    In the system of legal sources, apart from typical acts of local law created in accordance with the provisions of the Constitution, there are also acts that do not fall within the constitutional standards of law, although they concern, among other things, important social and economic issues. Such documents (planning acts, programmes, or strategies), although enacted by resolutions, often fail to contain generally binding regulations. Although this does not prevent them from having a binding force, they should nevertheless be made in compliance with the generally recognised rules of lawmaking, including the principles of proportionality, rationality, communicativeness and brevity. The generally applicable provisions often impose on local self-governments an obligation to create acts regulating important social issues, such as for instance programmes preventing and solving alcohol-related problems and determining the way of dealing with alcohol addiction in local conditions. However, these programmes are often developed in violation of the rules of a rational law-making process, and are frequently detached from the local reality, which makes them ineffective. Therefor it may be worth considering another way of responding to the problem of alcoholism, not necessarily by means of resolutions adopted annually. The obligation to develop a prevention and resolution programme addressing alcohol-related problems could be replaced, for example, by certain specific organisational measures, an educational programme, or the implementation of other measures. The law is not always the right way to achieve the goals set, but the above recommendation will requires statutory changes first, anyway.W systemie źródeł prawa wyróżnić można, obok typowych aktów prawa miejscowego stanowionych zgodnie z przepisami Konstytucji, także takie akty, które nie mieszczą się w standardach konstytucyjnych źródeł prawa, choć dotyczą istotnych kwestii społecznych, gospodarczych itp. Tego rodzaju dokumenty (np. akty planowania, programy, strategie itp.), mimo że są uchwalane w drodze uchwał, nie zawsze zawierają przepisy powszechnie obowiązujące. Bez względu jednak na moc wiążącą aktu normatywnego prawodawca powinien przestrzegać powszechnie uznawanych reguł stanowienia prawa, w tym zasady proporcjonalności, racjonalności, komunikatywności i zwięzłości. Przepisy powszechnie obowiązujące często nakładają na samorządy lokalne obowiązek tworzenia aktów regulujących ważne kwestie społeczne, np. program profilaktyki i rozwiązywania problemów alkoholowych dotyczący zagadnienia sposobu postępowania ze zjawiskiem nałogu alkoholowego w warunkach lokalnych. Programy te często opracowywane są z naruszeniem reguł racjonalnego tworzenia prawa, a ponadto oderwane są od lokalnej rzeczywistości, co powoduje, że są one nieskuteczne. Warto zatem rozważyć możliwość innego sposobu reagowania na zjawisko alkoholizmu, nie za pomocą corocznie uchwalanych uchwał. Obowiązek sporządzania programu profilaktyki i rozwiązywania problemów alkoholowych mógłby zostać zastąpiony np. odpowiednimi działaniami organizacyjnymi, edukacyjnymi itp. Prawo nie zawsze jest właściwym sposobem osiągania zakładanych celów. Wymaga to jednak zmian ustawowych

    CrossCheck:toward passive sensing and detection of mental health changes in people with schizophrenia

    Early detection of mental health changes in individuals with serious mental illness is critical for effective intervention. CrossCheck is the first step towards the passive monitoring of mental health indicators in patients with schizophrenia and paves the way towards relapse prediction and early intervention. In this paper, we present initial results from an ongoing randomized control trial, where passive smartphone sensor data is collected from 21 outpatients with schizophrenia recently discharged from hospital over a period ranging from 2-8.5 months. Our results indicate that there are statistically significant associations between automatically tracked behavioral features related to sleep, mobility, conversations, smartphone usage and self-reported indicators of mental health in schizophrenia. Using these features we build inference models capable of accurately predicting aggregated scores of mental health indicators in schizophrenia with a mean error of 7.6% of the score range. Finally, we discuss results on the level of personalization that is needed to account for the known variations within people. We show that by leveraging knowledge from a population with schizophrenia, it is possible to train accurate personalized models that require fewer individual-specific data to quickly adapt to new user

    Este proyecto de investigación se cuestiona sobre la estructura organizativa de la Educación Técnica Profesional en escuelas de la ciudad de Misiones y las características didácticas y pedagógicas que asumen los procesos de enseñanza y aprendizaje en esta modalidad educativa, para el campo de las Ciencias Económicas. La reestructuración del Sistema Educativo supone el establecimiento de acuerdos y redefiniciones al interior de las escuelas, en relación con la norma que propicia la vinculación de la educación, el trabajo y la producción, y la realización de prácticas educativas que permitan a los estudiantes el manejo de tecnologías o brinden la experiencia adecuada a su formación y orientación vocacional. Por ello realizaremos una revisión crítica de las documentaciones, marcos legales y orientaciones curriculares, así como de su impacto en las prácticas educativas desde la perspectiva de los actores, teniendo presente las peculiaridades de esta modalidad formativa

    Characterization of Passive CMOS Strip Sensors

    Recent advances in CMOS imaging sensor technology , e.g. in CMOS pixel sensors, have proven that the CMOS process is radiation tolerant enough to cope with certain radiation levels required for tracking layers in hadron collider experiments. With the ever-increasing area covered by silicon tracking detectors cost effective alternatives to the current silicon sensors and more integrated designs are desirable. This article describes results obtained from laboratory measurements of silicon strip sensors produced in a passive p-CMOS process. Electrical characterization and charge collection measurements with a 90Sr source and a laser with infrared wavelength showed no effect of the stitching process on the performance of the sensor.Comment: 6 pages, 16 figure

    ASSOCIATION OF THE GLUCOCORTICOID RECEPTOR GENE POLYMORPHISMS AND THEIR INTERACTION WITH STRESSFUL LIFE EVENTS IN POLISH ADOLESCENT GIRLS WITH ANOREXIA NERVOSA

    Background: Disturbances in stress response mechanisms and hypothalamic-pituitary-adrenal axis (HPA) functioning are considered important factors involved in the pathophysiology of anorexia nervosa (AN). Thus, genetic variations in the end effector of HPA - glucocorticoid receptor gene and relationships to stressful life events (SLE) may be connected to a higher risk of illness. The aim of the study was examining the association between glucocorticoid receptor gene (NR3C1) polymorphisms and risk factors among stressful life events in AN patients. Subjects and methods: This study comprised 256 patients with AN and 167 control subjects. The questionnaires examining brief history of the mother’s pregnancy and long-acting stress factors, as well as life events checklist to assess stressful life events during the 6 months prior to hospitalization were used. The eight common SNPs (rs6198, rs6191, rs6196, rs258813, rs33388, rs41423247, rs56149945 and rs10052957) of NR3C1 gene were genotyped. Results: The association of five polymorphisms (rs6191, rs258813, rs33388, rs41423247 and rs10052957) and one complex allele (TCAGT) of NR3C1 gene with increased risk of AN were found. However, no significant correlations between early, longacting and predicting hospitalization SLE and any of the analyzed polymorphisms were observed. Conclusions: The results confirm that the NR3C1 gene is associated with AN risk regardless of the type of stressful triggering factors

    Investigation of the effects of phthalates on in vitro thyroid models with RNA-Seq and ATAC-Seq

    IntroductionPhthalates are a class of endocrine-disrupting chemicals that have been shown to negatively correlate with thyroid hormone serum levels in humans and to cause a state of hyperactivity in the thyroid. However, their mechanism of action is not well described at the molecular level.MethodsWe analyzed the response of mouse thyroid organoids to the exposure to a biologically relevant dose range of the phthalates bis(2-ethylhexyl) phthalate (DEHP), di-iso-decylphthalate (DIDP), di-iso-nonylphthalate (DINP), and di-n-octylphthalate (DnOP) for 24 h and simultaneously analyzed mRNA and miRNA expression via RNA sequencing. Using the expression data, we performed differential expression and gene set enrichment analysis. We also exposed the human thyroid follicular epithelial cell line Nthy-ori 3-1 to 1 µM of DEHP or DINP for 5 days and analyzed changes in chromatin accessibility via ATAC-Seq.ResultsDose-series analysis showed how the expression of several genes increased or decreased at the highest dose tested. As expected with the low dosing scheme, the compounds induced a modest response on the transcriptome, as we identified changes in only mmu-miR-143-3p after DINP treatment and very few differentially expressed genes. No effect was observed on thyroid markers. Ing5, a component of histones H3 and H4 acetylation complexes, was consistently upregulated in three out of four conditions compared to control, and we observed a partial overlap among the genes differentially expressed by the treatments. Gene set enrichment analysis showed enrichment in the treatment samples of the fatty acid metabolism pathway and in the control of pathways related to the receptor signalling and extracellular matrix organization. ATAC-Seq analysis showed a general increase in accessibility compared to the control, however we did not identify significant changes in accessibility in the identified regions.DiscussionWith this work, we showed that despite having only a few differentially expressed genes, diverse analysis methods could be applied to retrieve relevant information on phthalates, showing the potential of in vitro thyroid-relevant systems for the analysis of endocrine disruptors

    Type and duration of subsyndromal symptoms in youth with bipolar I disorder prior to their first manic episode

    Objectives: The aim of the present study was to systematically evaluate the prodrome to mania in youth. Methods: New-onset/worsening symptoms/signs of \u3e= moderate severity preceding first mania were systematically assessed in 52 youth (16.2 +/- 2.8 years) with a research diagnosis of bipolar I disorder (BD-I). Youth and/or caregivers underwent semi-structured interviews, using the Bipolar Prodrome Symptom Scale-Retrospective. Results: The mania prodrome was reported to start gradually in most youth (88.5%), with either slow (59.6%) or rapid (28.8%) deterioration, while a rapid-onset-and-deterioration prodrome was rare (11.5%). The manic prodrome, conservatively defined as requiring \u3e= 3 symptoms, lasted 10.3 +/- 14.4 months [95% confidence interval (CI): 6.3-14.4], being present for \u3e= 4 months in 65.4% of subjects. Among prodromal symptoms reported in \u3e= 50% of youth, three were subthreshold manic in nature (irritability: 61.5%, racing thoughts: 59.6%, increased energy/activity: 50.0%), two were nonspecific (decreased school/work functioning: 65.4%, mood swings/lability: 57.7%), and one each was depressive (depressed mood: 53.8%) or subthreshold manic/depressive (inattention: 51.9%). A decreasing number of youth had \u3e= 1 (84.6%), \u3e= 2 (48.1%), or \u3e= 3 (26.9%) \u27specific\u27 subthreshold mania symptoms (i.e., elation, grandiosity, decreased need for sleep, racing thoughts, or hypersexuality), lasting 9.5 +/- 14.9 months (95% CI: 5.0-14.0), 3.5 +/- 3.5 months (95% CI: 2.0-4.9), and 3.0 +/- 3.2 months (95% CI: 1.0-5.0) for \u3e= 1, \u3e= 2, or \u3e= 3 specific symptoms, respectively. Conclusions: In youth with BD-I, a relatively long, predominantly slowonset mania prodrome appears to be common, including subthreshold manic and depressive psychopathology symptoms. This suggests that early clinical identification and intervention may be feasible in bipolar disorder. Identifying biological markers associated with clinical symptoms of impending mania may help to increase chances for early detection and prevention before full mania

    Association of Genetic Markers with CSF Oligoclonal Bands in Multiple Sclerosis Patients

    Objective:to explore the association between genetic markers and Oligoclonal Bands (OCB) in the Cerebro Spinal Fluid (CSF) of Italian Multiple Sclerosis patients.Methods:We genotyped 1115 Italian patients for HLA-DRB1*15 and HLA-A*02. In a subset of 925 patients we tested association with 52 non-HLA SNPs associated with MS susceptibility and we calculated a weighted Genetic Risk Score. Finally, we performed a Genome Wide Association Study (GWAS) with OCB status on a subset of 562 patients. The best associated SNPs of the Italian GWAS were replicated in silico in Scandinavian and Belgian populations, and meta-analyzed.Results:HLA-DRB1*15 is associated with OCB+: p = 0.03, Odds Ratio (OR) = 1.6, 95% Confidence Limits (CL) = 1.1-2.4. None of the 52 non-HLA MS susceptibility loci was associated with OCB, except one SNP (rs2546890) near IL12B gene (OR: 1.45; 1.09-1.92). The weighted Genetic Risk Score mean was significantly (p = 0.0008) higher in OCB+ (7.668) than in OCB- (7.412) patients. After meta-analysis on the three datasets (Italian, Scandinavian and Belgian) for the best associated signals resulted from the Italian GWAS, the strongest signal was a SNP (rs9320598) on chromosome 6q (p = 9.4×10-7) outside the HLA region (65 Mb).Discussion:genetic factors predispose to the development of OCB
