PRADA: Predictable Allocations by Deferred Actions

    Get PDF
Modern hard real-time systems still employ static memory management. However, dynamic storage allocation (DSA) can improve the flexibility and readability of programs as well as drastically shorten their development times. But allocators introduce unpredictability that makes deriving tight bounds on an application\u27s worst-case execution time even more challenging. Especially their statically unpredictable influence on the cache, paired with zero knowledge about the cache set mapping of dynamically allocated objects leads to prohibitively large overestimations of execution times when dynamic memory allocation is employed. Recently, a cache-aware memory allocator, called CAMA, was proposed that gives strong guarantees about its cache influence and the cache set mapping of allocated objects. CAMA itself is rather complex due to its cache-aware implementations of split and merge operations. This paper proposes PRADA, a lighter but less general dynamic memory allocator with equally strong guarantees about its influence on the cache. We compare the memory consumption of PRADA and CAMA for a small set of real-time applications as well as synthetical (de-) allocation sequences to investigate whether a simpler approach to cache awareness is still sufficient for the current generation of real-time applications

Website Design and Trust Elements: A/B Testing on a Start-up\u27s Website

    Get PDF
Start-ups are young companies that are hardly known, especially during their early stages, by the relevant stakeholders. A start-up\u27s website is, therefore, often the first point of contact for potential customers, investors, or partners. Such a website usually explains the new product or service and presents the founding team with its competencies. The user\u27s perception of the website and its design can be crucial in determining whether the user is interested in getting in touch with the start-up or even considering the purchase of the respective product or service. User’s trust in the website and its operator is essential for this. The so-called trust elements, such as logos, testimonials, or seals, are intended to create trust on websites. So far, the influence of these elements on user behaviour has hardly been empirically proven in a real-life context. Therefore, we have applied the method of A/B testing to the website of a fictive start-up. Trust elements were placed on one variant of the website (A), whereas on the other variant, there were none (B). The experiment shows that the duration of the user sessions does not differ between the two variants. However, more requests were made on the website variant with trust elements

Facile Production of the Pseudomonas aeruginosa Virulence Factor LasB in Escherichia coli for Structure-Based Drug Design

    Get PDF
The human pathogen Pseudomonas aeruginosa has a number of virulence factors at its disposal that play crucial roles in the progression of infection. LasB is one of the major virulence factors and exerts its effects through elastolytic and proteolytic activities aimed at dissolving connective tissue and inactivating host defense proteins. LasB is of great interest for the development of novel pathoblockers to temper the virulence, but access has thus far largely been limited to protein isolated from Pseudomonas cultures. Here, we describe a new protocol for high-level production of native LasB in Escherichia coli. We demonstrate that this facile approach is suitable for the production of mutant, thus far inaccessible LasB variants, and characterize the proteins biochemically and structurally. We expect that easy access to LasB will accelerate the development of inhibitors for this important virulence factor

FORCE ANALYSIS OF THE UNDERWATER STATIONARY RUNNIG

    Get PDF
It aimed to analyze the vertical component of the ground reaction force in the underwater stationary running. The sample was composed by 6 subjects divided in two groups (Male Group and Female Group). The underwater stationary running was performed in two immersion levels: in the hip level and in the xiphoid process level. An underwater force plate was used. For data analysis descriptive statistics was used. The mean values of vertical GRF were 2,08BW for the MG and 1,69BW for the FG in the hip level; 1,15BW for the MG and 1,12BW for the FG in the xiphoid process level. The results showed the vertical component of the GRF is affected by the immersion level and by the frequency of the activity. Both factors should be considered by professionals who work with therapeutic or physical conditioning programs using the underwater stationary running

How Heterogeneous Pore Scale Distributions of Wettability Affect Infiltration into Porous Media

    Get PDF
Wettability is an important parameter that significantly determines hydrology in porous media, and it especially controls the flow of water across the rhizosphere—the soil-plant interface. However, the influence of spatially heterogeneous distributions on the soil particles surfaces is scarcely known. Therefore, this study investigates the influence of spatially heterogeneous wettability distributions on infiltration into porous media. For this purpose, we utilize a two-phase flow model based on Lattice-Boltzmann to numerically simulate the infiltration in porous media with a simplified geometry and for various selected heterogeneous wettability coatings. Additionally, we simulated the rewetting of the dry rhizosphere of a sandy soil where dry hydrophobic mucilage depositions on the particle surface are represented via a locally increased contact angle. In particular, we can show that hydraulic dynamics and water repellency are determined by the specific location of wettability patterns within the pore space. When present at certain locations, tiny hydrophobic depositions can cause water repellency in an otherwise well-wettable soil. In this case, averaged, effective contact angle parameterizations such as the Cassie equation are unsuitable. At critical conditions, when the rhizosphere limits root water uptake, consideration of the specific microscale locations of exudate depositions may improve models of root water uptake

An Efficient Way to Screen Inhibitors of Energy-Coupling Factor (ECF) Transporters in a Bacterial Uptake Assay

    Get PDF
Herein, we report a novel whole-cell screening assay using Lactobacillus casei as a model microorganism to identify inhibitors of energy-coupling factor (ECF) transporters. This promising and underexplored target may have important pharmacological potential through modulation of vitamin homeostasis in bacteria and, importantly, it is absent in humans. The assay represents an alternative, cost-effective and fast solution to demonstrate the direct involvement of these membrane transporters in a native biological environment rather than using a low-throughput in vitro assay employing reconstituted proteins in a membrane bilayer system. Based on this new whole-cell screening approach, we demonstrated the optimization of a weak hit compound (2) into a small molecule (3) with improved in vitro and whole-cell activities. This study opens the possibility to quickly identify novel inhibitors of ECF transporters and optimize them based on structure–activity relationships
