidupan manusia pasti selalu berkenaan dengan politik, baik sebagai subjek yang berpolitik maupun sebagai objek yang digerakan oleh politik itu sendiri. Dengan jelas dapat dikatakan bahwa dinamika kehidupan manusia dalam suatu negara pasti berada dalam sirkulasi subjek dan objek sekaligus. Idealnya, baik yang dipercayakan sebagai pemimpin politik maupun sebagai masyarakat yang dikendalikan oleh politik itu sendiri, keduanya adalah subjek (pelaku) politik. Artinya, mereka yang dipercayakan mengarahkan dan mengatur politik negara jelas adalah subjek yang selalu bergelut dengan politik secara konkret , akan tetapi masyarakat umum juga adalah subjek, artinya segala aspek kehidupan dan aktivitas masyarakat membawa pengaruh bagi dunia politik baik  secara langsung maupun tidak langsung, atau dengan kata lain dapat dikatakan bahwa setiap aktivitas masyarakat baik dalam dunia pendidikan, agama, sosial, hukum, dan lain sebagainya menjadi tanggung jawab politik untuk menata, mengendalikan, dan mengarahkan semua aspek kehidupan tersebut demi kebaikan bersama, sehingga dengan singkat dapat dikatakan bahwa masyarakat adalah penentu politik itu sendiri dan sebagai fungsi kontrol politik, baik secara langsung maupun tidak langsung

    EEG potentials associated with artificial grammar learning in the primate brain

    AbstractElectroencephalography (EEG) has identified human brain potentials elicited by Artificial Grammar (AG) learning paradigms, which present participants with rule-based sequences of stimuli. Nonhuman animals are sensitive to certain AGs; therefore, evaluating which EEG Event Related Potentials (ERPs) are associated with AG learning in nonhuman animals could identify evolutionarily conserved processes. We recorded EEG potentials during an auditory AG learning experiment in two Rhesus macaques. The animals were first exposed to sequences of nonsense words generated by the AG. Then surface-based ERPs were recorded in response to sequences that were ‘consistent’ with the AG and ‘violation’ sequences containing illegal transitions. The AG violations strongly modulated an early component, potentially homologous to the Mismatch Negativity (mMMN), a P200 and a late frontal positivity (P500). The macaque P500 is similar in polarity and time of occurrence to a late EEG positivity reported in human AG learning studies but might differ in functional role

    Antibody Fc Glycosylation Discriminates Between Latent and Active Tuberculosis

    Background. Mycobacterium tuberculosis remains a global health problem and clinical management is complicated by difficulty in discriminating between latent infection and active disease. While M. tuberculosis-reactive antibody levels are heterogeneous, studies suggest that levels of IgG glycosylation differ between disease states. Here we extend this observation across antibody domains and M. tuberculosis specificities to define changes with the greatest resolving power. Methods. Capillary electrophoretic glycan analysis was performed on bulk non-antigen–specific IgG, bulk Fc domain, bulk Fab domain, and purified protein derivative (PPD)- and Ag85A-specific IgG from subjects with latent (n = 10) and active (n = 20) tuberculosis. PPD-specific isotype/subclass, PPD-specific antibody-dependent phagocytosis, cellular cytotoxicity, and natural killer cell activation were assessed. Discriminatory potentials of antibody features were evaluated individually and by multivariate analysis. Results. Parallel profiling of whole, Fc, and Fab domain-specific IgG glycosylation pointed to enhanced differential glycosylation on the Fc domain. Differential glycosylation was observed across antigen-specific antibody populations. Multivariate modeling highlighted Fc domain glycan species as the top discriminatory features, with combined PPD IgG titers and Fc domain glycans providing the highest classification accuracy. Conclusions. Differential glycosylation occurs preferentially on the Fc domain, providing significant discriminatory power between different states of M. tuberculosis infection and disease

    Coordinated Fc-effector and neutralization functions in HIV-infected children define a window of opportunity for HIV vaccination

    OBJECTIVES: Antibody function has been extensively studied in HIV-infected adults but is relatively understudied in children. Emerging data suggests enhanced development of broadly neutralizing antibodies (bNAbs) in children but Fc effector functions in this group are less well defined. Here, we profiled overall antibody function in HIV-infected children. DESIGN: Plasma samples from a cross-sectional study of 50 antiretroviral therapy-naive children (aged 1-11 years) vertically infected with HIV-1 clade A were screened for HIV-specific binding antibody levels and neutralizing and Fc-mediated functions. METHODS: Neutralization breadth was determined against a globally representative panel of 12 viruses. HIV-specific antibody levels were determined using a multiplex assay. Fc-mediated antibody functions measured were antibody-dependent: cellular phagocytosis (ADCP); neutrophil phagocytosis (ADNP); complement deposition (ADCD) and natural killer function (ADNK). RESULTS: All children had HIV gp120-specific antibodies, largely of the IgG1 subtype. Fifty-four percent of the children exhibited more than 50% neutralization breadth, with older children showing significantly broader neutralization activity. Apart from ADCC, observed only in 16% children, other Fc-mediated functions were common (>58% children). Neutralization breadth correlated with Fc-mediated functions suggesting shared determinants of enhanced antibody function exist. CONCLUSIONS: These results are consistent with previous observations that children may develop high levels of neutralization breadth. Furthermore, the striking association between neutralization breadth and Fc effector function suggests that HIV vaccination in children could yield multifunctional antibodies. Paediatric populations may therefore provide an ideal window of opportunity for HIV vaccination strategies

    Fidelity trade-off for finite ensembles of identically prepared qubits

    We calculate the trade-off between the quality of estimating the quantum state of an ensemble of identically prepared qubits and the minimum level of disturbance that has to be introduced by this procedure in quantum mechanics. The trade-off is quantified using two mean fidelities: the operation fidelity which characterizes the average resemblance of the final qubit state to the initial one, and the estimation fidelity describing the quality of the obtained estimate. We analyze properties of quantum operations saturating the achievability bound for the operation fidelity versus the estimation fidelity, which allows us to reduce substantially the complexity of the problem of finding the trade-off curve. The reduced optimization problem has the form of an eigenvalue problem for a set of tridiagonal matrices, and it can be easily solved using standard numerical tools.Comment: 26 pages, REVTeX, 2 figures.     Divergent Antibody Subclass and Specificity Profiles but Not Protective HLA-B Alleles Are Associated with Variable Antibody Effector Function among HIV-1 Controllers

    Understanding the coordination between humoral and cellular immune responses may be the key to developing protective vaccines, and because genetic studies of long-term HIV-1 nonprogressors have associated specific HLA-B alleles with spontaneous control of viral replication, this subject group presents an opportunity to investigate relationships between arms of the adaptive immune system. Given evidence suggesting that cellular immunity may play a role in viral suppression, we sought to determine whether and how the humoral immune response might vary among controllers. Significantly, Fc-mediated antibody effector functions have likewise been associated with durable viral control. In this study, we compared the effector function and biophysical features of HIV-specific antibodies in a cohort of controllers with and without protective HLA-B alleles in order to investigate whether there was evidence for multiple paths to HIV-1 control, or whether cellular and humoral arms of immunity might exhibit coordinated profiles. However, with the exception of IgG2 antibodies to gp41, HLA status was not associated with divergent humoral responses. This finding did not result from uniform antibody responses across subjects, as controllers could be regrouped according to strong differences in their HIV-specific antibody subclass specificity profiles. These divergent antibody profiles were further associated with significant differences in nonneutralizing antibody effector function, with levels of HIV-specific IgG1 acting as the major distinguishing factor. Thus, while HLA background among controllers was associated with minimal differences in humoral function, antibody subclass and specificity profiles were associated with divergent effector function, suggesting that these features could be used to make functional predictions. Because these nonneutralizing antibody activities have been associated with spontaneous viral control, reduced viral load, and nonprogression in infected subjects and protection in vaccinated subjects, understanding the specific features of IgGs with potentiated effector function may be critical to vaccine and therapeutic antibody development

    Global State Measures of the Dentate Gyrus Gene Expression System Predict Antidepressant-Sensitive Behaviors

    Background Selective serotonin reuptake inhibitors (SSRIs) such as fluoxetine are the most common form of medication treatment for major depression. However, approximately 50% of depressed patients fail to achieve an effective treatment response. Understanding how gene expression systems respond to treatments may be critical for understanding antidepressant resistance. Methods We take a novel approach to this problem by demonstrating that the gene expression system of the dentate gyrus responds to fluoxetine (FLX), a commonly used antidepressant medication, in a stereotyped-manner involving changes in the expression levels of thousands of genes. The aggregate behavior of this large-scale systemic response was quantified with principal components analysis (PCA) yielding a single quantitative measure of the global gene expression system state. Results Quantitative measures of system state were highly correlated with variability in levels of antidepressant-sensitive behaviors in a mouse model of depression treated with fluoxetine. Analysis of dorsal and ventral dentate samples in the same mice indicated that system state co-varied across these regions despite their reported functional differences. Aggregate measures of gene expression system state were very robust and remained unchanged when different microarray data processing algorithms were used and even when completely different sets of gene expression levels were used for their calculation. Conclusions System state measures provide a robust method to quantify and relate global gene expression system state variability to behavior and treatment. State variability also suggests that the diversity of reported changes in gene expression levels in response to treatments such as fluoxetine may represent different perspectives on unified but noisy global gene expression system state level responses. Studying regulation of gene expression systems at the state level may be useful in guiding new approaches to augmentation of traditional antidepressant treatments
