Control of somatosensory cortical processing by thalamic posterior medial nucleus: A new role of thalamus in cortical function

Less is known about paralemniscal thalamic nuclei function. In the vibrissae system, the posterior medial nucleus (POm) is the corresponding paralemniscal nucleus. POm neurons project to L1 and L5A of the primary somatosensory cortex (S1) in the rat brain. It is known that L1 modifies sensory-evoked responses through control of intracortical excitability suggesting that L1 exerts an influence on whisker responses. Therefore, thalamocortical pathways targeting L1 could modulate cortical firing. Here, using a combination of electrophysiology and pharmacology in vivo, we have sought to determine how POm influences cortical processing. In our experiments, single unit recordings performed in urethane- anesthetized rats showed that POm imposes precise control on the magnitude and duration of supra- and infragranular barrel cortex whisker responses. Our findings demonstrated that L1 inputs from POm imposed a time and intensity dependent regulation on cortical sensory processing. Moreover, we found that blocking L1 GABAergic inhibition or blocking P/Q-type Ca2+ channels in L1 prevents POm adjustment of whisker responses in the barrel cortex. Additionally, we found that POm was also controlling the sensory processing in S2 and this regulation was modulated by corticofugal activity from L5 in S1. Taken together, our data demonstrate the determinant role exerted by the POm in the adjustment of somatosensory cortical processing and in the regulation of cortical processing between S1 and S2. We propose that this adjustment could be a thalamocortical gain regulation mechanism also present in the processing of information between cortical areas.This work was supported by a grant from Ministerio de Economia y Competitividad (BFU2012-

Level TE9c of Sima del Elefante (Sierra de Atapuerca, Spain): A comprehensive approach
Level TE9c of the Sima del Elefante site (Sierra de Atapuerca, Spain) is one of the oldest sites with evidence of human occupation in western Europe. We began excavating level TE9c in 2003, and the work there continues today. The studies of the archaeology, palaeontology and geology from this locality have provided an indispensable dataset with which to capture a picture in the scenario of the origin of humans in Europe. Based on these data, we raise and discuss several topics, such as the possible origin of the lineage of the first hominins that inhabited western Europe; their capacity to have active hunting or scavenging abilities; whether their subsistence strategies were successful; and what the environment and habitats where these hominin groups settled was like. The aim of this paper is to present the results and discussions obtained from the research team and to establish the primary features of early human occupations in southwestern Europe. Tentatively, we may conclude, based on the events recorded at TE9c, that the first humans were in the Iberian peninsula at around 1.2 Ma they used the caves of the Sierra de Atapuerca as shelters probably during their hunting activities; the cavities were surrounded by Mediterranean forest, rivers and water ponds, and varied habitats as suggested by the rich and diverse assemblage of fossils of vertebrates (fish, amphibians and reptiles, birds, large and small mammals); where humans possibly caught what they found in the surroundings
Assessment of plasma chitotriosidase activity, CCL18/PARC concentration and NP-C suspicion index in the diagnosis of Niemann-Pick disease type C: A prospective observational study
Background: Niemann-Pick disease type C (NP-C) is a rare, autosomal recessive neurodegenerative disease caused by mutations in either the NPC1 or NPC2 genes. The diagnosis of NP-C remains challenging due to the non-specific, heterogeneous nature of signs/symptoms. This study assessed the utility of plasma chitotriosidase (ChT) and Chemokine (C-C motif) ligand 18 (CCL18)/pulmonary and activation-regulated chemokine (PARC) in conjunction with the NP-C suspicion index (NP-C SI) for guiding confirmatory laboratory testing in patients with suspected NP-C. Methods: In a prospective observational cohort study, incorporating a retrospective determination of NP-C SI scores, two different diagnostic approaches were applied in two separate groups of unrelated patients from 51 Spanish medical centers (n = 118 in both groups). From Jan 2010 to Apr 2012 (Period 1), patients with =2 clinical signs/symptoms of NP-C were considered ''suspected NP-C'' cases, and NPC1/NPC2 sequencing, plasma chitotriosidase (ChT), CCL18/PARC and sphingomyelinase levels were assessed. Based on findings in Period 1, plasma ChT and CCL18/PARC, and NP-C SI prediction scores were determined in a second group of patients between May 2012 and Apr 2014 (Period 2), and NPC1 and NPC2 were sequenced only in those with elevated ChT and/or elevated CCL18/PARC and/or NP-C SI =70. Filipin staining and 7-ketocholesterol (7-KC) measurements were performed in all patients with NP-C gene mutations, where possible. Results: In total across Periods 1 and 2, 10/236 (4%) patients had a confirmed diagnosis o NP-C based on gene sequencing (5/118 4.2%] in each Period): all of these patients had two causal NPC1 mutations. Single mutant NPC1 alleles were detected in 8/236 (3%) patients, overall. Positive filipin staining results comprised three classical and five variant biochemical phenotypes. No NPC2 mutations were detected. All patients with NPC1 mutations had high ChT activity, high CCL18/PARC concentrations and/or NP-C SI scores =70. Plasma 7-KC was higher than control cut-off values in all patients with two NPC1 mutations, and in the majority of patients with single mutations. Family studies identified three further NP-C patients. Conclusion: This approach may be very useful for laboratories that do not have mass spectrometry facilities and therefore, they cannot use other NP-C biomarkers for diagnosis

Gaia Focused Product Release: Radial velocity time series of long-period variables
The third Gaia Data Release (DR3) provided photometric time series of more
than 2 million long-period variable (LPV) candidates. Anticipating the
publication of full radial-velocity (RV) in DR4, this Focused Product Release
(FPR) provides RV time series for a selection of LPVs with high-quality
observations. We describe the production and content of the Gaia catalog of LPV
RV time series, and the methods used to compute variability parameters
published in the Gaia FPR. Starting from the DR3 LPVs catalog, we applied
filters to construct a sample of sources with high-quality RV measurements. We
modeled their RV and photometric time series to derive their periods and
amplitudes, and further refined the sample by requiring compatibility between
the RV period and at least one of the , , or
photometric periods. The catalog includes RV time series and variability
parameters for 9\,614 sources in the magnitude range , including a flagged top-quality subsample of 6\,093 stars
whose RV periods are fully compatible with the values derived from the ,
, and photometric time series. The RV time series
contain a mean of 24 measurements per source taken unevenly over a duration of
about three years. We identify the great most sources (88%) as genuine LPVs,
with about half of them showing a pulsation period and the other half
displaying a long secondary period. The remaining 12% consists of candidate
ellipsoidal binaries. Quality checks against RVs available in the literature
show excellent agreement. We provide illustrative examples and cautionary
remarks. The publication of RV time series for almost 10\,000 LPVs constitutes,
by far, the largest such database available to date in the literature. The
availability of simultaneous photometric measurements gives a unique added

Gaia Early Data Release 3: The celestial reference frame (Gaia-CRF3)
Context. Gaia-CRF3 is the celestial reference frame for positions and proper motions in the third release of data from the Gaia mission, Gaia DR3 (and for the early third release, Gaia EDR3, which contains identical astrometric results). The reference frame is defined by the positions and proper motions at epoch 2016.0 for a specific set of extragalactic sources in the (E)DR3 catalogue. Aims. We describe the construction of Gaia-CRF3 and its properties in terms of the distributions in magnitude, colour, and astrometric quality. Methods. Compact extragalactic sources in Gaia DR3 were identified by positional cross-matching with 17 external catalogues of quasi-stellar objects (QSO) and active galactic nuclei (AGN), followed by astrometric filtering designed to remove stellar contaminants. Selecting a clean sample was favoured over including a higher number of extragalactic sources. For the final sample, the random and systematic errors in the proper motions are analysed, as well as the radio-optical offsets in position for sources in the third realisation of the International Celestial Reference Frame (ICRF3). Results. Gaia-CRF3 comprises about 1.6 million QSO-like sources, of which 1.2 million have five-parameter astrometric solutions in Gaia DR3 and 0.4 million have six-parameter solutions. The sources span the magnitude range G = 13-21 with a peak density at 20.6 mag, at which the typical positional uncertainty is about 1 mas. The proper motions show systematic errors on the level of 12 ÎŒas yr-1 on angular scales greater than 15 deg. For the 3142 optical counterparts of ICRF3 sources in the S/X frequency bands, the median offset from the radio positions is about 0.5 mas, but it exceeds 4 mas in either coordinate for 127 sources. We outline the future of Gaia-CRF in the next Gaia data releases. Appendices give further details on the external catalogues used, how to extract information about the Gaia-CRF3 sources, potential (Galactic) confusion sources, and the estimation of the spin and orientation of an astrometric solution

Gaia Data Release 3: Mapping the asymmetric disc of the Milky Way
With the most recent Gaia data release the number of sources with complete 6D
phase space information (position and velocity) has increased to well over 33
million stars, while stellar astrophysical parameters are provided for more
than 470 million sources, in addition to the identification of over 11 million
variable stars. Using the astrophysical parameters and variability
classifications provided in Gaia DR3, we select various stellar populations to
explore and identify non-axisymmetric features in the disc of the Milky Way in
both configuration and velocity space. Using more about 580 thousand sources
identified as hot OB stars, together with 988 known open clusters younger than
100 million years, we map the spiral structure associated with star formation
4-5 kpc from the Sun. We select over 2800 Classical Cepheids younger than 200
million years, which show spiral features extending as far as 10 kpc from the
Sun in the outer disc. We also identify more than 8.7 million sources on the
red giant branch (RGB), of which 5.7 million have line-of-sight velocities,
allowing the velocity field of the Milky Way to be mapped as far as 8 kpc from
the Sun, including the inner disc. The spiral structure revealed by the young
populations is consistent with recent results using Gaia EDR3 astrometry and
source lists based on near infrared photometry, showing the Local (Orion) arm
to be at least 8 kpc long, and an outer arm consistent with what is seen in HI
surveys, which seems to be a continuation of the Perseus arm into the third
quadrant. Meanwhile, the subset of RGB stars with velocities clearly reveals
the large scale kinematic signature of the bar in the inner disc, as well as
evidence of streaming motions in the outer disc that might be associated with
spiral arms or bar resonances. (abridged
Identifying associations between diabetes and acute respiratory distress syndrome in patients with acute hypoxemic respiratory failure: an analysis of the LUNG SAFE database
Background: Diabetes mellitus is a common co-existing disease in the critically ill. Diabetes mellitus may reduce the risk of acute respiratory distress syndrome (ARDS), but data from previous studies are conflicting. The objective of this study was to evaluate associations between pre-existing diabetes mellitus and ARDS in critically ill patients with acute hypoxemic respiratory failure (AHRF). Methods: An ancillary analysis of a global, multi-centre prospective observational study (LUNG SAFE) was undertaken. LUNG SAFE evaluated all patients admitted to an intensive care unit (ICU) over a 4-week period, that required mechanical ventilation and met AHRF criteria. Patients who had their AHRF fully explained by cardiac failure were excluded. Important clinical characteristics were included in a stepwise selection approach (forward and backward selection combined with a significance level of 0.05) to identify a set of independent variables associated with having ARDS at any time, developing ARDS (defined as ARDS occurring after day 2 from meeting AHRF criteria) and with hospital mortality. Furthermore, propensity score analysis was undertaken to account for the differences in baseline characteristics between patients with and without diabetes mellitus, and the association between diabetes mellitus and outcomes of interest was assessed on matched samples. Results: Of the 4107 patients with AHRF included in this study, 3022 (73.6%) patients fulfilled ARDS criteria at admission or developed ARDS during their ICU stay. Diabetes mellitus was a pre-existing co-morbidity in 913 patients (22.2% of patients with AHRF). In multivariable analysis, there was no association between diabetes mellitus and having ARDS (OR 0.93 (0.78-1.11); p = 0.39), developing ARDS late (OR 0.79 (0.54-1.15); p = 0.22), or hospital mortality in patients with ARDS (1.15 (0.93-1.42); p = 0.19). In a matched sample of patients, there was no association between diabetes mellitus and outcomes of interest. Conclusions: In a large, global observational study of patients with AHRF, no association was found between diabetes mellitus and having ARDS, developing ARDS, or outcomes from ARDS.
