Service learning and alumni-granted industry-academic collaborations
Abstract. Service learning students can be seen as customers that make consumption for learning experiences from the service products. Service is hard to capture, and service production relies on the interaction between the user and the provider for simultaneous co-production. Also, knowledge and learning is difficult to procure, as it is intangible and rarely transmitted unless the consumer has specific needs that are not being adequately met by the provider. Incorporating the customer in innovation leads to novel ideas for service and also verifies the acceptance and usability of the service. To capture customer knowledge, the employee (and, by extension, firm) awareness of customer needs should be treated as the first priority.Keywords. Service-learning in higher education in Southeast Asia
Abstract. Although service-learning in higher education is a creation of the American society, it has found its way into many education systems outside of the United States. An excellent example of an area where service-learning is taking root is the southeast Asian region. Scholars believe that the slow but steady spread of service-learning as an educational innovation in Asia is symptomatic of modest changes in the conceptions of higher education. As the name suggests, service-learning is a form of education where students acquire real-life knowledge and skills through a wide range of experiences that benefit communities. In other words, service-learning combines education with community service, the aim of which is to enrich students while also meeting the needs of a society. Since service-learning has specific objectives and involves skills acquisition and critical analysis, it goes beyond the usual volunteering and or fundraising. As an active form of university engagement with surrounding communities, service-learning grounds students in local cultures while helping them pursue their educational goals. Keywords. Mitochondrial targeting of human NADH dehydrogenase (ubiquinone) flavoprotein 2 (NDUFV2) and its association with early-onset hypertrophic cardiomyopathy and encephalopathy
<p>Abstract</p> <p>Background</p> <p>NADH dehydrogenase (ubiquinone) flavoprotein 2 (NDUFV2), containing one iron sulfur cluster ([2Fe-2S] binuclear cluster N1a), is one of the core nuclear-encoded subunits existing in human mitochondrial complex I. Defects in this subunit have been associated with Parkinson's disease, Alzheimer's disease, Bipolar disorder, and Schizophrenia. The aim of this study is to examine the mitochondrial targeting of NDUFV2 and dissect the pathogenetic mechanism of one human deletion mutation present in patients with early-onset hypertrophic cardiomyopathy and encephalopathy.</p> <p>Methods</p> <p>A series of deletion and point-mutated constructs with the <it>c-myc </it>epitope tag were generated to identify the location and sequence features of mitochondrial targeting sequence for NDUFV2 in human cells using the confocal microscopy. In addition, various lengths of the NDUFV2 N-terminal and C-terminal fragments were fused with enhanced green fluorescent protein to investigate the minimal region required for correct mitochondrial import. Finally, a deletion construct that mimicked the IVS2+5_+8delGTAA mutation in <it>NDUFV2 </it>gene and would eventually produce a shortened NDUFV2 lacking 19-40 residues was generated to explore the connection between human gene mutation and disease.</p> <p>Results</p> <p>We identified that the cleavage site of NDUFV2 was located around amino acid 32 of the precursor protein, and the first 22 residues of NDUFV2 were enough to function as an efficient mitochondrial targeting sequence to carry the passenger protein into mitochondria. A site-directed mutagenesis study showed that none of the single-point mutations derived from basic, hydroxylated and hydrophobic residues in the NDUFV2 presequence had a significant effect on mitochondrial targeting, while increasing number of mutations in basic and hydrophobic residues gradually decreased the mitochondrial import efficacy of the protein. The deletion mutant mimicking the human early-onset hypertrophic cardiomyopathy and encephalopathy lacked 19-40 residues in NDUFV2 and exhibited a significant reduction in its mitochondrial targeting ability.</p> <p>Conclusions</p> <p>The mitochondrial targeting sequence of NDUFV2 is located at the N-terminus of the precursor protein. Maintaining a net positive charge and an amphiphilic structure with the overall balance and distribution of basic and hydrophobic amino acids in the N-terminus of NDUFV2 is important for mitochondrial targeting. The results of human disease cell model established that the impairment of mitochondrial localization of NDUFV2 as a mechanistic basis for early-onset hypertrophic cardiomyopathy and encephalopathy.</p
Hemispheric dispersion of radioactive plume laced with fission nuclides from the Fukushima nuclear event
Radioactivities of particulate 131I and 137Cs released from the Fukushima nuclear accident were monitored in a regional aerosol network including two high mountain sites (central Taiwan and Tibetan Plateau). The results were integrated with data measured elsewhere around the world, with special focus on the mid-latitudes. The hemispheric transport of the Fukushima radiation clouds (FRCs) by the westerlies took 3–4 km, whereas the second one up to 5 km or more. 131I and 137Cs were fractionated during transport, with 137Cs concentrated in the shallower layer, susceptible to depositional removal, while 131I moving faster and higher. This accident may be exemplified to identify some atmospheric processes on the hemispheric scale
Mutations in the PKM2 exon-10 region are associated with reduced allostery and increased nuclear translocation.
PKM2 is a key metabolic enzyme central to glucose metabolism and energy expenditure. Multiple stimuli regulate PKM2's activity through allosteric modulation and post-translational modifications. Furthermore, PKM2 can partner with KDM8, an oncogenic demethylase and enter the nucleus to serve as a HIF1α co-activator. Yet, the mechanistic basis of the exon-10 region in allosteric regulation and nuclear translocation remains unclear. Here, we determined the crystal structures and kinetic coupling constants of exon-10 tumor-related mutants (H391Y and R399E), showing altered structural plasticity and reduced allostery. Immunoprecipitation analysis revealed increased interaction with KDM8 for H391Y, R399E, and G415R. We also found a higher degree of HIF1α-mediated transactivation activity, particularly in the presence of KDM8. Furthermore, overexpression of PKM2 mutants significantly elevated cell growth and migration. Together, PKM2 exon-10 mutations lead to structure-allostery alterations and increased nuclear functions mediated by KDM8 in breast cancer cells. Targeting the PKM2-KDM8 complex may provide a potential therapeutic intervention
Knockdown of PsbO leads to induction of HydA and production of photobiological H2 in the green alga Chlorella sp. DT
Green algae are able to convert solar energy to H2 via the photosynthetic electron transport pathway under certain conditions. Algal hydrogenase (HydA, encoded by HYDA) is in charge of catalyzing the reaction: 2H+ + 2e− ↔ H2 but usually inhibited by O2, a byproduct of photosynthesis. The aim of this study was to knockdown PsbO (encoded by psbO), a subunit concerned with O2 evolution, so that it would lead to HydA induction. The alga, Chlorella sp. DT, was then transformed with short interference RNA antisense-psbO (siRNA-psbO) fragments. The algal mutants were selected by checking for the existence of siRNA-psbO fragments in their genomes and the low amount of PsbO proteins. The HYDA transcription and the HydA expression were observed in the PsbO-knockdown mutants. Under semi-aerobic condition, PsbO-knockdown mutants could photobiologically produce H2 which increased by as much as 10-fold in comparison to the wild type
Toll-like receptor 9 agonist enhances anti-tumor immunity and inhibits tumor-associated immunosuppressive cells numbers in a mouse cervical cancer model following recombinant lipoprotein therapy
BACKGROUND: Although cytotoxic T lymphocytes (CTLs) play a major role in eradicating cancer cells during immunotherapy, the cancer-associated immunosuppressive microenvironment often limits the success of such therapies. Therefore, the simultaneous induction of cancer-specific CTLs and reversal of the immunosuppressive tumor microenvironment may be more effectively achieved through a single therapeutic vaccine. A recombinant lipoprotein with intrinsic Toll-like receptor 2 (TLR2) agonist activity containing a mutant form of E7 (E7m) and a bacterial lipid moiety (rlipo-E7m) has been demonstrated to induce robust CTL responses against small tumors. This treatment in combination with other TLR agonists is able to eliminate large tumors. METHODS: Mouse bone marrow-derived dendritic cells (DCs) were employed to determine the synergistic production of pro-inflammatory cytokines upon combination of rlipo-E7m and other TLR agonists. Antigen-specific CTL responses were investigated using immunospots or in vivo cytolytic assays after immunization in mice. Mice bearing various tumor sizes were used to evaluate the anti-tumor effects of the formulation. Specific subpopulations of immunosuppressive cells in the tumor infiltrate were quantitatively determined by flow cytometry. RESULTS: We demonstrate that a TLR9 agonist (unmethylated CpG oligodeoxynucleotide, CpG ODN) enhances CTL responses and eradicates large tumors when combined with rlipo-E7m. Moreover, combined treatment with rlipo-E7m and CpG ODN effectively increases tumor infiltration by CTLs and reduces the numbers of myeloid-derived suppressor cells (MDSCs), tumor-associated macrophages (TAMs) and regulatory T cells (Tregs) in the tumor microenvironment. CONCLUSION: These findings suggest that the dramatic anti-tumor effects of the recombinant lipoprotein together with CpG ODN may reflect the amplification of CTL responses and the repression of the immunosuppressive environment. This promising approach could be applied for the development of additional therapeutic cancer vaccines
Weight Stigma Model on Quality of Life Among Children in Hong Kong: A Cross-Sectional Modeling Study
We proposed amodel to examine the relationship among different types of weight-related
stigmas and their relationship to quality of life (QoL). We recruited 430 dyads of
elementary school children [mean age = 10.07 years; nboy = 241 (56.0%); noverweight =
138 (32.1%)] and their parents. Parents completed QoL instruments about their children
assessing generic QoL and weight-related QoL. Children completed QoL instruments
assessing generic QoL and weight-related QoL and stigma scales assessing experienced
weight stigma, weight-related self-stigma, and perceived weight stigma. Experienced
weight stigma was significantly associated with perceived weight stigma, and in turn,
perceived weight stigma was significantly associated with weight-related self-stigma.
However, experienced weight stigma was not directly associated with weight-related
self-stigma. In addition, experienced stigma was negatively associated with both
child-rated and parent-rated QoL. Perceived weight stigma was associated only with
parent-rated weight-related QoL but not child-rated QoL. Self-stigma was associated
with child-rated QoL but not parent-rated QoL. Moreover, perceived weight stigma and
weight-related self-stigma were significant mediators in the association between body
weight and children’s QoL; experienced weight stigma was not a significant mediator.
The study findings can be used to inform healthcare providers about the relationship
among different types of stigmas and their influence on child-rated and parent-rated QoL
and help them develop interventions to address the global trend of overweight/obesity
in youth and pediatric populations
