    Population variation and differences in serum leptin independent of adiposity: a comparison of Ache Amerindian men of Paraguay and lean American male distance runners

    BACKGROUND: Serum leptin variation is commonly associated with fat percentage (%), body mass index (BMI), and activity. In this investigation, we report population differences in mean leptin levels in healthy men as well as associations with fat % and BMI that are independent of these factors and reflect likely variation resulting from chronic environmental conditions. METHODS: Serum leptin levels, fat %, and BMI were compared between lean American distance runners and healthy Ache Native Americans of Paraguay. Mean levels were compared as were the regressions between fat %, BMI, and leptin. Comparisons were performed between male American distance runners (n = 13, mean age 32.2 ± 9.2 SD) and highly active male New World indigenous population (Ache of Paraguay, n = 20, mean age 32.8 ± 9.2) in order to determine whether significant population variation in leptin is evident in physically active populations living under different ecological circumstances independent of adiposity and BMI. RESULTS: While the Ache were hypothesized to exhibit higher leptin due to significantly greater adiposity (fat %, Ache 17.9 ± 1.8 SD; runners 9.7 ± 3.2, p < 0.0001), leptin levels were nonetheless significantly higher in American runners (Ache 1.13 ng/ml ± 0.38 SD; runners 2.19 ± 1.15; p < 0.007). Significant differences in the association between leptin and fat % was also evident between Ache and runner men. Although fat % was significantly related with leptin in runners (r = 0.90, p < 0.0001) fat % was negatively related in Ache men (r = -0.50, p < 0.03). CONCLUSION: These results illustrate that chronic ecological conditions in addition to activity are likely factors that contribute to population variation in leptin levels and physiology. Population variation independent of adiposity should be considered to be an important source of variation, especially in light of ethnic and population differences in the incidence and etiology of obesity, diabetes, and other metabolic conditions

    Effect of Omega-3 Fatty Acids on Lipoprotein Profile and Particle Size in Hispanic Women

    The effect of increased Ω3 fatty acids on lipoprotein sub-fractions has not been well studied in Hispanic women, a group with a high prevalence of dyslipidemia and metabolic syndrome. The purpose of this randomized, placebo-controlled trial was to examine the effect of a 90-day (90D) combined EPA+DHA supplementation on lipoprotein profiles of Hispanic women. Twenty-seven non-diabetic women were randomly assigned to the Ω3 group (n=17, 1.9 g/day EPA+DHA) or the placebo group (n=10, 2 g/day oleic acid). At baseline (0D) and 90D a fasting blood sample was drawn for determination of lipoprotein profile and lipoprotein particle sub-fractions. For the Ω3 group, total cholesterol increased 6.5%, triglycerides were reduced 14.8%, LDL concentration increased 5.6%, and total LDL particle size concentration increased 8.2% (P \u3c 0.05). Large LDL particle size concentration increased 16% from 0D to 90D, this change was not significant (P = 0.051). At 90D, total cholesterol, LDLs, and LDL particle concentration were higher in the Ω3 group compared to placebo (P \u3c 0.05). Omega-3 supplementation resulted in reduced plasma TGs and an increase in large LDL particle size concentration, yet had no effect on HDL particle size or concentration. Further research is necessary to examine the effects of increased Ω3 fatty acid intake in Hispanic women on dyslipidemia and related co-morbidities

As a result of climate change, drought events are anticipated to increase in frequency and intensity even in regions where annual precipitation is unchanged, as well as where it is reduced. Many studies have been carried out on drought resistance of forage species (Holloway-Phillips and Brodribb 2011). However, for most experiments, it is unclear which mechanism(s) are responsible for variation in plant performance under moisture stress, whether drought response mechanisms were triggered, or whether differences in performance were simply the result of intrinsic plant vigour.     Cardiometabolic Plasticity in Response to a Short-Term Diet and Exercise Intervention in Young Hispanic and NonHispanic White Adults

    BACKGROUND: Young adult Mexican Americans (MA) exhibit lower insulin sensitivity (Si) than nonHispanic whites (NHW), even when controlling for fitness and adiposity. It is unclear if MA are as responsive to the same lifestyle intervention as NHW. OBJECTIVE: We developed a model to examine cardiometabolic plasticity (i.e., changes in Si and plasma lipids) in MA compared to NHW adults in response to a diet-exercise intervention. DESIGN: Sedentary subjects (20 NHW: 11F, 9M, 23.0 y, 25.5 kg/m(2); 17 MA: 13F, 4M, 22.7 y, 25.4 kg/m(2)) consumed their habitual diets and remained sedentary for 7 days, after which fasting blood samples were obtained, and a 3-h intravenous glucose tolerance test (IVGTT) was performed with the insulin area under the curve (IAUC) used to estimate Si. Subjects then completed a 7-day diet/exercise intervention (diet: low saturated fat, low added sugar, high fiber; exercise: cycling, six total sessions lasting 40-45 min/session at 65% VO(2) max). Pre-intervention tests were repeated. RESULTS: Pre intervention IAUC was 28% higher (p<0.05) in MA (IAUC pre  =  2298 µU*180 min/mL) than in NHW (IAUC = 1795 µU*180 min/mL). Following the intervention, there was a significant reduction in IAUC in MA (29%) and NHW (32%), however, the IAUC remained higher (p<0.05) for MA (post  = 1635 µU*180 min/mL) than for NHW (post = 1211 µU*180 min/mL). Pre test plasma lipids were not different in MA compared to NHW. Plasma cholesterol and TG concentrations significantly improved in both groups, but concentrations of low density lipoprotein-cholesterol and small dense LDL particles significantly improved only in the NHW. CONCLUSION: With a short-term diet-exercise intervention, the magnitude of improvements in Si and serum cholesterol and TG in Hispanics are similar to those in NHW. However, because at the outset MA were less insulin sensitive compared to NHW, within the short timeframe studied the ethnic gap in insulin sensitivity remained

ECMO, from a technical standpoint, is conceptually simple—however, it can be very challenging to implement therapy at the individual patient level as well as at hospital (or healthcare system) level. ECMO program development involves engagement of key stake-holders including physicians, nursing, and administrative leadership.     Plasma-borne indicators of inflammasome activity in Parkinson’s disease patients

    Parkinson’s disease (PD) is a neurodegenerative disorder characterized by motor and non-motor symptoms and loss of dopaminergic neurons of the substantia nigra. Inflammation and cell death are recognized aspects of PD suggesting that strategies to monitor and modify these processes may improve the management of the disease. Inflammasomes are pro-inflammatory intracellular pattern recognition complexes that couple these processes. The NLRP3 inflammasome responds to sterile triggers to initiate pro-inflammatory processes characterized by maturation of inflammatory cytokines, cytoplasmic membrane pore formation, vesicular shedding, and if unresolved, pyroptotic cell death. Histologic analysis of tissues from PD patients and individuals with nigral cell loss but no diagnosis of PD identified elevated expression of inflammasome-related proteins and activation-related “speck” formation in degenerating mesencephalic tissues compared with controls. Based on previous reports of circulating inflammasome proteins in patients suffering from heritable syndromes caused by hyper-activation of the NLRP3 inflammasome, we evaluated PD patient plasma for evidence of inflammasome activity. Multiple circulating inflammasome proteins were detected almost exclusively in extracellular vesicles indicative of ongoing inflammasome activation and pyroptosis. Analysis of plasma obtained from a multi-center cohort identified elevated plasma-borne NLRP3 associated with PD status. Our findings are consistent with others indicating inflammasome activity in neurodegenerative disorders. Findings suggest mesencephalic inflammasome protein expression as a histopathologic marker of early-stage nigral degeneration and suggest plasma-borne inflammasome-related proteins as a potentially useful class of biomarkers for patient stratification and the detection and monitoring of inflammation in PD

    A Hybrid Magneto-Optic Capacitive Memory with Picosecond Writing Time

    The long-term future of information storage requires the use of sustainable nanomaterials in architectures operating at high frequencies. Interfaces can play a key role in this pursuit via emergent functionalities that break out from conventional operation methods. Here, spin-filtering effects and photocurrents are combined at metal-molecular-oxide junctions in a hybrid magneto-capacitive memory. Light exposure of metal-fullerene-metal oxide devices results in spin-polarized charge trapping and the formation of a magnetic interface. Because the magnetism is generated by a photocurrent, the writing time is determined by exciton formation and splitting, electron hopping, and spin-dependent trapping. Transient absorption spectroscopy measurements show changes in the electronic states as a function of the magnetic history of the device within picoseconds of the optical pumping. The stored information is read using time-resolved scanning magneto optic Kerr effect measurements during microwave irradiation. The emergence of a magnetic interface in the picosecond timescale opens new paths of research to design hybrid magneto-optic structures operating at high frequencies for sensing, computing, and information storage

    Event-Related Potentials Dissociate Effects of Salience and Space in Biased Competition for Visual Representation

    BACKGROUND: Selective visual attention is the process by which the visual system enhances behaviorally relevant stimuli and filters out others. Visual attention is thought to operate through a cortical mechanism known as biased competition. Representations of stimuli within cortical visual areas compete such that they mutually suppress each others' neural response. Competition increases with stimulus proximity and can be biased in favor of one stimulus (over another) as a function of stimulus significance, salience, or expectancy. Though there is considerable evidence of biased competition within the human visual system, the dynamics of the process remain unknown. METHODOLOGY/PRINCIPAL FINDINGS: Here, we used scalp-recorded electroencephalography (EEG) to examine neural correlates of biased competition in the human visual system. In two experiments, subjects performed a task requiring them to either simultaneously identify two targets (Experiment 1) or discriminate one target while ignoring a decoy (Experiment 2). Competition was manipulated by altering the spatial separation between target(s) and/or decoy. Both experimental tasks should induce competition between stimuli. However, only the task of Experiment 2 should invoke a strong bias in favor of the target (over the decoy). The amplitude of two lateralized components of the event-related potential, the N2pc and Ptc, mirrored these predictions. N2pc amplitude increased with increasing stimulus separation in Experiments 1 and 2. However, Ptc amplitude varied only in Experiment 2, becoming more positive with decreased spatial separation. CONCLUSIONS/SIGNIFICANCE: These results suggest that N2pc and Ptc components may index distinct processes of biased competition--N2pc reflecting visual competitive interactions and Ptc reflecting a bias in processing necessary to individuate task-relevant stimuli

    Impacts of acoustic and gravity waves on the ionosphere

    The impact of regional-scale neutral atmospheric waves has been demonstrated to have profound effects on the ionosphere, but the circumstances under which they generate ionospheric disturbances and seed plasma instabilities are not well understood. Neutral atmospheric waves vary from infrasonic waves of &lt;20 Hz to gravity waves with periods on the order of 10 min, for simplicity, hereafter they are combined under the common term Acoustic and Gravity Waves (AGWs). There are other longer period waves like planetary waves from the lower and middle atmosphere, whose effects are important globally, but they are not considered here. The most ubiquitous and frequently observed impact of AGWs on the ionosphere are Traveling Ionospheric Disturbances (TIDs), but AGWs also affect the global ionosphere/thermosphere circulation and can trigger ionospheric instabilities (e.g., Perkins, Equatorial Spread F). The purpose of this white paper is to outline additional studies and observations that are required in the coming decade to improve our understanding of the impact of AGWs on the ionosphere.</p
