A bifibrational reconstruction of Lawvere's presheaf hyperdoctrine

Combining insights from the study of type refinement systems and of monoidal closed chiralities, we show how to reconstruct Lawvere's hyperdoctrine of presheaves using a full and faithful embedding into a monoidal closed bifibration living now over the compact closed category of small categories and distributors. Besides revealing dualities which are not immediately apparent in the traditional presentation of the presheaf hyperdoctrine, this reconstruction leads us to an axiomatic treatment of directed equality predicates (modelled by hom presheaves), realizing a vision initially set out by Lawvere (1970). It also leads to a simple calculus of string diagrams (representing presheaves) that is highly reminiscent of C. S. Peirce's existential graphs for predicate logic, refining an earlier interpretation of existential graphs in terms of Boolean hyperdoctrines by Brady and Trimble. Finally, we illustrate how this work extends to a bifibrational setting a number of fundamental ideas of linear logic.Comment: Identical to the final version of the paper as appears in proceedings of LICS 2016, formatted for on-screen readin

FUS-CHOP promotes invasion in myxoid liposarcoma through a SRC/FAK/RHO/ROCK-dependent pathway

Deregulated SRC/FAK signaling leads to enhanced migration and invasion in many types of tumors. In myxoid and round cell liposarcoma (MRCLS), an adipocytic tumor characterized by the expression of the fusion oncogene FUS-CHOP, SRC have been found as one of the most activated kinases. Here we used a cell-of-origin model of MRCLS and an MRCLS cell line to thoroughly characterize the mechanisms of cell invasion induced by FUS-CHOP using in vitro (3D spheroid invasion assays) and in vivo (chicken chorioallantoic membrane model) approaches. FUS-CHOP expression activated SRC-FAK signaling and increased the invasive ability of MRCLS cells. In addition, FAK expression was found to significantly correlate with tumor aggressiveness in sarcoma patient samples. The involvement of SRC/FAK activation in FUS-CHOP–mediated invasion was further confirmed using the SRC inhibitor dasatinib, the specific FAK inhibitor PF-573228, and FAK siRNA. Notably, dasatinib and PF573228 could also efficiently block the invasion of cancer stem cell subpopulations. Downstream of SRC/FAK signaling, we found that FUS-CHOP expression increases the levels of the RHO/ROCK downstream effector phospho-MLC2 (T18/S19) and that this activation was prevented by dasatinib or PF573228. Moreover, the ROCK inhibitor RKI-1447 was able to completely abolish invasion in FUS-CHOP–expressing cells. These data uncover the involvement of SRC/FAK/RHO/ROCK signaling axis in FUS-CHOP–mediated invasion, thus providing a rationale for testing inhibitors of this pathway as potential novel antimetastatic agents for MRCLS treatmentPeer ReviewedPostprint (author's final draft

Tras 20 meses de evolución la paciente está asintomática sin evidencia de progresión de la enfermedadWe report a patient with back pain and neurological symptoms several months of evolution to the test image was wrongly diagnosed with spinal metastases without prior confirmation with biopsy. The progressive deterioration of neurological and new imaging confirmed the diagnosis of spinal tuberculosis. The patient was treated by curettage, debridement, anterior interbody fusion and internal fixation in addition to medical treatment. There neurological recovery of sensory function, motor and sphincter almost complete month after the intervention. After 20 months of evolution, the patient is asymptomatic without evidence of disease progressio

Prostate cancer, PI3K, PTEN and prognosis

Loss of function of the PTEN tumour suppressor, resulting in dysregulated activation of the phosphoinositide 3-kinase (PI3K) signalling network, is recognized as one of the most common driving events in prostate cancer development. The observed mechanisms of PTEN loss are diverse, but both homozygous and heterozygous genomic deletions including PTEN are frequent, and often accompanied by loss of detectable protein as assessed by immunohistochemistry (IHC). The occurrence of PTEN loss is highest in aggressive metastatic disease and this has driven the development of PTEN as a prognostic biomarker, either alone or in combination with other factors, to distinguish indolent tumours from those likely to progress. Here, we discuss these factors and the consequences of PTEN loss, in the context of its role as a lipid phosphatase, as well as current efforts to use available inhibitors of specific components of the PI3K/PTEN/TOR signalling network in prostate cancer treatment.</jats:p

A Fuzzy Syllogistic Reasoning Schema for Generalized Quantifiers

    Get PDF
In this paper, a new approximate syllogistic reasoning schema is described that expands some of the approaches expounded in the literature into two ways: (i) a number of different types of quantifiers (logical, absolute, proportional, comparative and exception) taken from Theory of Generalized Quantifiers and similarity quantifiers, taken from statistics, are considered and (ii) any number of premises can be taken into account within the reasoning process. Furthermore, a systematic reasoning procedure to solve the syllogism is also proposed, interpreting it as an equivalent mathematical optimization problem, where the premises constitute the constraints of the searching space for the quantifier in the conclusion.Comment: 22 pages, 6 figures, journal pape

Flue gas adsorption by single-wall carbon nanotubes: A Monte Carlo study

    Get PDF
Adsorption of flue gases by single-wall carbon nanotubes (SWCNT) has been studied by means of Monte Carlo simulations. The flue gas is modeled as a ternary mixture of N2, CO2, and O2, emulating realistic compositions of the emissions from power plants. The adsorbed flue gas is in equilibrium with a bulk gas characterized by temperature T, pressure p, and mixture composition. We have considered different SWCNTs with different chiralities and diameters in a range between 7 and 20 Ã…. Our results show that the CO2 adsorption properties depend mainly on the bulk flue gas thermodynamic conditions and the SWCNT diameter. Narrow SWCNTs with diameter around 7 Ã… show high CO2 adsorption capacity and selectivity, but they decrease abruptly as the SWCNT diameter is increased. For wide SWCNT, CO2 adsorption capacity and selectivity, much smaller in value than for the narrow case, decrease mildly with the SWCNT diameter. In the intermediate range of SWCNT diameters, the CO2 adsorption properties may show a peculiar behavior, which depend strongly on the bulk flue gas conditions. Thus, for high bulk CO2 concentrations and low temperatures, the CO2 adsorption capacity remains high in a wide range of SWCNT diameters, although the corresponding selectivity is moderate. We correlate these findings with the microscopic structure of the adsorbed gas inside the SWCNTs.Peer reviewe
