    A Comparison of Salinity Effects from Hurricanes Dolly (2008) and Alex (2010) in a Texas Lagoon System

    A comparison of salinity effects from hurricanes Dolly (2008) and Alex (2010) in a Texas lagoon system.Hurricanes are not uncommon along the Gulf of Mexico coast, but there are few studies of the effects they have on coastal embayments. Hurricanes Dolly (2008) and Alex (2010) were both Category 2 storms affecting the Lower Laguna Madre (LLM) of Texas. Surveys were performed to assess poststorm water quality after landfall of both storms at up to 18 sample stations. The main difference between storm effects was salinity reduction because of stormwater input from the watershed. Effects from Hurricane Dolly were of short duration and small magnitude, whereas the effects from Hurricane Alex were extensive and lasted more than a month. Differences in spatial patterns in salinity were significantly more pronounced across the LLM than were temporal differences. Precipitation of 50–100 cm caused stormwater discharge to exceed 1000 m s−1 to the LLM during the Alex event and depressed salinity over more than three-fourths (ca. 500 km2) of the estuary for 2 months. Storm-related effects on water-column physiochemistry were persistently lowest near freshwater drains (Arroyo Colorado). Salinity remained less than 5 for more than 2 months during the Alex freshet. Freshwater input from Hurricane Dolly was relatively minor because the storm precipitation was largely restricted to the small Arroyo Colorado watershed. Effects from Alex were delayed but were greater because of the bulk of the precipitation falling in the Rio Grande/Rio Bravo drainage basin in México. The greatest impact from that freshwater disturbance was the loss of seagrasses after prolonged exposure to hyposalinity. Hurricanes Dolly and Alex both affected the LLM but with contrasting impacts that reflected spatial and meteorological differences between the two storms

    Measurement of Intracellular Fluorescence of Human Monocytes Relative to Oxidative Metabolism

    Human monocytes (MN) produce O2â and H2O2 when stimulated by agonists. Dichlorofluorescin diacetate (DCFHâ DA) has been used as a substrate for measuring intracellular oxidant production in neutrophils. DCFHâ DA is hydrolyzed by esterases to dichlorofluorescin (DCFH), which is trapped within the cell. This nonfluorescent molecule is then oxidized to fluorescent dichlorofluorescin (DCF) by action of cellular oxidants. DCFHâ DA can not be appreciably oxidized to a fluorescent state without prior hydrolysis. We have examined the utility of DCFHâ DA for the assessment of monocyte oxidative responses. The levels of intracellular fluorescence measured by flow cytometry were considerably less than expected from reported levels of O2â â production or chemiluminescence assays. Compared with neutrophils, monocytes produced minimal increases in DCF fluorescence after stimulation with phorbol myristate acetate as measured by flow cytometry, but both cell types showed increases in fluorescence when bulk cell suspensions were measured by spectrofluorometry. To determine the intracellular location of the DCFH, bulk fluorescence measurements were made on both whole and sonicated cell preparations. When intact mononuclear cells were preloaded with DCFHâ DA, then sonicated and oxidized with added excess H2O2, the increase in fluorescence was only 30% of the fluorescence of mononuclear cell sonicates to which DCFHâ DA was added and oxidized in a similar manner. These results suggest that a portion of the DCFHâ DA incorporated by intact cells, is not susceptible to oxidation by the added H2O2. Addition of NaOH to induce hydrolysis of any residual DCFHâ DA in the sonicates of DCFHâ DAâ loaded intact mononuclear cells resulted in a further increase in fluorescence upon addition of H2O2, suggesting that a significant portion of the DCFHâ DA was not hydrolyzed despite ample uptake of this dye by these cells. In contrast, no further increase in fluorescence was observed in sonicates of DCFHâ DAâ loaded intact neutrophils, suggesting complete hydrolysis of all incorporated DCFHâ DA to DCFH. When monocytes were allowed to phagocytose DCFHâ DAâ loaded Staphylococcus aureus, intracellular fluorescence was measurable by flow cytometry, indicating intracellular oxidation of the fluorochromes. We therefore propose that in monocytes the mechanism of intracellular processing of these fluorochromes differs from that in neutrophils owing to differences in intracellular localization of fluorochromes, site of oxidant production, and/or accessibility of the DCFHâ DA to esterolysis.Peer Reviewedhttps://deepblue.lib.umich.edu/bitstream/2027.42/141433/1/jlb0304.pd

    Services for Adults with an Autism Spectrum Disorder

    Objective: The need for useful evidence about services is increasing as larger numbers of children identified with an autism spectrum disorder age toward adulthood. The objective of this review was to characterize the topical and methodological aspects of research on services for supporting success in work, education, and social participation among adults with an autism spectrum disorder and to propose recommendations for moving this area of research forward. Method: Review of literature published in English from 2000 to 2010.Results: We found that the evidence base about services for adults with an ASD is underdeveloped and can be considered a field of inquiry that is relatively unformed. Extant research does not reflect the demographic or impairment heterogeneity of the population, the range of services that adults with autism require in order to function with purposeful lives in the community, and the need for coordination across service systems and sectors. Conclusions: Future studies must examine issues related to cost and efficiency given the broader sociopolitical and economic context of service provision. Furthermore, future research needs to consider how demographic and impairment heterogeneity have implications for building an evidence base that will have greater external validity

    Gene expression studies of developing bovine longissimus muscle from two different beef cattle breeds

    Background: The muscle fiber number and fiber composition of muscle is largely determined during prenatal development. In order to discover genes that are involved in determining adult muscle phenotypes, we studied the gene expression profile of developing fetal bovine longissimus muscle from animals with two different genetic backgrounds using a bovine cDNA microarray. Fetal longissimus muscle was sampled at 4 stages of myogenesis and muscle maturation: primary myogenesis (d 60), secondary myogenesis (d 135), as well as beginning (d 195) and final stages (birth) of functional differentiation of muscle fibers. All fetuses and newborns (total n = 24) were from Hereford dams and crossed with either Wagyu (high intramuscular fat) or Piedmontese (GDF8 mutant) sires, genotypes that vary markedly in muscle and compositional characteristics later in postnatal life. Results: We obtained expression profiles of three individuals for each time point and genotype to allow comparisons across time and between sire breeds. Quantitative reverse transcription-PCR analysis of RNA from developing longissimus muscle was able to validate the differential expression patterns observed for a selection of differentially expressed genes, with one exception. We detected large-scale changes in temporal gene expression between the four developmental stages in genes coding for extracellular matrix and for muscle fiber structural and metabolic proteins. FSTL1 and IGFBP5 were two genes implicated in growth and differentiation that showed developmentally regulated expression levels in fetal muscle. An abundantly expressed gene with no functional annotation was found to be developmentally regulated in the same manner as muscle structural proteins. We also observed differences in gene expression profiles between the two different sire breeds. Wagyu-sired calves showed higher expression of fatty acid binding protein 5 (FABP5) RNA at birth. The developing longissimus muscle of fetuses carrying the Piedmontese mutation shows an emphasis on glycolytic muscle biochemistry and a large-scale up-regulation of the translational machinery at birth. We also document evidence for timing differences in differentiation events between the two breeds. Conclusion: Taken together, these findings provide a detailed description of molecular events accompanying skeletal muscle differentiation in the bovine, as well as gene expression differences that may underpin the phenotype differences between the two breeds. In addition, this study has highlighted a non-coding RNA, which is abundantly expressed and developmentally regulated in bovine fetal muscle

    Density of states in d-wave superconductors disordered by extended impurities

    The low-energy quasiparticle states of a disordered d-wave superconductor are investigated theoretically. A class of such states, formed via tunneling between the Andreev bound states that are localized around extended impurities (and result from scattering between pair-potential lobes that differ in sign) is identified. Its (divergent) contribution to the total density of states is determined by taking advantage of connections with certain one-dimensional random tight-binding models. The states under discussion should be distinguished from those associated with nodes in the pair potential.Comment: 5 pages, 1 figur

    Characterization of the near-Earth Asteroid 2002NY40

    In August 2002, the near-Earth asteroid 2002 NY40, made its closest approach to the Earth. This provided an opportunity to study a near-Earth asteroid with a variety of instruments. Several of the telescopes at the Maui Space Surveillance System were trained at the asteroid and collected adaptive optics images, photometry and spectroscopy. Analysis of the imagery reveals the asteroid is triangular shaped with significant self-shadowing. The photometry reveals a 20-hour period and the spectroscopy shows that the asteroid is a Q-type

    Low-energy quasiparticle states, arising from scattering by single-particle potentials in d-wave superconductors, are addressed. Via a natural extension of the Andreev approximation, the idea that sign-variations in the superconducting pair-potential lead to such states is extended beyond its original setting of boundary scattering to the broader context of scattering by general single-particle potentials, such as those due to impurities. The index-theoretic origin of these states is exhibited via a simple connection with Witten's supersymmetric quantum-mechanical model.Comment: 5 page

    Pneumonia in Bighorn Sheep: Testing the Super-Spreader Hypothesis

    Following introduction of pneumonia, disease can persist in bighorn sheep (Ovis canadensis) populations for decades as annual or sporadic pneumonia epidemics in lambs.  Recurring years of depressed recruitment due to high rates of pneumonia-induced mortality in juveniles is a major obstacle to population recovery.  Management strategies for resolving this problem have so far been elusive. We are investigating the feasibility of removing individual “super-spreaders” to improve lamb survival.  Individual variation in infection and transmission is well documented in human diseases (e.g. “Typhoid Mary”).  We are testing the hypothesis that pneumonia epidemics in lambs are initiated by transmission of pathogens from a few “chronic-shedder” ewes. We have completed the first year of a 5-year project in the Hells Canyon region of Idaho, Oregon, and Washington, and in a captive population at South Dakota State University. Through repeated testing of free-ranging individuals in Hells Canyon, we have identified individual differences in shedding of Mycoplasma ovipneumoniae, a primary pathogen in the bighorn sheep respiratory disease complex.  We also found that when penned separately in captivity, lambs of ewes that consistently tested positive (chronic shedders) were infected and died of pneumonia, whereas lambs born to ewes from an infected population that tested negative (non-shedders), were not infected and survived.  Over the next 4 years we plan to 1) continue and expand testing of free-ranging and captive animals, 2) determine whether removal of chronic-shedder ewes improves lamb survival in free-ranging populations, 3) expand and replicate chronic-shedder commingling experiments in captivity, and 4) establish and monitor a new population founded with non-shedders from an infected population

    Multichannel quantum-defect theory for ultracold atom-ion collisions

    We develop an analytical model for ultracold atom-ion collisions using the multichannel quantum-defect formalism. The model is based on the analytical solutions of the r^-4 long-range potential and on the application of a frame transformation between asymptotic and molecular bases. This approach allows the description of the atom-ion interaction in the ultracold domain in terms of three parameters only: the singlet and triplet scattering lengths, assumed to be independent of the relative motion angular momentum, and the lead dispersion coefficient of the asymptotic potential. We also introduce corrections to the scattering lengths that improve the accuracy of our quantum-defect model for higher order partial waves, a particularly important result for an accurate description of shape and Feshbach resonances at finite temperature. The theory is applied to the system composed of a 40Ca+ ion and a Na atom, and compared to numerical coupled-channel calculations carried out using ab initio potentials. For this particular system, we investigate the spectrum of bound states, the rate of charge-transfer processes, and the collision rates in the presence of magnetic Feshbach resonances at zero and finite temperature.Comment: 39 pages, 21 figure
