    The Effect of 17α-Ethynilestradiol and GPER1 Activation on Body and Muscle Growth, Muscle Composition and Growth-Related Gene Expression of Gilthead Seabream, Sparus aurata L

    Endocrine-disrupting chemicals include natural and synthetic estrogens, such as 17α- ethynilestradiol (EE2 ), which can affect reproduction, growth and immunity. Estrogen signalling is mediated by nuclear or membrane estrogen receptors, such as the new G-protein-coupled estrogen receptor 1 (GPER1). The present work studies the effect of EE2 and G1 (an agonist of GPER1) on body and muscle parameters and growth-related genes of 54 two-year-old seabreams. The fish were fed a diet containing EE2 (EE2 group) and G1 (G1 group) for 45 days and then a diet without EE2 or G1 for 122 days. An untreated control group was also studied. At 45 days, the shortest body length was observed in the G1 group, while 79 and 122 days after the cessation of treatments, the shortest body growth was observed in the EE2 group. Hypertrophy of white fibers was higher in the EE2 and G1 groups than it was in the control group, whereas the opposite was the case with respect to hy- perplasia. Textural hardness showed a negative correlation with the size of white fibers. At the end of the experiment, all fish analyzed in the EE2 group showed a predominance of the gonadal ovarian area. In addition, the highest expression of the mafbx gene (upregulated in catabolic signals) and mstn2 (myogenesis negative regulator) was found in EE2 -exposed fish.Versión del editor2,46

    Tamoxifen disrupts the reproductive process in gilthead seabream males and modulates the effects promoted by 17alpha-ethynylestradiol

    17α-ethynylestradiol (EE2), which is used in oral contraceptives and hormone replacement therapy, is a well documented estrogenic endocrine disruptor and an aquatic contaminant. In the present study, adult male specimens of the marine hermaphrodite teleost gilthead (Sparus aurata L.) were fed a diet containing tamoxifen (Tmx), an estrogen receptor ligand used in cancer therapy, alone or combined with EE2, for 25 days and then fed a commercial diet for a further 25 days (recovery period). The effects of short (5 days) and long (25 days) treatments on several reproductive and gonad immune parameters and the reversibility of the disruptive effects after the recovery period were examined. Our data showed that Tmx acted as an estrogenic endocrine disruptor as revealed by the increase in the hepatic transcription of the vitellogenin gene in males, the serum levels of 17β-estradiol and the gonad expression levels of the estrogen receptor α and G protein-coupled estrogen receptor genes, and the recruitment of leukocytes into the gonad, a well known estrogenic-dependent process in gilthead seabream males. On the other hand, Tmx also increased sperm concentration and motility as well as the serum levels of androgens and the expression levels of genes that codify for androgenic enzymes, while decreasing the expression levels of the gene that code for gonadal aromatase. When applied simultaneously, Tmx and EE2 could act in synergy or counteract, each other, depending on the parameter measured.     Exposure to 17α-ethynylestradiol (EE2, 5 μg/g food) impairs some reproductive events in the protandrous gilthead seabream and a short recovery period does not allow full recovery. In this study, spermiating seabream males in the second reproductive cycle (RC) were fed a diet containing 5 or 2.5 μg EE2/g food for 28 days and then a commercial diet without EE2 for the remaining RC. Individuals were sampled at the end of the EE2 treatment and then at the end of the RC and at the beginning of the third RC, 146 and 333 days after the cessation of treatment, respectively. Increased hepatic transcript levels of the gene coding for vitellogenin (vtg) and plasma levels of Vtg indicated both concentrations of EE2 caused endocrine disruption. Modifications in the histological organization of the testis, germ cell proliferation, plasma levels of the sex steroids and pituitary expression levels of the genes coding for the gonadotropin β-subunits, fshβ and lhβ were detected. The plasma levels of Vtg and most of the reproductive parameters were restored 146 days after treatments.     17α-ethynilestradiol (EE2) and tamoxifen (Tmx) are pollutants world-wide distributed in aquatic environments. Gilthead seabream, Sparus aurata L., is highlighted as a species model of intensively culture in anthropogenic disturbed environments. The effects of these pollutants on gilthead seabream reproduction and some immune responses have been described but, the humoral innate antimicrobial activities have never received attention. In this work we analysed the latest in the plasma of gilthead seabream males of different ages and reproductive stages treated with 0, 2.5, 5 or 50 μg EE2 or 100 μg Tmx g−1 food during different times of exposure and of reverting to commercial diet (recovery). The peroxidase and protease activities decreased as the spermatogenesis of the first reproductive cycle (RC) proceeded in control fish. However, only protease and antiprotease activities showed different level at different stages of the second RC in control fish, but showed scarce disruption in fish treated with EE2 or Tmx. Peroxidase and bactericide activities are more sensitive to EE2, than to Tmx. The effects induced by EE2 varied depending on the activity analyzed, the dose and the time of exposure and the reproductive stage and the age of the specimens.Versión del editor2,92

    Cannabinoid receptor CB2 ablation protects against TAU induced neurodegeneration.

    Tauopathies are a group of neurodegenerative diseases characterized by the alteration/aggregation of TAU protein, for which there is still no effective treatment. Therefore, new pharmacological targets are being sought, such as elements of the endocannabinoid system (ECS). We analysed the occurrence of changes in the ECS in tauopathies and their implication in the pathogenesis. By integrating gene expression analysis, immunofluorescence, genetic and adeno-associated virus expressing TAU mouse models, we found a TAU-dependent increase in CB2 receptor expression in hippocampal neurons, that occurs as an early event in the pathology and was maintained until late stages. These changes were accompanied by alterations in the endocannabinoid metabolism. Remarkably, CB2 ablation in mice protects from neurodegeneration induced by hTAU P301L overexpression, corroborated at the level of cognitive behaviour, synaptic plasticity, and aggregates of insoluble TAU. At the level of neuroinflammation, the absence of CB2 did not produce significant changes in concordance with a possible neuronal location rather than its classic glial expression in these models. These findings were corroborated in post-mortem samples of patients with Alzheimer’s disease, the most common tauopathy. Our results show that neurons with accumulated TAU induce the expression of the CB2 receptor, which enhances neurodegeneration.     Biofortification with magnesium nanofertilizer on bioactive compounds and antioxidant capacity in green beans

    The use of nanofertilizers has the potential to be used to enrich edible organs with nutrients (biofortification) and improve the biosynthesis of bioactive compounds and their antioxidant capacity. Therefore, the objective of this study was to evaluate the effect of biofortification with magnesium (Mg) nanofertilizer on the accumulation of bioactive compounds and antioxidant capacity in green bean cv. Strike compared to a conventional fertilizer (Mg sulfate). Two sources of Mg were applied via foliar: Nanofertilizer and Mg Sulfate at doses of 0, 50, 100, 200, and 300 mg/L of Mg. The accumulation of total polyphenols, flavonoids, anthocyanins, bioactive compounds, and antioxidant capacity was evaluated in pods. The results obtained in this research confirm the effect of green bean pods biofortified with Mg nanofertilizers on the production and accumulation of bioactive compounds and antioxidant capacity, improving the nutrition and nutraceutical quality of green beans. The 50 mg/L dose of Mg nanofertilizer was the most effective treatment to increase bioactive compounds and antioxidant capacity compared to high doses of Mg sulfate (300 mg/L).     Catalog for the ESPRESSO blind radial velocity exoplanet survey

    One of the main scientific drivers for ESPRESSO,\'Echelle SPectrograph, is the detection and characterization of Earth-class exoplanets. With this goal in mind, the ESPRESSO Guaranteed Time Observations (GTO) Catalog identifies the best target stars for a blind search for the radial velocity (RV) signals caused by Earth-class exoplanets. Using the most complete stellar catalogs available, we screened for the most suitable G, K, and M dwarf stars for the detection of Earth-class exoplanets with ESPRESSO. For most of the stars, we then gathered high-resolution spectra from new observations or from archival data. We used these spectra to spectroscopically investigate the existence of any stellar binaries, both bound or background stars. We derived the activity level using chromospheric activity indexes using log(RHK)log(R'_{HK}), as well as the projected rotational velocity v sin i\textit{v sin i}. For the cases where planet companions are already known, we also looked at the possibility that additional planets may exist in the host's habitable zone using dynamical arguments. We estimated the spectroscopic contamination level, v sin i\textit{v sin i}, activity, stellar parameters and chemical abundances for 249 of the most promising targets. Using these data, we selected 45 stars that match our criteria for detectability of a planet like Earth. The stars presented and discussed in this paper constitute the ESPRESSO GTO catalog for the RV blind search for Earth-class planets. They can also be used for any other work requiring a detailed spectroscopic characterization of stars in the solar neighborhood.Comment: Corrected a typo in references. Corrected typo in table B.
