    In vivo and in situ rhizosphere respiration in Acer saccharum and Betula alleghaniensis seedlings grown in contrasting light regimes

    A perfusive method combined with an open-system carbon dioxide measurement system was used to assess rhizosphere respiration of Acer saccharum Marsh. (sugar maple) and Betula alleghaniensis Britton (yellow birch) seedlings grown in 8-1 pots filled with coarse sand. We compared in vivo and in situ rhizosphere respiration between species, among light regimes (40, 17 and 6% of full daylight) and at different times during the day. To compute specific rhizosphere respiration, temperature corrections were made with either species-specific coefficients (Q10) based on the observed change in respiration rate between 15 and 21°C or an arbitrarily assigned Q10 of 2. Estimated, species-specific Q10 values were 3.0 and 3.4 for A. saccharum and B. alleghaniensis, respectively, and did not vary with light regime. Using either method of temperature correction, specific rhizosphere respiration did not differ either between A. saccharum and B. alleghaniensis, or among light regimes except in A. saccharum at 6% of full daylight. At this irradiance, seedlings were smaller than in the other light treatments, with a larger fine root fraction of total root dry mass, resulting in higher respiration rates. Specific rhizosphere respiration was significantly higher during the afternoon than at other times of day when temperature-corrected on the basis of an arbitrary Q10 of 2, suggesting the possibility of diurnal variation in a temperature-independent component of rhizosphere respiration

    Comparative kinetic analysis of two fungal β-glucosidases

    <p>Abstract</p> <p>Background</p> <p>The enzymatic hydrolysis of cellulose is still considered as one of the main limiting steps of the biological production of biofuels from lignocellulosic biomass. It is a complex multistep process, and various kinetic models have been proposed. The cellulase enzymatic cocktail secreted by <it>Trichoderma reesei </it>has been intensively investigated. β-glucosidases are one of a number of cellulolytic enzymes, and catalyze the last step releasing glucose from the inhibitory cellobiose. β-glucosidase (BGL1) is very poorly secreted by <it>Trichoderma reesei </it>strains, and complete hydrolysis of cellulose often requires supplementation with a commercial β-glucosidase preparation such as that from <it>Aspergillus niger </it>(Novozymes SP188). Surprisingly, kinetic modeling of β-glucosidases lacks reliable data, and the possible differences between native <it>T. reesei </it>and supplemented β-glucosidases are not taken into consideration, possibly because of the difficulty of purifying BGL1.</p> <p>Results</p> <p>A comparative kinetic analysis of β-glucosidase from <it>Aspergillus niger </it>and BGL1 from <it>Trichoderma reesei</it>, purified using a new and efficient fast protein liquid chromatography protocol, was performed. This purification is characterized by two major steps, including the adsorption of the major cellulases onto crystalline cellulose, and a final purification factor of 53. Quantitative analysis of the resulting β-glucosidase fraction from <it>T. reesei </it>showed it to be 95% pure. Kinetic parameters were determined using cellobiose and a chromogenic artificial substrate. A new method allowing easy and rapid determination of the kinetic parameters was also developed. β-Glucosidase SP188 (K<sub>m </sub>= 0.57 mM; K<sub>p </sub>= 2.70 mM) has a lower specific activity than BGL1 (K<sub>m </sub>= 0.38 mM; K<sub>p </sub>= 3.25 mM) and is also more sensitive to glucose inhibition. A Michaelis-Menten model integrating competitive inhibition by the product (glucose) has been validated and is able to predict the β-glucosidase activity of both enzymes.</p> <p>Conclusions</p> <p>This article provides a useful comparison between the activity of β-glucosidases from two different fungi, and shows the importance of fully characterizing both enzymes. A Michaelis-Menten model was developed, including glucose inhibition and kinetic parameters, which were accurately determined and compared. This model can be further integrated into a cellulose hydrolysis model dissociating β-glucosidase activity from that of other cellulases. It can also help to define the optimal enzymatic cocktails for new β-glucosidase activities.</p

    Improper colouring of weighted grid and hexagonal graphs

    International audienceWe study a weighted improper colouring problem motivated by a frequency allocation problem. It consists of associating to each vertex a set of p(v) (weight) distinct colours (frequencies), such that the set of vertices having a given colour induces a graph of degree at most k (the case k = 0 corresponds to a proper coloring). The objective is to minimize the number of colors. We propose approximation algorithms to compute such colouring for general graphs. We apply these to obtain good approximation ratio for grid and hexagonal graphs. Furthermore we give exact results for the 2-dimensional grid and the triangular lattice when the weights are all the same

    International audienceThis paper deals with the design of on board networks in satellites (also called Traveling wave tube Amplifiers (TWTA)). These networks should connect signals arriving on some ports of the satellite to amplifiers, even in case of failures of some amplifiers. They are made of links and expensive switches each with 4 links. So, the aim is to design networks having as few switches as possible and satisfying the following property: \emph{there exist pp edge-disjoint paths from the pp signals arriving on p+λp + \lambda ports (inputs) to any set of pp amplifiers (outputs) chosen from the p+kp+k total number of outputs}. We call such networks \emph{valid (p,λ,k)(p,\lambda,k)-networks} and want to determine the minimum number of switches N(p,λ,k)\mathcal{N}(p, \lambda,k) of such networks. By symmetry we suppose λk\lambda \leq k. We give tight results for small values of kk and asymptotic results when k=O(logp)k = O(\log p) which are tight when k=Θ(λ)k=\Theta(\lambda) and when λ=0\lambda=0

    Chimie dynamique: reconnaissance de nucléobases par des récepteurs synthétiques et isomérie cis-trans d'hydrazones acylées.Ce travail traite du développement des systèmes moléculaires qui peuvent s'adapter à l'addition de substances qui agissent comme un gabarit. Cette approche permet d'isoler une espèce majeure à partir d'un mélange de composés par le biais de la chimie combinatoire dynamique (CCD). La première partie de ma thèse de doctorat inclus l'utilisation d'un ADN simple brin (ADNsb) comme un gabarit pour le transfert d'information par auto-assemblage de récepteurs sans avoir besoin d'enzyme. De nouveaux récepteurs de l'adénine et de la guanine (pinces A et G) solubles dans l'eau ont été conçues dans ce but. Une approche utilisant la résonance magnétique nucléaire (RMN) a été utilisée pour déterminer l'affinité de liaison comme preuve d'une reconnaissance spécifique et efficace. Une évaluation dans l'eau par dichroïsme circulaire (CD) et mesure de la température de fusion par UV (Tm) a été réalisée. Cela a permis de tester respectivement la capacité d'auto-assemblage entre les pinces et un modèle ADNsb, et la force du processus de coopérativité. La deuxième partie de ce travail est axée sur le tri spontanné de motifs pyridine acylhydrazone et sur les configurations intéressantes qu'ils adoptent. Nous avons étudié la synthèse d'une série de motifs pyridine acylhydrazone: dimère, trimères et pentamères. Des études RMN ont permis d'évaluer les changements dans l'équilibre configurationnel cis / trans de ces systèmes dynamiques. Les études ont montré que l'équilibre attendu est biaise la cis acylhydrazone pyridine isomère a été observée par diffraction des rayons X.Dynamic chemistry: nucleobase recognition by synthetic receptors and cis-trans acylhydrazone isomerism. This work deals with the development of molecular systems which can adapt upon the addition of substances that act as templates. This approach enables one major species to be identified from a mixture of compounds through the use of dynamic combinatorial chemistry (DCC). The first part of my PhD included the use of a single stranded DNA (ssDNA) as a template for information transfer via the self-assembly of receptors without the need for enzymes. New water soluble adenine and guanine receptors (A and G clamps) were designed and synthesised for this purpose. Nuclear magnetic resonance (NMR) titration studies were carried out to calculate the binding affinity and as a proof of specific and efficient recognition. An assessment in water via circular dichroism (CD) and UV temperature melting (Tm) studies was carried out. This tested the ability for self-assembly between the clamps and a ssDNA template and the strength of the cooperative process respectively. The second part of my PhD focused on the self-sorting of acylhydrazone pyridine motifs and the interesting configurations they adopt. The feasibility to synthesise these acylhydrazone pyridine motifs (dimer, trimers and pentamers) was investigated. X-ray and NMR studies showed that the equilibrium was found to be biased in an unusual way, and the cis acylhydrazone pyridine isomer was observed.BORDEAUX1-Bib.electronique (335229901) / SudocSudocFranceF
