    Betsy - A BPEL Engine Test System

    More than five years have passed since the final release of the long-desired OASIS standard of a process language for web service orchestration, the Web Services Business Process Execution Language (BPEL). The aim of this standard is to establish a universally accepted orchestration language that forms a core part of current service-oriented architectures and, because of standardisation, avoids vendor lock-in. High expectations, in academia and practice alike, have been set on it. By now, several fully conformant and highly scalable engines should have arrived in the market. The perception of many however, is that standard conformance in current engines is far from given. It is our aim to shed light on this situation. In this study, we present the tool betsy, a BPEL Engine Test System that allows for a fully-automatic assessment of the standard conformance of a given BPEL engine. We use it to examine the five most important open source BPEL engines available today. Betsy comes with a large set of engineindependent conformance test cases for assessing BPEL standard conformance. This enables us to give a view of the state of the art in BPEL support

    Chronic Immune Activation in HIV-1 Infection Contributes to Reduced Interferon Alpha Production via Enhanced CD40:CD40 Ligand Interaction

    Although a signature of increased interferon (IFN-)alpha production is observed in HIV-1 infection, the response of circulating plasmacytoid dendritic cells (PDC) to Toll-like receptor ligand stimulation is substantially impaired. This functional PDC deficit, which we specifically observed in HIV-1 infected individuals with less than 500 CD4+ T cells/µl, is not well understood. We provide evidence that the peripheral IFN-alpha production in HIV-1 infection is actively suppressed by the enhanced interaction of CD40 ligand (CD40L), a member of the tumor necrosis factor family, and its receptor CD40, which are both upregulated upon immune activation. Plasma levels of soluble CD40L were significantly higher in untreated HIV-1 infected individuals (n = 52) than in subjects on long-term antiretroviral therapy (n = 62, p<0.03) and in uninfected control donors (n = 16, p<0.001). Concomitantly, cell-associated CD40L and the expression of the receptor CD40 on the PDC were significantly upregulated in HIV-1 infection (p<0.05). Soluble and cell-associated CD40L inhibited the PDC-derived IFN-alpha production by CpG oligodeoxynucleotides dose-dependently. This suppressive effect was observed at much lower, physiological CD40L concentrations in peripheral blood mononuclear cells (PBMC) of HIV-1 infected individuals compared to controls (p<0.05). The CpG-induced IFN-alpha production in PBMC of HIV-1 infected donors was directly correlated with PDC and CD4+ T cell counts, and inversely correlated with the viral loads (p<0.001). In HIV-1 infected donors with less than 500 CD4+ T cells/µl, the CpG-induced IFN-alpha production was significantly correlated with the percentage of CD40-expressing PDC and the level of CD40 expression on these cells (p<0.05), whereas CD40L plasma levels played a minor role. In addition, low-dose CD40L contributed to the enhanced production of interleukin 6 and 8 in PBMC of HIV-1 infected donors compared to controls. Our data support the conclusion that the chronic immune activation in HIV-1 infection impairs peripheral PDC innate immune responses at least in part via enhanced CD40:CD40L interactions

    Cytochrome P450 2B6 (CYP2B6) and constitutive androstane receptor (CAR) polymorphisms are associated with early discontinuation of efavirenz-containing regimens

    Objectives Cytochrome P450 2B6 (CYP2B6) is responsible for the metabolic clearance of efavirenz and single nucleotide polymorphisms (SNPs) in the CYP2B6 gene are associated with efavirenz pharmacokinetics. Since the constitutive androstane receptor (CAR) and the pregnane X receptor (PXR) correlate with CYP2B6 in liver, and a CAR polymorphism (rs2307424) and smoking correlate with efavirenz plasma concentrations, we investigated their association with early (<3 months) discontinuation of efavirenz therapy. Methods Three hundred and seventy-three patients initiating therapy with an efavirenz-based regimen were included (278 white patients and 95 black patients; 293 male). DNA was extracted from whole blood and genotyping for CYP2B6 (516G → T, rs3745274), CAR (540C → T, rs2307424) and PXR (44477T → C, rs1523130; 63396C → T, rs2472677; and 69789A → G, rs763645) was conducted. Binary logistic regression using the backwards method was employed to assess the influence of SNPs and demographics on early discontinuation. Results Of the 373 patients, 131 withdrew from therapy within the first 3 months. Black ethnicity [odds ratio (OR) = 0.27; P = 0.0001], CYP2B6 516TT (OR = 2.81; P = 0.006), CAR rs2307424 CC (OR = 1.92; P = 0.007) and smoking status (OR = 0.45; P = 0.002) were associated with discontinuation within 3 months. Conclusions These data indicate that genetic variability in CYP2B6 and CAR contributes to early treatment discontinuation for efavirenz-based antiretroviral regimens. Further studies are now required to define the clinical utility of these association

    Das Übereinkommen der Vereinten Nationen über die Rechte von Menschen mit Behinderungen sowie das dazu gehörende Fakultativprotokoll zum Übereinkommen der Vereinten Nationen über die Rechte von Menschen mit Behinderungen trat am 26.03.2009 ohne Einschränkung als innerstaatliches deutsches Recht in Kraft. Sich mit diesen vielschichtigen Themenkomplexen auseinanderzusetzen war das Ziel der Tagung „Herausforderung Inklusion: Schule – Unterricht – Profession“, die am 27. und 28. März 2014 an der Otto-Friedrich-Universität in Bamberg stattfand und sowohl betroffene Eltern, als auch Praktikerinnen und Praktiker und Wissenschaftlerinnen und Wissenschaftler zur Diskussion einlud. Der vorliegende Band geht auf diese Tagung zurück und möchte die vielfältigen Facetten der theoretischen, konzeptuellen und didaktisch-methodischen Zugänge im Kontext der derzeit geführten Debatten und Argumentationsmuster um ein inklusives Bildungswesen abbilden und neue Perspektiven für Forschung, Disziplin und Profession anregen. Demzufolge liegt der Fokus der Beiträge zum einen auf der Auseinandersetzung mit theoretischen Zugängen zur Inklusion sowie der Diskussion von nationalen und internationalen empirischen Erkenntnissen aus Studien der Inklusionsforschung, zum anderen auf der Präsentation von inklusiven Konzepten einer Schulentwicklung, (fach)didaktisch-methodischen Überlegungen und Modellprojekten aus der Praxis

    Measuring the Installability of Service Orchestrations Using the SQuaRE Method

    Service-oriented software consists of middleware, such as application servers and runtime engines, into which service applications are deployed. This middleware is often complex and difficult to install. The deployment of services requires the crafting of deployment descriptors and packaging of applications. As a consequence, the installation of service-oriented software systems can be a daunting task. Installability, however, is an important influencer of the portability of software. Portability in turn is one of the main goals of service orchestration languages based on open standards. In this paper, we investigate the installability of service orchestrations based on the Systems and software Quality Requirements and Evaluation (SQuaRE) method, the new series of software qual- ity standards currently under development by the ISO/IEC. We develop a measurement framework based on SQuaRE and tailored to evaluating the installability of service orchestrations and their runtimes. We validate the measurement framework theoretically and show its applicability in a case study

    Open Source versus Proprietary Software in Service-Orientation : The Case of BPEL Engines

    It is a long-standing debate, whether software that is developed as open source is generally of higher quality than proprietary software. Although the open source community has grown immensely during the last decade, there is still no clear answer. Service-oriented software and middleware tends to rely on highly complex and interrelated standards and frameworks. Thus, it is questionable if small and loosely coupled teams, as typical in open source software development, can compete with major vendors. Here, we focus on a central part of service-oriented software systems, i.e., process engines for service orchestration, and compare open source and proprietary solutions. We use the Web Services Business Process Execution Language (BPEL) and compare standard conformance and its impact on language expressiveness in terms of workflow pattern support of eight engines. The results show that, although the top open source engines are on par with their proprietary counterparts, in general proprietary engines perform better
