    Enhancing the Nutritional Quality of Flour Tortillas: An Investigation of Consumer Receptivity to a Fortified Tortilla Product

    The majority of U.S. adults consume less than the recommended amount of whole grains, dietary fiber, and vegetables on a daily basis. Increased intake of these foods is linked with reduced risks of developing chronic diseases, such as heart attack, cardiovascular disease, and stroke. With increased prevalence of such chronic health conditions and inadequate intakes of whole grains and vegetables, the need for an appealing enriched food product is evident. Over the past decade, the tortilla industry has been the fastest-growing sector of the U.S. baking industry; therefore, enhancing the nutritional content of tortillas could result in healthier food products acceptable to consumers. This research aimed to determine the effects of altering the flour component and adding a legume ingredient, specifically lentils, on the sensory attributes and overall characteristics of flour tortillas. Sorghum, an ancient whole grain variety, is rich in antioxidants, dietary fiber, and iron. Lentils, a type of grain legume, are high in fiber, B vitamins, minerals, proteins, and complex carbohydrates. Ten phases of bench-top product development using standardized preparation techniques were completed, resulting in two optimal fortified tortilla product formulations. Sensory evaluations were conducted among college students and instructors (n=35) for sensory attributes and overall likeability of four samples: two fortified tortilla products, a commercial tortilla made with refined flour, and a commercial health-enhanced tortilla. Results indicate that fortified tortilla products enhanced with pureed cooked lentils and sorghum flour are acceptable among the sampled population. Future research opportunities include conducting sensory evaluations among a larger population, optimizing product formulation for use in commercial production, extending product shelf life, and further enhancing nutritional qualities of tortillas by reducing sodium and fat content

    Bat response to differing fire severity in mixed-conifer forest California, USA

    Wildlife response to natural disturbances such as fire is of conservation concern to managers, policy makers, and scientists, yet information is scant beyond a few well-studied groups (e.g., birds, small mammals). We examined the effects of wildfire severity on bats, a taxon of high conservation concern, at both the stand (<1 ha) and landscape scale in response to the 2002 McNally fire in the Sierra Nevada region of California, USA. One year after fire, we conducted surveys of echolocation activity at 14 survey locations, stratified in riparian and upland habitat, in mixed-conifer forest habitats spanning three levels of burn severity: unburned, moderate, and high. Bat activity in burned areas was either equivalent or higher than in unburned stands for all six phonic groups measured, with four groups having significantly greater activity in at least one burn severity level. Evidence of differentiation between fire severities was observed with some Myotis species having higher levels of activity in stands of high-severity burn. Larger-bodied bats, typically adapted to more open habitat, showed no response to fire. We found differential use of riparian and upland habitats among the phonic groups, yet no interaction of habitat type by fire severity was found. Extent of high-severity fire damage in the landscape had no effect on activity of bats in unburned sites suggesting no landscape effect of fire on foraging site selection and emphasizing stand-scale conditions driving bat activity. Results from this fire in mixed-conifer forests of California suggest that bats are resilient to landscape-scale fire and that some species are preferentially selecting burned areas for foraging, perhaps facilitated by reduced clutter and increased post-fire availability of prey and roost

    Molecular evidence for resurrection of 'Plesiochorus elongatus' (Digenea : Gorgoderidae) : an urinary bladder parasite of sea turtles

    Trematodes of the genus Plesiochorus were recovered from the urinary bladder of a stranded female adult loggerhead sea turtle, Caretta caretta, on a beach in Rio de Janeiro State, Brazil. Morphological analysis of the specimens revealed characteristics resembling the sub-species Plesiochorus cymbiformis elongatus rather than the recently synonymised Plesiochorus cymbiformis. Molecular phylogenetic analysis of the ITS2 region also showed that P. c. elongatus was distinct from P. cymbiformis and related taxa. Further analysis of the ITS2 revealed substantial differentiation between P. cymbiformis from the USA and Brazil and the newly sequenced P. c. elongatus from Brazil, while a previously unspecified Plesiochorus sp. from the USA closely related to the novel Brazilian P. c. elongatus was reconciled as a USA isolate of P. c. elongatus. Based on both the morphological and molecular data it is suggested that P. c. elongatus should be referred to as Plesiochorus elongatus and be considered as the second species in the genus

    Exceptional chemotherapy response in metastatic colorectal cancer associated with hyper-indel-hypermutated cancer genome and comutation of POLD1 and MLH1

    Purpose A 73-year-old woman with metastatic colon cancer experienced a complete response to chemotherapy with dose-intensified irinotecan that has been durable for 5 years.Wesequenced her tumor and germ line DNA and looked for similar patterns in publicly available genomic data from patients with colorectal cancer. Patients and Methods Tumor DNA was obtained from a biopsy before therapy, and germ line DNA was obtained from blood. Tumor and germline DNA were sequenced using a commercial panel with approximately 250 genes. Whole-genome amplification and exome sequencing were performed for POLE and POLD1. A POLD1 mutation was confirmed by Sanger sequencing. The somatic mutation and clinical annotation data files from the colon (n = 461) and rectal (n = 171) adenocarcinoma data sets were downloaded from The Cancer Genome Atlas data portal and analyzed for patterns of mutations and clinical outcomes in patients withPOLE- and/orPOLD1- mutated tumors. Results The pattern of alterations included APC biallelic inactivation and microsatellite instability high (MSI-H) phenotype, with somatic inactivation of MLH1 and hypermutation (estimated mutation rate > 200 per megabase). The extremely high mutation rate led us to investigate additional mechanisms for hypermutation, including loss of function of POLE. POLE was unaltered, but a related gene not typically associated with somatic mutation in colon cancer, POLD1, had a somatic mutation c.2171G > A[p.Gly724Glu]. Additionally, we noted that the high mutation rate was largely composed of dinucleotide deletions. A similar pattern of hypermutation (dinucleotide deletions, POLD1 mutations, MSI-H) was found in tumors from The Cancer Genome Atlas. Conclusion POLD1 mutation with associated MSI-H and hyper-indel-hypermutated cancer genome characterizes a previously unrecognized variant of colon cancer that was found in this patient with an exceptional response to chemotherapy

    Granzyme A-producing T helper cells are critical for acute graft-versus-host disease

    Acute graft-versus-host disease (aGVHD) can occur after hematopoietic cell transplant in patients undergoing treatment for hematological malignancies or inborn errors. Although CD4+ T helper (Th) cells play a major role in aGVHD, the mechanisms by which they contribute, particularly within the intestines, have remained elusive. We have identified a potentially novel subset of Th cells that accumulated in the intestines and produced the serine protease granzyme A (GrA). GrA+ Th cells were distinct from other Th lineages and exhibited a noncytolytic phenotype. In vitro, GrA+ Th cells differentiated in the presence of IL-4, IL-6, and IL-21 and were transcriptionally unique from cells cultured with either IL-4 or the IL-6/IL-21 combination alone. In vivo, both STAT3 and STAT6 were required for GrA+ Th cell differentiation and played roles in maintenance of the lineage identity. Importantly, GrA+ Th cells promoted aGVHD-associated morbidity and mortality and contributed to crypt destruction within intestines but were not required for the beneficial graft-versus-leukemia effect. Our data indicate that GrA+ Th cells represent a distinct Th subset and are critical mediators of aGVHD

    The role of ZntA in Klebsiella pneumoniae zinc homeostasis

    Klebsiella pneumoniae is an opportunistic Gram-negative pathogen that is a leading cause of healthcare-associated infections, including pneumonia, urinary tract infections, and sepsis. Essential to the colonization and infection by K. pneumoniae is the acquisition of nutrients, such as the transition metal ion zinc. Zinc has crucial structural and catalytic roles in the proteome of all organisms. Nevertheless, in excess, it has the potential to mediate significant toxicity by dysregulating the homeostasis of other transition elements, disrupting enzymatic processes, and perturbing metalloprotein cofactor acquisition. Here, we sought to elucidate the zinc detoxification mechanisms of K. pneumoniae, which remain poorly defined. Using the representative K. pneumoniae AJ218 strain, we showed that the P-type ATPase, ZntA, which is upregulated in response to cellular zinc stress, was the primary zinc efflux pathway. Deletion of zntA rendered K. pneumoniae AJ218 highly susceptible to exogenous zinc stress and manifested as an impaired growth phenotype and increased cellular accumulation of the metal. Loss of zntA also increased sensitivity to cadmium stress, indicating a role for this efflux pathway in cadmium resistance. Disruption of zinc homeostasis in the K. pneumoniae AJ218 ΔzntA strain also impacted manganese and iron homeostasis and was associated with increased production of biofilm. Collectively, this work showed the critical role of ZntA in K. pneumoniae zinc tolerance and provided a foundation for further studies on zinc homeostasis and the future development of novel antimicrobials to target this pathway. IMPORTANCE: Klebsiella pneumoniae is a leading cause of healthcare-associated infections, including pneumonia, urinary tract infections, and sepsis. Treatment of K. pneumoniae infections is becoming increasingly challenging due to high levels of antibiotic resistance and the rising prevalence of carbapenem-resistant, extended-spectrum β-lactamases producing strains. Zinc is essential to the colonization and infection by many bacterial pathogens but toxic in excess. This work described the first dissection of the pathways associated with resisting extracellular zinc stress in K. pneumoniae. This study revealed that the P-type ATPase ZntA was highly upregulated in response to exogenous zinc stress and played a major role in maintaining bacterial metal homeostasis. Knowledge of how this major bacterial pathogen resists zinc stress provided a foundation for antimicrobial development studies to target and abrogate their essential function.Eve A.     Association between walking pace and stroke incidence: findings from the UK Biobank prospective cohort study

    Background and Purpose— Stroke incidence in younger and middle-aged people is growing. Despite this, its associations in this subset of the stroke population are unknown, and prevention strategies are not tailored to meet their needs. This study examined the association between self-reported walking pace and incident stroke. Methods— Data from the UK Biobank were used in a prospective population-based study. Three hundred and sixty-three thousand, one hundred and thirty-seven participants aged 37 to 73 years (52% women) were recruited. The associations of self-reported walking pace with stroke incidence over follow-up were investigated using Cox proportional-hazard models. Results— Among 363,137 participants, 2705 (0.7%) participants developed a fatal or nonfatal stroke event over the mean follow-up period of 6.1 years (interquartile range, 5.4–6.7). Slow walking pace was associated with a higher hazard for stroke incidence (hazard ratio [HR], 1.45 [95% CI, 1.26–1.66]; P&lt;0.0001). Stroke incidence was not associated with walking pace among people &lt;65 years of age. However, slow walking pace was associated with a higher risk of stroke among participants aged ≥65 years (HR, 1.42 [95% CI, 1.17–1.72]; P&lt;0.0001). A higher risk for stroke was observed on those with middle (HR, 1.28 [95% CI, 1.01–1.63]; P=0.039) and higher (HR, 1.29 [95% CI, 1.05–1.69]; P=0.012) deprivation levels but not in the least deprived individuals. Similarly, overweight (HR, 1.30 [95% CI, 1.04–1.63]; P=0.019) and obese (HR, 1.33 [95% CI, 1.09–1.63]; P=0.004) but not normal-weight individuals had a higher risk of stroke incidence. Conclusions— Slow walking pace was associated with a higher risk of stroke among participants over 64 years of age in this population-based cohort study. The addition of the measurement of self-reported walking pace to primary care or public health clinical consultations may be a useful screening tool for stroke risk

    Integrating Biology and Access to Care in Addressing Breast Cancer Disparities: 25 Years’ Research Experience in the Carolina Breast Cancer Study

    Purpose of Review: To review research on breast cancer mortality disparities, emphasizing research conducted in the Carolina Breast Cancer Study, with a focus on challenges and opportunities for integration of tumor biology and access characteristics across the cancer care continuum. Recent Findings: Black women experience higher mortality following breast cancer diagnosis, despite lower incidence compared to white women. Biological factors, such as stage at diagnosis and breast cancer subtypes, play a role in these disparities. Simultaneously, social, behavioral, environmental, and access to care factors are important. However, integrated studies of biology and access are challenging and it is uncommon to have both data types available in the same study population. The central emphasis of phase 3 of the Carolina Breast Cancer Study, initiated in 2008, was to collect rich data on biology (including germline and tumor genomics and pathology) and health care access in a diverse study population, with the long-term goal of defining intervention opportunities to reduce disparities across the cancer care continuum. Summary: Early and ongoing research from CBCS has identified important interactions between biology and access, leading to opportunities to build greater equity. However, sample size, population-specific relationships among variables, and complexities of treatment paths along the care continuum pose important research challenges. Interdisciplinary teams, including experts in novel data integration and causal inference, are needed to address gaps in our understanding of breast cancer disparities

    Remote practicals in the time of coronavirus, a multidisciplinary approach

    Due to the COVID-19 pandemic, universities across the world have curtailed face to face teaching. Associated with this is the halt to the delivery of the practical experience required of engineering students. The Multidisciplinary Engineering Education (MEE) team at The University of Sheffield have responded to this problem in an efficient and effective way by recording laboratory experiences and putting videos, quizzes and data online for students to engage with. The focus of this work was on ensuring all Learning Outcomes (LOs) for modules and courses were preserved. Naturally, practical skills cannot be easily provided using this approach, but it is an effective way of getting students to interact with real data, uncertainty and equipment which they cannot access directly. A number of short case studies from across the range of engineering disciplines are provided to inspire and guide other educators in how they can move experiments on line in an efficient and effective manner. No student feedback is available at the time of writing, but anecdotal evidence is that this approach is at least acceptable for students and a way of collecting future feedback is suggested. The effort expended on this approach and the artefacts produced will support student learning after the initial disruption of the lockdown has passed

    D* Production in Deep Inelastic Scattering at HERA

    This paper presents measurements of D^{*\pm} production in deep inelastic scattering from collisions between 27.5 GeV positrons and 820 GeV protons. The data have been taken with the ZEUS detector at HERA. The decay channel D+(D0Kπ+)π+D^{*+}\to (D^0 \to K^- \pi^+) \pi^+ (+ c.c.) has been used in the study. The e+pe^+p cross section for inclusive D^{*\pm} production with 5<Q2<100GeV25<Q^2<100 GeV^2 and y<0.7y<0.7 is 5.3 \pms 1.0 \pms 0.8 nb in the kinematic region {1.3<pT(D±)<9.01.3<p_T(D^{*\pm})<9.0 GeV and η(D±)<1.5| \eta(D^{*\pm}) |<1.5}. Differential cross sections as functions of p_T(D^{*\pm}), η(D±),W\eta(D^{*\pm}), W and Q2Q^2 are compared with next-to-leading order QCD calculations based on the photon-gluon fusion production mechanism. After an extrapolation of the cross section to the full kinematic region in p_T(D^{*\pm}) and η\eta(D^{*\pm}), the charm contribution F2ccˉ(x,Q2)F_2^{c\bar{c}}(x,Q^2) to the proton structure function is determined for Bjorken xx between 2 \cdot 104^{-4} and 5 \cdot 103^{-3}.Comment: 17 pages including 4 figure
