Requirement of the FATC domain of protein kinase Tel1 for localization to DNA ends and target protein recognition
Two large phosphatidylinositol 3-kinase-related protein kinases (PIKKs), ATM and ATR, play a central role in the DNA damage response pathway. PIKKs contain a highly conserved extreme C-terminus called the FRAP-ATM-TRRAP-C-terminal (FATC) domain. In budding yeast, ATM and ATR correspond to Tel1 and Mec1, respectively. In this study, we characterized functions of the FATC domain of Tel1 by introducing substitution or truncation mutations. One substitution mutation, termed tel1-21, and a truncation mutation, called tel1- Î"C, did not significantly affect the expression level. The tel1-21 mutation impaired the cellular response to DNA damage and conferred moderate telomere maintenance defect. In contrast, the tel1-Î"C mutation behaved like a null mutation, conferring defects in both DNA damage response and telomere maintenance. Tel1-21 protein localized to DNA ends as effectively as wild-type Tel1 protein, whereas Tel1-Î"C protein failed. Introduction of a hyperactive TEL1-hy mutation suppressed the tel1-21 mutation but not the tel1-Î"C mutation. In vitro analyses revealed that both Tel1-21 and Tel1-Î"C proteins undergo efficient autophosphorylation but exhibit decreased kinase activities toward the exogenous substrate protein, Rad53. Our results show that the FATC domain of Tel1 mediates localization to DNA ends and contributes to phosphorylation of target proteins. © 2015 Ogi, Goto, Ghosh, et al
Radiation performance of AlGaAs concentrator cells and expected performance of cascade structures
Aluminum gallium arsenide, GaAs, silicon and InGaAs cells have been irradiated with 1 MeV electrons and 37 MeV protons. These cells are candidates for individual cells in a cascade structure. Data is presented for both electron and proton irradiation studies for one sun and a concentration level of 100X AMO. Results of calculations on the radiation resistance of cascade cell structures based on the individual cell data are also presented. Both series connected and separately connected structures are investigated
Abundances of Disk Planetary Nebulae in M31 and the Radial Oxygen Gradient
We have obtained spectra of 16 planetary nebulae in the disk of M31 and
determined the abundances of He, N, O, Ne, S and Ar. Here we present the median
abundances and compare them with previous M31 PN disk measurements and with PNe
in the Milky Way. We also derive the radial oxygen gradient in M31, which is
shallower than that in the Milky Way, even accounting for M31's larger disk

Regional variation in digital cushion pressure in the forefeet of horses and elephants
In this study, we seek to understand how the digital cushion morphologies evident in horse and elephant feet influence internal and external foot pressures. Our novel use of invasive blood pressure monitoring equipment, combined with a pressure pad and force plate, enabled measurements of (ex vivo) digital cushion pressure under increasing axial loads in seven horse and six elephant forefeet. Linear mixed effects models (LMER) revealed that internal digital cushion pressures increase under load and differ depending on region; elephant feet experienced higher magnitudes of medial digital cushion pressure, whereas horse feet experienced higher magnitudes of centralised digital cushion pressure. Direct comparison of digital cushion pressure magnitudes in both species, at equivalent loads relative to body weight, revealed that medial and lateral pressures increased more rapidly with load in elephant limbs. Within the same approximate region, internal pressures exceeded external, palmar pressures (on the sole of the foot), supporting previous Finite Element (FE) predictions. High pressures and large variations in pressure may relate to the development of foot pathology, which is a major concern in horses and elephants in a captive/domestic environment
Quantum Monte Carlo method using phase-free random walks with Slater determinants
We develop a quantum Monte Carlo method for many fermions that allows the use
of any one-particle basis. It projects out the ground state by random walks in
the space of Slater determinants. An approximate approach is formulated to
control the phase problem with a trial wave function . Using
plane-wave basis and non-local pseudopotentials, we apply the method to Si
atom, dimer, and 2, 16, 54 atom (216 electrons) bulk supercells. Single Slater
determinant wave functions from density functional theory calculations were
used as with no additional optimization. The calculated binding
energy of Si2 and cohesive energy of bulk Si are in excellent agreement with

Crystal growth in fused solvent systems
The successful nucleation of bismuth germanate, B12GeO20 on a high quality seed and the growth of regions of single crystals of the same orientation of the seed are reported. Lead germanate, Pb5Ge3O11 was also identified as a ferroelectric crystal with large electrooptic and nonlinear optic constants. Solvent criteria, solvent/development, and crystal growth are discussed, and recommendations for future studies are included
