    Stochastic modelling of regional archaeomagnetic series

    SUMMARY We report a new method to infer continuous time series of the declination, inclination and intensity of the magnetic field from archeomagnetic data. Adopting a Bayesian perspective, we need to specify a priori knowledge about the time evolution of the magnetic field. It consists in a time correlation function that we choose to be compatible with present knowledge about the geomagnetic time spectra. The results are presented as distributions of possible values for the declination, inclination or intensity. We find that the methodology can be adapted to account for the age uncertainties of archeological artefacts and we use Markov Chain Monte Carlo to explore the possible dates of observations. We apply the method to intensity datasets from Mari, Syria and to intensity and directional datasets from Paris, France. Our reconstructions display more rapid variations than previous studies and we find that the possible values of geomagnetic field elements are not necessarily normally distributed. Another output of the model is better age estimates of archeological artefacts

    SummaryCalcitonin (CT) is a known potent inhibitor of bone resorption but its effect on cartilage enzymatic degradation has been incompletely studied. Salmon CT, at a concentration of 0, 0.1, 0.25, 0.5, 2.5 and 50 ng/ml, was added at 24 or 72 h to the culture medium of chondrocytes from human osteoarthritic hips and knees. The spontaneous collagenolytic activity, measured using a radiolabeled type II collagen, was inhibited by CT in a dose-dependent manner. However, CT had no effect on the total collagenolytic activity assayed after APMA activation. Stromelysin and plasmin activity, measured by degradation of casein and a synthetic substrate, were also unaffected by CT. Chondrocyte phospholipase A2 activity, assayed using a labeled specific substrate, was decreased by CT. Chondrocyte pre-incubation with CT significantly decreased the cell binding of labeled TNFα, but did not affect IL-1ÎČ cell binding. Attachment of chondrocytes on fibronectin was markedly stimulated by CT, while attachment to type II collagen was not. Significant effects were obtained using at least 2 or 5 ng/ml of CT. CT appears to decrease collagenolytic activity by decreasing its activation and/or increasing its inhibition by tissue inhibitors of metalloproteinases (TIMP). CT might act on osteoarthritic chondrocyte activation via mechanisms such as phospholipase A2 activity, human necrosis factor-α or fibronectin receptor expression

    Sponge-Inspired Dibromohemibastadin Prevents and Disrupts Bacterial Biofilms without Toxicity

    Since the banning of several families of compounds in antifouling (AF) coatings, the search for environmentally friendly AF compounds has intensified. Natural sources of AF compounds have been identified in marine organisms and can be used to create analogues in laboratory. In a previous study, we identified that dibromohemibastadin-1 (DBHB) is a promising AF molecule, leading to the inhibition of the activity of phenoloxidase, an enzyme involved in the attachment of mussels to surfaces. This paper describes the activity of the DBHB on biofilm formation and its detachment and on bacterial adhesion and communication: quorum sensing. DBHB has an anti-biofilm activity without affecting adhesion of marine and terrestrial bacteria at a dose of 10 ”M. Moreover, DBHB activity on quorum sensing (QS) is demonstrated at doses of 8 and 16 ”M. The activity of DBHB on QS is compared to kojic acid, a quorum sensing inhibitor already described. This compound is a promising environmentally friendly molecule potentially useful for the inhibition of microfouling

    ASSESSMENT OF URINARY HYDROXYPYRIDINIUM CROSS-LINKS MEASUREMENT IN OSTEOARTHRITIS

    The aim of this study is to re-evaluate urinary collagen cross-links, previously proposed as markers of osteoarthritis (OA). The urinary excretion of collagen cross-links, pyridinoline (PYD) and deoxypyridinoline (DPD), was measured using high-performance liquid chromatography (HPLC) in 114 patients with OA, 19 patients with rheumatoid arthritis (RA) and 40 healthy subjects. An increase in PYD and DPD, expressed per millimole of creatinine, was confirmed in RA. However, PYD and DPD in patients with hip OA, knee OA and polyOA were similar, and did not differ from controls. In patients with radiographic end-stage OA, PYD and DPD were significantly higher than in patients with an early OA, but not significantly higher than in controls. The PYD/DPD ratio did not vary with the OA stage. Thus, urinary collagen cross-links are not elevated in OA, but could reflect bone sclerosis and/or erosion in late O

    Antioxidant bioactivity of extracts from beach cast leaves of posidonia oceanica (L.) delile

    The marine environment is a generous source of biologically active compounds useful for human health. In 50 years, about 25,000 bioactive marine compounds have been identified, with an increase of 5% per year. Peculiar feature of algae and plants is the production of secondary metabolites, such as polyphenols, synthesized as a form of adaptation to environmental stress. Posidonia oceanica is a Mediterranean endemic and dominant seagrass and represents a biologically, ecologically and geologically important marine ecosystem. Within this study, methanolic and ethanolic extracts were generated from fresh and dried Posidonia oceanica leaves, with the aim to employ and valorize the beach cast leaves. The best yield and antioxidant activity (polyphenols content equal to 19.712 \ub1 0.496 mg GAE/g and DPPH IC50 of 0.090 \ub5g/\ub5L.) were recorded in 70% ethanol extracts (Gd-E4) obtained from leaves dried for two days at 60\u25e6C and ground four times. HPLC analyses revealed the presence of polyphenols compounds (the most abundant of which was chicoric acid) with antioxidant and beneficial properties. Bioactive properties of the Gd-E4 extracts were evaluated in vitro using fibroblast cells line (HS-68), subjected to UV induced oxidative stress. Pre-treatment of cells with Gd-E4 extracts led to significant protection against oxidative stress and mortality associated with UV exposure, thus highlighting the beneficial properties of antioxidants compounds produced by these marine plants against photo damage, free radicals and associated negative cellular effects. Beach cast leaves selection, processing and extraction procedures, and the in vitro assay results suggested the potentiality of a sustainable approach for the biotechnological exploitation of this resource and could serve a model for other marine resources

    Alignment of the medial tibial plateau affects the rate of joint space narrowing in the osteoarthritic knee

    SummaryObjectiveTo determine, in serial fixed-flexion (FF) radiographs of subjects with knee osteoarthritis (KOA), the importance of, and basis for, the effect of alignment of the medial tibial plateau (MTP), as determined by the inter-margin distance (IMD), on joint space narrowing (JSN).MethodsBaseline and 12-month X-rays of 590 knees with Kellgren and Lawrence grade (KLG) 2/3 OA from the public-release dataset of the Osteoarthritis Initiative (OAI) were assigned to subgroups based upon IMD at baseline (IMDBL) and the difference between IMDBL and IMD12mos. Relationships of JSN to IMDBL and to the difference between IMDBL and IMD12mos were evaluated.ResultsIn all 590 knees, mean JSN was 0.13±0.51mm (P<0.0001) and MTP alignment and replication of IMDBL in the 12-month film were, in general, poor. JSN was significantly (P=0.012) more rapid in Subgroup A (IMD≀1.70mm at both time points) than in Subgroup B (both IMDs>1.70mm): 0.15±0.43; 0.08±0.47. Within Subgroup B we identified a subset, Subgroup B1, in which, although alignment was poor at both time points, the large IMDBL was, by chance, highly reproduced by IMD12mos (difference between the two IMDs=0.01±0.27mm, NS). JSN in Subgroup B1 was 0.06±0.41mm and did not differ from that in other knees of Subgroup B (P=0.87). The standardized response mean (SRM) in all 590 knees and Subgroups A, B and B1 was 0.25, 0.34, 0.17 and 0.06, respectively. Independent of IMDBL, JSN correlated significantly with the difference between the IMDs in the two radiographs (r=0.17, P=0.0001).ConclusionSkewed MTP alignment in serial films and poor replication of IMDBL in the follow-up exam affect JSN measurement. The magnitude of change in joint space width (JSW) related to the poor quality of alignment that is common with the FF view jeopardizes accurate evaluation of JSN

    Characterization of nitrotyrosine as a biomarker for arthritis and joint injury

    Objectives: To characterize the utility of nitrotyrosine (NT) as a biomarker for arthritis and joint injury. Design: Synovial fluid, plasma, and urine from patients diagnosed with osteoarthritis (OA), rheumatoid arthritis (RA), anterior cruciate ligament (ACL) injury, meniscus injury and pseudogout, and knee-healthy volunteers were analyzed for concentrations of NT, nitrate and nitrite (NOx), matrix metalloproteinase (MMP)-3, MMP-1, MMP-9, more than 40 chemokines and cytokines. Results: In OA, plasma and synovial fluid NT were increased versus healthy volunteers. Synovial fluid to plasma NT ratios were elevated in OA patients. Synovial fluid from patients with ACL and meniscus injury and pseudogout had increased levels of NT (P < 0.001). In these samples, NT levels significantly correlated with ARGS-aggrecan neoepitope generated by aggrecanase cleavage of aggrecan (P <= 0.001), cross-linked C-telopeptides of type II collagen (P < 0.001), MMP-1 (P = 0.008), and MMP-3 (P <= 0.001). In RA, plasma NT decreased following 6 months of anti-tumor necrosis factor (TNF) treatment. For every 1.1% change in log(10) NT, there was a 1.0% change in the log(10) disease activity scores (DAS28-3 CRP). Both predicted and observed DAS28-3 CRP showed a robust linear relationship with NT. RA plasma NT positively correlated with CRP, MMP-3 and interferon gamma-induced protein 10. Conclusions: NT may serve as a useful biomarker for arthritis and joint injury. In RA, NT is highly correlated with several biomarkers and clinical correlates of disease activity and responds to anti-TNF therapy. (C) 2012 Osteoarthritis Research Society International.     Antifouling activity of novel polyisoprene-based coatings made from photocurable natural rubber derived oligomers

    Natural rubber is a renewable resource with a potential as precursor of a very wide range of novel polymers, including polyisoprene-based surfaces with antifouling (AF) activity. In this work, new ionic and non-ionic coatings were prepared by the photocrosslinking reaction of photosensitive cis-1,4- oligoisoprenes, bearing a variable number of ammonium groups. The photochemical crosslinking was achieved using radical (via acrylate groups) or cationic (via epoxy groups) processes. Surface properties of these coatings were studied by static contact angle measurements and AFM imaging. Assessment of bioactivity demonstrated that most of the resulting coatings showed AF potential against fouling organisms: growth inhibition of marine bacteria (Pseudoalteromonas elyakovii, Shewanella putrefaciens, Cobetia marina, Polaribacter irgensii, Vibrio aestuarianus) and fungi (Halosphaeriopsis mediosetigera, Asteromyces cruciatus, Lulworthia uniseptata, Zalerion sp., Monodictys pelagica); decreased adhesion of microalgae (Navicula jeffreyi, Cylindrotheca closterium, Chlorarachnion globosum, Pleurochrysis roscoffensis, Exanthemachrysis gayraliae, Amphora coffeaeformis); inhibition of attachment and/or germination of spores of Ulva intestinalis. The best AF activity was obtained with the ionic surfaces. These new coatings prepared from precursors obtained from natural rubber are in essence active by contact. As the biocidal functions are fixed covalently to the polymer chain, detectable release of biocidal products in the marine ecosystem is prevented so that a valuable environment-friendly alternative for new AF coatings is hereby proposed
