Preparation and properties of model glasses of historic enamels of the Green Vault Museum, Dresden
Many enamelled art objects of the Green Vauk Museum, Dresden, have been suffering a severe deterioration of the enamel layers during the last 20 to 30 years. The authors obtained three splinters of about 200 mg weight each of green, blue, and opaque turquoise enamel of the Aurangzeb ensemble. Model glasses of these originals were prepared which should match them as closely as possible. The investigations showed that the simple degree of network polymerization related to the SiO2/Na2O ratio is insufficient to understand their chemical properties even if one takes components such as MgO, CaO or AI2O3 also into consideration. To understand further structural influences IR, UV/VIS, and EPR spectroscopy studies were performed on the enamels, and it could be evidenced that components such as MnO, Fe2O3, CuO, CoO, PbO and SnO2 not only act as colorants or opacifiers but also as stabilizers of these materials. Taking this into account the chemical and physical behavior of the respective enamels in the museum can be elucidated

The appreciation of Rembrandt van Rijn has gone through remarkable changes. The 18th century academic criticism regarded him as typical example of the gifted but uneducated "pictor vulgaris". During the 19th century Rembrandt at first became a model for the anti-academic realists, thereupon a prototype of the "modern artist". The present thesis analyses the literature on Rembrandt since about 1850, differentiating between two stages of the artists reappreciation. In France and the Netherlands he was at first discribed as the selfconfident exponent of contemporary bourgeoisie. Towards the end of the 19th century the conception of Rembrandt as an unappreciated genius evolved. He was now considered as someone who followed his persuasions against the judgement of his fellow-citizens. The methodologically reflected study focusses on describing the rhetorical pattern of this modern idea of artistic subjectivity
Toxicological and endocrinolgical aspects of cytochrome P450 in breast and lung
Cytochrome P450 (CYP) enzymes have well characterized functions in
dispositionand pharmacokinetics of xenobiotics as well as in biosynthesis
and/or metabolismof endogenous compounds, such as steroid hormones, bile
acids, arachidonic acid andits derivatives. Cytochrome P450 mediated
metabolic activation is known to be a prerequisitefor the cytotoxic and
mutagenic activity of many environmental toxins and
procarcinogens.Xenobiotic and steroid-metabolizing P450 enzymes are found
in many tissues in thebody, although at lower levels than in the liver
and adrenal glands. It is the majorhypothesis of this thesis that
cell-specific expression of P450 can influence hormonemetabolism and
xenobiotic toxicity in target cells. In order to test this theorywe have
investigated P450 mediated activation of procarcinogens in the lung and
characterizedthe P450 content of rat and human breast.
Environmental factors have been implicated in the etiology of both lung
and breastcancer. Heterocyclic amines are a group of dietary
procarcinogens, that produce tumorsat multiple sites in experimental
animals and that have been proposed to play a rolein human cancer. The
capacity of rat and mouse lung microsomes to activate the
heterocyclicamines IQ, MelQx and PhlP was demonstrated using the Ames
mutagenicity test. Withinhibitory antibodies it was shown that CYP2A3
mediated a large part of the activationof IQ in rat lung microsomes.
CYP2A3 has been detected in rat lung and nasal mucosa, but not in the
liver and thus these studies underscore the importance of
studyingextrahepatic reactions.
The mammary gland is characterized by its hormonal regulation of growth
and differentiation. In studies on rat breast, we could show that the P450
profile also varies with ageand endocrine status of the animals.
Quantitation of enzyme levels was achieved afterpartial purification of
P450 and expression of P450 enzymes was analysed by Westernblot of the
protein in combination with RT PCR of mRNA. The studied P450 enzymeswere
distinctly expressed in virgin (CYP2A3, 2D6, 3A), pregnant (CYPIA1),
lactating(CYP IAI ) and post lactating (CYP2B and 3A) rat breast, whereas
other forms (CYP2EI,4A, aromatase) were found in all samples.
Immunohistochemical localization revealedthat CYP2A3 was specifically
expressed in a limited number of epithelial cells inthe ducts of
6-week-old rat breast. This finding permits future studies to
evaluatewhether these cells are at risk for genotoxic effects of
procarcinogens that areactivated by CYP2A3.
Analysis of P450 profiles in human breast samples from reduction
mammaplastiesclearly showed the usefulness of this tissue source for
characterization of humanbreast P450. A large interindvidual variation
was found in the expression of CYPIAI,2A6, 2B6 and 3A. These P450s are
known to metabolize many xenobiotics and drugs, such as tamoxifen as well
as steroid hormones. As in the rat CYP2E I, 4A and aromatasewere detected
in most human samples. In addition, CYP I B I and 2C were expressedin all
samples.
These studies clearly reveal cellular and developmental (in rat) as well
as interindividual(in human) differences in the P450 profile and indicate
that P450 forms in the mammarygland may play a role in in situ activation
of procarcinogens and metabolism of hormonesas well as antihormones.
