    Without trying to explain away an error that gross, we can only say that it is the kind of error that occurred only once, and occurred in spite of our editorial method rather than because of it. Most of the other outright errors were less egregious: misspelled names; failure to annotate at it appeared in the original, though we doubt that history was changed by the omission of that particular punctuation. We would still continue to correct obvious typographical errors in typewritten or printed documents. Maybe a type does reveal something deeply hidden, but is it deeply hidden in the author or the stenographer? Only a psycho-historian can analyze all the typos of a lifetime and reach conclusions as to their psychological significance, and for that he would surely want to see the originals, to see if the typist was agitated enough to punch out all the O\u27s

    We might as well begin on a note of candor by admitting the worst error we ever made, for of all the things that we would have done differently this surely heads the list. On the errata page of volume 8 appears the note: Volume 4, p. 309, n. 1. The man wrongly identified as Robert Brown Elliott [a black man] was actually William Elliott, a white man. The letter to BTW, Apr. 25,1898, was from Rev. G. M. Elliott of Beaufort, S.C. Not only had we confused a black man with a white man and another black man, but in a display of erudition we gratuitously had mentioned a letter that Elliott-the wrong Elliott-wrote to Booker T. Washington fourteen years after our own annotation indicated his death date. And they say that dead men tell no tales. At least there were no errata in our erratum. Such a compounding of errors could only have been achieved by a committee. For most of our annotations, we are able to trace back responsibility by checking the raw data notes, but in this case these had mysteriously disappeared. It all reminds us of the famous Nast cartoon about the Tweed ring. It shows a circle of bloated politicians. The caption reads, Who Stole the People\u27s Money? Each man is pointing his finger at the man on his left. That is our worst error yet, but we still have to do the cumulative index with its infinite possibilities.     Metabolic Effects of Avocado/Soy Unsaponifiables on Articular Chondrocytes

    Avocado/soy unsaponifiable (ASU) components are reported to have a chondroprotective effect by virtue of anti-inflammatory and proanabolic effects on articular chondrocytes. The identity of the active component(s) remains unknown. In general, sterols, the major component of unsaponifiable plant material have been demonstrated to be anti-inflammatory in vitro and in animal models. These studies were designed to clarify whether the sterol content of ASU preparations were the primary contributors to biological activity in articular chondrocytes. ASU samples were analyzed by high pressure liquid chromatography (HPLC) and GC mass spectrometry. The sterol content was normalized between diverse samples prior to in vitro testing on bovine chondrocytes. Anabolic activity was monitored by uptake of 35-sulfate into proteoglycans and quantitation of labeled hydroxyproline and proline content after incubation with labeled proline. Anti-inflammatory activity was assayed by measuring reduction of interleukin-1 (IL-1)-induced synthesis of PGE2 and metalloproteases and release of label from tissue prelabeled with S-35.All ASU samples exerted a similar time-dependent up-regulation of 35-sulfate uptake in bovine cells reaching a maximum of greater than 100% after 72 h at sterol doses of 1–10 μg/ml. Non-collagenous protein (NCP) and collagen synthesis were similarly up-regulated. All ASU were equally effective in dose dependently inhibiting IL-1-induced MMP-3 activity (23–37%), labeled sulfate release (15–23%) and PGE2 synthesis (45–58%). Up-regulation of glycosaminoglycan and collagen synthesis and reduction of IL-1 effects in cartilage are consistent with chondroprotective activity. The similarity of activity of ASU from diverse sources when tested at equal sterol levels suggests sterols are important for biologic effects in articular chondrocytes

    Performance of Abbott Architect, Ortho Vitros, and Euroimmun Assays in Detecting Prior SARS-CoV-2 Infection [preprint]

    Background: Several serological assays have been developed to detect anti-SARS-CoV-2 IgG antibodies, but evidence about their comparative performance is limited. We sought to assess the sensitivity of four anti-SARS-CoV-2 IgG enzyme-linked immunosorbent assays (ELISA) in individuals with evidence of prior SARS-CoV-2 infection. Methods: We obtained sera from 36 individuals with PCR-confirmed SARS-CoV-2 infection between March and May 2020. We evaluated samples collected at around 21 days (±14 days) after their initial PCR test using 3 commercially available ELISA assays, two anti-spike (Ortho-Clinical Diagnostics Vitros, and Euroimmun) and one anti-nucleocapsid (Abbott Architect), and a Yale-developed anti-spike ELISA test. We determined the sensitivity of the tests and compared their results. The Euroimmun and Yale ELISA had an equivocal and indeterminate category, which were considered as both negative and positive. Results: Among the 36 individuals with SARS-CoV-2 infection, mean age was 43 (±13) years and 19 (53%) were female. The sensitivities of the tests were not significantly different (Abbott Architect, Ortho Vitros, Euroimmmun, and Yale assays: 86% (95% confidence interval [CI], 71-95), 94% (95% CI, 81-99), 86% (95% CI, 71-95), and 94% (95% CI, 81-99), respectively; p-value=0.464). The sensitivities of the Euroimmun and Yale ELISA tests increased when the equivocal/indeterminate results were considered positive (97% [95% CI, 85-100] and 100% [95% CI, 90-100], respectively), but were not significantly different from other tests (p=0.082). The cross-correlation coefficient ranged from 0.85-0.98 between three anti-spike protein assays (Ortho Vitros, Euroimmun, Yale) and was 0.58-0.71 between the three anti-spike protein assays and the anti-nucleocapsid assay (Abbott). Conclusion: The sensitivities of four anti-SARS-CoV-2 protein assays did not significantly differ, although the sample size was small. Sensitivity also depended on the interpretation of equivocal and indeterminate results. The strongest correlations were present for the three anti-spike proteins assays. These findings suggest that individual test characteristics and the correlation between different tests should be considered when comparing or aggregating data across different populations studies for serologic surveillance of past SARS-CoV-2 infection

    Observations of Particle Loss due to Injection-Associated Electromagnetic Ion Cyclotron Waves

    We report on observations of electromagnetic ion cyclotron (EMIC) waves and their interactions with injected ring current particles and high energy radiation belt electrons. The magnetic field experiment aboard the twin Van Allen Probes spacecraft measured EMIC waves near L = 5.5–6. Particle data from the spacecraft show that the waves were associated with particle injections. The wave activity was also observed by a ground-based magnetometer near the spacecraft geomagnetic footprint over a more extensive temporal range. Phase space density profiles, calculated from directional differential electron flux data from Van Allen Probes, show that there was a significant energy-dependent relativistic electron dropout over a limited L-shell range during and after the EMIC wave activity. In addition, the NOAA spacecraft observed relativistic electron precipitation associated with the EMIC waves near the footprint of the Van Allen Probes spacecraft. The observations suggest EMIC wave-induced relativistic electron loss in the radiation belt

    α Cell Function and Gene Expression Are Compromised in Type 1 Diabetes.

    Many patients with type 1 diabetes (T1D) have residual β cells producing small amounts of C-peptide long after disease onset but develop an inadequate glucagon response to hypoglycemia following T1D diagnosis. The features of these residual β cells and α cells in the islet endocrine compartment are largely unknown, due to the difficulty of comprehensive investigation. By studying the T1D pancreas and isolated islets, we show that remnant β cells appeared to maintain several aspects of regulated insulin secretion. However, the function of T1D α cells was markedly reduced, and these cells had alterations in transcription factors constituting α and β cell identity. In the native pancreas and after placing the T1D islets into a non-autoimmune, normoglycemic in vivo environment, there was no evidence of α-to-β cell conversion. These results suggest an explanation for the disordered T1D counterregulatory glucagon response to hypoglycemia. Cell Rep 2018 Mar 6; 22(10):2667-2676

    A population-based study of tumor gene expression and risk of breast cancer death among lymph node-negative patients

    INTRODUCTION: The Oncotype DX assay was recently reported to predict risk for distant recurrence among a clinical trial population of tamoxifen-treated patients with lymph node-negative, estrogen receptor (ER)-positive breast cancer. To confirm and extend these findings, we evaluated the performance of this 21-gene assay among node-negative patients from a community hospital setting. METHODS: A case-control study was conducted among 4,964 Kaiser Permanente patients diagnosed with node-negative invasive breast cancer from 1985 to 1994 and not treated with adjuvant chemotherapy. Cases (n = 220) were patients who died from breast cancer. Controls (n = 570) were breast cancer patients who were individually matched to cases with respect to age, race, adjuvant tamoxifen, medical facility and diagnosis year, and were alive at the date of death of their matched case. Using an RT-PCR assay, archived tumor tissues were analyzed for expression levels of 16 cancer-related and five reference genes, and a summary risk score (the Recurrence Score) was calculated for each patient. Conditional logistic regression methods were used to estimate the association between risk of breast cancer death and Recurrence Score. RESULTS: After adjusting for tumor size and grade, the Recurrence Score was associated with risk of breast cancer death in ER-positive, tamoxifen-treated and -untreated patients (P = 0.003 and P = 0.03, respectively). At 10 years, the risks for breast cancer death in ER-positive, tamoxifen-treated patients were 2.8% (95% confidence interval [CI] 1.7–3.9%), 10.7% (95% CI 6.3–14.9%), and 15.5% (95% CI 7.6–22.8%) for those in the low, intermediate and high risk Recurrence Score groups, respectively. They were 6.2% (95% CI 4.5–7.9%), 17.8% (95% CI 11.8–23.3%), and 19.9% (95% CI 14.2–25.2%) for ER-positive patients not treated with tamoxifen. In both the tamoxifen-treated and -untreated groups, approximately 50% of patients had low risk Recurrence Score values. CONCLUSION: In this large, population-based study of lymph node-negative patients not treated with chemotherapy, the Recurrence Score was strongly associated with risk of breast cancer death among ER-positive, tamoxifen-treated and -untreated patients

    Borderline hypertension was the topic of one of the “Hypertension seminars” arranged by the Hypertension Section at the Östra Hospital, GÖteborg, Sweden. On that occasion Professor Stevo Julius, Ann Arbor, Michigan, USA, was an invited guest. During the seminar, various aspects of borderline hypertension were discussed, e.g. the natural history, hemodynamics and management of this condition.
