    Understanding about determinant factors of poverty will help policy maker to ensurethat the poor get benefit from the economic policy. In general, this study aim toanalyze the changes of determinant factors of poverty before and after theimplementation of fiscal decentralization. Using the model, this study found that incommunity factor there are some variables have change from 1999 to 2002. One ofthese variables is road infrastructure. In 2002 (after fiscal decentralization period),the quality of road was worse than before so the impact is the poverty was increase.This study shows that if both central and local government concern with povertyreduction then they have to notice some sectors such as agriculture, education, familyhealth, and infrastructure.     Generating a study at STT Wastukancana Purwakarta is strongly important to get an effective way to study. As the allocation of rooms to the schedule made is still manual, it is possible there are some room service clashes. To solve that problem, a study scheduling using computerization is needed in order to make no clash of room service. The use of genetic algorithm to solve the problem is able to get the optimal solution in generating the schedule with chromosome representative as an integer from each of data primary key, the beginning of population initialization, the selection with rank-selection method, two-points crossover method, and mutation. From the test, the result points out that the optimal schedule with 416 pengampu data is generated when a number of population are 30, a number of generations are 150, the crossover probability value (Pc) is 0.4, and the mutation probability value (Pm) is 0.37

    Salah satu prinsip perkawinan Islam adalah Menguatkan ikatan perkawinan agar berlangsung selama-selamanya. Karena itu, segala usaha harus dilakukan agar persekutuan dalam ikatan perkawinan itu dapat terus berlanjutan. Namun dalam perjalanan mengarungi bahtera rumah tangga, tidak selamanya mulus seperti yang diharapkan, pasti akan menhadapi sebuah rintangan yang menjadi permasalahan dalam rumah tangga. Apabila permasalahan itu tidak dapat diselesaikan dengan baik akan menimbulkan perselisihan dan akhir dengan perceraian. Apabila rumah tangga sudah tidak dapat dipertahankan dan bila mempertahankan menimbulkan perselisihan dan penderitaan yang berpangjangan. Agama Islam tidak Mengikat mati perkawinan akan tetapi tidak pula mempermudahkan. Majelis Agama Islam Wilayah Patani telah menerima memeriksa dan memutus setiap perkara yang masuk di Majelis Agama Islam Wilayah Patani, khususnya perkara cerai gugat yang merupakan perkara tertinggi sebanyak 1182 perkara dari tahun 2016-2018 dan terus mengalami peningkatan. Melihat data tersebut penyusun tertarik untuk meneliti Perceraian Dalam Perkawinan dikarenakan Kekerasan Dalam Rumah Tangga di Majelis Agama islam Wilayah Patani. Adapun rumusan masalah yang menjadi fokos dalam penelitian ini adalah Faktor-faktor apa saja yang menyebabkan terjadi perceraian di Majelis Agama Islam Patani Selatan Thailand, Bagaimana Kewenangan terhadap perceraian akibat kekerasan dalam rumah tangga dan Bagaimana cara penyelesaian Majelis Agama Islam terhadap kasus kekerasan dalam rumah tangga. Metode yang digunakan dalam penlitian ini adalah pendekatan Kualitatif deskriptif dengan metode pengumpul data melalui observasi, wawancara dan dokumentasi. Hasil penelitian menunjukkan bahwa factor-faktor yang menyebab terjadinya cerai gugat yang salah satu tertinggi factor kekerasan dalam rumah tangga dan Majlis Agama Islam memiliki kewenangan yang hakiki dalam segala bidang yang bersangkutan mengenai persoalan agama termasuk di dalamnya cerai gugat. Namun, sebelum persoalan sampai kepada Majelis Agama Islam terlebih dahulu akan diselesaikan oleh imam masjid di tiap daerah

    Reverting estrogen-receptor-negative phenotype in HER-2-overexpressing advanced breast cancer patients exposed to trastuzumab plus chemotherapy

    INTRODUCTION: The amounts of estrogen receptor (ER) and progesterone receptor (PgR) in a primary tumor are predictive of the response to endocrine therapies of breast cancer. Several patients with ER-positive primary tumors relapse after adjuvant endocrine therapy with no ER expression in the recurrent tissue; much fewer with a recurrent disease after an ER-negative primary tumor may become endocrine responsive. These sequences of events indicate that a phenotype based on ER expression may not be a permanent feature of breast cancer. METHODS: Ten patients with advanced breast cancer whose tumors overexpressed HER-2, but not ER or PgR, were treated with weekly trastuzumab at standard doses with or without chemotherapy. RESULTS: Three out of 10 patients showed overexpression of ERs first appearing after 9, 12 and 37 weeks, respectively, from the initiation of trastuzumab. Two of these patients were subsequently treated with endocrine therapy alone: one of them received letrozole for 3 years without evidence of progression. CONCLUSION: Therapeutic targets enabling the appearance of an endocrine responsive disease may increase treatment options for patients with breast cancer. Furthermore, these clinical data suggest that an ER-negative phenotype is a multi-step process with a reversible repression modality, and that some ER-negative tumors may either revert to an ER-positive phenotype, allowing an endocrine treatment to be effective

    Asc1 Supports Cell-Wall Integrity Near Bud Sites by a Pkc1 Independent Mechanism

    Background: The yeast ribosomal protein Asc1 is a WD-protein family member. Its mammalian ortholog, RACK1 was initially discovered as a receptor for activated protein C kinase (PKC) that functions to maintain the active conformation of PKC and to support its movement to target sites. In the budding yeast though, a connection between Asc1p and the PKC signaling pathway has never been reported. Methodology/Principal Findings: In the present study we found that asc1-deletion mutant (asc1D) presents some of the hallmarks of PKC signaling mutants. These include an increased sensitivity to staurosporine, a specific Pkc1p inhibitor, and susceptibility to cell-wall perturbing treatments such as hypotonic- and heat shock conditions and zymolase treatment. Microscopic analysis of asc1D cells revealed cell-wall invaginations near bud sites after exposure to hypotonic conditions, and the dynamic of cells ’ survival after this stress further supports the involvement of Asc1p in maintaining the cell-wall integrity during the mid-to late stages of bud formation. Genetic interactions between asc1 and pkc1 reveal synergistic sensitivities of a double-knock out mutant (asc1D/pkc1D) to cell-wall stress conditions, and high basal level of PKC signaling in asc1D. Furthermore, Asc1p has no effect on the cellular distribution or redistribution of Pkc1p at optimal or at cell-wall stress conditions.     Functional interaction between mouse erbB3 and wild-type rat c-neu in transgenic mouse mammary tumor cells

    INTRODUCTION: Co-expression of several receptor tyrosine kinases (RTKs), including erbB2 and erbB3, is frequently identified in breast cancers. A member of the RTK family, the kinase-deficient erbB3 can activate downstream signaling via heterodimer formation with erbB2. We studied the expression of RTK receptors in mammary tumors from the wild-type (wt) rat c-neu transgenic model. We hypothesized that physical and functional interactions between the wt rat neu/ErbB2 transgene and mouse ErbB3-encoded proteins could occur, activating downstream signaling and promoting mammary oncogenesis. METHODS: Immunohistochemical and Western blot analyses were performed to study the expression of rat c-neu/ErbB2 and mouse erbB3 in mammary tumors and tumor-derived cell lines from the wt rat c-neu transgenic mice. Co-immunoprecipitation methods were employed to quantitate heterodimerization between the transgene-encoded protein erbB2 and the endogenous mouse erbB3. Tumor cell growth in response to growth factors, such as Heregulin (HRG), epidermal growth factor (EGF), or insulin-like growth factor-1 (IGF-1), was also studied. Post-HRG stimulation, activation of the RTK downstream signaling was determined by Western blot analyses using antibodies against phosphorylated Akt and mitogen-activated protein kinase (MAPK), respectively. Specific inhibitors were then used with cell proliferation assays to study the phosphoinositide-3 kinase (PI-3K)/Akt and MAPK kinase (MEK)/MAPK pathways as possible mechanisms of HRG-induced tumor cell proliferation. RESULTS: Mammary tumors and tumor-derived cell lines frequently exhibited elevated co-expression of erbB2 and erbB3. The transgene-encoded protein erbB2 formed a stable heterodimer complex with endogenous mouse erbB3. HRG stimulation promoted physical and functional erbB2/erbB3 interactions and tumor cell growth, whereas no response to EGF or IGF-1 was observed. HRG treatment activated both the Akt and MAPK pathways in a dose- and time-dependent manner. Both the PI-3K inhibitor LY 294002 and MEK inhibitor PD 98059 significantly decreased the stimulatory effect of HRG on tumor cell proliferation. CONCLUSION: The co-expression of wt rat neu/ErbB2 transgene and mouse ErbB3, with physical and functional interactions between these two species of RTK receptors, was demonstrated. These data strongly suggest a role for erbB3 in c-neu (ErbB2)-associated mammary tumorigenesis, as has been reported in human breast cancers

    Integrated-boost IMRT or 3-D-CRT using FET-PET based auto-contoured target volume delineation for glioblastoma multiforme - a dosimetric comparison

    <p>Abstract</p> <p>Background</p> <p>Biological brain tumor imaging using O-(2-[<sup>18</sup>F]fluoroethyl)-L-tyrosine (FET)-PET combined with inverse treatment planning for locally restricted dose escalation in patients with glioblastoma multiforme seems to be a promising approach.</p> <p>The aim of this study was to compare inverse with forward treatment planning for an integrated boost dose application in patients suffering from a glioblastoma multiforme, while biological target volumes are based on FET-PET and MRI data sets.</p> <p>Methods</p> <p>In 16 glioblastoma patients an intensity-modulated radiotherapy technique comprising an integrated boost (IB-IMRT) and a 3-dimensional conventional radiotherapy (3D-CRT) technique were generated for dosimetric comparison. FET-PET, MRI and treatment planning CT (P-CT) were co-registrated. The integrated boost volume (PTV1) was auto-contoured using a cut-off tumor-to-brain ratio (TBR) of ≥ 1.6 from FET-PET. PTV2 delineation was MRI-based. The total dose was prescribed to 72 and 60 Gy for PTV1 and PTV2, using daily fractions of 2.4 and 2 Gy.</p> <p>Results</p> <p>After auto-contouring of PTV1 a marked target shape complexity had an impact on the dosimetric outcome. Patients with 3-4 PTV1 subvolumes vs. a single volume revealed a significant decrease in mean dose (67.7 vs. 70.6 Gy). From convex to complex shaped PTV1 mean doses decreased from 71.3 Gy to 67.7 Gy. The homogeneity and conformity for PTV1 and PTV2 was significantly improved with IB-IMRT. With the use of IB-IMRT the minimum dose within PTV1 (61.1 vs. 57.4 Gy) and PTV2 (51.4 vs. 40.9 Gy) increased significantly, and the mean EUD for PTV2 was improved (59.9 vs. 55.3 Gy, p < 0.01). The EUD for PTV1 was only slightly improved (68.3 vs. 67.3 Gy). The EUD for the brain was equal with both planning techniques.</p> <p>Conclusion</p> <p>In the presented planning study the integrated boost concept based on inversely planned IB-IMRT is feasible. The FET-PET-based automatically contoured PTV1 can lead to very complex geometric configurations, limiting the achievable mean dose in the boost volume. With IB-IMRT a better homogeneity and conformity, compared to 3D-CRT, could be achieved.</p

    LNCaP Atlas: Gene expression associated with in vivo progression to castration-recurrent prostate cancer

    <p>Abstract</p> <p>Background</p> <p>There is no cure for castration-recurrent prostate cancer (CRPC) and the mechanisms underlying this stage of the disease are unknown.</p> <p>Methods</p> <p>We analyzed the transcriptome of human LNCaP prostate cancer cells as they progress to CRPC <it>in vivo </it>using replicate LongSAGE libraries. We refer to these libraries as the LNCaP atlas and compared these gene expression profiles with current suggested models of CRPC.</p> <p>Results</p> <p>Three million tags were sequenced using <it>in vivo </it>samples at various stages of hormonal progression to reveal 96 novel genes differentially expressed in CRPC. Thirty-one genes encode proteins that are either secreted or are located at the plasma membrane, 21 genes changed levels of expression in response to androgen, and 8 genes have enriched expression in the prostate. Expression of 26, 6, 12, and 15 genes have previously been linked to prostate cancer, Gleason grade, progression, and metastasis, respectively. Expression profiles of genes in CRPC support a role for the transcriptional activity of the androgen receptor (<it>CCNH, CUEDC2, FLNA, PSMA7</it>), steroid synthesis and metabolism (<it>DHCR24, DHRS7</it>, <it>ELOVL5, HSD17B4</it>, <it>OPRK1</it>), neuroendocrine (<it>ENO2, MAOA, OPRK1, S100A10, TRPM8</it>), and proliferation (<it>GAS5</it>, <it>GNB2L1</it>, <it>MT-ND3</it>, <it>NKX3-1</it>, <it>PCGEM1</it>, <it>PTGFR</it>, <it>STEAP1</it>, <it>TMEM30A</it>), but neither supported nor discounted a role for cell survival genes.</p> <p>Conclusions</p> <p>The <it>in vivo </it>gene expression atlas for LNCaP was sequenced and support a role for the androgen receptor in CRPC.</p
