    Workfare Programs in Rural America: Joblessness in Ohio\u27s Appalachian Counties

    In 1988 the Family Support Act was passed into law requiring welfare recipients to participate in work experience programs to receive their welfare benefits. This paper questions the effectiveness of mandatory workfare programs in rural impoverished regions of the United States. The Appalachian counties of Ohio are used as a case example to demonstrate the problems in implementing workfare programs in economically distressed regions where limited job opportunities exist. Implications for policy are examined, alternatives to mandatory work programs are discussed, and further research to determine the utility of workfare programs is called for

    HI intensity mapping with FAST

    We discuss the detectability of large-scale HI intensity fluctuations using the FAST telescope. We present forecasts for the accuracy of measuring the Baryonic Acoustic Oscillations and constraining the properties of dark energy. The FAST 1919-beam L-band receivers (1.051.05--1.451.45 GHz) can provide constraints on the matter power spectrum and dark energy equation of state parameters (w0,waw_{0},w_{a}) that are comparable to the BINGO and CHIME experiments. For one year of integration time we find that the optimal survey area is 6000ā€‰deg26000\,{\rm deg}^2. However, observing with larger frequency coverage at higher redshift (0.950.95--1.351.35 GHz) improves the projected errorbars on the HI power spectrum by more than 2Ā Ļƒ2~\sigma confidence level. The combined constraints from FAST, CHIME, BINGO and Planck CMB observations can provide reliable, stringent constraints on the dark energy equation of state.Comment: 7 pages, 3 figures, submitted to "Frontiers in Radio Astronomy and FAST Early Sciences Symposium 2015" conference proceedin

    Long-Term Estrogen Therapy Improves Vascular Function in Male to Female Transsexuals

    AbstractObjectives. This study sought to examine the effects of long-term estrogen therapy on vascular function in male to female transsexuals and to compare the findings with those observed in men and premenopausal women.Background. Gender differences in coronary artery disease have largely been attributed to the beneficial effects of estrogen on vascular function and plasma lipids in women. However, the effects of estrogen on the male vasculature have not been widely studied.Methods. We compared the effects of estrogen on vascular function in 14 male to female transsexuals, 14 age-matched men and 15 premenopausal women. Flow-mediated vasodilation and response to nitroglycerin were assessed in the brachial artery using noninvasive ultrasound.Results. Flow-mediated vasodilation was similar in transsexuals and women but greater than that in men ([mean Ā± SE] 11.5 Ā± 1.3% and 9.4 Ā± 1.1% vs. 5.2 Ā± 1.0% respectively, p < 0.005). Responses to nitroglycerin were also greater in transsexuals and women than in men (21.6 Ā± 1.7% and 21.0 Ā± 0.9% vs. 14.5 Ā± 1.2%, respectively, p = 0.0005). These differences persisted even after adjusting for vessel size. Despite similar total cholesterol levels, transsexuals had high density lipoprotein cholesterol levels similar to those in women and greater than those observed in men (1.76 Ā± 0.12 and 1.82 Ā± 0.11 mmol/liter vs. 1.35 Ā± 0.07 mmol/liter, respectively, p < 0.005). Moreover, triglyceride levels were greater in transsexuals than in men and women, and low density lipoprotein cholesterol (LDL-C) particle size was smaller (25.7 Ā± 0.2 nm vs. 26.2 Ā± 0.1 and 26.6 Ā± 0.1 nm, respectively, p = 0.0001). Serum testosterone (an index of estrogen therapy in transsexuals) was markedly suppressed in transsexuals and similar to that in women. Univariate analysis revealed that there was a strong inverse correlation between serum testosterone and flow-mediated vasodilation (rs= āˆ’0.48, p < 0.005). Multivariate analysis revealed that the best combination of predictors of flow-mediated vasodilation was serum testosterone, vessel size and LDL-C (R2= 0.3, p < 0.005).Conclusions. Long-term estrogen therapy appears to improve vascular function in male to female transsexuals and occurs despite higher triglyceride levels and the presence of small, dense LDL-C. The beneficial effects of estrogen are not gender specific or solely mediated through endothelium-derived nitric oxide.(J Am Coll Cardiol 1997;29:1437ā€“44

    Documenting and harnessing the biological potential of molecules in Distributed Drug Discovery (D3) virtual catalogs

    Virtual molecular catalogs have limited utility if member compounds are (i) difficult to synthesize or (ii) unlikely to have biological activity. The Distributed Drug Discovery (D3) program addresses the synthesis challenge by providing scientists with a free virtual D3 catalog of 73,024 easy-to-synthesize N-acyl unnatural Ī±-amino acids, their methyl esters, and primary amides. The remaining challenge is to document and exploit the bioactivity potential of these compounds. In the current work, a search process is described that retrospectively identifies all virtual D3 compounds classified as bioactive hits in PubChem-cataloged experimental assays. The results provide insight into the broad range of drug-target classes amenable to inhibition and/or agonism by D3-accessible molecules. To encourage computer-aided drug discovery centered on these compounds, a publicly available virtual database of D3 molecules prepared for use with popular computer docking programs is also presented

    Patient, companion, and oncologist agreement regarding information discussed during triadic oncology clinical interactions

    Background Although people with cancer want and need information from their oncologists, patients and oncologists often disagree about what information was discussed during clinical interactions. Most patients have companions present during oncology visits; we investigated whether companions process information more accurately than patients. Specifically, we examined whether patients and companions differed in agreement with oncologists about what was discussed. We also investigated the effect of topic on agreement and patient/companion selfā€reported understanding of discussions. Methods Patients with companions were invited to participate on first visits to a cancer center in Detroit, MI. Patients, companions, and oncologists independently completed questionnaires immediately following visits. Participants were asked whether five topics were discussed (diagnosis, prognosis, metastasis, treatment/treatment goals, and side effects) and, if discussed, what oncologists said. Participants were also asked to estimate their own and each other's understanding of discussions. Results A total of 66 patientā€“companionā€“oncologist triads participated. Agreement was higher regarding whether topics were discussed than what oncologists said. Agreement did not differ by dyad type. Patients, companions, and oncologists were equally likely to be the source of triadic disagreements. Agreement was high about diagnosis (>90%) but much lower about other topics, particularly side effects. Patients and companions reported greater understanding of discussions than oncologists estimated and more accurately estimated each other's understanding than did oncologists. Conclusions Companions and patients showed similar levels of agreement with oncologists about what they discussed during visits. Interventions are needed to improve communication of information to both patients and companions, especially about particular topics.     Loss of the insulin receptor in murine megakaryocytes/platelets causes thrombocytosis and alterations in IGF signalling.

    AIMS: Patients with conditions that are associated with insulin resistance such as obesity, type 2 diabetes mellitus, and polycystic ovary syndrome have an increased risk of thrombosis and a concurrent hyperactive platelet phenotype. Our aim was to determine whether insulin resistance of megakaryocytes/platelets promotes platelet hyperactivation. METHODS AND RESULTS: We generated a conditional mouse model where the insulin receptor (IR) was specifically knocked out in megakaryocytes/platelets and performed ex vivo platelet activation studies in wild-type (WT) and IR-deficient platelets by measuring aggregation, integrin Ī±IIbĪ²3 activation, and dense and Ī±-granule secretion. Deletion of IR resulted in an increase in platelet count and volume, and blocked the action of insulin on platelet signalling and function. Platelet aggregation, granule secretion, and integrin Ī±IIbĪ²3 activation in response to the glycoprotein VI (GPVI) agonist collagen-related peptide (CRP) were significantly reduced in platelets lacking IR. This was accompanied by a reduction in the phosphorylation of effectors downstream of GPVI. Interestingly, loss of IR also resulted in a reduction in insulin-like growth factor-1 (IGF-1)- and insulin-like growth factor-2 (IGF-2)-mediated phosphorylation of IRS-1, Akt, and GSK3Ī² and priming of CRP-mediated platelet activation. Pharmacological inhibition of IR and the IGF-1 receptor in WT platelets recapitulated the platelet phenotype of IR-deficient platelets. CONCLUSIONS: Deletion of IR (i) increases platelet count and volume, (ii) does not cause platelet hyperactivity, and (iii) reduces GPVI-mediated platelet function and platelet priming by IGF-1 and IGF-2

    Aerosol Profile Measurements from the NASA Langley Research Center Airborne High Spectral Resolution Lidar

    Since achieving first light in December of 2005, the NASA Langley Research Center (LaRC) Airborne High Spectral Resolution Lidar (HSRL) has been involved in seven field campaigns, accumulating over 450 hours of science data across more than 120 flights. Data from the instrument have been used in a variety of studies including validation and comparison with the Cloud- Aerosol Lidar and Infrared Pathfinder Satellite Observation (CALIPSO) satellite mission, aerosol property retrievals combining passive and active instrument measurements, aerosol type identification, aerosol-cloud interactions, and cloud top and planetary boundary layer (PBL) height determinations. Measurements and lessons learned from the HSRL are leading towards next-generation HSRL instrument designs that will enable even further studies of aerosol intensive and extensive parameters and the effects of aerosols on the climate system. This paper will highlight several of the areas in which the NASA Airborne HSRL is making contributions to climate science

    Assessment of the CALIPSO Lidar 532 nm Attenuated Backscatter Calibration Using the NASA LaRC Airborne High Spectral Resolution Lidar

    The Cloud-Aerosol Lidar with Orthogonal Polarization (CALIOP) instrument on the Cloud-Aerosol Lidar and Infrared Pathfinder Satellite Observations (CALIPSO) spacecraft has provided global, high-resolution vertical profiles of aerosols and clouds since it became operational on 13 June 2006. On 14 June 2006, the NASA Langley Research Center (LaRC) High Spectral Resolution Lidar (HSRL) was deployed aboard the NASA Langley B-200 aircraft for the first of a series of 86 underflights of the CALIPSO satellite to provide validation measurements for the CALIOP data products. To better assess the range of conditions under which CALIOP data products are produced, these validation flights were conducted under both daytime and nighttime lighting conditions, in multiple seasons, and over a large range of latitudes and aerosol and cloud conditions. This paper presents a quantitative assessment of the CALIOP 532 nm calibration (through the 532 nm total attenuated backscatter) using an internally calibrated airborne HSRL underflight data and is the most extensive study of CALIOP 532 nm calibration. Results show that average HSRL and CALIOP 532 nm total attenuated backscatter agree on average within 2.7% +/- 2.1% (CALIOP lower) at night and within 2.9 % +/- 3.9% (CALIOP lower) during the day., demonstrating the accuracy of the CALIOP 532 nm calibration algorithms. Additionally, comparisons with HSRL show consistency of the CALIOP calibration before and after the laser switch in 2009 as well as improvements in the daytime version 3 calibration scheme compared with the version 2 calibration scheme. Potential systematic uncertainties in the methodology relevant to validating satellite lidar measurements with an airborne lidar system are discussed and found to be less than 3.7% for this validation effort with HSRL. Results from this study are also compared to those from prior assessments of CALIOP calibration and attenuated backscatter
