    A STUDY ON THE RELATIONSHIP BETWEEN COLLEGE TEACHERS’ PERCEPTION OF PROFESSIONAL LEARNING COMMUNITIES AND SELF-EFFICACY

    The purpose of this quantitative survey study was to investigate the relationship between college teachers’ perceptions of the professional learning community and teacher self-efficacy. Drawing on quantitative data from a sample of 367 college teachers in Inner Mongolia in China during the 2023-2024 school year, the research investigated the impact of PLC participation on self-efficacy across different career stages and majors. The findings revealed a medium positive correlation between college teachers’ perception in PLCs and teacher self-efficacy. The regression model indicated that collective learning and application and shared personal practice significantly influenced the overall teacher self-efficacy, but shared and supportive leadership, shared values and vision, supportive conditions-structures and supportive conditions-relationships did not have a significant effect. The two predictors collectively explained 29.3% of the variance in total TESE scores. One-way ANOVA results showed there was no statistically significant difference in teacher self-efficacy based on college teachers’ professional ranks. The Independent Samples T-Test showed that there was a significant difference in TSES between normal major teachers and non-normal major teachers. The effect size of the difference was medium. Future studies should expand this research to include longitudinal studies on how PLC participation impact teacher self-efficacy over time and comparison studies on traditional PLCs and online PLC models to identify which characteristics are most effective in enhancing self-efficacy. What’s more, mixed-methods research design can explore both quantitative and qualitative aspects of self-efficacy, capturing the trends and experiences of teachers at different career stages. The research underscores the practical implications for educators, school leaders, and policymakers. By creating supportive work environment, designing effective professional development activities, supporting teacher’s professional growth, teachers’ self-efficacy and teaching quality can be effectively improved

    Human γδ T cells display the principal characteristics of professional antigen-presenting cells (APCs), in addition to playing a vital role in immunity through cytokine secretion and their cytotoxic activity. However, it is not clear whether γδ T cells perform APC-like functions under pathological conditions. In this study, we showed that, in contrast to peripheral-derived γδ T cells directly isolated from PBMCs of gastric cancer patients, tumor-activated γδ T cells not only killed tumor cells efficiently but also strongly induced primary CD4(+) and CD8(+)   αβ T cells proliferation and differentiation. More importantly, they abrogated the immunosuppression induced by CD4(+)CD25(+) Treg cells and induced the cytotoxic function of CD8(+)   αβ T cells from patients with gastric cancer. In conclusion, tumor-activated γδ T cells can induce adaptive immune responses through their APC-like functions, and these cells may be a potentially useful tool in the development of tumor vaccines and immunotherapy

    A Novel Derivative of the Natural Product Danshensu Suppresses Inflammatory Responses to Alleviate Caerulein-Induced Acute Pancreatitis

    Acute pancreatitis (AP), a common abdominal inflammatory disorder, is characterized by premature intracellular activation of digestive proteases within pancreatic acini and a consecutive systemic inflammatory response. Although the mechanism remains to be fully understood, inflammation is the main cause of pancreatic damage in AP. A novel compound [4-(2-acetoxy-3-((R)-3-(benzylthio)-1-methoxy-1-oxopropan-2-ylamino)-3-oxopropyl)-1,2-phenylene diacetate (DSC)], derived from danshensu, exhibits anti-inflammatory and anti-apoptotic properties in vitro. However, its potential beneficial effect in AP has not been demonstrated. This study aimed to investigate the effects and underlying mechanisms of DSC in experimental AP in mice. We found that DSC suppressed inflammatory responses in AP by inhibiting the activation of nuclear factor-κB (NF-κB), signal transducer and activator of transcription 3 (STAT3) and nucleotide-binding domain leucine-rich repeat containing family, pyrin domain-containing 3 (NLRP3) inflammasome. Furthermore, treatment with DSC modulated the infiltration of neutrophils and the phenotypes of macrophages in mice induced with AP. Interestingly, we found that the expression of nuclear factor-erythroid 2 related factor 2 (Nrf2) and its regulated antioxidant enzyme heme oxygenase-1 (HO-1), which modulate inflammatory activities, was significantly increased in DSC-treated groups. Together, our findings demonstrate that DSC alleviates pancreatic inflammation and damage in AP by inhibiting the activation of NF-κB, STAT3, and NLRP3 inflammasome and modulating immune cell responses

    Modulation of Gut Microbiota by Low Methoxyl Pectin Attenuates Type 1 Diabetes in Non-obese Diabetic Mice

    Intestinal homeostasis underpins the development of type 1 diabetes (T1D), and dietary manipulations to enhance intestinal homeostasis have been proposed to prevent T1D. The current study aimed to investigate the efficacy of supplementing a novel specific low-methoxyl pectin (LMP) dietary fiber in preventing T1D development. Female NOD mice were weaned onto control or 5% (wt/wt) LMP supplemented diets for up to 40 weeks of age, overt diabetes incidence and blood glucose were monitored. Then broad-spectrum antibiotics (ABX) treatment per os for 7 days followed by gut microbiota transfer was performed to demonstrate gut microbiota-dependent effects. Next-generation sequencing was used for analyzing the composition of microbiota in caecum. Concentration of short chain fatty acids were determined by GC-MS. The barrier reinforcing tight junction proteins zonula occludens-2 (ZO-2), claudin-1 and NOD like receptor protein 3 (NLRP3) inflammasome activation were determined by Western blot. The proportion of CD25(+)Foxp3(+)CD4(+) regulatory T cell (Foxp3(+) Treg) in the pancreas, pancreatic and mesenteric lymph nodes was analyzed by flow cytometry. We found that LMP supplementation ameliorated T1D development in non-obese diabetic (NOD) mice, as evidenced by decreasing diabetes incidence and fasting glucose levels in LMP fed NOD mice. Further microbiota analysis revealed that LMP supplementation prevented T1D-associated caecal dysbiosis and selectively enriched caecal bacterial species to produce more SCFAs. The LMP-mediated microbial balance further enhanced caecal barrier function and shaped gut-pancreatic immune environment, as characterized by higher expression of tight junction proteins claudin-1, ZO-2 in caecum, increased Foxp3(+) Treg population and decreased NLRP3 inflammasome activation in both caecum and pancreas. The microbiota-dependent beneficial effect of LMP on T1D was further proven by the fact that aberration of caecal microbiota by ABX treatment worsened T1D autoimmunity and could be restored with transfer of feces of LMP-fed NOD mice. These data demonstrate that this novel LMP limits T1D development by inducing caecal homeostasis to shape pancreatic immune environment. This finding opens a realistic option for gut microbiota manipulation and prevention of T1D in humans

    Changes of water clarity in large lakes and reservoirs across China observed from long-term MODIS

    Water clarity is a well-established first-order indicator of water quality and has been used globally by water regulators in their monitoring and management programs. Assessments of water clarity in lakes over large temporal and spatial scales, however, are rare, limiting our understanding of its variability and the driven forces. In this study, we developed and validated a robust Secchi disk depth (ZSD) algorithm for lakes across China based on two water color parameters, namely Forel-Ule Index (FUI) and hue angle α, retrieved from MODIS data. The MODIS ZSD model shows good results when compared with in-situ measurements from 17 lakes, with a 27.4% mean relative difference (MRD) in the validation dataset. Compared with other empirical ZSD models, our FUI and α-based model demonstrates improved performance and adaptability over a wide range of water clarity and trophic states. This algorithm was subsequently applied to MODIS measurements to provide a comprehensive assessment of water clarity in large lakes (N = 153) across China for the first time. The mean summer ZSD of the studied lakes between 2000 and 2017 demonstrated marked spatial and temporal variations. Spatially, the ZSD of large lakes presented a distinct spatial pattern of “high west and low east” over China. This spatial pattern was found to be associated with the significant differences in lake depth and altitude between west and east China while China's population, GDP, temperature, and precipitation distribution have also contributed to a certain extent. Temporally, the ZSD of most lakes increased during this period, with an overall mean rate of 3.3 cm/yr for all lakes. Here, 38.6% (N = 59) of the lakes experienced a significant increase in their ZSD value during the past 18 years while only 8.5% (N = 13) showed a significant decreasing trend. Significant increases in lake ZSD were observed in west China, which were found to correlate with the increase of air temperature and lake surface area. This is possibly a response of the lakes in west China to climate change. In the lake systems of east China, which are predominately used as a drinking water source, the increase in lake ZSD was found to be strongly correlated with changes in local GDP (gross domestic production), NDVI (normalized difference vegetation index) and lake surface area, suggesting a combined effect of the implemented management practices and climatic variability. The results of this study provide important information for water quality conservation and management in China, and also highlight the value of satellite remote sensing in monitoring water quality over lakes at a large scale and long-term

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Study on Lubrication Characteristics of Novel Forced Wave Generator of Harmonic Drive without Flexible Bearing

    In contrast to the conventional forced wave generator which consists of cam and flexible bearing in harmonic drive, the novel forced wave generator retains cam but cancels flexible bearing. In this article, the lubrication characteristics of the novel forced wave generator in harmonic drive is studied. First, an elliptical sliding bearing (ESB) model of simplified structure between the novel forced wave generator and the flex spline is established. Further, the computational fluid dynamics (CFD) method is employed to study the effect of some factors on the lubrication characteristics of the ESB model including elliptical gap ratio, width, and rotational speed. According to the analysis, the elliptical gap ratio has a great impact and its optimal value is 3, which is used in the design of the novel forced wave generator. Last, the practical design of the novel forced wave generator in harmonic drive is given, which can provide a basis for design and optimization of a forced wave generator without flexible bearing of the harmonic drive
