Adaptive Optics Imaging of IRAS 18276-1431: a bipolar pre-planetary nebula with circumstellar "searchlight beams" and "arcs"
We present high-angular resolution images of the post-AGB nebula
IRAS18276-1431 (also known as OH17.7-2.0) obtained with the Keck II Adaptive
Optics (AO) system in its Natural Guide Star (NGS) mode in the Kp, Lp, and Ms
near-infrared bands. We also present supporting optical F606W and F814W HST
images as well as interferometric observations of the 12CO(J=1-0), 13CO(J=1-0),
and 2.6mm continuum emission with OVRO. The envelope of IRAS18276-1431 displays
a clear bipolar morphology in our optical and NIR images with two lobes
separated by a dark waist and surrounded by a faint 4.5"x3.4" halo. Our Kp-band
image reveals two pairs of radial ``searchlight beams'' emerging from the
nebula center and several intersecting, arc-like features. From our CO data we
derive a mass of M>0.38[D/3kpc]^2 Msun and an expansion velocity v_exp=17km/s
for the molecular envelope. The density in the halo follows a radial power-law
proportional to r^-3, which is consistent with a mass-loss rate increasing with
time. Analysis of the NIR colors indicates the presence of a compact central
source of ~300-500K dust illuminating the nebula in addition to the central
star. Modeling of the thermal IR suggests a two-shell structure in the dust
envelope: 1) an outer shell with inner and outer radius R_in~1.6E16cm and
R_out>~1.25E17cm, dust temperature T_d~105-50K, and a mean mass-loss rate of
Mdot~1E-3Msun/yr; and 2) an inner shell with R_in~6.3E14cm, T_dust~500-105K,
and Mdot~3E-5Msun/yr. An additional population of big dust grains (radius
a>~0.4mm) with T_dust=150-20K and mass M_dust=(0.16-1.6)E-3 [D/3kpc]^2 Msun can
account for the observed sub-mm and mm flux excess. The mass of the envelope
enclosed within R_out=1.25E17cm derived from SED modeling is ~1[D/3kpc]^2 Msun.Comment: 46 pages, 14 figures, 3 tables, accepted for publication in ApJ.

Identification of Very Red Counterparts of SiO Maser and OH/IR Objects in the GLIMPSE Survey
Using the 3.6/4.5/5.8/8.0 micron images with 1.2 arcsec pixel resolution from
the Spitzer/GLIMPSE survey, we investigated 23 masing and 18 very red objects
that were not identified in the 2MASS survey. Counterparts for all selected
objects were found in the GLIMPSE images. Color indices in these IR bands
suggest the presence of a high-extinction layer of more than a few tenths of a
solar mass in front of the central star. Furthermore, radio observations in the
SiO and H2O maser lines found characteristic maser-line spectra of the embedded
objects, e.g., the SiO J=1-0 line intensity in the v=2 state stronger than that
of the v=1 state, or very widespread H2O maser emission spectra. This indicates
that these objects are actually enshrouded by very thick circumstellar matter,
some of which cannot be ascribed to the AGB wind of the central star.
Individually interesting objects are discussed, including two newly found water

Growth Spectrum Complexity Dictates Aromatic Intensity in Coriander (Coriandrum sativum L.)

Advancements in availability and specificity of light-emitting diodes (LEDs) have facilitated trait modification of high-value edible herbs and vegetables through the fine manipulation of spectra. Coriander (Coriandrum sativum L.) is a culinary herb, known for its fresh, citrusy aroma, and high economic value. Studies into the impact of light intensity and spectrum on C. sativum physiology, morphology, and aroma are limited. Using a nasal impact frequency panel, a selection of key compounds associated with the characteristic aroma of coriander was identified. Significant differences (P < 0.05) were observed in the concentration of these aromatics between plants grown in a controlled environment chamber under the same photosynthetic photon flux density (PPFD) but custom spectra: red (100%), blue (100%), red + blue (RB, 50% equal contribution), or red + green + blue (RGB, 35.8% red: 26.4% green: 37.8% blue) wavelengths. In general, the concentration of aromatics increased with increasing numbers of wavelengths emitted alongside selective changes, e.g., the greatest increase in coriander-defining E-(2)-decenal occurred under the RGB spectrum. This change in aroma profile was accompanied by significant differences (P < 0.05) in light saturated photosynthetic CO2 assimilation, water-use efficiency (Wi), and morphology. While plants grown under red wavelengths achieved the greatest leaf area, RB spectrum plants were shortest and had the highest leaf:shoot ratio. Therefore, this work evidences a trade-off between sellable commercial morphologies with a weaker, less desirable aroma or a less desirable morphology with more intense coriander-like aromas. When supplemental trichromatic LEDs were used in a commercial glasshouse, the majority of compounds, with the exception of linalool, also increased showing that even as a supplement additional wavelength can modify the aromatic profile increasing its complexity. Lower levels of linalool suggest these plants may be more susceptible to biotic stress such as herbivory. Finally, the concentration of coriander-defining aromatics E-(2)-decenal and E-(2)-hexenal was significantly higher in supermarket pre-packaged coriander leaves implying that concentrations of aromatics increase after excision. In summary, spectra can be used to co-manipulate aroma profile and plant form with increasing spectral complexity leading to greater aromatic complexity and intensity. We suggest that increasing spectral complexity progressively stimulates signaling pathways giving rise to valuable economic traits
The institutional shaping of management: in the tracks of English individualism
Globalisation raises important questions about the shaping of economic action by cultural factors. This article explores the formation of what is seen by some as a prime influence on the formation of British management: individualism. Drawing on a range of historical sources, it argues for a comparative approach. In this case, the primary comparison drawn is between England and Scotland. The contention is that there is a systemic approach to authority in Scotland that can be contrasted to a personal approach in England. An examination of the careers of a number of Scottish pioneers of management suggests the roots of this systemic approach in practices of church governance. Ultimately this systemic approach was to take a secondary role to the personal approach engendered by institutions like the universities of Oxford and Cambridge, but it found more success in the different institutional context of the USA. The complexities of dealing with historical evidence are stressed, as is the value of taking a comparative approach. In this case this indicates a need to take religious practice as seriously as religious belief as a source of transferable practice. The article suggests that management should not be seen as a simple response to economic imperatives, but as shaped by the social and cultural context from which it emerges
Genome-wide methylation analysis identifies epigenetically inactivated candidate tumour suppressor genes in renal cell carcinoma
Identification of candidate tumour suppressor genes frequently methylated in renal cell carcinoma
Promoter region hyermethylation and transcriptional silencing is a frequent cause of tumour suppressor gene (TSG) inactivation in many types of human cancers. Functional epigenetic studies, in which gene expression is induced by treatment with demethylating agents, may identify novel genes with tumour-specific methylation. We used high-density gene expression microarrays in a functional epigenetic study of 11 renal cell carcinoma (RCC) cell lines. Twenty-eight genes were then selected for analysis of promoter methylation status in cell lines and primary RCC. Eight genes (BNC1, PDLIM4, RPRM, CST6, SFRP1, GREM1, COL14A1 and COL15A1) showed frequent (30% of RCC tested) tumour-specific promoter region methylation. Hypermethylation was associated with transcriptional silencing. Re-expression of BNC1, CST6, RPRM and SFRP1 suppressed the growth of RCC cell lines and RNA interference knock-down of BNC1, SFRP1 and COL14A1 increased the growth of RCC cell lines. Methylation of BNC1 or COL14A1 was associated with a poorer prognosis independent of tumour size, stage or grade. The identification of these epigenetically inactivated candidate RCC TSGs can provide insights into renal tumourigenesis and a basis for developing novel therapies and biomarkers for prognosis and detection. Functional epigenomics approach to identify methylated candidate tumour suppressor genes in renal cell carcinoma
Promoter region hypermethylation and transcriptional silencing is a frequent cause of tumour suppressor gene (TSG) inactivation in many human cancers. Previously, to identify candidate epigenetically inactivated TSGs in renal cell carcinoma (RCC), we monitored changes in gene expression in four RCC cell lines after treatment with the demethylating agent 5-azacytidine. This enabled us to identify HAI-2/SPINT2 as a novel epigenetically inactivated candidate RCC TSG. To identify further candidate TSGs, we undertook bioinformatic and molecular genetic evaluation of a further 60 genes differentially expressed after demethylation. In addition to HAI-2/SPINT2, four genes (PLAU, CDH1, IGFB3 and MT1G) had previously been shown to undergo promoter methylation in RCC. After bioinformatic prioritisation, expression and/or methylation analysis of RCC cell linesÂąprimary tumours was performed for 34 genes. KRT19 and CXCL16 were methylated in RCC cell lines and primary RCC; however, 22 genes were differentially expressed after demethylation but did not show primary tumour-specific methylation (methylated in normal tissue (n=1); methylated only in RCC cell lines (n=9) and not methylated in RCC cell lines (n=12)). Re-expression of CXCL16 reduced growth of an RCC cell line in vitro. In a summary, a functional epigenomic analysis of four RCC cell lines using microarrays representing 11â000 human genes yielded both known and novel candidate TSGs epigenetically inactivated in RCC, suggesting that this is valid strategy for the identification of novel TSGs and biomarkers
Mouse Middle Ear Ion Homeostasis Channels and Intercellular Junctions
The middle ear contains homeostatic mechanisms that control the movement of ions and fluids similar to those present in the inner ear, and are altered during inflammation.The normal middle ear cavity is fluid-free and air-filled to allow for effective sound transmission. Within the inner ear, the regulation of fluid and ion movement is essential for normal auditory and vestibular function. The same ion and fluid channels active in the inner ear may have similar roles with fluid regulation in the middle ear.Middle and inner ears from BALB/c mice were processed for immunohistochemistry of 10 specific ion homeostasis factors to determine if similar transport and barrier mechanisms are present in the tympanic cavity. Examination also was made of BALB/c mice middle ears after transtympanic injection with heat-killed Haemophilus influenza to determine if these channels are impacted by inflammation.The most prominent ion channels in the middle ear included aquaporins 1, 4 and 5, claudin 3, ENaC and Na(+),K(+)-ATPase. Moderate staining was found for GJB2, KCNJ10 and KCNQ1. The inflamed middle ear epithelium showed increased staining due to expected cellular hypertrophy. Localization of ion channels was preserved within the inflamed middle ear epithelium.The middle ear epithelium is a dynamic environment with intrinsic mechanisms for the control of ion and water transport to keep the middle ear clear of fluids. Compromise of these processes during middle ear disease may underlie the accumulation of effusions and suggests they may be a therapeutic target for effusion control
Investigation of the role of SDHB inactivation in sporadic phaeochromocytoma and neuroblastoma
Germline mutations in the succinate dehydrogenase (SDH) (mitochondrial respiratory chain complex II) subunit B gene, SDHB, cause susceptibility to head and neck paraganglioma and phaeochromocytoma. Previously, we did not identify somatic SDHB mutations in sporadic phaeochromocytoma, but SDHB maps to 1p36, a region of frequent loss of heterozygosity (LOH) in neuroblastoma as well. Hence, to evaluate SDHB as a candidate neuroblastoma tumour suppressor gene (TSG) we performed mutation analysis in 46 primary neuroblastomas by direct sequencing, but did not identify germline or somatic SDHB mutations. As TSGs such as RASSF1A are frequently inactivated by promoter region hypermethylation, we designed a methylation-sensitive PCR-based assay to detect SDHB promoter region methylation. In 21% of primary neuroblastomas and 32% of phaeochromocytomas (32%) methylated (and unmethylated) alleles were detected. Although promoter region methylation was also detected in two neuroblastoma cell lines, this was not associated with silencing of SDHB expression, and treatment with a demethylating agent (5-azacytidine) did not increase SDH activity. These findings suggest that although germline SDHB mutations are an important cause of phaeochromocytoma susceptibility, somatic inactivation of SDHB does not have a major role in sporadic neural crest tumours and SDHB is not the target of 1p36 allele loss in neuroblastoma and phaeochromocytoma
âContextualizing and Critiquing the Fantastic Prosumer: Power, Alienation and Hegemonyâ
Abstract The âprosumerâ has emerged to become a central figure in contemporary culture. Through the melding of production with consumption, both mainstream and progressive analysts conceptualize prosumption to be a liberating, empowering and, for some, a prospectively revolutionary institution. In this article, these fantastic associations are critically assessed using an approach that situates prosumption activities, including contemporary online applications often referred to as âcocreationâ, in three social-historical contexts: capitalism as a political economy dominated by mediated abstractions; capitalist society as a hierarchical order; and alienation as a pervasive norm. Among other conclusions, we find that prosumption (particularly its Web 2.0 iterations), constitutes an emerging hegemonic institution; one that effectively frames and contains truly radical imaginations while also tapping into existing predilections for commodity-focused forms of self-realization
