Formability of the 5754-aluminum alloy deformed by a modified repetitive corrugation and straightening process
Sheets of 5754-aluminum alloy processed by a modified repetitive corrugation and straightening (RCS) process were tested in order to measure their formability. For this purpose, forming limit curves were derived. They showed that the material forming capacity decreased after being processed by RCS. However, they kept good formability in the initial stages of the RCS process. The formability study was complemented with microstructural analysis (derivation of texture) and mechanical tests to obtain the strain-rate sensitivity. The texture analysis was done by employing X-ray diffraction, obtaining pole figures, and the orientation distribution function. It was noticed that the initial texture was conserved after successive RCS passes, but the intensity dropped. RCS process did not induce ß-fiber, contrary to common deformation process. The strain-rate sensitivity coefficient was measured through tensile tests at different temperatures and strain rates; the coefficient of the samples processed after one and two passes were still relatively high, indicating the capacity to delay necking, in agreement with the good formability observed in the initial passes of the RCS processPeer ReviewedPostprint (published version
Chitosan-mediated shRNA knockdown of cytosolic alanine aminotransferase improves hepatic carbohydrate metabolism
Alanine aminotransferase (ALT) catalyses a transamination reaction that links carbohydrate and amino acid metabolism. In this study, we examined the effect of silencing cytosolic ALT (cALT) expression on the hepatic metabolism in Sparus aurata. A number of siRNA and shRNA designed to down-regulate cALT expression were validated in HEK-293 cells transfected with plasmids expressing S. aurata cALT or mitochondrial ALT (mALT) isoforms: ALT silencing significantly decreased the expression levels of S. aurata mRNA cALT1 to 62 % (siRNA) and 48 % (shRNA) of the values observed in control cells. The effect of cALT silencing was analysed in the liver of S. aurata 72 h after intraperitoneal injection of chitosan-tripolyphosphate (TPP) nanoparticles complexed with a plasmid encoding a shRNA to down-regulate cALT expression (pCpG-si1sh1). In fish fed diets with different ratio of protein to carbohydrate and treated with chitosan-TPP-pCpG-si1sh1, cALT1 and cALT2 mRNA levels significantly decreased irrespective of the diet. Consistently, ALT activity decreased in liver of treated animals. In the liver of S. aurata treated with chitosan-TPP-pCpG-si1sh1 nanoparticles, down-regulation of cALT expression increased the activity of key enzymes in glycolysis (6-phosphofructo-1-kinase and pyruvate kinase) and protein metabolism (glutamate dehydrogenase). Besides showing for the first time that administration of chitosan-TPP-pCpG-si1sh1 nanoparticles silences hepatic cALT expression in vivo, our data support that down-regulation of cALT could improve the use of dietary carbohydrates to obtain energy and spare protein catabolis
Green tea catechin inhibits fatty acid synthase without stimulating carnitine Palmitoyltransferase-1 or inducing weight loss in experimental animals
Background: The enzyme fatty acid synthase (FASN) is highly expressed in many human carcinomas and its inhibition is cytotoxic to human cancer cells. The use of FASN inhibitors has been limited until now by anorexia and weight loss, which is associated with the stimulation of fatty acid oxidation. Materials and Methods: The in vitro effect of (-)-epigallocatechin-3-gallate (EGCG) on fatty acid metabolism enzymes, on apoptosis and on cell signalling was evaluated. In vivo, the effect of EGCG on animal body weight was addressed. Results: EGCG inhibited FASN activity, induced apoptosis and caused a marked decrease of human epidermal growth factor receptor 2 (HER2), phosphatidylinositol 3-kinase (PI3K)/AKT and extracellular (signal)-regulated kinase (ERK) 1/2 proteins, in breast cancer cells. EGCG did not induce a stimulatory effect on CPT-1 activity in vitro (84% of control), or on animal body weight in vivo (99% of control). Conclusion: EGCG is a FASN inhibitor with anticancer activity which does not exhibit cross-activation of fatty acid oxidation and does not induce weight loss, suggesting its potential use as an anticancer drug
Microstructural evolution and mechanical behavior of an Al-6061 alloy processed by repetitive corrugation and straightening
The repetitive corrugation and straightening process is a severe plastic deformation technique that is particularly suited to process metallic sheets. With this technique, it is possible to develop nano/ultrafine-grained structured materials, and therefore, to improve some mechanical properties such as the yield strength, ultimate tensile strength, and fatigue lifetime. In this study, an Al-6061 alloy was subjected to the repetitive corrugation and straightening process. A new corrugation die design was proposed in order to promote a heterogeneous deformation into the metallic sheet. The evolution of the mechanical properties and microstructure obtained by electron backscatter diffraction of the alloy showed a heterogeneous distribution in the grain size at the initial cycles of the repetitive corrugation and straightening process. Uniaxial tensile tests showed a significant increase in yield strength as the number of repetitive corrugation and straightening passes increased. The distribution of the plastic deformation was correlated with the hardness distribution on the surface. The hardness distribution map matched well with the heterogeneous distribution of the plastic deformation obtained by finite element simulation. Mechanical, stress corrosion cracking and crystallographic study on flat components processed by two combined severe plastic deformation techniques
Although the current field of application of Al–alloy 7075 (AA7075) is vast, it is still limited due to some drawbacks, especially due to its susceptibility to stress corrosion cracking (SCC). This work aims to evaluate the microstructural, mechanical, and stress corrosion cracking (SCC) behaviors on an AA7075 in flat format deformed by a combination of repetitive corrugation and straightening (RCS) and accumulative roll bonding (ARB) techniques. Four different deformation routes were applied, namely: ARB (A), RCS (R), RCS + ARB (RA) and ARB + RCS (AR). As expected, the efficiency for grain refinement depends on the applied route, in terms of average grain size regarding the initial condition IC): AR > A > RA > R. All conditions resulted in unimodal and widened grain size distributions of micro-, submicro- and nano-metric dimensions. The study of crystallographic orientations showed that route R did not generate any new texture, whereas different preferred orientations were obtained for routes A, RA, and AR. The hardness and three-point bending tests showed an improvement of mechanical strength in the following order: AR > RA > A > R. The cracks per cm2 obtained in the corrosion study indicated that the best SCC resistance was R > A > AR > RA. Based on the above, the best combination of microstructural, mechanical, and SCC properties until one deformation pass was obtained by the single route of the ARB process.Peer ReviewedPostprint (published version
Repetitive corrugation and straightening effect on the microstructure, crystallographic texture and electrochemical behavior for the Al-7075 alloy
Anti-corrosion susceptibility is one of the top criteria for selecting metallic materials for several industrial applications. This work studies the corrosion performance on an Al-7075 alloy obtained by Repetitive Corrugation and Straightening (RCS). This processing method generated a microstructure formed by randomly distributed micro-, submicro-, and nano- metric grain sizes. The samples exhibited a drop in corrosion resistance for a longer duration in the electrolyte and higher deformation. However, the samples processed by RCS showed better electrochemical stability in comparison with the non-deformed condition. The improvement of electrochemical stability could be associated with the particular microstructure generated during the RCS process.Postprint (published version
Clasificación de eritrocitos empleando modelos ocultos de Márkov
Se realiza un estudio del desempeño de los modelos ocultos de Márkov (HMM) en la clasificación morfológica supervisada de eritrocitos en muestras de sangre periférica de pacientes con anemia drepanocítica. Los contornos se representan de forma novedosa considerando las diferencias angulares en la curvatura de los puntos del mismo. El entrenamiento de cada modelo se realiza tanto con la descripción normal de los contornos como con la representación de la rotación de los mismos, para garantizar una mayor estabilidad en los parámetros estimados. Se desarrolla un proceso de validación cruzada de 5x1 para estimación del error. Se obtienen las medidas de sensibilidad, precisión y especificidad de la clasificación. Los mejores resultados en cuanto a sensibilidad se obtienen al clasificar eritrocitos pertenecientes a dos clases: normales (96%) y elongados (99%). Al considerar además una clase de eritrocitos con otras deformaciones los mejores resultados se obtienen realizando el entrenamiento de los modelos con la rotación de todos los contornos, que alcanzó sensibilidades de normales (94%), elongados (82%) y con otras deformaciones (76%). FGF21 mediates the lipid metabolism response to amino acid starvation
Abstract Lipogenic gene expression in liver is repressed in mice upon leucine deprivation. The hormone fi broblast growth factor 21 (FGF21), which is critical to the adaptive metabolic response to starvation, is also induced under amino acid deprivation. Upon leucine deprivation, we found that FGF21 is needed to repress expression of lipogenic genes in liver and white adipose tissue, and stimulate phosphorylation of hormone-sensitive lipase in white adipose tissue. The increased expression of Ucp1 in brown adipose tissue under these circumstances is also impaired in FGF21- defi cient mice. Gene markers of dietary macronutrient composition and growth in the skeletal muscle of gilthead sea bream (Sparus aurata)
To increase our current knowledge on the nutritional regulation of growth and gene expression pattern in fish skeletal muscle, the effect of dietary macronutrient composition was assessed on digestibility, nutrient retention, growth performance, and the mRNA levels of key genes involved in functionality, growth and development of the skeletal muscle in gilthead sea bream (Sparus aurata). Long-term starvation decreased the expression of myogenic regulatory factors such as Myod2, Myf5, myogenin (Myog) and Myf6 in the skeletal muscle of S. aurata. The supply of high or medium protein, low carbohydrate diets enhanced growth parameters, feed efficiency ratio, feed conversion ratio and significantly upregulated myod2. However, the supply of low protein, high carbohydrate diets restricted growth and stimulated the mRNA levels of myostatin, while downregulated follistatin (fst), igf1, mtor and rps6. Microarray analysis revealed igfals, tnni2, and gadd45a as gene markers upregulated by diets enriched with protein, lipids and carbohydrates, respectively. The results of the present study show that in addition to myod2, fst, igf1, mtor and rps6, the expression levels of igfals, tnni2 and remarkably gadd45a in the skeletal muscle can be used as markers to evaluate the effect of dietary macronutrient changes on fish growth and muscle development in S. aurata
