    Gene Delivery Technologies for Efficient Genome Editing: Applications in Gene Therapy

    Specific nucleases (SNs), including ZFNs, TALENs, and CRISPR (clustered regularly interspaced palindromic repeats), are powerful tools for genome editing (GE). These tools have achieved efficient gene repair and gene disruption of human primary cells. However, their efficiency and safety must be improved before translation into clinic. In particular, one of the main hurdles of GE technology is the delivery of the different components into the nucleus of target cells. Successful gene editing must be able to deliver the SNs and/or the donor DNA into a large number of target cells in order to have a therapeutic benefit. In addition, the delivery must be nontoxic and the SNs must be innocuous to the target cells. In this chapter, we will summarize the different ways to deliver SNs and donor DNA

    Assessment of the external load of amateur soccer players during four consecutive training microcycles in relation to the external load during the official match

    [EN] Aim: The aim of this study was to assess the external load of amateur male players during 4 consecutive training microcycles (M) at the beginning of the in-season according to the training session in absolute external load demands and in relation to the competition demands. Methods: Regional-level players (n = 10; age, 20.8 ± 1.7 years; height, 175.5 ± 3.8 cm; body mass, 69.7 ± 2.9 kg; soccer experience, 13.2 ± 2.5 years) were monitored using GPS devices during training sessions and matches. The external load variables measured were: duration (min); total distance covered (TD); distance covered at high-speed (HID, 14.4-19.8 km/h); distance covered at sprinting (SPD; >19.8 km/h); and distance covered in high intensity acceleration (ACD; >2.5 m/s2) and deceleration (DECD; <-2.5 m/s2). Results: The results indicated that the external load variables (time, HID, SPD, ACD, and DECD) were similar between the four microcycles. Greater (p<0.01) time, TD, HID and SPD were observed in match day (MD)-2 compared to MD+1, MD-3, and MD-1. Aside from training duration, all external loads variables (TD, HID, SPD, ACD, and DECD) were lower during training sessions compared to official matches (p<0.05). Conclusion: Amateur soccer players present relative stable external training loads across competitive microcycles, with the peak load observed two days before the official match. Besides this, the match constitutes the highest load during a typical competitive microcycle in this cohort of players.S

    'Enhancement of the crystallization process of TbxFe1-x thin films upon the formation of alpha-Tb phase'

    TbxFe1−x thin films deposited by sputtering on Mo were investigated structurally and magnetically. The microstructure consists of TbFe2 nanoparticles embedded in an amorphous matrix, and the Tb content can be correlated with an increase in the volume of these nanoparticles. Similar microstructure and behavior were found when TbFe2 was deposited on glass and on a Pt buffer layer. Nevertheless, thermal treatments promote a different effect, depending on the mechanical stiffness of the buffer layer. The layers deposited on Mo, a rigid material, show crystalline TbFe2 together with α-Tb phase upon thermal treatment. In contrast, TbFe2 does not crystallize properly on Pt, a material with a lower stiffness than Mo. Intermediate results were observed on the film deposited on glass. Experimental results show the impact of the buffer stiffness on the crystallization process. Moreover, the formation of α-Tb appears to be fundamental to crystallized TbFe2 on layers deposited on rigid buffer

    Evaluating a Speech Communication System for deaf people

    This paper describes the development of an Advanced Speech Communication System for Deaf People and its field evaluation in a real application domain: the renewal of Driver’s License. The system is composed of two modules. The first one is a Spanish into Spanish Sign Language (LSE: Lengua de Signos Española) translation module made up of a speech recognizer, a natural language translator (for converting a word sequence into a sequence of signs), and a 3D avatar animation module (for playing back the signs). The second module is a Spoken Spanish generator from sign-writing composed of a visual interface (for specifying a sequence of signs), a language translator (for generating the sequence of words in Spanish), and finally, a text to speech converter. For language translation, the system integrates three technologies: an example-based strategy, a rule-based translation method and a statistical translator. This paper also includes a detailed description of the evaluation carried out in the Local Traffic Office in the city of Toledo (Spain) involving real government employees and deaf people. This evaluation includes objective measurements from the system and subjective information from questionnaires. Finally, the paper reports an analysis of the main problems and a discussion about possible solutions

    Este artículo describe el desarrollo y la evaluación de un sistema de comunicación para personas sordas en un ámbito de aplicación específico: la renovación del permiso de conducir. El sistema de comunicación desarrollado está compuesto por dos módulos que permiten la comunicación en los dos sentidos. El primer módulo es un traductor de voz en castellano a Lengua de Signos Española (LSE) y está formado por un reconocedor de voz, un traductor de palabras en castellano a una secuencia de signos y un tercer módulo de representación de los signos mediante un agente animado. El segundo módulo es un generador de voz en castellano a partir de una secuencia de signos, y está formado por una interfaz donde se especifican los signos, un traductor (para convertir la secuencia de signos en una secuencia de palabras) y un conversor de texto a voz. En los dos módulos de traducción entre lenguas, se integran tres tecnologías: una basada en ejemplos, una basada en reglas y un traductor estadístico.     This paper describes the development of a Spanish Spanish Sign Language (LSE) translation system. Firstly, it describes the first Spanish Spanish Sign Language (LSE) parallel corpus focused on two specific domains: the renewal of the Identity Document and Driver’s License. This corpus includes more than 4,000 Spanish sentences (in these domains), their LSE translation and a video for each LSE sentence with the sign language representation. This corpus also contains more than 700 sign descriptions in several sign writing specifications. The translation system developed with this corpus consists of two modules: a Spanish into LSE translation module that is composed of a speech recognizer (for decoding the spoken utterance into a word sequence), a natural language translator (for converting a word sequence into a sequence of signs) and a 3D avatar animation module (for playing back the signs).     Eigenstate equivalent circuit for bi-periodic surfaces: Application to three parallel dipoles

    An equivalent circuit for the unit cell of bi-periodic surfaces is proposed. The equivalent circuit is general and simple. It is based on the decomposition of the eigenmodes of the structure and it takes into account the polarization of the incident wave. It consists of the interconnection of two different admittances and two transformers. The model of a unit cell composed of three parallel dipoles is extracted. The transformer of the proposed equivalent circuit models the rotation of the dipoles, while the admittances model their frequency behaviour (resonances). This equivalent circuit provides physical insight and, at the same time, it models in separate elements the contribution of the rotation and the dipole trio resonances.     Measuring odd- and even-mode propagation constants of coupled-lines by means of the lattice network

    A classic method for measuring the propagation constant of transmission lines is the one proposed by Bianco and Parodi more than four decades ago [1]. It is based on the invariance of the cross ratio of four complex numbers under a bilinear transformation. This means that it is possible to extract the value of the propagation constant (both real and imaginary parts) of an arbitrary transmission line (TL) by measuring, through an arbitrary (and unique) two-port, the input reflection coefficient of four TL sections of different lengths provided they are terminated by the same load. The accuracy of the original method can be greatly improved by measuring a higher number of TL sections and taking all the possible combinations of four lines.Universidad de Málaga.     Coupling a polarizable water model to the hydrated ion–water interaction potential: A test on the Cr3+ hydration

    A strategy to build interaction potentials for describing ionic hydration of highly charged monoatomic cations by computer simulations, including the polarizable character of the solvent, is proposed. The method is based on the hydrated ion concept that has been previously tested for the case of Cr3+ aqueous solutions [J. Phys. Chem. 100, 11748 (1996)]. In the present work, the interaction potential of [Cr(H2O6)]3+ with water has been adapted to a water model that accounts for the polarizable character of the solvent by means of a mobile charge harmonic oscillator representation (MCHO model) [J. Chem. Phys. 93, 6448 (1990)]. Monte Carlo simulations of the Cr3+ hexahydrate plus 512 water molecules have been performed to study the energetics and structure of the ionic solution. The results show a significant improvement in the estimate of the hydration enthalpy [ LlHhydr(Cr3+)=-1109.6:±70 kcal/mol] that now matches the experimental value within the uncertainty of this magnitude. The use of the polarizable water model lowers by �140 kcal/mol the statistical estimation of the [Cr(H2O6)]3+ hydration enthalpy compared to the nonpolarizable model. (-573 kcal/mol for the polarizable model vs -714 kcal/mol for the nonpolarizable one.)
