    Environmental Constraints Guide Migration of Malaria Parasites during Transmission

    Migrating cells are guided in complex environments mainly by chemotaxis or structural cues presented by the surrounding tissue. During transmission of malaria, parasite motility in the skin is important for Plasmodium sporozoites to reach the blood circulation. Here we show that sporozoite migration varies in different skin environments the parasite encounters at the arbitrary sites of the mosquito bite. In order to systematically examine how sporozoite migration depends on the structure of the environment, we studied it in micro-fabricated obstacle arrays. The trajectories observed in vivo and in vitro closely resemble each other suggesting that structural constraints can be sufficient to guide Plasmodium sporozoites in complex environments. Sporozoite speed in different environments is optimized for migration and correlates with persistence length and dispersal. However, this correlation breaks down in mutant sporozoites that show adhesion impairment due to the lack of TRAP-like protein (TLP) on their surfaces. This may explain their delay in infecting the host. The flexibility of sporozoite adaption to different environments and a favorable speed for optimal dispersal ensures efficient host switching during malaria transmission

    Performance of the multiband imaging photometer for SIRTF

    We describe the test approaches and results for the Multiband Imaging Photometer for SIRTF. To verify the performance within a `faster, better, cheaper' budget required innovations in the test plan, such as heavy reliance on measurements with optical photons to determine instrument alignment, and use of an integrating sphere rather than a telescope to feed the completed instrument at its operating temperature. The tests of the completed instrument were conducted in a cryostat of unique design that allowed us to achieve the ultra-low background levels the instrument will encounter in space. We controlled the instrument through simulators of the mission operations control system and the SIRTF spacecraft electronics, and used cabling virtually identical to that which will be used in SIRTF. This realistic environment led to confidence in the ultimate operability of the instrument. The test philosophy allowed complete verification of the instrument performance and showed it to be similar to pre-integration predictions and to meet the instrument requirements

    Factors influencing identification of and response to intimate partner violence: a survey of physicians and nurses

    BACKGROUND: Intimate partner violence against women (IPV) has been identified as a serious public health problem. Although the health care system is an important site for identification and intervention, there have been challenges in determining how health care professionals can best address this issue in practice. We surveyed nurses and physicians in 2004 regarding their attitudes and behaviours with respect to IPV, including whether they routinely inquire about IPV, as well as potentially relevant barriers, facilitators, experiential, and practice-related factors. METHODS: A modified Dillman Tailored Design approach was used to survey 1000 nurses and 1000 physicians by mail in Ontario, Canada. Respondents were randomly selected from professional directories and represented practice areas pre-identified from the literature as those most likely to care for women at the point of initial IPV disclosure: family practice, obstetrics and gynecology, emergency care, maternal/newborn care, and public health. The survey instrument had a case-based scenario followed by 43 questions asking about behaviours and resources specific to woman abuse. RESULTS: In total, 931 questionnaires were returned; 597 by nurses (59.7% response rate) and 328 by physicians (32.8% response rate). Overall, 32% of nurses and 42% of physicians reported routinely initiating the topic of IPV in practice. Principal components analysis identified eight constructs related to whether routine inquiry was conducted: preparedness, self-confidence, professional supports, abuse inquiry, practitioner consequences of asking, comfort following disclosure, practitioner lack of control, and practice pressures. Each construct was analyzed according to a number of related issues, including clinician training and experience with woman abuse, area of practice, and type of health care provider. Preparedness emerged as a key construct related to whether respondents routinely initiated the topic of IPV. CONCLUSION: The present study provides new insight into the factors that facilitate and impede clinicians' decisions to address the issue of IPV with their female patients. Inadequate preparation, both educational and experiential, emerged as a key barrier to routine inquiry, as did the importance of the "real world" pressures associated with the daily context of primary care practice

    Guidance for nuclear medicine staff on radiopharmaceuticals drug interaction

    Numerous drug interactions related to radiopharmaceuticals take place every day in hospitals many of which are not reported or detected. Information concerning this kind of reaction is not abundant, and nuclear medicine staff are usually overwhelmed by this information. To better understand this type of reaction, and to help nuclear medicine staff deal with it, a review of the literature was conducted. The results show that almost all of radiopharmaceuticals marketed around the world present drug interactions with a large variety of compounds. This suggests that a logical framework to make decisions based on reviews incorporating adverse reactions must be created. The review also showed that researchers undertaking a review of literature, or even a systematic review that incorporates drug interactions, must understand the rationale for the suggested methods and be able to implement them in their review. Additionally, a global effort should be made to report as many cases of drug interaction with radiopharmaceuticals as possible. With this, a complete picture of drug interactions with radiopharmaceuticals can be drawn.Diversos casos de interações medicamentosas com radiofármacos ocorrem diariamente na rotina hospitalar, contudo muitos deles não são notificados ou mesmo percebidos. Informações a respeito desse tipo de reação não é abundante e os profissionais da medicina nuclear muitas vezes estão assoberbados por essas informações. De modo a entender esse tipo de reação e auxiliar a medicina nuclear a lidar com essa situação uma revisão da literatura foi realizada. Os resultados mostraram que a totalidade dos radiofármacos comercializados no mundo apresentam interação medicamentosa com uma enorme variedade de outros medicamentos. Dessa forma sugere-se que revisões sobre radiofármacos inclua um capítulo sobre efeitos adversos. Além disso, um esforço mundial para notificar efeitos adversos deve ser realizado, pois somente dessa forma se terá um quadro real da situação referente interações medicamentosas com radiofármacos

    Combined Information from Retardation Factor (Rf) Values and Color Reactions on the Plate Greatly Enhances the Identification Power of Thin-Layer Chromatography in Systematic Toxicological Analysis

    A numerical color coding system has been developed to describe the colors of spots obtained after using location reagents in thin-layer chromatography (TLC). This system makes color reactions on the plate amenable to computer handling, so that the retardation factor (Rf) values plus color reactions can be used for identification of unknown substances in toxicological analysis. It is based on a series of four color reactions carried out in sequence on the same plate, and encoding of the observed color is done by means of a wheel of reference colors. The combined information of Rf values plus color reactions resulted in an eightfold increase in identification power in comparison with the information provided by the Rf value alone. Moreover, a single TLC system now provides a three- to four-times higher identification power than a single gas chromatography system like OV-1. The identification power of TLC can be enhanced even further by running two or more systems in parallel

Complexity metrics have been used in software engineering and business process management (BPM) to capture the degree of complexity of conceptual models. A vast array of metrics has been proposed for processes in BPM. The recent introduction of the Decision Model and Notation (DMN) standard provides opportunities to shift towards the Separation of Concerns paradigm when it comes to modelling processes and decisions. However, unlike for processes, no studies exist that address the representational complexity of DMN decision models. In this paper, we provide a first set of ten complexity metrics for the decision requirements level of the DMN standard by gathering insights from the process modelling and software engineering fields. Additionally, we offer a discussion on the evolution of those metrics and we provide directions for future research on DMN compexity.status: publishe

    Plasmodium sporozoite motility is essential for establishing malaria infections. It depends on initial adhesion to a substrate as well as the continuous turnover of discrete adhesion sites. Adhesion and motility are mediated by a dynamic actin cytoskeleton and surface proteins. The mode of adhesion formation and the integration of adhesion forces into fast and continuous forward locomotion remain largely unknown. Here, we use optical tweezers to directly trap individual parasites and probe adhesion formation. We find that sporozoites lacking the surface proteins TRAP and S6 display distinct defects in initial adhesion; trap(-) sporozoites adhere preferentially with their front end, while s6(-) sporozoites show no such preference. The cohesive strength of the initial adhesion site is differently affected by actin filament depolymerization at distinct adhesion sites along the parasite for trap(-) and s6(-) sporozoites. These spatial differences between TRAP and S6 in their functional interaction with actin filaments show that these proteins have nonredundant roles during adhesion and motility. We suggest that complex protein–protein interactions and signaling events govern the regulation of parasite gliding at different sites along the parasite. Investigating how these events are coordinated will be essential for our understanding of sporozoite gliding motility, which is crucial for malaria infection.
